Dietary Effect of Puer Tea Extract on the Body Weight in Rats

  • Puer tea is a traditional beverage originating from Yunnam area of China. It supplies basic nutrients such as vitamin C. It has been well reported that daily drinking of Puer tea can help the digestion and ease the stomachache after food intake. Puer tea also contains various polyphenols which may exert antibacterial activity against Staphylococcus aureus and Listeria monocytogenes. Because of these functional effects on digestive system we suspected if Puer tea can display any dietary effect or decrease the obesity after long-term drinking. We employed 6-week old SD rats as experimental animal and treated them with extract of Puer tea in relation to the body weights. Rats were divided into 5 groups (NC, PC, E, E+P, E+P5). NC group was experimental control and rest of them are as follows: water only (PC), water with exercise (E), water with exercise and Puer tea extract (E+P), water with exercise and 5X extract of Puer tea (E+P5). Feeding was carried out every day for 5 weeks by oral administration. Reduction rate of body weights was highest in E group. Relative ratio of losing weight was as follows: PC group (100.78%), E group (95.57%), E+P group (94.53%) and E+P5 group (74.22%), respectively. Exercise was more helpful to control the body weight. The result strongly suggests that Puer tea is highly effective to control the body weight and could be used for pharmaceutical purpose to treat obesity without side effects.

  • An investigation was conducted to evaluate the effects of supplementing Alisma canaliculatum, Viscum album and Cornus officinalis probiotics on the growth performance and immune response in growing pigs. This experiment was conducted using 120 pigs (crossing of Landrace${\times}$Yorkshire and castrated) which were assigned to 5 treatments in 3 replications with 8 pigs per replications. The dietary treatments were NC group (without antibiotics), PC group (basal+Oxytetracycline 50ppm), AC group (basal+A. canaliculatum 0.5%), VA group (basal+V. album 0.5%) and COP group (basal+C. officinalis probiotics 0.5%). The initial body weights of pigs were 35kg on average and the experiment lasted for 9 weeks. The experimental animals were kept in the pens following a completely randomized design. They were provided the diets adequate for grower stage as recommended by NRC (ME:3,265 kcal/kg and CP:16%). COP fed pigs showed lower weight gain up to 6 weeks of age compared to NC group and other groups without significant differences (P>0.05). The carcass weights of pigs fed VA and COP were significantly higher compared to NC group (P<0.05), Back fat thicknesses groups fed three different additives were higher than NC group and lower then PC group (P<0.05). Crude fat contents in loin meat were significantly lower in groups fed three different additives while moisture contents of those three groups were higher than other groups (P<0.05). The thiobarbituric acid reaction substance (TBARS) value measured at fresh and $2^{nd}$ weeks was lower in additives fed groups but no statistical differences were observed among the treatments (P>0.05). Significantly highest PUFA (16.42g/100g) and ${\omega}$-3 fatty acids (ALA, EPA and DHA) content of meat were observed in COP fed pigs compared to NC group (P>0.05), which might mean that three additives function to enhance serum IgG in pigs. In consequence, it can be suggested that AC, VA and COP may have a potential to replace antibiotics as growth promoter and immune enhancer in the diets for growing pigs.

  • Bis(N-Methylphenazinium)-Tetracyanopalladate(II) Hydrate $(C_{13}H_{11}N_2){_2}[Pd(CN)_4]{\cdot}H_2O$의 단위 착이온 및 결정의 구조들을 X-선 회절법으로 연구하였다. 이 결정은 사방정계이고 공간군은 $P2_1/b$(군번호=14)이다. 단위세포 a=9.783(4), b=10.788(4), c=13.666(4) ${\AA},\;{\beta}=104.59(5),\;V=1392.9{\AA}{^3},\;Z=2,\;Dc=1.476gcm^{-3},\;F(000)=632,\;{\mu}=7.05cm^{-1}$이다. 구조분석은 중금속법으로 풀었으며, 최소자승법으로 정밀화 하였고, 최종 신뢰도 값들은 1930 회절반점에 대하여 R=0.0257, Rw=0.0732, Rall=0.0283 및 S=1.07 이었다. 두 이온들은 근본적인 평면구조로써, 이들의 충진구조는 착음이온을 두개의 양이온들이 거의 평행하게 둘러싸고 있는 삼중체들을 형성하고 있다. 양이온과 음이온들의 이면각은 $10.16(4)^{\circ}$인 삼중체들이 b축을 따라서 배열되어 있다, 삼중체내 및 삼면체간의 면간 거리들은 3.419(3)와 $3.402(4){\AA}$이었다.

  • Ethylenediammonium chromate, $C_2H_{10}N_2{\cdot}CrO_4$ 결정구조는 a=6.667(2), b=8.845(2), c=11.827(2) ${\AA}$, 사방정계(Orthorhombic), 공간군 $P2_12_12_1$, 공간군 번호 19이며, Z=4, V=697.4(3) ${\AA}{^3},\;Dc=1.696gcm^{-3},\;{\mu}=1.594mm^{-1}$이었다. 구조분석은 중금속법으로 풀었으며, 최소 자승법으로 정밀화하였고, 최종 신뢰도 값들은 1204개의 회절반점에 대하여 $R_1=0.0254,\;R_w=0.070,\;R_{all}=0.0255$, S=1.133이었다. 두가지 이온들의 자료들은 기존 유사물질들의 자료에 상응하였으며, Ethylenediammonium 이온은 trans 구조로써 주위의 음이온과 사이에 많은 수소결합으로 연결되어 있다.

Bacillus sp. A-6의 Xylanase의 정제와 특성 (Purification and Characterization of Xylanase from Bacillus sp. A-6)

  • Bacillus sp. A-6의 배양액의 상등액으로부터 한외여과와 5 mM sodium acetate, pH 5.0 용액으로 평형화된 SP-Sepharose column을 사용한 이온교환 크로마토그래피에 의해 xylanase를 정제하였다. Column에 흡착된 xylanase는 0.05 M NaCl 이하의 농도에서 용출되었다. 용출된 xylanase가 SDS-PAGE에서 단일 펩티드 밴드로 분리되어 순수함을 확인하였으며 oat spelt xylan을 기질로한 zymogram에서 xylan을 분해하는 밴드로 나타났다. Xylanase의 분자량은 SDS-PAGE에서 15,000이었고 겔여과 크로마토그래피에서 14,100 이었다. 박층막 크로마토그래피에서 xylanase가 oat spelt xylan을 xylobiose와 xylooligosaccharide로 분해함을 보였다. Xylanase를 가열할 때에 상대활성도가 $40^{\circ}C$에서 7시간 후에 80%로 감소하였으며 $60^{\circ}C$에서는 1시간 후에 40% 이하로 감소하였다.

Effects of Different Levels of Concentrate in the Diet on Physicochemical Traits of Korean Native Black Goat Meats

  • The effects of feeding of diets based on roughage supplemented with concentrate on the carcass and meat quality attributes of Korean black goats were investigated. The 40 male goats at 5 mon age were divided into four treatment groups; T1 (1.5%), T2 (2.0%), T3 (2.5%) and T4 (ad libitum) feeding of concentrate with ad libitum rice straw. Forty bucks at the end of the feeding were fasted for 12 h before slaughter and were then slaughtered. For carcass measurement, cold buck carcasses were examined after 24 h of chilling at $5^{\circ}C$. The carcasses were chilled in refrigerator at $2{\pm}2^{\circ}C$ for 24 h, then the longissimus dorsi muscle was removed from the carcasses. The samples were separately vacuum packaged, frozen and stored at $-20^{\circ}C$ for up to 1 mon, prior to physicochemical evaluations. Carcass yields were greatly affected by rate of concentrate feeding. T4 showed the heaviest live and carcass weights and the highest dressing and fat percentages (p<0.05). The moisture contents were lower in T4 compared with other treatments, while the crude protein and fat contents were higher (p<0.05). mono-unsaturated fatty acid in T4 was significantly higher than others, which showed the highest percentages of oleic acid (C18:1). For the sensory test, T4 was tenderer and produced better flavor scores than others. Increasing the level of concentrate in the diet resulted in an improvement in growth performance, and carcass and physicochemical characteristics of goat meats.

고지방식이 유도 비만생쥐에 대한 영계출감탕(苓桂朮甘湯) 가(加) 황기(黃芪)의 항비만 효과 (The Anti-obesity Effects of Younggyechulgam-tang-ga Hwanggi on Obesity in Mice Induced by High Fat Diet)

  • Objectives This study was conducted to experimentally evaluate the effects of Younggyechulgam-tang-ga Hwanggi(YGT) on obesity in mice induced by high fat diet. Methods The experiment was conducted with 4-week-old male mice divided into 5 groups. They were a normal diet group(Nor), a high fat diet group(Veh), a positive drug control group-orlistat 40 mg/kg(Oris), a 1.08 g/kg group(YGTL), and a 2.16 g/kg group(YGTH), and were tested for five weeks. Changes in antioxidant activity, body weight, organ weight, ROS, AST, ALT, TC, TG, HDL-C, LDL-C and lipid metabolism protein were checked. Results YGTL and YGTH group significantly reduced body weight compared to Veh group. YGTH group significantly reduced visceral fat weights compared to Veh group. In blood biochemistry analysis, ROS, AST, ALT, TC, TG and LDL-C in YGTL and YGTH group were significantly lower than Veh group. HDL-C increased significance in YGTL and YGTH group. In antioxidation protein analysis, Catalase, GPx and HO-1 have increased significantly in YGTL and YGTH group. YGTH group have increased $PPAR-{\alpha}$, p-AMPK compared to Veh group. but decreased FAS. SREBP-1, p-ACC levels in YGTL and YGTH group were decreased compared to Veh group, however CPT-1, UCP-2 levels in YGTL and YGTH group were increased compared to Veh group. Conclusions YGT has anti-obesity effects by regulating lipolysis and antioxidation in a diet-induced obesity model. Additional clinical studies are needed.

사람의 p53 유전자와 Glutathione S-Transferase와의 융합 단백질의 대장균에서의 발현 (Expression of Human p53 Gene as Glutathione S-transferase Fusion Proteins in Escherichia coli)

  • p53 유전자의 변화는 인간의 여러 암에서 가장 흔하게 발견되며 종양세포내에서는 이러한 변형된 p53 단백질의 양의 증가가 초래된다. 세포내에 축적된 p53 단백질의 발견은 인간의 암증세를 판단할 유용한 기중이 되기도 한다. 본 연구에서는 이러한 면역조직화학 검사에 쓰일 수 있는 폴리클로날 항체를 만들기 위햐여 사람의 p53 유전자를 glutathione S-transferase 와의 융합 단백질의 형태로서 대장균내에서 발현시켰다. p53 의 아미노산 1-158번을 코딩하고 있는 NeoI fragment 와 아미노산 159-393 번을 코딩하는 NocI-BamHI fragment 를 BamHI linker 를 이용하여 in frame 으로 pGEX-2T 의 BamHI 자리에 삽입하여 재조합 플라스미드 pGTNS 와 pGTNL 을 각각 만들었다. 또 PCR 에 의한 증폭에 의햐여 아미노산 38-145번을 코딩하는 유전자 부위를 증폭하였으며 BamHI 과 PvuII 로 절단하여 pGEX-2T의 BamHI 과 SmaI 자리에 삽입함으로써 pGTBP 를 제조하였다. 이들 재조합 균주들을 IPTG 로 4시간 induction 한 후 세포 추출물로부터 glutathione Sepharose bead 를 이용하여 융합단백질을 분리하였다. Bead 에 결합된 단백질은 10% SDS-polyacrylamide gel 에서 전기영동하였으며, 각각의 분자량은 54 kDa, 53 kDa 와 40 kDa 였다. 이러한 방법으로 1리터 배양으로부터 약 1mg 의 단백질을 정제하였다.

율무청국장이 고지방식이를 급여한 흰쥐의 체중변화 및 지질대사에 미치는 영향 (Effects of Korean Traditional Cheonggukjang Added with Job's Tears on Body Weight Gains and Lipid Metabolism in Rats Fed High-Fat Diet)

  • 율무청국장이 고지방식이를 섭취한 흰쥐의 체중 및 체지방, 혈청과 간, 분변의 지질농도 및 혈당농도 등에 미치는 영향을 조사하고자 SD계 수컷 흰쥐를 대상으로 고지방식이 (D12492, 45 kcal fat)에 청국장 및 율무청국장(율무 함량 20%) 분말을 각각 10%, 20%씩 중량비로 첨가하여 5주간 급여한 결과는 다음과 같다. 체중증가량은 군별 유의성이 없었으나 식이효율은 고지방식이 대조군에 비해 율무청국장 분말 20% 첨가군에서 유의적으로 낮았으며(p<0.05), 지방조직의 무게는 실험군 간에 유사한 수준을 나타내었다. 배변량 및 간 조직의 총콜레스테롤과 중성지방 함량은 군별 유사한 경향을 나타내었으며, 분변의 총콜레스테롤 함량은 군별 유의적 차이가 없었으나 중성지방 함량은 대조군에 비해 실험식이 첨가 수준에 따라 유사하거나 증가하는 경향을 보였다. 혈청 총콜레스테롤, HDL-콜레스테롤 및 LDL-콜레스테롤 농도는 군별 유의적인 차이가 없었으나 동맥경화지수와 심혈관위험지수는 청국장 분말 10%, 20% 및 율무청국장 분말 20% 첨가군에서 대조군보다 유의적으로 낮았으며(p<0.05), 중성지방 농도는 대조군에 비해 청국장 및 율무청국장 분말 20% 첨가군에서 유의적으로 감소하였다(p<0.05). 한편, 혈당 농도는 실험군 간에 유의적인 차이가 없는 것으로 드러났다. 이상에서 율무청국장 식이는 식이효율의 감소, 중성지방의 배설량 증가, 그리고 혈청 중성지방 농도의 감소 및 동맥경화지수와 심혈관위험지수의 감소 등의 효과를 확인할 수 있었다.

흰쥐에서 후두하경로를 통한 하수체 적출술의 개량과 하수체 적출 흰쥐를 이용한 bGH 효능시험 (Improved Hypophysectomy Technique by Sublaryngeal A, pp.oach and the Efficacy of bGH in Hypophysectomized Female Rats)

  • Improved hypophysectomy technique and the efficacy of bGH in hypophysectomized female rats were studied as a basis for the function of endocrine gland and hormones. 1. With improved sublaryngeal a, pp.oach by hypophysectomy, 34 rats out of 40 were successfully hypophysectomized(more than 80% of success). The surgical time for hypophysectomy per rat was taken about 3.0$\pm$1.0 min. 2. For the effect of hypophysectomy and bGH on body weight gain among the 5 gruops of A, B, C, D, and E, the weight gains for A, B, C, D, and E, were 93g, 9.2g, 10.7g, 25.1g and 36.2g, respectively, which implied that hypophysectomy had a major effect on body weight. And also, the effect of bGH was paralleled with the level of bGH. There were significant difference between control and hypophysectomized groups, and between the bGH treated and bGH non-treated groups after hypophysectomy(p<0.01). 3. For the effect of hypophysectomy and bGH on the length of femur, tibia, humerus, and anterbrachi, longest was for A, shortest for B, and longer in order of C, D, and E. The A group was significantly longer than B, C, D, and E groups. The B group was shorter than C, D, and E groups(p<0.01). 4. For the effect of hypophysectomy and bGH on the weight of femur, tibia, humerus, and antebrachi, heaviest was for A, lightest for B, and heavier in order of C, D, and E. The A group was significantly heavier than B, C, D, and E groups. The B group was lighter than C, D, and E groups(p<0.01). 5. In changes of body length, and weights of liver and heart, A group was most drastically increased. Among the hypophysectomized groups, the change was increased in order of B, C, D, and E. Especially, the change of A group was significantly greater than B, C, D and E gruops. However, no significant differences were found among B, C, D, and E groups. 6. The hypophysectomy in rat had a significant effect on body growth. The effect of hormone was remarkably different depending upon the level of hormone. It was found that 300$\mu\textrm{g}$ of bGH/kg of body wt/day was slightly lower for normal growth.
