  • The activity of $Ca^{2+}$calmodulin (CaM)-dependent protein kinase Ia (CaM kinase Ia) is shown to be regulated through direct phosphorylation by CaM kinase I kinase (CaMK IK). In the present study, three distinct CaMKIK peaks were separated from Q-Sepharose colunm chromatography of pig brain homogenate using a Waters 650 Protein Purification System. The purified CaMKIK from the major peak potently and rapidly enhanced CaM kinase Ia activity, reaching a maximal stimulation within 2min at the concentrations of 12-15nM. The activated state of CaM kinase Ia is characterized by a markedly enhanced $V_{max}4 as well as significantly decreased $K_m\;and\;K_a$ values toward peptide substrate and CaM, respectively. These observations suggest the activation process of CaM kinase Ia. The phosphorylation of CaM kinase Ia by CaMKIK may induce its conformational change responsible for the alterations in the kinetic properties, which ultimately leads to the rapid enzyme activation.

Effects of Ethylene and $Ca^{2+}$ on Activity of Phenylalanine Ammonia-Lyase in Glucan-Treated Daucus carota

  • Involvement of ethylene and Ca2+ on the induction of phenylalanine ammonia-lyase (PAL) was investigated in Daucus carota L. suspension culture system. Ethylene production started to increase about 3 h after glucan treatment. And the maximal induction of ethylene was preceded by PAL induction by 30 min. After the treatment of ethrel, PAL activity was increased. When cells were treated with glucan and Co2+, PAL activity was simultaneously reduced. Ethylene production was reduced dramatically in calcium-free medium, even though glucan was treated. PAL activity and ethylene producton was inhibited conspicuously when ethylene glycolbis($\beta$-aminoethyl ether) N,N,N',N'-tetraacetic acid (EGTA) was treated with glucan. Verapamil and trifluoperazine also inhibited PAL activity. When cells were treated with calcium ionophore A23187, PAL activity was increased in nontreated medium. We report here PAl activity is increased in related to ethylene production and involvement of Ca2+ in glucan-treated carrot suspension cells.

trans-Cinnamaldehyde 유도체의 항진균활성 검색 (Antifungal Activities of trans-Cinnamaldehyde Derivatives)

  • Antifungal activities of trans-cinnamaldehyde (CA) derivatives including commercial CA derivatives as well as synthesized CA derivatives against various human pathogenic fungi were investigated. Among the derivatives tested, -chlorocinnamaldehyde, $\alpha$-bromocinnamaldehyde and 7-phenyl-2,4,6-heptatrienal were more potent than CA in antifungal activity, $\alpha$-Bromocinnamaldehyde was the most effective in inhibiting the growth of representative fungi of dermatomycosis with minimum inhibitory cocentration(MIC) of 0.61~9.76$\mu\textrm{g}$/ml . In the structure-activity relationship, introduction of the chlorine and bromine group into the C-2 of CA resulted in the decrease of MIC. Derivative with more double bonds exhibited the increase of antifungal activity against various pathogenic fungi.

Modulation of $Ca^{2+}-Activated$ Potassium Channels by cGMP-Dependent Signal Transduction Mechanism in Cerebral Arterial Smooth Muscle Cell of the Rabbit

  • The present investigation tested the hypothesis that the activation of protein kinase G (PKG) leads to a phosphorylation of $Ca^{2+}-activated$ potassium channel $(K_{Ca}\;channel)$ and is involved in the activation of $K_{Ca}$ channel activity in cerebral arterial smooth muscle cells of the rabbit. Single-channel currents were recorded in cell-attached and inside-out patch configurations of patch-clamp techniques. Both molsidomine derivative 3-morpholinosydnonimine-N-ethylcarbamide $(SIN-1,\;50\;{\mu}M)$ and 8-(4-Chlorophenylthio)-guanosine-3',5'-cyclic monophosphate $(8-pCPT-cGMP,\;100\;{\mu}M),$ a membrane-permeable analogue of cGMP, increased the $K_{Ca}$ channel activity in the cell-attached patch configuration, and the effect was removed upon washout of the drugs. In inside-out patches, single-channel current amplitude was not changed by SIN-1 and 8-pCPT-cGMP. Application of ATP $(100\;{\mu}M),$ cGMP $(100\;{\mu}M),$ ATP+cGMP $(100\;{\mu}M\;each),$ PKG $(5\;U/{\mu}l),$ ATP $(100\;{\mu}M)+PKG\;(5\;U/{\mu}l),$ or cGMP $(100\;{\mu}M)+PKG\;(5\;U/{\mu}l)$ did not increase the channel activity. ATP $(100\;{\mu}M)+cGMP\;(100\;{\mu}M)+PKG\;(5\;U/{\mu}l)$ added directly to the intracellular phase of inside-out patches increased the channel activity with no changes in the conductance. The heat-inactivated PKG had no effect on the channel activity, and the effect of PKG was inhibited by 8-(4-Chlorophenylthio)-guanosine-3',5'-cyclic monophosphate, Rp-isomer $(Rp-pCPT-cGMP,\;100\;{\mu}M),$ a potent inhibitor of PKG or protein phosphatase 2A (PP2A, 1 U/ml). In the presence of okadaic acid (OA, 5 nM), PP2A had no effect on the channel activity. The $K_{Ca}$ channel activity spontaneously decayed to the control level upon washout of ATP, cGMP and PKG, and this was prevented by OA (5 nM) in the medium. These results suggest that the PKG-mediated phosphorylations of $K_{Ca}$ channels, or some associated proteins in the membrane patch increase the activity of the $K_{Ca}$ channel, and the activation may be associated with the vasodilating action.

Mouse Egg의 안정막전압과 Pump 활동에 대한 양이온의 효과 (Effect of Cations on Resting Potential and Pump Activity of Unfertilized Mouse Eggs)

  • The present study was performed to observe the effects of cations on resting membrane potential and pump activity in the unfertilized eggs of ICR strain mice. After an induction of superovulation, the fresh eggs with zona pellucida were collected and the membrane potentials were recorded. Recordings of membrane potential in this study was obtained from the physiological conditions ($37^{\circ}C$ and 4mM Ca in standard solution), differently from the another reports with unphysiological conditions (room temprature and high Ca in standard solution) for a stable and long-lasting observations. Presented data was obtained within 6 hours after collection from the oviduct. The results observed are as follows, 1) Resting potential of the unfertilized eggs was $-25.8{\pm}3.8mV$ $(Mean{\pm}Se,\;n=31)$. 2) As the K ion concentration was increased, resting membrane potential was depolarized but showed hyperpolarization with $K^{+}$ below 25mM. 3) Alteration of the resting membrane potential for the changes of $Na^{+}$ concentration were hardly observed, while resting potential was hyperpolarized as $Ca^{2+}$ concentration was increased. 4) Pump activity as transient or prolonged hyperpolarization was $-2.29{\pm}0.75mV$ $(Mean{\pm}Se,\;n=16)$, the hyperpolarization was increased in both amplitude and duration under the 10mM $Ca^{2+}$ solution. 5) Hyperpolarization due to pump activity was decreased or disappeared by $5{\times}10^{-5}\;M$ ouabain treatment and could not be observed under the both Na-free and Ca-free solutions. 6) Above results are likely to suggest that the resting potential of the mouse unfertilized eggs is affected to mainly by Ca-dependent K conductance and Na-Ca exchange mechanism and that there is pump activity coupling between $K{+}$, $Na^{+}$ and $Ca^{2+}$.

Antibacterial Activities of Peptides Designed as Hybrids of Antimicrobial Peptides

  • CA(1-8)ME(1-12), the CA-ME hybrid peptide of the amino terminal segments of cecropin A (CA) and melittin (ME), has been reported to have a broad spectrum and improved potency without a hemolytic property. In order to obtain new synthetic peptides with powerful antibacterial activity without hemolytic activity, several hybrid peptides were designed from the sequences of CA, ME, magainin 2, bombinin and lactoferricin. All hybrid peptides were constructed to form an amphipathically basic-flexible-hydrophobic structure and synthesized by the solid phase method. Their hemolytic activities against human red blood cells and antibacterial activities against both Gram-positive and Gram-negative bacteria were detennined. CA(1-8)MA(1-12), CA(1-8)BO(1-12), MA(10-17)ME(1-12) and LF(20-29)ME(1-12) showed comparable activities with broad spectra against both Gram-positive and Gram-negative bacteria relative to CA(1-8)ME(1-12) but without hemolytic properties. These hybrid peptides, therefore, could be useful as model peptides to design a novel peptide with improved antibacterial activity and study on structure-activity relationships of antimicrobial peptides.

Synthetic Lead Compounds Modulate Activity of Large-conductance $Ca^{2+}$-activated Potassium Channels Expressed in Xenopus Oocytes

  • Large-conductance $Ca^{2+}$-activated potassium channels ($BK_{Ca}$ are a widely distributed and play key roles in various cell functions. In nerve cells, $BK_{Ca}$ channels shorten the duration of action potentials and block $Ca^{2+}$ entry thereby repolarizing excitable cells after excitation. $BK_{Ca}$ channel opening has been postulated to confer neuroprotection during stroke, and has attracted attention as a means for therapeutic intervention in asthma, hypertension, convulsions, and traumatic brain injury. Several natural and synthetic compounds including a steroid hormone, $\beta$-estradiol, have been identified as the activators of $BK_{Ca}$ channels. Based on the structural features of the previously reported activators of $BK_{Ca}$ channels, we designed several lead compounds, synthesized chemically, and tested their functional activity on cloned $BK_{Ca}$ channels. The $\alpha$ subunit of rat $BK_{Ca}$ channel was expressed alone or with different $\beta$ subunits in Xenopus oocytes and the effects of the compounds were tested electrophysiological means. One of the lead compounds affected the activity of the $\alpha$ subunit of $BK_{Ca}$ channel in a $\beta$ subunit-specific manner. While the activity of B $K_{ca}$ channel $\alpha$ subunit was Potentiated, the channel composed of $\alpha$ and $\beta$1 subunits were inhibited by this compound. We are currently investigating the mechanism of the $\beta$ subunit-dependent effects and planning to localize the receptor site of the lead compound.f the lead compound.

Novel $Ca^{2+}$-ATPase Found in the Human Milk Membrane Fraction

  • Calcium-stimulated ATPase ($Ca^{2+}$-ATPase) which has optimal pH value at 7.0 was found in the membrane fraction of human milk, and its enzymatic properties were studied. The purified $Ca^{2+}$-ATPase required 0.45 mM Ca ion for maximal activity. Among the nucleosides, $Ca^{2+}$-ATPase showed a higher substrate specificity to ATP and UTP than to CTP and GTP. $Ca^{2+}$-ATPase had apparent Km value of 0.065, and V max of 7.63 mol ATP hydrolyzed/mg pro-tein per min, respectively. $Ca^{2+}$-ATPase was potently inhibited by lanthanide, vanadate, and p-chloromercuribenzoate, and inactivated by EDTA, and CDTA and EGTA, but were unaffected by N-ethylmaleimide, $NaN_3$, ouabain, or oligomycin, and was completely inactivated by heating at $60^{\circ}C$ for 10 min. This enzyme activity was concentrated in the membrane fraction of the cream and skim milk membrane, but not founded in bovine milk.

Caffeic Acid의 항산화 활성 및 Amyloid beta와 LPS에 의한 C6 Glial 세포의 산화적 스트레스 보호 효과 (Antioxidant Activity and Protective Effect of Caffeic Acid against Oxidative Stress Induced by Amyloid Beta and LPS in C6 Glial Cells)

  • This study was investigated the radical scavenging effect and the protective activity of caffeic acid (CA) against oxidative stress. CA showed strong 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and hydroxyl radical ( OH) scavenging activity, showing 42.00% and 87.22% at 5 μM concentration of DPPH and ·OH scavenging activity, respectively. Furthermore, we studied protective activity of CA from amyloid beta (A${\beta}$25-35) and lipopolysaccharide (LPS) induced neuronal cell damage and neuronal inflammation using C6 glial cells. The treatment of A${\beta}$25-35 to C6 glial cell showed declines in cell viability and high generation levels of reactive oxygen species (ROS). However, the treatment of CA increased cell viability. The treatment of 5 ${{\mu}M}$ CA led to the elevation of cell viability from 59.28% to 81.22%. In addition, the production of ROS decreased cellular levels of ROS by the treatment of CA. The treatment of LPS to C6 glial cells increased significant elevation of nitric oxide (NO) production, while CA decreased NO production significantly. The production of NO increased by the treatment of LPS to 131.08%, while CA at the concentration of 1 ${{\mu}M}$ declined the NO production to 104.86%. The present study indicated thatCA attenuated A${\beta}$25-35-induced neuronal oxidative stress and inflammation by LPS, suggesting as a promising agent for the neurodegenerative diseases.

Role of Regulators of G-Protein Signaling 4 in $Ca^{2+}$ Signaling in Mouse Pancreatic Acinar Cells

  • Regulators of G-protein signaling (RGS) proteins are regulators of $Ca^{2+}$ signaling that accelerate the GTPase activity of the G-protein ${\alpha}$ -subunit. RGS1, RGS2, RGS4, and RGS16 are expressed in the pancreas, and RGS2 regulates G-protein coupled receptor (GPCR)-induced $Ca^{2+}$ oscillations. However, the role of RGS4 in $Ca^{2+}$ signaling in pancreatic acinar cells is unknown. In this study, we investigated the mechanism of GPCR-induced $Ca^{2+}$ signaling in pancreatic acinar cells derived from $RGS4^{-/-}$ mice. $RGS4^{-/-}$ acinar cells showed an enhanced stimulus intensity response to a muscarinic receptor agonist in pancreatic acinar cells. Moreover, deletion of RGS4 increased the frequency of $Ca^{2+}$ oscillations. $RGS4^{-/-}$ cells also showed increased expression of sarco/endoplasmic reticulum $Ca^{2+}$ ATPase type 2. However, there were no significant alterations, such as $Ca^{2+}$ signaling in treated high dose of agonist and its related amylase secretion activity, in acinar cells from $RGS4^{-/-}$ mice. These results indicate that RGS4 protein regulates $Ca^{2+}$ signaling in mouse pancreatic acinar cells.
