Characteristics and Screening of Antioxidative Activity for the Fruit by Rubus coreanus Miq. Clones (복분자딸기 클론별 과실특성과 항산화 활성 탐색)

  • Fruit characteristics, saccharinity, and antioxidative activity of selected clones of Rubus coreanus, grown in Korea, well-known as edible and medical resources, frequently used and which has an excellent pharmacological activity are examined and analyzed. The results obtained from this study can be summarized as follows; Result of surveying characteristics for the fruit and saccharinity variation by collection time, there are some meaningful differences between examined clones and the collection time, exception of the fruit width. From the clones, SG 3 is the biggest in size, the highest in the saccharinity out of five clones, on the other hand, CJ 18 and SH 3 are the opposite. Meanwhile, comparing examined saccharinity factor with maximum temperature and precipitation out of daily climates at Suwon Meteorological Observatory at the same period, on the day the highest temperature is $29.5^{\circ}C$, saccharinity distinctively ascends, but on the day the precipitation is recorded 121.5 mm and next day, the saccharinity descends. Also, five-day saccharinity until the precipitation lower than 0.1 mm per day shows from 10.4 to 11.5 Brix higher than the average saccharinity of 9.7 Brix. Fifteen clones including CA 6 about fruit characteristics in terms of collection layer and saccharinity variation were examined, there is significantly difference among examined clones, but about the collection layer, only saccharinity factor has meaningful significance. For clones, MJ 11 is the biggest and excellent character of 15 clones but the saccharinity, 10.3 Brix lows a bit, MC 9 and HAE 5 are the worst fruit in character but saccharinity is 13.4 Brix and 10.6 Brix which is higher than the average saccharinity of 9.8 Brix. About saccharinity for the collection layer, the fruit average saccharinity, collected from the upper layer which is good at lightness and temperature condition, is 10.2 Brix which shows higher than the one of the fruit, 9.7 Bix and 9.5 Brix collected from the middle or the lower layer. The saccharinity of fruit collected from the middle or the lower layer is lower than the average saccharinity of 9.8 Brix out of 15 surveyed clones. Surveying antioxidative activity resulted from free radical scavenging activity about 30 clones of fruit in total, there is difference between surveyed clones. Especially, Immatured fruit extracts exhibited higher free radical scavenging activity of 94.5 to 97.5% which indicates the similarity to Ascorbic acid, unlike other clones, on the concentration of 250 to 1,000 ppm.

  • This inquiry was conducted to develop DeNOx catalyst for LNT. In order to develop appropriate catalysts, four catalysts, which do not use PGM (Platinum Group Metal), were carefully selected : Al/Co/Mn, Al/Co/Ni/Mn, Al/Co/Mn/Ca, Al/Co/Ni mixed metal oxides during preliminary experiments. Also, XRD, EDS, SEM, BET and TPD tests were carried as well to evaluate both physicochemical properties of such four catalysts. As a result of the experiment, four catalysts were composed of spinel-shaped crystals and had more than enough pore volume and size to have oxidation-reduction reaction of NOx gases. Additionally, through TPD test, all four types of catalysts were proved to possibly have an oxidation-reduction acid site and NO oxidation activities similar to commercial catalysts. Based on the results above, if we have further change in the composition components and active ingredients according to the catalysts that were chosen in this investigation, then we are more welcomed to expect to have an enhanced DeNox catalyst for LNT.

Study on the Antioxidant and Anti-inflammatory Effects of Extracts of Callus Cultures, pericarp, flesh, fruit of Trapa Japonica (능실 열매의 부위별 추출물 및 캘러스배양 추출물의 항산화, 항염증 효과 연구)

  • The purpose of this study was to evaluate the antioxidant and anti-inflammatory effects of extracts of callus cultures, pericarp, flesh, fruit of Trapa Japonica. The toxicity of extracts from Trapa Japonica pericarp investigated using the RAW 264.7 cell showed 45.8 ± 1.5% of cell survival rate. And the results of investigation using HaCaT cells showed a 51.1 ± 1.0% cell viability at 100 ㎍/㎖ in the pericarp extract and lower cell viability at higher concentrations. The total content of polyphenol pericarp extract was 213.20 ± 15.78 mg/g, while the total content of flavonoid was 29.30 ± 3.24mg. And the total content of polyphenol callus cultures extract was 205.20 ± 18.97 mg/g, while the total content of flavonoid was 237.4 ± 7.43 mg. With a concentration level of 0.005 ~ 1000 ㎍/㎖ extract of Trapa Japonica pericarp the range of removal of 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radicals was 67.53 ± 1.5 % ~ 75.75 ± 0.5 % respectively and the range of removal of extract of Trapa Japonica callus cultures extract was also 3.1 ± 0.1 % ~ 77.32 ± 0.5 % respectively. As a result of measuring the Nitric Oxide(NO) generation amount of all Trapa Japonica extract 6.25, 12.5, 25, 50 ㎍/㎖ concentration exhibited significant (p < 0.05, p < 0.01) decreases.

Manufacture of Nutritionally Balanced 'Sunsik' for the Moderns: Its Quality Characteristics (현대인을 위한 기능성 영양균형선식의 제조와 품질)

  • This study was conducted to investigate the quality characteristics of functional 'sunsik' as the nutritionally balanced diet for the moderns. The major nutrient of nutritionally balanced 'sunsik' was carbohydrate, while contained lower contents of protein and lipid than general 'sunsik'. Sucrose was the major free sugar of nutritionally balanced 'sunsik', and major organic acids were malic, tartaric, oxalic and citric acid. Free amino acids were predominantly serine, arginine, alanine and glutamic acid. Linolenic acid $(12.52\%)$ was contained in the nutritionally balanced 'sunsik'. Ca, Mg, K, Na and Fe were found as major minerals. Antioxidant activity was about $29\%$ compared to BHA and BHT(200 ppm), and about $40\%$ of $\alpha-tocopherol$(400 ppm). Sensory evaluation of nutritional balanced 'sunsik' gained higher sensory score in color, cooked aroma, flavor, chewiness and overall acceptance, compared to the general 'sunsik'.

Analysis on the Components of the Elaeagnus multiflora Thunb. Leaves (뜰보리수 잎의 유용성분 분석)

  • This study analyzed components of the leaves of Elaeagnus multiflora as part of studies on the nutritional and functional materials of fruits and leaves of this plant. The moisture content of the leaves was 71.6% and the carbohydrate, crude protein, lipid and ash contents were 24.1, 1.4, 0.4 and 2.5%, respectively. Concentrations of reducing sugars, soluble proteins and polyphenols were 460.0, 503.3 and 805.6 mg/100 g, respectively. Fructose was the dominant free sugar, and arabinose, maltose, glucose, and a small amount of trehalose were also detected. Malic acid was the main organic acid in E. multiflora leaves, and acetic acid, citric acid, lactic acid, and succinic acid were also present. E. multiflora leaves were high in K, Ca and Mg. Of hydrolyzed amino acids, alanine was present at the highest concentration (112.0 mg/100 g), with threonine, leucine, valine and phenylalanine being the next most common. Glutamic acid and ornithine were the dominant free amino acid and amino acid derivative, respectively.

Physicochemical Properties of Flowering Cherry (Prunus serrulata L.) Fruits According to Cultivars (버찌의 품종별 이화학적 특성)

  • Physicochemical properties of flowering cherry (Prunus yedoensis, Prunus sargentii, Prunus lannesiana, Prunus cerasus) fruits were evaluated. The moisture contents were the highest in fruits of Prunus cerasus; crude protein ranged from $0.19{\sim}1.49%$ and crude fat ranged from $1.00{\sim}2.66%$. The fruits of Prunus lannesiana included higher contents of crude ash compared with those of other cultivars. The mineral contents of Prunus lannesiana fruits were the highest in Ca, K, Mg, Fe, and Mn. The amino acid contents of only methionine was the highest in fruits of Prunus sargentii (1.64 mg/L). Other amino acids were the highest in fruits of Prunus cerasus. The Brix value was the highest in fruits of Prunus yedoensis, followed by fruits of Prunus sargentii, Prunus lannesiana, and Prunus cerasus. Organic acid contents ranged from $3.42{\sim}4.18%$. Anthocyanin contents were the highest in fruits of Prunus sargentii in methanol extract. Therefore oriental cherry commensurates with development of nature pigment and functional foods. Especially, Prunus cerasus was greater than other cultivars.

Effects of Bambusae Caulis in Taeniam Extract on the UVB-induced Cell Death, Oxidative Stress and Matrix Metalloproteinase 1 Expression in Keratinocytes (각질세포에서 자외선B가 유도한 세포 사멸, 산화적 스트레스 및 matrix metalloproteinase 1 발현에 대한 죽여추출물의 영향)

  • Ultraviolet radiation (UV) is a major cause of skin photoaging, and effective UV protecting agents are needed for the skin health and beauty. This study was undertaken to examine the effects of Bambusae caulis in Taeniam extract (BCTE) on UVB-induced cell death, oxidative stress and matrix metalloproteinase 1 (MMP1) expression in cell-based assays. HaCaT human keratinocytes were exposed to UVB in the presence of BCTE at different concentrations and resulting changes in cell viability and biochemical events were determined. The results showed that BCTE enhanced the viabilities of UVB-exposed cells, and attenuated apoptotic events such as cleavage of procaspase 3 to its active form, and the increase of Bax to Bcl-2 ratios. BCTE also attenuated the reactive oxygen generation and lipid peroxidation in cells exposed to UVB. Additionally, it attenuated the expression of matrix metalloproteinase 1 and the phosphorylation of c-Jun N-terminal kinase stimulated by UVB. Conclusively, the present study demonstrated that BCTE pro tected skin cells from the UVB-induced cell death, oxidative stress and MMP1 expression, suggesting its potential use as a cosmetic ingredient mitigating some features of the skin photoaging.

Application of Antimicrobial Peptides against Microcystis aeruginosa to Control Harmful Algal Blooms (항균 펩타이드를 이용한 녹조현상 원인종 Microcystis aeruginosa의 제어)

  • Microcystis aeruginosa, a freshwater cyanobacteria species known to be one of the most predominant species responsible for cyanobacterial harmful algal blooms (CyanoHABs). It has been frequently associated with the contamination of neurotoxins and peptide hepatotoxins, such as microcystin and lipopolysaccharides-LPSs. CyanoHABs control technologies so far put in place do not provide a fundamental solution and cause secondary pollution linked with the control measures. For this study, algicidal peptides, which have been reported to be non-toxic and to have antimicrobial properties, were employed for the development of novel eco-friendly control against CyanoHABs. The four peptides (CMA1, CMA2, HPA3P, and HPA3NT3) selected in this study showed significant algicidal effects against M. aeruginosa cells inducing cell aggregation and flotation. Moreover, the newly generated peptides (K160242-5) with certain modifications also displayed high algicidal activity. The algicidal activity of the peptides was found to depend on the concentrations and structures of each of amino acid. The results of this study suggested a novel possibility of CyanoHABs control using the non-toxic algicidal peptides.

Study on Reinforcing Skin Barrier and Anti-aging of Exosome-like Nanovesicles Isolated from Malus domestica Fruit Callus (사과 캘러스로부터 분리된 엑소좀-유사 Nanovesicles 의 피부 장벽 및 피부 노화 방지 개선 연구)

  • Plant-derived exosome-like nanovesicles (PELNs) are known to include various biological activities and possess high biocompatibility. Because PELNs can influence immune responses, cell differentiation, and proliferation regulation, they can be applied in multiple industries. However, the studies on the skin physiological of exosome-like nanovesicles derived from plant callus are insignificant compared to nanovesicles derived from mammalian cells. In this study, callus was induced from apple fruit (Malus domestica), and exosome-like nanovesicles (ACELNs) were isolated for improving skin barrier and anti-aging. The yield of ACELNs was 6.42 × 109 particles/mL, and the particle size was ranged from 100 to 200 nm. HDF cells and HaCaT cells were concentration-dependent, increased in cell, and decreased in cytotoxicity. The cornified envelope formation was significantly increased compared to the control group. The COL1A1 expression and the FBN1 expression in HDF cells were increased. In addition, the ACELNs promoted collagen biosynthesis in UVA-irradiated HDF cells. These results might be considered as potential materials that could improve skin barrier and prevent skin aging.

  • This study was carried out by investigating the changes of physico-chemical and mechanical properties of Gwanghwal and Poseung series in order to obtain basic information on the soil improvement and management. 1. In Gwanghwal series, the content of salt in soil and ground water before rice transplanting at the 21st year after reclamation were 0.29 and 1.02 percent respectively, and the safty cultivation of paddy rice was possible after 33 years of reclamation in both soil series. 2. Clay contents. atterberg limits and clay activity according to the age of reclaimation showed negative correlation. but sand content and N-value were positive in them. 3. Cone index was lower than $3kg/cm^2$ for the first 5 years after reclaimation in both soil series, and the formation of hard pan were 33rd and 49th year after reclamation in Gwanghwal and Poseung series respectively, and its thickness was more in Poseung than in Gwanghwal series. 4. Shearing resistance decreased with the age of reclaimation in both soil series. and friction resistance increased in Gwanghwal than Poseung siries. 4. Soil pH. available, $SiO_2$, $Mg^2$, $K^+$, $Na^+$ and C. E. C showed negative correlation. with the year cultivated paddy rice. but O. M. ava ilable $P_2O_5Ca^{2+}$ and T-N had positive one in both soil series. Contents of O. M and $Ca^{2+}$ and T-N had positive one in both soil series. Contents of O. M and $Ca^{2+}$ were high in Gwanghwal, but C. E. C. was in Poseung series.
