산호로부터 2개의 푸란노세스키테르펜의 분리와 구조 결정 (Isolation and Structure Determination of Two Furanosesquiterpenes from the Soft Coral Sinularia lochmodes)

  • 하와이 Pohnpei에서 채집한 soft coral Sinularia lochmodes로부터 2개의 furanosesquiterpenes인 (5'E)-5-(2',6'-dimethylocta-5',7'-dienyl) furan-3-carboxylic acid (1)와 (1'E,5'E)-5-(2',6'-dimethylocta-l',5',7'-trienyl) furan-3-carboxylic acid (2)가 검출되었다. 이들의 구조를 $^1H$ , $^{13}C$ NMR, Homo-COSY, $^1H$-$^{13}C$ (1 bond) Heteronuclear Multiple Quantum Coherence Spectroscopy (HMQC), $^1H$-$^{13}C$ (2 and 3 bond) Heteronuclear Multiple Bond Coherence Spectroscopy (HMBC), Electron Impact Mass Spectroscopy (EI-ms) 및 Infrared Spectroscopy (IR)에 의해 밝혔다.

포도의 품종별 휘발성 향기성분 분석 (Volatile Flavor Components in Various Varieties of Grape(Vitis vinifera L.))

  • SDE 방법으로 추출한 블랙올림피아, 캠벨, 델라웨어의 향기성분을 분석하여 각각 77종, 72종, 74종의 휘발성 향기성분을 분리.동정하였다. 블랙올림피아의 주요향기성분으로 (E)-2-hexenal(20.36%), diethylacetal(18.03%), hexanal, ethyl acetate가 다량 함유되어 있었다. 캠벨에서는 에스터류로써 30.81%를 나타낸 ethyl acetate가 가장 높은 함량을 보였으며, ethyl (E,E)-2,4-decadienoate와 ethyl butanoate가 각각 7.96%, 2.59%를 차지하였고, 알코올류와 알데이드류는 주로 $C_{6}$ 화합물이 다량 함유되었다. 델라웨어에서는 블랙올림피아나 켐벨과는 달리 알코올류가 높은 함량을 나타내었으며, 주요 성분은 (E)-2-hexenal (21.07%)과 (E)-2-hexen-1-ol(19.43%) 이었다. 세 종류의 품종에서 공통적으로 hexanal, (E)-2-hexenal, hexanol, (E)-2-hexen-1-ol, ethyal acetate 둥이 다량 동정되어 $C_{6}$-화합물이 포도의 주된 향기성분으로 확인되었으나 테르펜올류는 델라웨어에서만 미량으로 검출되었다. 품종별 휘발성 향기성분의 총함량으로 블랙올림피아, 캠벨, 델라웨어에서 각각 13.7 mg/kg, 48.5 mg/kg,15.3 mg/kg이 정량분석 되었다.

Temperature Effect on the Functional Expression of Human Cytochromes P450 2A6 and 2E1 in Escherichia coli

  • Human cytochromes P450 (GYP) 2A6 and 2E1 are of great interest because of their important roles in the oxidation of numerous drugs and carcinogens. Bacterial expression systems, especially Escherichia coli cells, have been widely used for the production of various GYP enzymes in order to obtain high yield of proteins. The expression methods usually employ longer culture time (30-72 h) at lower temperature (usually under $30^{\circ}C$). Expression levels of GYPs 2A6 and 2E1 at $37^{\circ}C$ were compared to those at $28^{\circ}C$, which is a usual temperature used in most bacterial expression systems for human GYP expression. Within 18 h the expression levels of GYPs 2A6 and 2E1 reached up to 360 and 560 nmol per liter culture at $37^{\circ}C$, respectively, which are compatible with those of 36 h culture at $28^{\circ}C$. The activities of GYPs expressed at $37^{\circ}C$ were also comparable to those expressed at $28^{\circ}C$. The present over-expression system can be useful for rapid production of large amounts of active human GYPs 2A6 and 2E1 in E. coli.

  • To extend the knowledge of triple bonding between group 6 transition metal and heavier group 14 elements, the structural and bonding aspects of ($η^5-C_5H_5$)$(CO)_2$M≡ER (M = Cr, Mo, W; E = Si, Ge, Sn, Pb) are investigated by hybrid density functional calculations at the B3PW91 level. Substituent effects are also investigated with R = H, Me, $SiH_3$, Ph, $C_6H_3-2,6-Ph_2$, $C_6H_3-2,6-(C_6H_2-2,4,6-Me_3)_2$, and $C_6H_3-2,6-(C_6H_2-2,4,6- iPr_3)_2$.

알릴아민 항진균제의 합성과 생물학적 평가 (Synthesis and Biological Evaluation as a Potential Allylamine Type Antimycotics)

  • Structure-activity relationship studies of allylamine type of antimycotics were carried out to evaluate the effect of naphthyl and methyl portion of naftifine. Compounds with 4-fluorophenyl(2a-5a), 2-fluorophenyl(2b-5b), 2,4-dichlorophenyl(2c-5c), 2,6-dichlorophenyl(2d-5d), 4-nitrophenyl(2e-5e), and 2,3-dihydro-benzo[1,4]dioxan-6-yl( 2f-5f) instead of naphthyl group with hydrogen(3a-3f), methyl(4a-4f), and ethyl(5a-5f) in the place of methyl in naftifine were synthesized and tested their in vitro anti-fungal activity against five different fungi. Eight compounds(3a, 5a, 3c, 4c, 4d, 5d, 5e, and 4f) showed significant antifungal activity against T. mentagrophytes. (E)-N-Ethyl-(3-phenyl-2-propenyl)-4-nitro-benzenemethaneamine(5e) displayed moderate antifungal activity against all five different fungi.

$\omega$3계 및 $\omega$6계 지방산 식이가 흰쥐의 모유와 혈청의 지방산 성분 및 비타민 E 수준에 미치는 영향 (Effects of $\omega$3 and $\omega$6 Polyunsaturated Fatty Acid Diets on Fatty Acid Composition and Vitamin E Levels in Milk and Serum of the Rat)

  • The effects of feeding diets with different fatty acids on the composition of fatty acids and vitamin E status in maternal milk & serum and pup's serum were studied. Dietary fats(10% by wt) include on oil(CO), soybean oil(SO), perilla seed oil(PO : about 60% , C18 : 3 $\omega$3) and fish oil(FO : rich in C20 : 5$\omega$3, eicosapentaenoic acid = EPA & 22 : 6$\omega$3, docosahexaenoic acid = DHA), Sprague-Dawley rats weighing 200-250g, were fed experimental diets from pregnancy through lactation period. Maternal milk was obtained by gentle squeezing after 30 minutes of oxytocin(0.2 IU, intraperitoneal) injection. The fatty acid compositions of milk and serum were analyzed at day-2 and day-15. The concentrations of vitamin E in maternal milk and serum and pup's serum were also analyzed. The groups of CO, SO and PO which had no DHA in their diet, contained DHA in their milk, The rations of EPA+DHA/arachidonic acid(AA) were higher in PO group than those in either CO or SO group. This seemed to be due not only to more conversion from C18 : 3$\omega$3 to C20 : 5$\omega$3 and C22 : 6$\omega$3 but also to inhibition of C18 : 2$\omega$6 conversion to C20 : 4$\omega$6. More DHA was found in day-2 milk than in day-15 milk. It was also noted that milk contained more DHA was found in day-2 milk than in day-15 milk. It was also noted that milk contained more DHA than serum and this difference was larger in day-2 than in day-15 milk. Even though the concentrations of vitamin E both in maternal serum and milk were lower in PO and FO groups fed highly unsaturated fat than in CO or SO groups, pup's serum did not show a significant difference among all the experimental groups indicating that the pups man secure their essential nutrients by the biomagnification mechanism.

국내산 복숭아의 품종별 휘발성 향기성분 (Volatile Flavor Components in Various Varieties of Peach(Prunus persica L.) Cultivated in Korea)

  • 국내에서 생식용 및 가공용으로 널리 재배되고 있는 5종류 품종, 백도ㆍ천도ㆍ유명ㆍ대구보 및 황도의 복숭아의 휘발성 향기성분을 SDE 방법으로 추출하고, 이를 GC와 GC/MS를 사용하여 분리 확인하였다. 복숭아의 품종별 즉 백도, 천도, 유명, 대구보 및 황도에서 83, 85, 70, 74, 66종의 휘발성 향기성분이 각각 동정되었다. 확인된 성분들은 29 알코올, 27 케톤, 18 알데히드, 9 에스터, 5 에테르, 3 유기산. 6 테르펜과 그 유도체 및 26 기타성분으로 구성되었다. 각각의 품종에서 (E)-2-hexenal이 가장 많은 함량을 차지하였으며, 품종별 약간의 차이는 있었지만 ethylacetate, hexanal, o-xylene, (E)-2-hexen-1-ol, benzaldehyde, r-decalactone과 r-dodecalactone 역시 다량 확인되었다. $C_{6}$-C$_{12}$까지의 r-와 $\delta$-lactone류, 특히 이들 lactone류 중에서 다량 검출된 r-decalactone과 r-dodecalactone이 포함된 화합물그룹과 불포화지방산의 과산화물인 $C_{6}$-aldehyde와 $C_{6}$-alcohol이 복숭아의 휘발성 향기성분에 큰 비중을 차지하였다.

중등단계 직업교육에서의 발명·지식재산 교육내용에 대한 담당 교사의 교육요구도 우선 순위 분석 (An Analysis on the Priority of Educational Needs of Teachers in Charge of Educational Contents of Invention Intellectual Property in Secondary Vocational Education)

  • 본 연구의 목적은 중등단계 직업교육에서의 발명 지식재산 교육내용에 대한 교사의 교육요구도 우선 순위를 분석하여, 중등단계 직업교육에서의 발명 지식재산교육이 효과적으로 이루어 질 수 있는 토대가 되는 교원의 역량 개발을 위한 직무 연수 프로그램 개발에 기초 자료를 제공하는 것이다. 이를 위해서 담당 교사 인식에 기반한 중등단계 직업교육에서의 발명 지식재산 교육내용에 대한 교육요구도와 이러한 교육요구도의 우선 순위를 분석하여 제시하였다. 본 연구의 구체적인 결과를 제시하면 다음과 같다. 첫째, 중등단계 직업교육에서 발명 지식재산 교육내용에 대한 담당 교사의 교육 요구도 평균은 5.02로 나타났다. 교육요구도가 전체 평균보다 높은 교육내용 항목은 23개이고, 이들 항목과 항목별 평균은 F4(특허 청구 범위) 6.72, F5(명세서 수정 및 보충) 6.46, F2(특허 도면 작성) 6.39, F3(특허명세서와 요약서 작성) 6.31, A5(발명 기법과 활동) 6.27, E6(발명 디자인 프로젝트) 6.15, H3(발명품 사업화) 5.97, F1(특허 정보 및 출원) 5.90, E5(디자인 권리화) 5.78, E3(발명 디자인의 디자인 과정) 5.77, A4(발명과 문제 해결) 5.57, G2(특허 조사 및 분류) 5.47, C2(발명 문제 해결 사고 기법) 5.45, E4(발명 디자인 제품 제작) 5.45, B5(발명특허 프로젝트) 5.42, A2(창의성 개발) 5.26, C4(발명 문제 해결 프로젝트) 5.26, H4(발명품 마케팅) 5.26, H2(발명품 사업성 분석) 5.20, D4(발명과 경영) 5.16, C3(문제 해결 활동) 5.14, E2(발명 디자인의 발상과 표현) 5.11, B3(발명기법의 실제) 5.08 순으로 나타났다. 둘째, 중등단계 직업교육에서 발명 지식재산 교육내용에 대한 담당 교사의 교육 요구도 우선 순위는 1순위에 해당하는 교육내용 항목은 13개, 2순위는 10개, 3순위는 17개로 나타났다. 1순위에 해당하는 교육내용 항목은 A4(발명과 문제 해결), A5(발명 기법과 활동), B5(발명특허 프로젝트), C2(발명 문제 해결 사고 기법), C4(발명 문제 해결 프로젝트), E3(발명 디자인 과정), E4(발명 디자인 제품 제작), E5(디자인 권리화), E6(발명 디자인 프로젝트), F1(특허 정보 및 출원), F2(특허 도면 작성), F3(특허명세서와 요약서 작성), H3(발명품 사업화)이다. 2순위에 해당하는 교육내용 항목은 A2(창의성 개발), B3(발명기법의 실제), C3(문제 해결 활동), D4(발명과 경영), E2(발명 디자인의 발상과 표현), F4(특허 청구 범위), F5(명세서 수정 및 보충), G2(특허 조사 및 분류), H2(발명품 사업성 분석), H4(발명품 마케팅)이다. 3순위는 1, 2 순위를 제외한 교육내용이다.

An Investigation of Characteristics of Chinese Bellflower (Platycodon grandiflorum A.) Cultivated Soil

  • In order to understand the characteristics of soil according to the cultivation environment of Chinese bellflower (Platycodon grandiflorum A.), soil chemical properties of 12 collected soil samples from 6 cultivated fields in Okcheon, Chungbuk province in August. 2017 were analyzed. The soil pH was distributed within the range of 4.61 to 5.25 at all cultivation years and E.C (Electric Conductivity) and T-N (Total Nitrogen) of the cultivation year were not significant. Available $P_2O_5$ was higher than the average for medicinal crops and P. grandiflorum in Korea and C.E.C (Cation Exchange Capacity) was inconsistent for each cultivation year. In particularly, it was validated that the content of exchangeable cations K, Ca, Ma, and Na in this experiment was similar to that of C.E.C according to the cultivation years, because C.E.C had a high correlation with the exchangeable cations. For the available $P_2O_5$, as affected by trans-planting, 5Y-NT-H (cultivated 5 years and non-transplanted) had 58 mg/kg, while 5Y-T-H (cultivated 5 years and transplanted) had 246 mg/kg. The soil pH was found to be lower (acidic) in diseased soils than healthy soils. E.C was confirmed to be was higher in diseased soils than healthy soils except for the one cultivated for 2 years. The contents of T-N and available $P_2O_5$ were higher in diseased soil except for the one cultivated for 5 years and 11 years. The exchangeable cation K and Na tended to be higher in diseased soils rather than that in healthy soils, and the exchangeable cation Ca and Mg contents were higher in healthy soils than in diseased soils. The C.E.C of the soil was lower than that of healthy soils in all of the years except for the one which was cultivated for 5 years (transplanted).

Elimination Reactions of (E)-2,4,6-Trinitrobenzaldehyde O-Aryloximes Promoted by R3N/R3NH+ in 70 mol% MeCN(aq). Effect of β-Aryl Group the Nitrile-Forming Transition-State

  • Nitrile-forming eliminations from $(E)-2,4,6-(NO_2)_3C_6H_2CH=NOC_6H_4-2-X-4-NO_2$ (1) promoted by $R_3NH/R_3NH^+$ in 70 mol % MeCN(aq) have been studied kinetically. When X = $NO_2$, the reactions exhibited second-order kinetics as well as Br$\"{o}$nsted ${\beta}$ = 0.63 and ${\mid}{\beta}_{lg}{\mid}$ = 0.34-0.46, and an E2 mechanism is evident. As the leaving group was made poorer (X = H, Cl, and $CF_3$), Br$\"{o}$nsted ${\beta}$ value increased from 0.63 to 0.85-0.89 without much change in the ${\mid}{\beta}_{lg}{\mid}$ value E2, indicating that structure of the transition state changed to an E1cb-like with extensive $C_{\beta}-H$ bond cleavage, significant negative charge development at the ${\beta}$-carbon, and limited $C_{\alpha}$-OAr bond cleavage.
