The Effects of Green Tea Catechins on Vascular Smooth Muscle Tension and 45 $Ca^{2+}$ Uptake (녹차 카테킨류의 혈관장력 및 $Ca^{2+}$유입에 미치는 영향)

  • The objective of this study is to investigate the direct effects of green tea catechins(GTC) on vascular smooth muscle tension and 45Ca2+ Uptake in rat aorta. The methods used in this study are isometric tension measurements using physiograph, Lanthanum method for 45Ca2+(2 uCi/ml) uptake measurement in rat aorta. GTC modified tension induced by 40 mM KCl or 1 uM norepinephrine in rat aorta. Low concentrations of GTC(<0.5mg/ml) increased tension by 40 mM KCl or 1 uM norepinephrine, individually. However, high conecentration of GTC(>0.5 mg/ml) inhibiited tension by 40 mM KCl or 1 uM norepinephrine, individually. GTC increased 45Ca uptake induced by 40 mM KCl in a dose-dependent manner. From these results, GTC has the dual actions in vascular smooth muscle in vitro. Low concentrations of GTC augments tension by K or norepinephrine. However, high concentrations of GTC inhibits tension by K or norepinephrine GTC may have Ca2+ channel activation, action, which may result in unphysiological vasodilation by Ca2+ overload in vascular smooth muscle.

$[^3H]$ Ouabain Binding and Effect of Ouabain on $^{45}Ca^{2+}$-Uptake in Rat Cardiac Myocytes (쥐 심근 세포의 $[^3H]$ Ouabain 결합과 $^{45}Ca^{2+}}$섭취에 미치는 Ouabain의 영향)

  • Specific [$^{3}H$] ouabain binding and $Ca^{2+}$ -uptake were measured to elucidate the role of high affinity [$^{3}H$] ouabain binding site in rat cardiac myocytes which contain 65% of rod cells. High affinity [$^{3}$H] ouabain binding site, which is about 3% of total pump sites, with apparent dissociation constant ($K_{D}$) of $1.1{\times}10^{-7}M$ and maximum binding site concentration (Bmax) of 1.2 pmol/mg protein ($1.754{\times}10^{5}cells$) were identified. At the concentration of $10^{-7}M$ to $10^{-4}M$, ouabain produced concentration dependent increase in $Ca^{2+}$-uptake of myocytes. The effect of ouabain on $Ca^{2+}$-uptake was not effected by membrane depolarization (elevated K+ in incubation medium) or verapamil. These results suggest that in rat ventricular myocytes the ouabain receptor complex to high affinity site may increase Na+ - $Ca^{2+}$ exchange across the sarcolemmal membrane by inhibition of Na+, K+ - ATPase.

Interactions between Estradiol-17 ${\beta}-BSA$ and Calcitropic Hormones in $Ca^{2+}$ Uptake in Renal Proximal Tubule Cells

  • The aim of the present study was to investigate the interaction of $estradiol-17{\beta}-bovine$ serum albumin $(E_2-BSA)$ and calcitropic hormones, such as parathyroid hormone, calcitonin, and vitamin D, in regulation of $Ca^{2+}$ uptake in primary cultured renal proximal tubule cells. Statistically significant increase in $Ca^{2+}$ uptake was found from 2 hours after $(E_2-BSA)\;(10^{-9}\;M)$ treatment, while $estradiol-17{\beta}\;(10^{-9}\;M)$ did not affect. Treatment of the cells with $(E_2-BSA)\;(10^{-9}\;M)$ together with parathyroid hormone (PTH) $(10^{-8}\;M),$ vitamin D $(10^{-8}\;M),$ or calcitonin $(10^{-8}\;M)$ significantly stimulated $Ca^{2+}$ uptake by 32.50%, 29.30%, or 27.75%, respectively, compared with the control. However, calcitropic hormones did not exhibit any synergistic effect on the E2-BSA-induced stimulation. $E_2-BSA$ significantly increased cAMP generation and PKC activity. The stimulatory effect of cotreatment of $E_2-BSA$ and PTH or vitamin D was blocked by SQ22536 (an adenylate cyclase inhibitor) and staurosporine (a PKC inhibitor), but the effect of cotreatment of $E_2-BSA$ and calcitonin was not blocked. Furthermore, 8-Br-cAMP and TPA (an artificial PKC promoter) increased $Ca^{2+}$ uptake by 25.51% and 16.47%, respectively, compared with the control. In conclusion, $E_2-BSA$ combined with calcitropic hormones regulated $Ca^{2+}$ uptake partially via cAMP and PKC-dependent mechanisms in renal proximal tubule cells.

Characteristics of Salt Adsorption by Calcium Alginate Beads (칼슘알긴산비드에 의한 염분의 흡착특성)

  • The adsorption characteristics of sodium chloride into Ca-alginate beads have been investigated and the result were as follows: Sodium chloride uptake by Ca-alginate beads increased with time. The highest uptake volume of sodium chloride was 4.2g after 10 minutes. The uptake volume by Fe, Ca, Ba, and Sr-alginate beads was 5.6g, 4.2g, 4.2g and 4.0g, respectively but in case of Fe-alginate beads, the induced hydrogel beads were very fragile and the strength of Fe-alginate beads were weaker than Ca- and Ba-alginate beads. Mg-alginate bead was not formed and Ca-, Ba- and Sr-alginate beads had a similar uptake volume about 4.2g, respectively. The uptake volume of sodium chloride by CaCl$_2$concentration(0.1M. 0.2M and 1M), curing solution, was 4.8, 4.2g and 4.1g, respectively. The uptake volume by sodium alginate concentration(0.6%, 1% and 2%) was 2.8g, 4.0g, and 4.4g, respectively and Ca-alginate bead size was not effected in uptake sodium chloride. The uptake rate on initial sodium chloride concentration(4%, 8%, 12% and 16%) was 30%, 28%, 27% and 25%, respectively. The uptake rate on basic pH(10.0) was higher than when compared to other neutral pH(6.8) and acidic pH(4.0). The initial uptake velocity of sodium chloride from immobilization beads with salt resistant bacteria was lower than that of non-immobilization beads. The uptake rate of sodium chloride was decreased according to elongation of curing time. Reusability of Ca-alginate beads was possible but according to reutilization, the salt uptake volume of beads was also decreased. The uptake volume of sodium chloride from Doengjang by Ca-alginate beads on time course(3, 6, 12, and 24 hour) was revealed 5g, 6g, 7g and 7g, respectively.

The Effect of $Ca^{2+}$ and Its Specific Time in the Maturation of Mammalian Oocyte (포유류의 난자성숙과 성숙과정에서 $Ca^{2+}$의 영향과 그 작용시기)

  • Follicular oocytes were released from the graafian follicles of ovaries from 3-4 weeks old mice. The spontaenous maturation of these follicular oocyes was inhibited by the treatment of dbcAMP and progesterone and these oocytes were cultured for 2-8hr in the Modified Hank's balanced salt solution(MHBS). Ethylenediaminetetraaceticacid(EDTA) and calmoudulin antagonist, trifluoperazine (TFP) were treated to the culture medium in order to investigate whether these chemical agents inhibit calcium uptake into the oocyte and oocyte maturation. $^{45}Ca^{2+}$, 10-${\mu}$Ci/ml was added to the culture medium during the culture period. $^{45}Ca^{2+}$uptake into the oocytes was examined whether and when various kind of oocyte maturation inhibiting agents inhibit or stimulate the influx of calcium into oocytes. Dibutyryl cAMP and progesterone decrease $^{45}Ca^{2+}$uptake into the oocytes and synergistic inhibiting effect of dbcAMP and progesterone was prominent at much lower dosages. Calcium uptake into oocytes seems to be higher during first 2 hour culture period rather than next 4hr culture. After 8hr culture, calcium uptake level of the oocytes which GVBD already took place gradually approached to the level of those which were maintained at GV by the treatment of dbcAMP and progesterone. However, $^{45}Ca^{2+}$uptake into the GV maintained oocytes did not change at all even after 8hr culture period. In addition, calcium chelating agent, EDTA inhibited calcium uptake into oocytes as well as nuclear maturation of oocytes. Lower dosage used in the present study did not inhibit calcium uptake as well as oocyte maturation.

  • It was reported from our laboratory that the rate of deterioration of the force of contraction was slower in heart from Panax ginseng extract treated rats. Present investigation was designed to elucidate the mechanism of the slow deterioration of contractility of ginseng treated hearts. Therefore, $^{45}Ca^{2+}$ Uptake by sarcoplasmic reticulum (SR) isolated from ginseng treated rate and control rats was studied. Rate weighing 150-250g were administered orally with ginseng ethanol extract (100mg/kg) for 10 days. Cardiac SR was isolated by differential centrifugation and $^{45}Ca^{2+}$ uptake was assessed by the Millipore method. Freshly isolated SR from treated as well as control animals did not show any differences, but after incubation for 30 and 60 min at 37.deg.C, $^{45}Ca^{2+}$ uptake of control animal SR was found to be greatly depressed. The SR of treated animal possessed a greater degree of resistance to incubation. Thus it can be concluded that ginseng may have an ability to sustain the normal function of the heart by sustaining Ca accumulation by SR involved with the excitationcontraction coupling processes.

Effects of the Oriental Medicinal Prescriptions, Jahyulyangeuntang and Yanghyuljangeunkeonbohwan, on Calcium Absorption in the Human Colon Carcinoma Cell Line (Caco-2 Cells) (자혈양근탕 및 양혈장근건보환이 인체 소장 상피세포주 (Caco-2) 모델에서 칼슘 흡수에 미치는 영향)

  • Effects of the two oriental medicinal prescriptions, Jahyulyangeuntang (JH) and Yanghyuljangeunkeonbohwan (YH), on intestinal calcium absorption were examined in the human colon carcinoma tell line, Caco-2 cells. Intestinal calcium absorption was evaluated at the level of Ca uptake into the cells across the brush border membranes, as well as at the level of net Ca transport (implying the amount of intestinal Ca transported into the blood stream). When the Caco-2 cells were incubated for 4, 8, 16 and 24 days post seeding, the cells were differentiated continuously, and showed progressively increased activities of Ca uptake (1.13 $\pm$ 0.04, 1.19 $\pm$ 0.02, 1.94 $\pm$ 0.03, and 2.40 $\pm$ 0.12 nmole.mg protein$^{-1}$ .30 min$^{-1}$ , respectively). Pretreatment of confluent Caco-2 cells with 50 $\mu\textrm{g}$/ml of YH for 24 hours resulted in a 30% increase in Ca uptake (p < 0.07), while pretreatment of the cells with the same concentration of JH for 6 hours resulted in a 24% increase (p < 0.05) in Ca uptake, compared to the value for the control cells (2.34 $\pm$ 0.10 nmole.mg protein$^{-1}$ .30 min$^{-1}$ ). When the cells were pretreated with varied concentrations (5-100 $\mu\textrm{g}$/ml) of the test samples for 6 hours, maximal increases in Ca uptake were observed in the cells pretreated with 100 $\mu\textrm{g}$/ml of YH (a 23% increase), and 50 $\mu\textrm{g}$/ml of JH (a 28% increase), respectively : however, no influence was seen on the net Ca transport activity. These results show that pretreatment with JH or YH, the two oriental medicinal prescriptions commonly used for improvement of bone metabolism, could possibly increase Ca accumulation inside the cells. but not the intestinal Ca net transport in vitro.

Effect of t-butylhydroperoxide on $Na^+-dependent$ Glutamate Uptake in Rabbit Brain Synaptosome

  • The effect of an organic peroxide, t-butylhydroperoxide (t-BHP), on glutamate uptake was studied in synaptosomes prepared from cerebral cortex. t-BHP inhibited the $Na^+-dependent$ glutamate uptake with no change in the $Na^+-independent$ uptake. This effect of t-BHP was not altered by addition of $Ca^{2+}$ channel blockers (verapamil, diltiazem and nifedipine) or $PLA_2$ inhibitors (dibucaine, butacaine and quinacrine). However, the effect was prevented by iron chelators (deferoxamine and phenanthroline) and phenolic antioxidants (N,N'-diphenyl-phenylenediamine, butylated hydroxyanisole, and butylated hydroxytoluene). At low concentrations (<1.0 mM), t-BHP inhibited glutamate uptake without altering lipid peroxidation. Moreover, a large increase in lipid peroxidation by $ascorbate/Fe^{2+}$ was not accompanied by an inhibition of glutamate uptake. The impairment of glutamate uptake by t-BHP was not intimately related to the change in $Na^+-K+-ATPase$ activity. These results suggest that inhibition of glutamate uptake by t-BHP is not totally mediated by peroxidation of membrane lipid, but is associated with direct interactions of glutamate transport proteins with t-BHP metabolites. The $Ca^{2+}$ influx through $Ca^{2+}$ channel or $PLA_2$ activation may not be involved in the t-BHP inhibition of glutamate transport.

  • The Ca uptake by the fragmented sarcoplasmic reticulum of the rabbit skeletal muscle was measured under various concentrations of K, Mg, Caffeine, procaine and quinine. The ATPase activity of this reticular membrane was measured under the same conditions simultaneously. The saturation of Ca uptake was almost completed within 1 minute. The Ca uptake was inhibited by high concentrations of K (above 50 mM) and Mg (above 1 mM)in the absence of ATP. When ATP is present, however, the Ca uptake did not reflect the concentration of K, while it increased greatly as the concentration of Mg was increased. Caffeine and procaine caused the inhibition of Ca uptake in the presence of ATP, but quinine did not. The ATPase activity of the membrane was little affected by the concentration of K, while it was enhanced in the presence of Mg. Caffeine, procaine and quinine did not influence the ATPase activity.

Role of $Ca^{2+}$ in the Stimulation of Glucose Transport by Insulin in Adipocytes

  • We investigated the role of $Ca^{2+}$ and protein kinases/phosphatases in the stimulatory effect of insulin on glucose transport. In isolated rat adipocytes, the simple omission of $CaCl_2$ from the incubation medium significantly reduced, but did not abolish, insulin-stimulated 2-deoxy glucose (2-DG) uptake. Pre-loading adipocytes with intracellular $Ca^{2+}$ chelator, 5,5'-dimethyl bis (o-aminophenoxy)ethane-N,N,N'N' tetraacetic acetoxymethyl ester (5,5'-dimethyl BAPTA/AM) completely blocked the stimulation. Insulin raised intracellular $Ca^{2+}$ concentration $([Ca^{2+}]_i)$ about 1.7 times the basal level of $72{\pm}5$ nM, and 5,5'-dimethyl BAPTA/AM kept it constant at the basal level. This correlation between insulin-induced increases in 2-DG uptake and $[Ca^{2+}]_i$ indicates that the elevation of $[Ca^{2+}]_i$ may be prerequisite for the stimulation of glucose transport. Studies with inhibitors (ML-9, KN-62, cyclosporin A) of $Ca^{2+}-calmodulin$ dependent protein kinases/phosphatases also indicate an involvement of intracellular $Ca^{2+}.$ Additional studies with okadaic acid and calyculin A, protein phosphatase-1 (PP-1) and 2A (PP-2A) inhibitors, indicate an involvement of PP-1 in insulin action on 2-DG uptake. These results indicate an involvement of $Ca^{2+}-dependent$ signaling pathway in insulin action on glucose transport.
