Anthers contanining uninucleate microspores of eight cultivars of pepper were cultured on MS medium supplemented with 0.004mg/L 2,4-D and 0.1mg/L kinetin, 3% sucrose and 0.2% Gelrite, kept at $35^{\circ}C$ for 24 h, and then cultured at $25^{\circ}C$ with a photoperiod of 16 h daylight for 40 days. Frequency of embryoid and callus formation was varied with cultivars. Embryoid formation was found in Cheongyang and Fushimi Amanaga, while callus formation was in California Wonder, Fushimi Amanaga and Geoseong. In anther culture medium supplemented with 1% activated charcoal, embryoid formation was found with 0.5% of frequency only in Cheongyang, while no callus formation was found. In 1/2MS medium, frequency of embryoid formation in Shishitou, Yatsufusa and Taka no Tsume was 1.2%, 0.4% and 0.4%, respectively. On the other hand, in 1/2 B5 medium, frequency of callus formation in Yatsufusa and Taka no Tsume was 2.8% and 2.7%, respectively. Embryoids transferred to hormone-free MS medium were developed to plantlets and acclimatized. The number of chromosomes in the root tip cells of the haploid plant was 2n=$\times$=12 in cv. Cheongyang.
Purpose: Bone mineral density (BMD) measurements need to be precise enough to be capable of detecting small changes in bone mass of rats. Using a regular dual-energy X-ray absorptiometry (DXA), we measured many BMD of various skeletal sites in rats to examine precision of DXA in relation to the repositioning on the bones of rats. Materials and Methods: Using DXA and small animal software, scans were performed 4 times in all 12 male rats without repositioning (Group 1a). Another four scans for 6 of 12 rats were done with repositioning between scans (Group 2). Customized regions of interest (ROIs), encapsulate the right hind limb, L1-4, skull and pelvic bones were drawn at each measurement. The precision of the measurements was evaluated by measuring the coefficient of variation (CV) of four measurements of BMD at each skeletal site of all rats with or without repositioning. Significance of differences between group 1b (six rats out of group 1a, which were come under group 2) and group2 were evaluated with Wilcoxon Signed Rank Sum Test. Results: CVs obtained at different skeletal sites of all measurements in Group 1b and 2. It was $3.51{\pm}1.20$, $ 2.62{\pm}1.20$ for the hindlimb (p=0.173), $3.83{\pm}2.02$, $4.59{\pm}2.02$ for L1-4 (p=0.600), $3.73{\pm}1.87$, $1.53{\pm}0.89$ for skull (p=0.046), and $2.92{\pm}0.60$, $1.45{\pm}0.60$ for pelvic bones (p=0.075). Conclusion: Our study demonstrates that the DXA technique has the precision necessary when used to assess BMD for various skeletal sites in rats regardless of repositioning.
Comprehensive studies on seedling production and transplanting cultivation of waxy corn are necessary to fulfill the comsumer's preference for the high quality whole ear waxy corn and its year-round supply for higher price, and to escape marketing in a short period at harvest season. Two waxy corn hybrids (cv. Chalok# 1 and Chalok# 4) were used to study the effects of seeding date, nursery days and plug cell size on growth and yield of waxy corn, and to clarify the reliable transplanting date at the Experimental Field of the National Institute of Crop Science in 2007. The number of days from seeding to silking was increased as nursery days were extended, but the number of days from transplanting to silking was shortened comparing to the direct seeding. Number of days from seeding to silking was shorter by 9~12 days for Chalok# 1 than for Chalok# 4, while the number of days from seeding to silking was shorter by 12~15 days in the second cropping than in the first cropping. Culm length of transplanted waxy corns was decreased as the nursery days were extended. Culm length of seeded Chaok# 1 and Chaok# 4 were decreased by 17% and 24%, respectively, in the second cropping compared to those in the first cropping. Ear length was somewhat decreased as the nursery days were extended. Chalok# 1 in the second cropping the degree of decrease was much higher in second cropping than in first cropping of Chalok# 1. Comparing to the first cropping, number of marketable ears per 10a of Chalok# 1 decreased as high as 64%, while that of seeded Chalok# 4 decreased mere 12% in the second cropping.
Total phenolics and flavonoids, and the antioxidant capacity of plum cultivar wines (Prunus salicina L. cv. Soldam and P. salicina L. cv. Formosa) were determined using spectrophotometric methods. The total phenolic and flavanoid contents of Soldam wine were $478.4\;{\pm}\;5.6\;mg$ GAE and $202.4\;{\pm}\;7.5\;mg$ CE per L,respectively, and in Formosa wine were $200.6\;{\pm}\;7.5\;mg$ GAE and $64.4\;{\pm}\;6.8\;mg$ CE per L, respectively. Neutral and acidic phenolics in Soldam wine were extracted with ethyl acetate and 0.01 N HCl, respectively. In the 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging assay, neutral phenolics (64.5 EDA%) had $3{\sim}4$ times higher antioxidant activity than acidic phenolics (21.5 EDA%) and other related phenolic compounds such as chlorogenic acid (15.5 EDA%) and quercetin (24.6 EDA%) at a concentration of $100\;{\mu}g/mL$. The antiviral activities of neutral and acidic phenolics in Soldam wine were investigated in vitro using a virus-induced cytopathic effect (CPE) inhibition assay. Results showed that neutral and acidic phenolics at concentrations of $100\;{\mu}g/mL$ inhibited porcine epidemic diarrhea virus (PEDV) replication at rates of 78.12% and 58.37%, respectively. The inhibition rate of 10 g/mL neutral phenolics (69.42%) was higher than that of ribavirin as an antiviral reagent (57.86%). At concentrations of $100\;{\mu}g/mL$ or less, neutral and acidic phenolics of Soldam wine had no cytotoxic effect against vero cells.
Red and blue lights are effective wavelengths for photosynthesis in plants. In this study, we determined the effects of various combined ratios of red to blue LEDs on the quality of cherry tomato seedlings prior to transplantation, and their subsequent effects on the yield and quality of tomato fruits after transplanting. Two-week-old cherry tomato seedlings (Solanum lycopersicum cv. 'Cuty') were cultivated under various combined ratios of red (R; peak wavelength 655 nm) to blue (B; 456 nm) LEDs [red:blue = 41:59 (59B), 53:47 (47B), 65:35 (35B), 74:26 (26B), 87:13 (13B), or 100:0 (0B)] and fluorescent lamps and raised for 27 days. The cherry tomato seedlings were subsequently transplanted into a venlo-type greenhouse and cultivated for 75 days. At the seedling stage, the shoot fresh weight of seedlings in all RB combined treatments, except 0B and 59B, was higher than that of the control after 27 days of LED treatment. Shoot dry weight and leaf area also showed trends similar to that of shoot fresh weight. The stem length was significantly higher in 13B, 26B, and 35B treatments compared with the control and other treatments. In particular, the stem length of 26B plants was approximately 3.2 times longer than that of 59B plants. At 37 days after transplanting, the number of nodes was significantly higher in 26B and 47B plants, and the plant height of 26B plants was significantly higher than that of control and 59B plants. Total fruit yield in 26B plants, which was the highest, was approximately 1.6 and 1.8 times higher than that in control and 59B plants, respectively. Thus, the results of this study indicate that various combined ratios of red to blue LEDs directly affected to the growth of cherry tomato seedlings and may also affect parameters of reproductive growth such as fruit yield after transplantation.
The mushrooms collected at seven areas of Mt. Chiak in 2002 and 2003 were classified to analyse the distribution and species diversity. Frequency (number of mushroom : N), number of species (S), relative species density (RSD), similarity index (C), richness index (R1), variety index (V1), evenness index (E2), and dominance index(D1) were investigated. Total N and S was 143 and 84, respectively. The RSD was 0.179 ~ 0.226 of the 7 areas. The yearly C of the total area (0.213) was 8.2%. more higher than the average C of 7 areas (0.131). The order in the coefficient of variation (CV) of the indicator for 7 areas was N (10.5%) > D1 (9.2%) > V1 (8.9%) > S (8.5%) > R1 (7.4%) > E2 (2.2%). The average R1 of the 7 areas was 5.36 with the range from 4.85 to 6.01, and 16.72 for the total area. The average V1 of the 7 areas was 16.24 with the range from 14.44 to 18.66, and 68.82 for the total area. The average E2 of the 7 areas was 0.95 with the range from 0.926 to 0.982, and 0.819 for the total area. The average D1 of the 7 areas was 0.071 with the range from 0.055 to 0.073, and 0.081 for the total area. The correlation between N and 5 kinds of diversity indicator (S, R1, V1, E2, D) was not statistically significant, but the correlation between R1, E2 and D1 was statistically significant each other.
To identify nutritional and therapeutic properties of the new Korean red waxy sorghum cultivar ‘Hwanggeumchalsusu (HGC)’, we assayed the antioxidative effects and neuraminidase inhibitory activity. A methanol and 70% ethanol extract of HGC exhibited strong antioxidative effects (IC50 values of 83.2±2.7 for DPPH) and 85.6±2.4 μg/ml for ABTS) and neuraminidase (ND) inhibitory activity (IC50 values of 1.8±0.1 from extracted with methanol and 3.4±0.1 μg/ml from extracted with 70% ethanol) compared with that of the control, noncolored sorghum cultivar ‘Huinchalsusu (HC)’ (IC50> 200 μg/ml). We isolated nine polyphenols, Gallic acid (1), Protocatecuic acid (2), p-Hydroxy benzoic acid (3), Vanillic acid (4), Caffeic acid (5), Ferulic acid (6), Luteolinidin (7), Apigeninidin (8), Luteolin (9), from the HGC - methanol extract, to determine whether they were the active components Luteolinidin of one kind of polyphenols from the HGC, exhibited significant antioxidative effects (IC50 values of 10.9±0.5 μM for DPPH and 8.6 0.6 μM for ABTS) and neuraminidase (ND) inhibitory activity (IC50 values of 26.3±0.6) showed noncompetitive inhibition model. The binding affinity of the ND inhibitors in molecular docking experiments correlated with their ND inhibitory activities. These results suggest that HGC may be utilized to serve as a potential effective antioxidant and inhibitor of ND.
This study was conducted to evaluate the residual effects of basic oxygen furnace (BOF) slag applied in rice paddy fields as soil conditioner one year before. The experimental fields of Lim et al. (2000) located in Youjung and Nampyung were used for this purpose. Both variety (Oryza sativa L. cv. Dongjinbyeo) and cultural practices were the same as those in Lim et al. (2000). Soil chemical properties, plant height, number of tillers per plant, yield and yield components were observed. The temporal variation of treatment mean value in soil chemical properties appeared to be similar trends in both Youjung and Nampyung experimental fields. Soil pH and Ca content were still significantly higher than those in control treatment up to July of the second season, but decreased progressively as time passed. However, the effects lasted longer as slag rate became higher. BOF slag seems to have residual effects as a soil conditioner or Ca fertilizer in soil for two years. BOF slag rate of $4Mg\;ha^{-1}$ raised soil pH almost the same as lime rate of $2Mg\;ha^{-1}$. Content of $SiO_2$ in soil applied slag appeared to be higher compared with control. Fe and Mg content in soil with slag treatment was significantly higher than that of control in 1997, but it was almost the same level as that of control in 1998. In YouJung experimental field, rough rice yield of slag teatment became higher as slage rate incresed. Slag rate of $12Mg\;ha^{-1}$ showed the highest rough rice yield of $5,400kg\;ha^{-1}$ among treatment, which was 14% higher than that of control with $4,720kg\;ha^{-1}$. Slag rate of $12Mg\;ha^{-1}$ showed relatively higher plant height and higher number of tillers at the early growth stage compared with other treatments. In NamPyung experimental field, rough rice yield was the highest at the plot of lime rate $2Mg\;ha^{-1}$ and became higher as slag rate increased. There were no significant differences in rough rice yield between lime treatment and slag treatments. Slag rate of $12Mg\;ha^{-1}$ showed the highest rough rice yield of $7,170kg\;ha^{-1}$ among slag treatment, which was 8% significantly higher than that of control with $6,670kg\;ha^{-1}$. Slag rate of $12Mg\;ha^{-1}$ showed relatively slower growth in plant height at the early growth stage, but superior growth at the later growth stage, and significantly higher number of spiklets per panicle and 1000-grain weight than that of control.
The growth and contents of anthocyanins and ascorbic acid in lettuce(Lactuca sativa L., 'Jeokchima') as affected by supplemental UV-A LED irradiation under different light quality and photoperiod conditions were analyzed in this study. Five light qualities, namely B (blue LED), R (red LED), BUV (blue LED+UV-A LED), RUV (red LED+UV-A LED) and Control (white fluorescent lamps) with photoperiods of 12/12 hours (day/night), 16/8 hours, or 20/4 hours were provided to investigate the effects of light quality and photoperiod on the growth and accumulation of anthocyanins and ascorbic acid in lettuce leaves. As measured 28 days after transplanting, the number of leaves, leaf length, leaf width, leaf area, shoot fresh weight and dry weight of lettuce were significantly affected by light quality and photoperiod. The number of leaves, leaf length, leaf width, leaf area, shoot fresh weight and dry weight of lettuce grown under R treatment increased with increasing light period. By contrast, leaf development was inhibited, but chlorophyll content increased, under B treatment. Supplemental UV-A irradiation significantly decreased leaf length, leaf width, leaf area and shoot fresh weight. Anthocyanins in lettuce increased significantly with decreasing dark period under B treatment. A synergistic effect of supplemental UV-A LED irradiation on anthocyanins accumulation was found for lettuce leaves grown under R treatment but not B treatment. Ascorbic acid in lettuce was greatly affected by photoperiod. Ascorbic acid content at BUV and RUV treatments increased by 20-30% compared to without UV-A LED irradiation. From these results, it was concluded that growth and contents of anthocyanins and ascorbic acid in lettuce are significantly affected by supplemental UV-A LED irradiation. The results obtained in this study will be informative for efforts to improve the nutritional value of leafy vegetables grown in plant factories.
It has been well established that aluminum (Al) inhibits root tip growth rapidly in acid soil. We report the correlation between Al induced growth inhibition and impaired $H^+-flux$ in mung bean (Vigna radiate L. cv. Kumsung). The root growth inhibition was dependent on Al concentration (0, 10, 25, 50, $100{\mu}M$) and exposure time (12 and 24 h). Using Hematoxylin staining, it was observed that the root damage was occurred preferentially in regions with high Al accumulation. Using the pH indicator, it was shown that the surface pH of root tip was strongly alkalized in the control whereas changed only slightly in the $50{\mu}M$ Al-treated root. The $H^+-ATPase$ activity of plasma membrane vesicles was inhibited by 56% in the Al-treated roots compared to control root. Decrease in the amount of the plasma membrane $H^+-ATPase$ (100 kDa) translation in the plant roots under Al stress was demonstrated by Western blot analysis. These results indicate that the dynamics of $H^+-flux$ across the root tip play an important role in root growth under Al stress.
