A double-stranded RNA (dsRNA) mycovirus was detected in malformed fruiting bodies of Pleurotus ostreatus strain ASI2792, one of bottle cultivated commercial strains of the edible oyster mushroom. The partial RNA-dependent RNA polymerase (RdRp) gene of the P. ostreatus ASI2792 mycovirus (PoV-ASI2792) was cloned, and a cDNA sequences alignment revealed that the sequence was identical to the RdRp gene of a known PoSV found in the P. ostreatus strain. To investigate the symptoms of PoV-ASI2792 infection by comparing the isogenic virus-free P. ostreatus strains with a virus-infected strain, isogenic virus-cured P. ostreatus strains were obtained by the mycelial fragmentation method for virus curing. The absence of virus was verified with gel electrophoresis after dsRNA-specific virus purification and Northern blot analysis using a partial RdRp cDNA of PoV-ASI2792. The growth rate and mycelial dry weight of virus-infected P. ostreatus strain with PoV-ASI2792 mycovirus were compared to those of three virus-free isogenic strains on 10 different media. The virus-cured strains showed distinctly higher mycelial growth rates and dry weights on all kinds of experimental culture media, with at least a 2.2-fold higher mycelial growth rate on mushroom complete media (MCM) and Hamada media, and a 2.7-fold higher mycelial dry weight on MCM and yeast-malt-glucose agar media than those of the virus-infected strain. These results suggest that the infection of PoV mycovirus has a deleterious effect on the vegetative growth of P. ostreatus.
Single-minded 1 gene (SIM1) is a homolog of Drosophila SIM1 gene which plays a key role in the midline cell lineage of the central nervous system and is implicated in the regulation of feeding behavior and obesity in the human and mouse. In this study, porcine SIM1 gene was firstly mapped to chromosome 1p13 using radiation hybrid (RH) mapping and two polymorphisms were detected at position 607 (A/G) in SIM1 intron7 and position 780 (C/T) in SIM1 exon8. The last substitution was genotyped in a 364 F2 animal-population and an association analysis of these genotypes was performed with growth, carcass and meat quality traits by the statistical animal model. The results showed the significant influence of the SIM1 genotype on growth (p<0.05): live weight at birth, later period of growth and average daily gain; and effects on carcass composition (p<0.05): weight of head and buck kneed foreleg, backfat depth, loin eye area, carcass leaf fat and ham fat weights; and traits related to intramuscular fat content (p<0.05).
서로 비슷한 어종으로 약간의 저차 가공시에도 육안적 규별이 어려운 12개 어종의 등전점 전기영동 결과는 다음과 같다. 3종의 갑각류에서 대하와 중하는 밀새우와 다른 단백질 pattern이 pH 4.5 이하에서와 pH 5~7.5에서 서로 구별이 가능하였으며, pH 범위를 변경했을때, 중하와 대하 사이의 구별은 pH 5 부근에 있는 단백질띠로 나타났지만 크기나 형태적으로 거의 같은 대하의 치어에 대한 조사가 필요한 것으로 생각되었다. 민어속 어류에서도 참조기와 항강달이는 비슷한 등전점을 가지는 단백질의 분포가 pH 5~8 사이로 나타났다. 이 분포대에서 돌가자미와 도다리에서도 비슷한 현상을 보였으며, pH 6~7, pH 8~9 범위에서 박대와 도다리는 서로 다른 단백질 분포를 보였고, 참가자미와 돌가자미는 pH 4.5, pH 6.0과 pH 8.0이하에서 뚜렷이 다른 분포를 보였다. pH범위를 변경하여도 단백질 pattern의 구별이 어려운 참조기와 황강달이에서의 2차원 전기영동은 pH 5.0rhk pH 6.0부근에 있는 단백질들에서 추정분자량 11,700과 87,000으로서 등전점 전기영동시 pH 범위 변경 및 2차원 전개 전기영동을 병용할 경우 서해산 어종(중하, 대하, 밀새우, 참조기, 수조기, 황강달이, 참가자미, 도다리, 박대, 돌가자미, 병어, 아홉등가리)의 판별에 구성 단백질의 등전점 전기영동이 신속하고 효과적임을 알 수 있었다.
Objective: An experiment was to evaluate the interplay of dietary lipid sources and feeding regime in the transition from sow milk to solid food of abruptly weaned piglets. Methods: Soon after weaning, 144 piglets were selected and were trained over a 15 day period to experience gradually reducing dietary fat content from 12% to 6% for lard (L), soybean oil (S), and coconut oil (C) and their feeding behavior and diet preference then tested in a behavior observation experiment. Another 324 weaned piglets were used in three consecutive feeding experiments to measure the effect of different dietary fats on performance and feed choice in the four weeks after abrupt weaning. The lipid sources were used as supplements in a 3% crude fat corn/soya basal diet, with 6% of each being included to form diets 9C, 9S, and 9L respectively, and their effects on performance measured. Combinations of these diets were then further compared in fixed blends or free choice selection experiments. Results: Piglets pre-trained to experience reducing lipid inclusion showed different subsequent preferences according to lipid source, with a preference for lard at 9%, soybean oil at 3%, and coconut oil at 6% inclusion rate (p<0.001). Following abrupt weaning, whilst after 4 weeks those fed 9C had the heaviest body weights (18.13 kg, p = 0.006). Piglets fed a fixed 1:1 blend of 9C+9S had a poorer feed conversion ratio (FCR = 1.80) than those fed a blend of 9C+9L (FCR = 1.4). The 9C and 9L combination groups showed better performance in both fixed blend and free choice feeding regimes. Conclusion: After abrupt weaning, they still have dependence on high oleic acid lipids as found in sow milk. A feeding regime offering free choice combination of lipids might give the possibility for piglets to cope better with the transition at weaning, but further research is needed.
In this study, the effects of granite-grit offered free-choice on voluntary intakes of grit and subsequently on some morphologic traits of the digestive tract and performance of broilers were investigated. A total of 240 7d-old male broiler chicks (Ross 308) were allotted randomly to 10 floor pens supplied with wood shavings. The experiment lasted for 42 days. There were 2 dietary treatments, each consisting of 5 replicates. Each replicate consisted of 24 birds kept in an experimental unit with a floor size of $2{\times}2m$. Dietary treatments consisted of control (C) in which broilers were fed standard broiler rations, and acid insoluble granite-grit choice (AIGG) in which broilers were fed standard broiler rations and grit in separate troughs. Mean amounts consumed varied quite widely from week to week, but on average broilers ate 3.41 g per d per bird during the experimental period. Birds had a higher voluntary intake of granite-grit at an early age (7 to 21 d of age) than later (22 to 42 d of age). The voluntary intake of granite-grit of AIGG broilers increased (p<0.05) from 2.7 g/day at 7-14 d to 4.4 g at 15-21 d of age, and then it decreased to 3.4, 3.2 and 3.4 g/day between 22-28, 29-35 and 36-42 d of age, respectively. This level of grit intake increased (p<0.05) weights of empty gizzard (0.97 vs. 1.30 g), edible inner organs (3.51 vs. 3.69 g), and length of gut (8.86 vs. 9.01 cm) as a proportion of body weight and the content of insoluble ash (8.4 vs. 42.2 g/kg) in the faeces compared to the control group. Feeding free-choice grit had little or no effect on final live weights (2,542 vs. 2,543 g), daily gains (69 vs. 69 g), carcass weights (1,924 vs. 1,911), dressing percentages (75.6% vs. 75.1%) and feed efficiencies (1.69 vs. 1.66). Birds given grit did not gain more weight than those not given grit but they tended to have (p<0.07) lower feed intake (116.7 vs. 114.5 g), and consequently lower protein and energy intake. In conclusion, the granite-grit consumed voluntarily by broilers increased gut length and empty gizzard weight without affecting growth performance of broilers. Thus, it can be assumed that the voluntary consumption of granite-grit was too low to affect performance.
Three plasmids derived from the ${\beta}-agarase$ gene (PjaA) of Pseudomonas sp. W7 were expressed in Escherichia coli AD494(DE3) pLysS with lactose as an inducer. These products corresponded to the complete (PjaA) and the two C-terminal truncated (PjaAI and PjaAII) forms of ${\beta}-agarase$. The PjaAI and the PjaAII were originated from exonuclease L treatment from PjaA by deleting 127 and 182 amino acid residues-encoded nucleic acids at 3' region, respectively. The molecular weights of the purified proteins were 71 kDa, 58 kDa, and 50 kDa on SDS-PAGE, respectively. The $K_m$ value of PjaAI was lower than that of the PjaA, and the catalytic efficiency ($k_{cat}/K_m$) of PjaAI was increased to 5 times. The enzyme of PjaAI retained more than 90% activity at $50^{\circ}C$. In contrast to the PjaAI, the remaining activity of the PjaA was only 20% at the same temperature.
가다랭이 통조림을 제조할 때 대량 폐기되는 내장을 유효하게 이용할 목적으로 어류 내장 silage 제조를 위한 유산균주의 배양특성 및 저장기간에 따른 원료의 성상변화를 조사한 결과, MRS 배지에서 배양한 L. bulgaricus 및 L. plantarum 균주는 $pH\;5.5\~6.5$, 배양온도 $35^{\circ}C$ 부근에서 산생성 및 증식이 양호하였으며, 가다랭이 내장 자숙액에 사용된 당질원인 당밀 dextrose의 적정 첨가농도는 가다랭이 내장 중량 당 각각 $10\%$와 $7\%$ 정도인 것으로 나타났다. 저장기간에 따라 산첨가 silage의 pH는 초기 4.0에서 저장 42일째 pH 4.5로 다소 증가하였으며, 유산균 발효 silage는 초기 pH 5.9에서 점차 감소하여 pH 약 4.0까지 저하되었다. silage의 초기 VBN 함량은 약 $62\~65\;mg/100g$에서 저장기간에 따라 점차 증가하여 저장 42일후 산첨가 silage는 112.6\;mg/100g이었고, L. bulgarius 및 L. plantarum 발효 silage는 각각 139.9 mg/100g와 155.0 mg/100g 이었다. L. plantarum 발효 silage의 저장중 경시별 아미노질소 함량은 저장 4일째 1391.3 mg/100g으로서 약 $81\%$의 가수분해율을 나타내었다. 또한 L. plantarum을 이용하여 제조한 발효 silage의 초기 생균수는 $2.7\times10^8/g$에서 저장 35일째는 $2.3\times10^7/g$으로 나타났다.
한국산과 중국산 녹두 종자에서 0.15 M NaCl/0.1 M sodium phosphate buffer (pH7.0)에 의한 추출, $(NH_4)_2SO_4$ 침전, 최종적으로 Sephadex G-100을 이용한 affinity chromatography에 의해 lectin을 분리한 다음, 이들의 생화학적 특성을 조사, 비교하였다. 사람의 적혈구는 trypsin의 처리 유무와 상관없이 응집반응이 일어나지 않으며, 토끼의 적혈구에서는 trypsin을 처리한 경우에만 응집반응이 일어났다. 두 녹두 종자 lectin의 분자량은 SDS-PAGE를 통해 54 kDa와 28 kDa로 확인되었다. 한국산 녹두 종자 lectin의 최적 반응 온도는 $60^{\circ}C$이며, 중국산 녹두 종자 lectin의 경우는 $50^{\circ}C$로 나타났다. 종자 lectin이 열에 가장 안정한 온도는 한국산의 경우는 $50^{\circ}C$이며, 중국산의 경우 $40-50^{\circ}C$로 밝혀졌다. 한국산 녹두 종자 lectin은 pH 3.2에서 가장 높은 활성을 보였으며, 중국산 녹두 종자 lectin은 pH 6.2에서 가장 높은 활성을 보였다. 변성제를 처리했을 때, thiourea와 guanidine-HCl과는 혈액 응집이 일어나지 않아 변성작용이 일어남을 알 수 있었고, urea와는 혈액 응집이 일어나지 않아 변성작용이 일어나지 않음을 알 수 있었다. D-glucose외 6가지의 탄수화물에 대한 특이성이 나타나지 않았다. 또한 두 녹두 종자의 lectin은 $Ca^{2+}$, $Co^{2+}$, $Cu^{2+}$, $Cu^{2+}$, $Mg^{2+}$ 및 $Mn^{2+}$ 등의 금속이온에 대한 특이성이 없었다.
이 연구의 목적은 agarase를 생산하는 새로운 해양 박테리아를 분리하는 것이다. Agarase는 agarose를 가수분해하여 다양한 생리활성을 가지는 agarooligosaccharides 또는 neoagarooligosaccharides를 생성할 수 있다. 전라남도의 여수연안에서 채취한 해수로부터 DH-3 균주를 분리하였다. DH-3의 배양액으로부터 얻었으며, 특성연구에 사용하였다. 20, 30, 40, 50, 60 및 $70^{\circ}C$에서의 온도별 상대활성은 각각 50, 55, 70, 100, 90 및 50%였다. pH 5, 6, 7 및 8에서의 pH별 상대활성은 각각 75, 100, 90, 75%였다. 본 효소는 20 mM Tris-HCl 완충액에서 pH 6.0 및 $50^{\circ}C$에서 최대활성을 나타내었다. 본 효소는 한천이 액체상태로 존재하는 $50^{\circ}C$에서 최대활성을 나타내었으므로 산업적으로 유용한 효소로 판단된다. 본 효소는 20, 30 및 $40^{\circ}C$에서 2시간 동안 열처리하여도 80% 이상의 잔존활성을 보였다. TLC 분석결과, Persicobacter sp. DH-3 균주의 한천분해효소는 네오한천올리고당인 neoagarohexaose와 neoagarotetraose를 생성하였으므로 ${\beta}$-agarase로 확인되었다. Zymogram 분석결과, Persicobacter sp. DH-3는 적어도 45, 70 및 140 kDa 크기의 3종류 이상의 한천분해효소를 생산하는 것을 확인할 수 있었다. 그러므로, Persicobacter sp. DH-3 유래 agarase는 기능성을 가진 neoagarooligosaccharides의 생산에 활용될 수 있을 것으로 기대된다.
This study was conducted to investigate the effect of dietary supplementation of wild grape (Vitis coignetiae) on egg qualities. Laying hens were randomly assigned to three different dietary groups containing 0, 0.25, and 0.5% of wild grape and fed for 8 weeks, respectively. Eggs were collected after feeding period and stored at $4^{\circ}C$ for 7 days. Egg quality traits and cholesterol level of egg yolk were measured at 0 and 7 days of storage. There were no significant differences in total cholesterol content of egg yolk and egg shell thickness among the treatments. However, egg weights of wild grape-fed groups significantly increased compared to that of control. Dietary supplementation of 0.25% wild grape increased the shell and yolk colors compared to the control. Dietary supplementation of 0.5% wild grape significantly increased albumen height and Haugh unit and decreased egg shell hardness and pH values at day 0. However, no differences were found after 7 days of storage. Consequently, the dietary supplementation of wild grape improved the egg qualities on some extent including egg weights, shell and yolk color, albumen height, and Haugh unit.
