  • The heat-storage characteristics accompanied by exothermic reaction at the regeneration of $Ca(OH)_2$ in tile heat-storage mode of a chemical heat pump system using a $Ca(OH)_2/CaO$ reversible thermochemical reaction was examined in a lab-scale unit. In this heat-storage mode, the particle bed of CaO could be regenerated by heating the $Ca(OH)_2$ packed bed to the higher temperature at which the equilibrium pressure in the reactor is greater than the water vapor pressure in the condenser. The results are i) the dehydration, thermal decomposition, rate of $Ca(OH)_2$ was higher at the lower part of particle bed than at the upper part, ii) in the reactor, the dehydration was proceeded along radial and axial direction, from inner part to the outer part, which explains heat transfers from the center to wall and from the tenter to lower or upper part of reactor.

  • It has been shown in this and earlier investigation that the turtle bladder mucosa has three main cell types on their mucosal surface. They are the granular cells, ${\alpha}$ CA cells, and ${\beta}$ CA cells. The three major transport mechanisms that occurs in the turtle bladder are sodium reabsorption, proton secretion, and bicarbonate secretion. In the present work the trans-port mechanisms by bladder epithelial cells of freshwater turtle, Pseudemys scripta, are summarized as follows. 1. The granular cells play an important role in sodium transport, while the ${\alpha}$ and ${\beta}$ CA cells do not appear to play a determining role in sodium transport. 2. It appears that the active sodium transport in the granular cells occurs in two-step process, implying that first, sodium diffuses into the cells, followed by an energy-dependent efflux step, which is catalyzed by the ouabain-sensitive Na-K ATPase. 3. The ${\alpha}$ type of CA cells are responsible for the proton secretion using the proton pump on the apical plasma membrane, while the ${\beta}$ type of CA cells are believed to be responsible for bicarbonate secretion. 4. When looked at under freeze-fracture electron microscopy, the apical plasma membrane of ${\alpha}$ cells have a characteristic population of rod-shaped intramembranous particles which are believed to be components of the proton pumps. Conversely, ${\beta}$ type of CA cells show rod-shaped particles in their basolateral plasma membranes, which is consistent with the proton absorptive, bicarbonate secretory mechanism. 5. In the turtle bladder, the ${\alpha}$ and ${\beta}$ type of cells are believed to be both responsible for proton transport, but in opposite directions.

  • To develope chemical heat pump using available energy sources such as solar heat and many kinds of waste thermal energy we have studied the enhancement effect of inserted fins in cylindical packed bed reactor. Two dimensionnal(radial and circumferential) partial differential eqaetions, concerning heat and masstransfer in CaO packed bad, are solved numerically to describe the characteristics of the reaction of fins inserted reactor and heat transfer. The results obtained by numerical analysis about two dimensional profiles of temperature and conversion in the reactant in the packed bed and exothermic heat amount released from the reactor are follows; -. The insertion of fins in reactor can redue the reaction completion time by half. -. The rate of thermochemical reaction depends of the temperature and concentration and it is also governed by the boundary conditions and heat transfer rate in the particle packed bed.

  • This study was carried out to examine the $H^+$ transport mechanism by observing the properties of cellular membrane having an ${\alpha}$ type of carbonic anhydrase (CA)-containing cells in turtle urinary bladder. The urinary bladder consists of a heterogenous population of cells. As a result of fine observation with traditional thin-section electron microscopy. the bladder epithelium has three different cell types on mucosal surface. They are a basal cell, a granular cell and a third type of CA-rich cell. The CA-rich cells are divided into two distinct smaller groups within them and called them ${\alpha}$ type and ${\beta}$ type of CA cells. The ${\alpha}$ type of CA cells are responsible for the proton secretion using the proton pumps on the apical plasma membrane, while the ${\beta}$ type of CA cells secrete bicarbonate via an oppositely-directed proton pumps in their basolateral plasma membrane. After performing the freeze-fracture technique, it was shown that there were distributed a large number of intramembranous particles having a special structure on the apical membrane of ${\alpha}$ type of CA-rich cells in the process of their $H^+$ secretion. In turtle bladder ${\alpha}$ type of CA-rich cells, this particle was the only prominent structure in the apical membrane. These intramembrane rod-shaped particles probably represent the integral membrane components of the proton pump. This result may explain that carbonic anhydrase within epithelial cell of urinary bladder takes part in formation of $H^+$ and bicarbonate, that active transport of $H^+$ is done, and that the reabsorption of bicarbonate suggests transport mechanism containing $H^+$ secretion. However, it seems that more studies are required for considering their regular transport pathway.

  • In this paper, with water-LiCl-$CaCl_2-Zn(NO_3)_2$ mixture which utilizes solar energy at the evaporator heat source, a thermodynamic analysis was performed to provide design data for a double-effect absorption heating system. A comparative study of the water-LiCl-$CaCl_2-Zn(NO_3)_2$ mixture against the water-LiBr pair was conducted by a computer simulation. The computer simulation is based on mass, material and heat balance equations for each part of the system. Coefficients of performance and flow ratios for effects of different operating temperatures are investigated. It is found that the heating COP is higher for the water-LiCl-$CaCl_2-Zn(NO_3)_2$ mixture than for the water-LiBr pair, and FR is lower for the former.

  • The effects of scorpion (Leiurus quinquestriatus hebraeus, Lqh) venom were evaluated on the activities of microsomal $Ca^{2+}-ATPase$ and $Ca^{2+}$ release channel prepared from the epithelial cells of pig airway. Whole venom of Lqh $(120\;{\mu}g/ml)$ increased the activity of microsomal $Ca^{2+}-ATPase$ about 32% in the tight-sealed microsomes and about 28% in the Triton X-100-treated or $Ca^{2+}$ ionophore A23187-treated leaky microsomes. Thapsigargin, a specific antagonist of $Ca^{2+}-ATPase$, inhibited 42% of total ATPase activity and also completely blocked the effects of Lqh venom, suggesting that Lqh venom directly activiates the microsomal $Ca^{2+}-ATPase$. In order to determine if Lqh venom increases the microsomal uptake of $^{45}Ca^{2+}$, Lqh venom was added in the uptake medium. The Lqh venom increased microsomal $^{45}Ca^{2+}$ uptake up to ${\sim}20%$ and the increase was only observed when heparin, an antagonist of $InsP_3$ receptor channel, was added in the uptake medium. Lqh venom in the absence of heparin unexpectedly decreased the rate and the amount of $^{45}Ca^{2+}$ uptake. These results were explained by simultaneous increases in $^{45}Ca^{2+}$ release as well as $^{45}Ca^{2+}$ uptake by Lqh venom. Lqh venom itself increased the release of $^{45}Ca^{2+}$ as much as $^{45}Ca^{2+}$ release by $4\;{\mu}m\;InsP_3$, implying that Lqh venom also activates $InsP_3$ receptor, microsomal $Ca^{2+}$ release channel. Based on these results, we suggest that the Lqh venom consists of at least two components; one activates the $InsP_3$ receptor and the other avates the $Ca^{2+}-ATPase$. Currently we a investigating the chemical and electrophysiological properties of the active components of Lqh venom.

  • In order to investigate the mechanisms of epithelial ion transports, microsomes of soybean roots were prepared and the activity of microsomal ATPases was measured by an enzyme-coupled assay. The effects of various ions were evaluated on the total activity of microsomal ATPases and the average activity was 190 nmol/min/mg protein in the control solution containing $10\;mM\;Na^+\;and\;120\;mM\;K^+$. The activities were increased to 150% and decreased to 63% of the control activity in the solution containing $130\;mM\;K^+$ without $Na^+$ and in the solution containing $130\;mM{\;}Na^+$ without $K^+$, respectively. In general, the activity of microsomal ATPase was increased by$K^+$ in a concentration-dependent manner The activity was also increased at lower pH and relatively higher activities were observed in the pH range of $6{\sim}7$. However, the activity was decreased at weak alkaline $pH\;and{\sim}80%$ of the activity was inhibited at pH 9. Since intracellular $Ca^{2+}$ has been known to control the activity of various enzymes, we have investigated the effects of intra-and extrarnicrosomal $Ca^{2+}$ on the activity of microsomal ATPases. The maximal activity was obtained at the extrarnicrosomal $Ca^{2+}$ concentrations below 1 nM. The activity was gradually decreased by increasing $‘Ca^{2+}’$ concentration and 50% inhibition was observed at ${\sim}500{\;}{\mu}M{\;}Ca^{2+}$. The increase in luminal $Ca^{2+}$ concentration also inhibited the activity of microsomal ATPase. When the influx of external $Ca^{2+}$ was induced by $Ca^{2+}$ ionophore A23187 treatment, the activity was decreased by 30%; however, it was recovered by EGTA-induced chelation of $Ca^{2+}$. These results suggest that the presence of $Ca^{2+}$ regulation sites on both cytoplasmi and luminal sides of microsomal ATPases.

  • Phospholamban is the regulator of $Ca^{2+}-ATPase$ in cardiac sarcoplasmic reticulum(SR). The mechanism of regulation appears to involve inhibition by dephosphorylated phospholamban. Phosphorylation of phospholamban relieves this inhibition. Recently, there has been a report that the cytoplasmic domain (amino acids 1-25) of phospholamban is insufficient to inhibit the $Ca^{2+}$ pump. To explore the domains of phospholamban responsible for $Ca^{2+}-ATPase$ inhibitory activity, we examined the effect of a synthetic phospholamban peptide consisting of amino acid residues 1-25 on $Ca^{2+}$ uptake by reconstituted skeletal SR $Ca^{2+}-ATPase$. The $Ca^{2+}-ATPase$ of skeletal SR was purified and reconstituted in proteoliposomes containing phosphatidylcholine (PC) or phosphatidylcholine: phosphatidylserine (PC:PS). Inclusion of a phospholamban peptide in PC proteoliposomes was associated with significant inhibition of the initial rates of $Ca^{2+}$ uptake at pCa 6.0, and phosphorylation of this peptide by the catalytic subunit of cAMP-dependent protein kinase reversed the inhibitory effect on the $Ca^{2+}$ pump. Similar effects of phospholamban peptide were also observed using PC:PS proteoliposomes. Based on these results, we could conclude that the cytoplasmic domain of phospholamban, containing the phosphorylation sites, by itself is sufficient to inhibit the $Ca^{2+}$ pump of SR.
