Effects of Rhizobium Inoculation on the changes of Ureide-N and Amide-N Concentration in Stem and Root exudate of Soybean Plant (대두근류균(大豆根瘤菌) 접종(接種)이 뿌리와 줄기 즙액중(汁液中) Amide-N 및 Ureide-N 농도(濃度)에 미치는 영향(影響))

  • A series of green house experiment was conducted to find but the effect of fertilizer application and inoculation of rhizobium on the changes of amide-N, ureide-N and $NH_4-N$ concentration in stem and root exudates of soybean plant growth. The results obtained were summarized as follows ; 1. Five strains of indigenous Rhizobium japonicum-nitrogen fixing activity($C_2H_2$-reducing activity) was more than 6.4 to 20.1 nmole/hr/tube-were identified from 37 soil samples in 22 areas of farmers field throughout country. 2. These identified 5 strains of rhizobium were obtained high nitrate reductase but low ammonium and nitrite oxidase activities. Among 5 strains of rhizobium the Rhizobium japonicum RjK-134 was applied for this green house experiment. 3. Dry matter yield was increased by the combination of inoculation of Rhizobium japonicum RjK-134 with no fertilizer and without nitrogen fertilizer application. However, dry matter yield was decreased with application of N and NPK with inoculation of rhizobium. 4. The concentrations of amide-N and ureide-N were increased in xylem sap than that of root exudate and higher concentration was obtained ar 30 days after planting than flowering stage (45 days after planting). 5. The combination of NPK application with inoculation of Rhrizobium japonicum RjK-134 enhanced the increase of amide-N and ureide-N concentration in xylem sap and root exudate. 6. High ammonium-N concentration in xylem sap and root exudate were obtained in combination with without-fertilizer under no inoculation of rhizobium and N and NPK application with inoculation of rhizobium.

STUDIES ON THE VARIATION OF MICROFLORA DURING THE FERMENTATION OF ANCHOVY, ENGRAULIS JAPONICA (멸치 젓갈 숙성에 따른 미생물상의 변화에 대하여)

  • Identification and change of microflora during the fermentation of anchovy Engraulis japonica, under the halophilic circumstance were investigated. The change of salinity and pH in meat and juice which decide the environment for microorganism and decomposition of nitrogenous compound which functions as a nutrient source were also discussed by measuring the content of total-N, amino-N, nonprotein-N, TMA and VBN, The fresh anchovy was mixed with rock salt (20 percent w/w) and stocked for six months. Through the fermentation lag phase of viable cells extended for 20 days that was obviously larger compared with other circumstances, hereafter increased to reach the maximum value of $5\times10^4$ total count per gram at 35 day stock. The stationary phase proceeded for 25 days. 540 strains were isolated and among them 11 genus of bacteria, 3 genus of yeasts, were identified and other 2 yeast strains of unidentified. At the initial stage of fermentation, Pseudomonas, and Helobacterium prevalently grew, at the middle stage, they disappeared rapidly and Pediococcus and yeasts completely dominated, where they are assumed to get directly involved with fermentation of fish, The PH value tended to decrease in the progress of fermentation and at 100 day stock it showed the minimum value of 5.5 to 5.6 in both meat and juice. The highest salinity of meat decreased to 18 percent, while in juice it decreased to 28 percent since 50 days stock. The content of total-N in meat gradually decreased to 2.8 percent, while in juice it increased to 2.3 percent at 100 day stock, However nonprotein-N was 1.8 percent and amino-N was 1.1 Percent. Since 100 days stock, the increasing rate of amino-M is too low it could be judged to entered the final stage of fermentation, In the first 20 days stock, the increase of VBN and TMA can be explained by the growth of putrefactive bacteria such as pseudomonas on the meat before salts penetrate into the fish meat, while reincrement after 100 days stock, is explained by decomposition of free amino acid due to the reactions of bacteria and enzymes.

Quality Characteristics and Inhibition Activity against Helicobacter pylori KCCM 40449 of Liquorice Yogurts Manufactured by Exopolysaccharide Producing Lactic Acid Bacteria (Exopolysaccharide 생성 유산균을 이용한 감초 추출물 첨가 Yoghurt의 품질특성 및 Helicobacter pylori KCCM 40449 억제활성)

  • This study was carried out to fortify the antimicrobial activity of yoghurt by adding liquorice extract to it. The liquorice extracts (1 mg/mL) showed relatively high antibacterial activity against H. pylori KCCM 40449 (p < 0.05). The solvent liquorice extracts of minimal inhibitory concentrations (MIC) against H. pylori KCCM 40449 were 25- 100 ${\mu}g$/mL. Lactobacillus amylovorus DU-21 with high EPS production ability were inoulated to milk after the addition of different amounts of liquorice extracts (0.0%, 0.05%, 0.1% and 0.2%). The physico-chemical characteristics of yoghurts added with liquorice extracts were examined. The initial pH, titratable acidity, viscosity and viable cell counts of the yoghurt added liquorice extracts were 3.41-3.51, 1.021-1.091%, 1,686-1,930 cp and 9.41-9.38 Log CFU/mL, respectively. The viscosity and syneresis of yoghurt were better than that of the control. Antimicrobial activity against H. pylori KCCM 40449 increased with increasing addition of liquorice extract. However, the sensory score of yoghurt added with different amounts of liquorice extracts was lower than that of the control (p < 0.05). As a result of the sensory evaluations, the flavor, taste, texture, color and overall acceptability of the yoghurt with 0.05% liquorice extract were found to be much better than those of the other groups (p < 0.05). Overall, the optimal amount of liquorice extract added in the manufacture of yoghurt was 0.05% of the total weight. Further studies on increment of antimicrobial activity and palatability of liquorice extract added yoghurt are necessary.

The Responses of Particulate Phosphorus Exposed to the Fresh Water in Marine Sediment (담수화로 인한 퇴적물 내 입자성 인의 거동에 관한 실험적 연구)

  • We incubated marine columnar sediments at $25^{\circ}C$ for 230 days to simulate the responses of phosphorus in the sediment which was exposed to freshwater. The incubation was composed of three different treatments (FW: freshwater, FWA: freshwater under anoxic condition, and SW: seawater as a Control). Six particulate fractions of phosphorus in sediment were obtained through sequential extraction and, for comparison, phosphate concentrations in porewater and superlying water were also determined. After the incubation, evidently higher concentrations of phosphate were found in FW and FWA compared to SW. Mass extinction of living organisms in marine sediment from freshwater shock and consequent decay of their corps probably contributed such high phosphate spike in the overlying water. Higher concentrations of BD-P(lron-bound P) were found in FW compared to SW. After exposure to the freshwater, we could determine that penetration depth of dissolved oxygen in marine sediment will be deeper. A result of increases of ferrous compounds in freshwater where contained less sulfide has been obtained. Because of these phenomena, BD-P was increased in FW. On the contrary, BD-P was decreased in FWA since poor dissolved oxygen concentration. In FWA, total amount of Leachable P(SUM of LOP) has been remarkably increased through the experiment, which strongly suggested the easy conversion of the leachable P into reactive P. This experiment has shown that most of diverse P species in marine sediment were leachable under freshwater and low oxygen condition. Therefore reclamation of natural tidalfalt and consequent freshwater introduction seems to trigger the conversion of diverse P-species to leachable P in the marine sediments, which will exert high benthic load of phosphate to the overlying water.

Production and Characterization of Extracellular Polysaccharide Produced by Pseudomonas sp. GP32 (Pseudomonas sp. GP32에 의해 생산된 세포 외 다당류의 생산 및 특성)

  • A strain GP32 which produces a highly viscous extracellular polysaccharide was conducted with soil samples and identified as Pseudomonas species. The culture flask conditions for the production of extracellular polysaccharide by Pseudomonas sp. GP32 were investigated. The most suitable carbon and nitrogen source for extracellular polysaccharide production were galactose and (NH4)2SO4. The optimum carbon/nitrogen ratio for the production of extracellular polysaccharide was around 50. The optimum pH and temperature for extracellular polysaccharide production was 7.5 and 32℃, respectively. In batch fermentation using a jar fermentor, the highest extracellular polysaccharide content (15.7 g/l) was obtained after 70 hr of cultivation. The extracellular polysaccharide produced by Pseudomonas sp. GP32 (designated Biopol32) was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography. Biopol32, which has an estimated molecular weight of over 3×107 datons, is a novel polysaccharide derived from sugar components consisting of galactose, glucose, gulcouronic acid and galactouronic acid in an approximate molar ratio of 1.85 : 3.24 : 1.00 : 1.42. The solution of Biopol32 showed non-Newtonian characteristics. The viscosity of Biopol32 exhibited appeared to be higher at all concentration compared to that of zooglan from Zoogloea ramigera. An analysis of the flocculating efficiency of Biopol32 in industry wastewater (food, textile, and paper wastewater) revealed chemical oxygen demand (COD) reduction rates 58.4-67.3% and suspended solid (SS) removal rates 82.6-91.3%. Based on these results, Biopol32 is a possible candidate for industrial applications such as wastewater treatment.

  • A microbiological nitrate determination method by E. coli is modified in Korea, using K12 wildtype, KCTC 1116, for the quantitative reduction of $NO_3{^-}$ to $NO_2{^-}$. The nitrate in plant, soil or water sample is determined spectrophotometrically after being diazotized with sulfaniamide and N-(1-naphthl)-ethlenediamine. The modified E. coli cell method and principle for nitrate determination using Korean wildtype E. coli strain is described, and cell culture and preparation of stock suspension for E. coli as well. This modified E. coli cell method can be managed simply and fast, it is suitable for the investigation of the large serials, it can be also automated and has a high degree of sensitivity up to 0.01ppm $NO_3{^-}-N$ in the sample solution. The applicability of the modified E. coli cell method has been tested for plant, soil and water analysis on a wide range of different samples. Recovery rates of added nitrate have been determined and comparisons with other standard nitrate analytical procedures have been carried out. The results with the modified E. coli cell method show high correlation ($r^2=0.98$) with those gained by the standard analytical procedures. The advantages and disadvantages of the method are also discussed to other nitrate determination methods.

Age Variation in Extractive Nitrogenous Constituents of the Cultured Ascidian, Halocynthia roretzi Muscle (양식산 우렁쉥이, Halocynthia roretzi 함질소 엑스성분의 연령차)

  • The muscle extracts of the ascidian, Halocynthia roretzi cultured for two and three years old on the southern coast near Chungmu and the eastern coast near Pohang of Korea, were analyzed for extractive nitrogen (EN), free amino acids (FAA), combined amino acids (CAA), nucleotides and related compounds (NRC), quaternary ammonium bases and guanidino compounds using specimens collected in February 1989 and in April 1989, and compared for those contents with each other. As for the amount of EN, no remarkable difference was found between two- and three-year-old samples collected at St. 1 in the spring and winter seasons, while at St. 2 in the spring season the two-year-old sample was distinctly lower than the three-year-old one. Taurine, proline, glutamic acid, glycine and alanine were the major FAA in every sample. The amount of taurine, the most prominent FAA, was higher in three-year-old sample than in two-year-old one regardless of sampling station and season. Most of the other major FAA showed a similar tendency to EN at both sampling stations in both seasons. Adenosine 5'-triphosphate (ATP), adenosine 5'-diphosphate (ADP), adenosine 5'-monophosphate (AMP), inosine 5'-monophosphate (IMP), inosine (Ino) and hypoxanthine (Hyp) were detected in all the samples and ATP, ADP and AMP were the major ingredients. The amounts of total NRC were in parallel with those of EN and total FAA. As for the contents of betaines, two- and three-year-old samples collected in the winter season exhibited a great discrepancy each other, the former being clearly lower than the latter, but no remarkable difference was observed between two samples of two groups in the spring season. In proximate composition of the muscles, the two-year-old sample was considerably higher in moisture content and lower in protein and glycogen contents than the three-year-old one at St. 2 in the spring season. The large discrepancies observed between two- and three-year-old samples from St. 2 seems to be attributable to the difference in size of samples rather than to the difference in age.

Evaluation of time-dependent antimicrobial effect of sodium dichloroisocyanurate (NaDCC) on Enterococcus faecalis in the root canal (이염화이소시아뉼산나트륨 제재의 근관 내 사용 시 시간에 따른 E. faecalis에 대한 항균성 평가)

  • The purpose of this study was to assess the antibacterial effect of sodium dichloroisocyanurate (NaDCC), sodium hypochlorite (NaOCl), and chlorhexidine (CHX) on Enterococcus faecalis and to evaluate and to compare the time-dependant antimicrobial effect of NaDCC with NaOCl and CHX in the root canal in vitro before and after instrumentation. Extracted Human single teeth were prepared by serial instrumentation technique. The samples were autoclaved and contaminated for 3 days with E. faecalis monocultures. The teeth were then divided into 4 groups Each group was irrigated and inserted with 2% NaOCl, 2% NaDCC, 2% CHX and steri)ized saline. After 6, 12, 24, 72h, and 1 week incubation, sterilized paper point was inserted into the root canal. Paper points containing root canal contents were then placed on the agar plate. And then each root cana) was prepared with #4 and #5 GG (Gates-Glidden) drill. The debris were collected in the sterilized microtube and the plates were incubated at $37^{\circ}C$ in an increased $CO_2$ atmosphere. After 24h incubation the growth of bacteria around the paper points were measured. NaOCl and NaDCC solution shows similar antimicrobial effect for E. faecalis at 6, 12, 24, 72h and 1 week. In centrol group, irrigated with sterilized saline, no antimicrobial effect was observed. The results are in agreement with other investigators, who have shown the bactericidal property and possibility of NaDCC as a root canal irrigation solution. Thus it seems that NaDCC solutions can be clinically applied into the root canal within 1 week after dilution.

Comparison of Extractive Nitrogenous Constituents in Cultured and Wild Olive Flounder (Paralichthys olivaceus) Muscle (양식 및 자연산 넙치의 함질소엑스성분 비교)

  • The nitrogenous compounds in the muscle extracts of cultured and wild olive flounder, Paralichthys olivaceous, were analyzed. The analyzed coumpounds were extractive nitrogen, free amino acids, oligopeptides, ATP and its related ompounds, quaternary ammonium bases, and guanidino compounds. The distribution pattern of these compounds in cultured and wild fish was found to be very similar. Although the ATP and its related compounds and creatine in the muscle of cultured fish were slightly abundant than those in the muscle of wild one, the extractive nitrogen, total free amino acid, oligopeptides, and TMAO were found to be slightly rich in the muscle of wild fish than those in the muscle of cultrued one. The moisture content of cultured fish was relatively lower but the protein and fat contents of cultured one were higher than those of wild fish. However the differences in the proximate composition, extractive nitrogen and nitrogenous compounds between two fishes were not significantly different.

The Effects of Salinity Stress on the Antioxidant and Anti-inflammatory Activities of Crepidiastrum sonchifolium (염분 스트레스가 고들빼기의 항산화 및 항염증 활성에 미치는 영향)

  • This study was conducted to investigate the effect of salinity stress on the antioxidant and anti-inflammatory activities of Crepidiastrum sonchifolium. The plant was treated with NaCl at concentrations of 0, 50, 100, and 200mM for 6 weeks. After treatment, the whole plant was collected, and 70% ethanol extracts were prepared. The DPPH radical scavenging activity was highest in the order of NaCl treatment concentrations of 0, 100, and 50mM, while the 200mM treatment group showed the lowest radical scavenging. The total phenol and total flavonoid contents showed very similar results to the antioxidant activity depending on the NaCl concentration, confirming that the phenolic compounds of the plant can contribute to the antioxidant capacity against salinity stress. In addition, the investigation of the effect of NaCl-treated C. sonchifolium extract on the inhibition of NO production in the LPS-stimulated mouse macrophage cell line (Raw 264.7) revealed that NO production significantly decreased in the 1,000㎍/mL treatment group across all NaCl concentration groups. But, the high concentration (1,000㎍/mL) treatment of the 100mM and 200mM NaCl treatment groups was found to have a negative effect on cell survival. These results suggest that radical scavenging activity is highest in healthy plants and that they produce antioxidants to respond to NaCl salinity stress up to 100mM. However, a high NaCl concentration of 200mM has a negative effect on the physiological activity of the plants. Compared with the results of the previously reported growth index, it is thought that the growth and physiological activity of plants can be positively affected in an NaCl treatment environment of 50mM or less.
