The inhibitory activity of ent-norsecurinine alkaloid was evaluated against spore germination of some plant pathogenic fungi(Curvularia maculans, Curvularia species, C. palliscens, Colletotrichum gloeosporioides, Colletotrichum species, Afternaria solani, A. brassicae, Fusarium udum, Helminthosporium echinoclova and H. penniseti). It inhibited spore germination of all the test fungi. C. maculans, C. species, and C. palliscens were the most sensitive as complete inhibition of spore germination was observed at 1000 ppm. A. solani was not inhibited by this chemical.
Composite adsorbents were prepared by mixing water plant sludge with phenolic resin having the ratio of 1 : 1, 1 : 2, and 1 : 3 respectively, curing from $100^{\circ}C$ to $170^{\circ}C$ under $N_2$ atmosphere, and then activating with $N_2$ at $700^{\circ}C$. Thermal property, specific surface area and morphology of the composite adsorbents as well as their precursors were measured by TGA, BET and SEM respectively. Removal efficiency of the composite adsorbents to ${NH_4}^+$ and TOC was compared with those of commercial zeolite and activated carbon. The adsorbents presented very promising TOC removal efficiency of 98%, which was identical level to that of commercial activated carbon while they displayed removal efficiency, only 32%, of ${NH_4}^+$. Therefore, this composite adsorbent considered as the alternative material of commercial activated carbon, used as an expensive removal agent of organic substances and THM in water treatment plant and it also suggested a possibility of practical application in other processes.
The optimal culture conditions were studied for plant regeneration from mesophyll protoplasts of Solanum sisymbriifolium. Axenic seedlings of S. sisymbriifolium were used as a explant for protoplast culture. Many viable protoplasts were isolated by incubating leaf slices in an enzyme solution containing 0.25% Meicerase and 0.05% Macerozyme for 16 hr at $25^{\circ}C$ without shaking. Protoplast density of $5.0{\times}10^4\;ml^{-1}$ in Kao medium containing 5.0 mg/L NAA, 1.0 mg/L 2,4-D and 1.0 mg/L BA was optimal for colony formation. Most colonies were formed when protoplasts were cultured at $25^{\circ}C$ after initial culture at $30^{\circ}C$ for one week. On the MS agar medium with 1.0 mg/L zeatin, 38.4% of protoplast-derived calli differentiated shoots. These shoots rooted on 1/2MS medium with 5.0 g/L sucrose and 2.5 g/L gellan gum, and developed into whole plants.

It is now generally accepted that a phosphoinositide cycle is involved in the transduction of a variety of signals in plant cells. In animal cells, the hydrolysis of phosphatidyl-4,5-bisphosphate catalysed by phosphatidylinositol - specific phospholipase C yields to D-myo-inositol - 1,4,5-trisphosphate and diacylglycerol, which are well known second messengers. The binding of InsP$_3$to a receptor located on the endoplasmic reticulum triggers a calcium release from the endoplasmic reticulum. We have detected and partially characterised key components of phosphoinositide signaling. First, tobacco microsomal fraction and plasma membrane PI-PLC. Consecutively, using a radioligand binding assay we have identified a $Ca^{2+}$ -dependent high affinity InsP$_3$binding site in microsomal membrane fraction vesicle preparation and then we have measured inositol-1,4,5-trisphosphate induced calcium release from tobacco microsomal fraction. These findings suggest that phosphoinositide signaling system is present and operates in the tobacco suspension culture.e.
Giemsa C-banded karyotypes of A. thunbergii, A. deltoide-fistulosum, A. cyaneum and A. cyaneum var. deltoides were analyzed, and the interspecific relationships were investigated on the basis of the C-banding patterns. The banding pattern of each species was unique and made possible an easy and clear separation among them. A. thunbergii and A. deltoide-fistulosum revealed very close banding relationship but a. cyaneum and A. cyaneum var. deltoides showed significant difference in banding pattern in spite of their close plant systematic position.
A series of experiments were carried out to know the effects of pollen stage, cold shock temperature and duration, and media for callus and green plant induction in rice anther culture. The results indicated that: (a) uninucleate stage of pollen was the most suitable stage for effective callus induction, (b) cold shock temperature of 8$^{\circ}C$ and 12$^{\circ}C$ was appeared to be proper temperature for callus induction, (c) callus induction rate was increased in the eight to 12 days long cold storage, (d) the medium N6 was better than that of N6D for callus induction, (e) green plant induction was better in both 4$^{\circ}C$ and 8$^{\circ}C$ than that of 12$^{\circ}C$ cold shock, (f) green plant frequency was higher in eight to 12 days long cold storage and (g) green plant frequency was doubled in the MS medium when compared with N6 medium.
This study describes an efficient protocol for Agrobacterium tumefaciens-mediated transformation of two subgroups of genotype AAA bananas (Musa acuminata cv. Pei Chiao and Musa acuminata cv. Gros Michel). Instead of using suspension cells, cauliflower-like bud clumps, also known as multiple bud clumps (MBC), were induced from sucker buds on MS medium containing $N^6$-Benzylaminopurine (BA), Thidiazuron (TDZ), and Paclobutrazol (PP333). Bud slices were co-cultivated with A. tumefaciens C58C1 or EHA105 that carry a plasmid containing Arabidopsis root-type ferredoxin gene (Atfd3) and a plant ferredoxin-like protein (pflp) gene, respectively. These two strains showed differences in transformation efficiency. The EHA105 strain was more sensitive in Pei Chiao, 51.3% bud slices were pflp-transformed, and 12.6% slices were Atfd3-transformed. Gros Michel was susceptible to C58C1 and the transformation efficiency is 4.4% for pflp and 13.1% for Atfd3. Additionally, gene integration of the putative pflp was confirmed by Southern blot. Resulting from the pathogen inoculation assay, we found that the pflp transgenic banana exhibited resistance to Fusarium oxysporum f. sp. cubense tropical race 4. This protocol is highly advantageous to banana cultivars that have difficulties in setting up suspension cultures for the purpose of quality improvement through genetic transformation. In addition, this protocol would save at least 6 months in obtaining explants for transformation and reduce labor for weekly subculture in embryogenic cell suspension culture systems.

Each commercial cyromazine insecticide has different HPLC conditions. The aim of this study was to establish a standardized chromatographic method for the determination of cyromazine in commercial insecticides. The separation was achieved on two C18 columns - Waters® Bondapak C (4×300 nm i.d., 10 ㎛) and X bridge (4.6×250 nm i.d., 5 ㎛) using a mobile phase composed of water/methanol/ethanolamine (76:24:0.1, v/v), with UV detection at wavelengths 230 nm and 254 nm. A total of six commercial cyromazine insecticides were analyzed. In this study, the optimal high-performance liquid chromatography conditions for the analysis of cyromazine were as follows: a mobile phase of water/methanol/ethanolamine (76:24:0.1, v/v) at a flow rate of 1.0 mL/min and a detection wavelength of 230 nm using a X bridge C18 column (4.6×250 nm i.d., 5 ㎛) at a column temperature of 25℃. The calibration curve was linear in the concentration range of 5~50 ㎍/mL, with a correlation coefficient of 0.99995. The cyromazine detection limit was 0.2 ㎍/mL, and the limit of quantification was 0.59 ㎍/mL. The percentage recovery ranged from 99.8% to 101.0% for cyromazine, and the relative standard deviation was not over 2.0%. The cyromazine concentration ranged from 92.7% to 109.4% and was within the acceptable range (90~120%) for the percent of the labeled amount. This method was found to be suitable for determining cyromazine in commercial insecticides.
This study was conducted to investigate the suitable conditions for seed germination of Rosa davurica PALL. In the characteristics of the fruit and seed of Rosa davurica, fruit length and width was 1.3cm and 0.9cm , respectively, and seed number was eighty-nine. Artificial and low temperature storage at 4$^{\circ}C$ increased the rate of split achene pericarp until 46.6% and storage at 15$^{\circ}C$ incubator decreased the rate of split achene pericarp (10.5%). The rate of seed germination of split achene pericarp in control at 15$^{\circ}C$ was 90% and of non-split achene in GA$_3$100ppm at 15$^{\circ}C$ was 36.8%. Average germination day of split achene pericarp seeds in GA$_3$150ppm at 2$0^{\circ}C$ was 4.2 days and non-split achene pericarp in GA$_3$100ppm at 2$0^{\circ}C$ was 7.3 days. Seed germination was not different between various concentrations of GA$_3$ treatments in split achene pericarp seeds but the rate of germination was more reduced in high concentration of 200ppm. Only the treatments of GA$_3$ was increased germination rate at 4$^{\circ}C$ and immature seed of Rosa davurica.

This experiment was carried out to select the best adaptable varieties of Tall fescue in mountainous pastures of Taekwalyon area. The evaluation was based on the data of varietial differences in the dry weight of plant@W) and yield components of the 2nd growth. Nine varieties of Tall fescue examined were Barcel, Barvetia, Demeter, Enforcer, Forager, Fuego, Johnstone, Safe and Stef. And, the final cutting times were Sept. 30 ($C_1$), Oct.14 ($C_2$),Oct. 28 ($C_3$), and Nov. 11($C_4$). The reults were summarized as follows: 1. Barcel and Forager with many tillers per plant(NT) showed a high dry weight of plant@W) of the 2nd growth. 2. The dry weight of plant(DW) of the 2nd growth indicated significantly positive correlation with the number of tillers per plant(NT), but there was not significant correlation with the dry weight of tiller(WT). 3. The maximum dry weight of plant@W) of the 2nd growth was obtained at $C_3$(28 Oct.). 4. The dry weight of the 2nd growth showed an inverse relations with the dry weight of the 1st growth as affected by various final cutting time in growth period of autumn.
