Development of Analysis Method for Cholesterol in Infant Formula by Direct Saponification (직접 검화법을 이용한 조제분유의 콜레스테롤 분석법 개발)

  • An improved cholesterol analysis method was developed for powdered infant formula by gas chromatographic separation after liquid-liquid extraction and partition. In the official Korea Food Standard method for cholesterol analysis, the water phase and solvent phase were not well separated in the case of emulsified foods such as powdered infant formulas and baby foods. For the rapid and simple sample preparation method, an optimized direct saponification condition was established for heating temperature, heating time, and KOH concentration. From the results, the optimum conditions were as follows: heating temperature $90^{\circ}C$, heating time 60 min, and 16 M KOH 10 mL for a 2 g infant formula sample; improved separation condition for gas chromatography was as follows: the initial oven condition was $250^{\circ}C$ for 25 min, the oven temperature was increased to $290^{\circ}C$ by $10^{\circ}C$/min ratio, and finally the oven temperature remained at $290^{\circ}C$for 9 min. The developed method could be implemented for the study of cholesterol, providing the advantages of reduced inspection time and cost in emulsified foods such as infant formula.

Development of a Kit for Diagnosing AtCYP78A7 Protein in Abiotic-tolerant Transgenic Rice Overexpressing AtCYP78A7 (AtCYP78A7 과발현 환경스트레스 내성 형질전환 벼의 단백질 진단 키트 개발)

  • Quantitative determination of the protein expression levels is one of the most important parts in assessment of the safety of foods derived from genetically modified (GM) crops. Overexpression of AtCYP78A7, a gene encoding cytochrome P450 protein, has been reported to improve tolerance to abiotic stress, such as drought and salt stress, in transgenic rice (Oryza sativa L.). In the present study, an enzyme-linked immunosorbent assay (ELISA) kit for diagnosing AtCYP78A7 protein including AtCYP78A7-specific monoclonal antibody was developed. GST-AtCYP78A7 recombinant protein was induced and purified by affinity column. Four monoclonal antibodies (mAb 6A7, mAb 4C2, mAb 11H6, and mAb 7E8) against recombinant protein were also produced and biotinylated with avidin-HRP. After pairing test using GST-AtCYP78A7 protein and lysate of rice samples, mAb 4C2 and mAb 7E8 were selected as a capture antibody and a detecting antibody, respectively, for ELISA kit. Product test using rice samples indicated that percentages of detected protein in total protein were greater than 0.1% in AtCYP78A7-overexpressing transgenic rice (Line 10B-5 and 18A-4), whereas those in negative control non-transgenic rice (Ilpum and Hwayoung) were less than 0.1%. The ELISA kit developed in this study can be useful for the rapid detection and safety assessment of transgenic rice overexpressing AtCYP78A7.

The Runoff Characteristics of Non-point Pollution Sources in Industrial Complex(II): Focusing on the Outflow Characteristics of the C Industrial Complex by Rainfall Event (산업단지 비점오염원의 유출특성(II): C산업단지의 강우사상별 유출특성을 중심으로)

  • In this study, rainfall water outlet water quality monitoring was performed on the C industrial complex to evaluate the characteristics of non-point pollutant runoff from the industrial complex during rainfall and to use it as basic data for calculating the load and unit of non-point pollutant. As a result of calculating EMC according to the outflow amount by rainfall event, the 1st rainfall showed EMCs ranges of BOD, CODMn, SS, T-N, and T-P of 1.32~48.76, 3.32~43.75, 2.89~199.43, 2.76~8.93, 0.08~068, and the 2nd rainfall was 0.5~2.9, 2.71~7.13, 2.82~174.94, 1.33~4.03, 0.01~1.28 mg/L, respectively. As a result of calculating the ratio of cumulative outflow and cumulative pollution load, most of the pollution load was less than the rainfall outflow, but over time, the initial washing phenomenon occurred as the ratio of cumulative rainfall outflow and cumulative pollution load increased to more than 1.

Analysis of Co- and Post-Seismic Displacement of the 2017 Pohang Earthquake in Youngilman Port and Surrounding Areas Using Sentinel-1 Time-Series SAR Interferometry (Sentinel-1 시계열 SAR 간섭기법을 활용한 영일만항과 주변 지역의 2017 포항 지진 동시성 및 지진 후 변위 분석)

  • Ports are vital social infrastructures that significantly influence both people's lives and a country's economy. In South Korea, the aging of port infrastructure combined with the increased frequency of various natural disasters underscores the necessity of displacement monitoring for safety management of the port. In this study, the time-series displacements of Yeongilman Port and surrounding areas in Pohang, South Korea, were measured by applying Permanent Scatterer Interferometric Synthetic Aperture Radar (PSInSAR) to Sentinel-1 SAR images collected from the satellite's ascending (February 2017-July 2023) and descending (February 2017-December 2021) nodes, and the displacement associated with the 2017 Pohang earthquake in the port was analyzed. The southern (except the southernmost) and central parts of Yeongilman Port showed large displacements attributed to construction activities for about 10 months at the beginning of the observation period, and the coseismic displacement caused by the Pohang earthquake was up to 1.6 cm of the westward horizontal motion and 0.5 cm of subsidence. However, little coseismic displacement was observed in the southernmost part of the port, where reclamation was completed last, and in the northern part of the oldest port. This represents that the weaker the consolidation of the reclaimed soil in the port, the more vulnerable it is to earthquakes, and that if the soil is very weakly consolidated due to ongoing reclamation, it would not be significantly affected by earthquakes. Summer subsidence and winter uplift of about 1 cm have been repeatedly observed every year in the entire area of Yeongilman Port, which is attributed to volume changes in the reclaimed soil due to temperature changes. The ground of the 1st and 2nd General Industrial Complexes adjacent to Yeongilman Port subsided during the observation period, and the rate of subsidence was faster in the 1st Industrial Complex. The 1st Industrial Complex was observed to have a westward horizontal displacement of 3 mm and a subsidence of 6 mm as the coseismic displacement of the Pohang earthquake, while the 2nd Industrial Complex was analyzed to have been little affected by the earthquake. The results of this study allowed us to identify the time-series displacement characteristics of Yeongilman Port and understand the impact of earthquakes on the stability of a port built by coastal reclamation.

Analysis of Modality and Procedures for CCS as CDM Project and Its Countmeasures (CCS 기술의 CDM 사업화 수용에 대한 방식과 절차 분석 및 대응방안 고찰)

  • Carbon dioxide, emitted by human activities since the industrial revolution, is regarded as a major contributor of global warming. There are many efforts to mitigate climate change, and carbon dioxide capture and geological storage (CCS) is recognized as one of key technologies because it can reduce carbon dioxide emissions from large point sources such as a power station or other industrial installation. The inclusion of CCS as clean development mechanism (CDM) project activities has been considered at UNFCCC as financial incentive mechanisms for those developing countries that may wish to deploy the CCS. Although the Conference of the Parties serving as the Meeting of the Parties to the UNFCCC's Kyoto Protocol (CMP), at Cancun in December 2010, decided that CCS is eligible as CDM project activities, the issues identified in decision 2/CMP.5 should be addressed and resolved in a satisfactory manner. Major issues regarding modalities and procedure are 1) Site selection, 2) Monitoring, 3) Modeling, 4) Boundaries, 5) Seepage Measuring and Accounting, 6) Trans-Boundary Effects, 7) Accounting of Associated Project Emissions (Leakage), 8) Risk and Safety Assessment, and 9) Liability Under the CDM Scheme. The CMP, by its decision 7/CMP.6, invited Parties to submit their views to the secretariat of Subsidiary Body for Scientific and Technological Advice (SBSTA), SBSTA prepared a draft modalities and procedure by exchanging views of Parties through workshop held in Abu Dhabi, UAE (September 2011). The 7th CMP (Durban, December 2011) finally adopted the modalities and procedures for CCS as CDM project activities (CMP[2011], Decision-/CMP.7). The inclusion of CCS as CDM project activities means that CCS is officially accredited as one of $CO_2$ reducing technologies in global carbon market. Consequently, it will affect relevant technologies and industry as well as law and policy in Korea and aboard countries. This paper presents a progress made on discussion and challenges regarding the issue, and aims to suggest some considerations to policy makers in Korea in order to demonstrate and deploy the CCS project in the near future. According to the adopted modalities and procedures for CCS as CDM project activities, it is possible to implement relevant CCS projects in Non-Annex I countries, including Korea, as long as legal and regulatory frameworks are established. Though Korea enacted 'Framework Act on Low Carbon, Green Growth', the details are too inadequate to content the requirements of modalities and procedures for CCS as CDM project. Therefore, it is required not only to amend the existing laws related with capture, transport, and storage of $CO_2$ for paving the way of an prompt deployment of CCS CDM activities in Korea as a short-term approach, but also to establish the united framework as a long-term approach.

Characteristics of Seepage Water and Groundwater in a Coastal LPG Storage Cavern of Jeonnam (전남 해안 LPG 저장공동 유출수와 주변 지하수의 수질특성)

  • Water curtain of an underground LPG storage cavern is a facility to prevent leakage of high pressure gases, for which groundwater should flow freely towards the cavern and groundwater level also must be stably maintained. In this study, in order to evaluate qualities of seepage water and surrounding groundwater of an underground LPG storage cavern in Yeosu, 4 rounds of samplings, field measurements and laboratory analyses (February, May, August, October of 2007) were conducted. According to field measurements, pH was weak acidic to neutral but it gradually increased with time. Electrical conductivity (EC) of groundwater near a salt stack showed very high values between 10.47 and 38.50 mS/cm. Dissolved oxygen (DO) showed a very wide range of 0.20~8.74 mg/L and a mean of oxidation-reduction potential (ORP) was 159 mV, which indicated an oxidized condition. Levels of $Fe^{2+}$ and $Mn^{2+}$ were mostly less than 3 mg/L. All of seepage waters showed a Na-Cl type while only groundwater near the salt stack showed a Na-Cl type with a high total dissolved solid. The other groundwaters exhibited typical $Ca-HCO_3$ types. Levels of aerobic bacteria were mostly very high (573-39,520 CFU/mL). Based on the analyses of these hydrochemistry and biological characteristics, it is concluded that there are no particular problems in groundwater and seepage water, which not causing a trouble in the cavern operation. However, both for control of bio-clogging and for sustainable operation of the water curtain system, a regular hydrochemical and microbiological monitoring is required for the seepage water and surrounding groundwater.

Parametric and Non-parametric Trend Analysis of Groundwater Data Obtained from National Groundwater Monitoring Stations (국가 지하수관측소 지하수위, 전기전도도 및 수온자료에 대한 모수적 및 비모수적 변동 경향성 분석)

  • Trends of variation in groundwater levels, electrical conductivities and water temperatures obtained from the national groundwater monitoring stations (95 shallow and 169 deep wells) of Korea were evaluated. For the analysis, both parametric (linear regression) and non-parametric (Mann-Kendall test, Sen's test) methods were adopted. Results of linear regression analysis indicated that about 50% of the monitoring wells showed increasing trends of groundwater levels, electrical conductivities, and water temperatures and the others showed decreasing trends. However, the non-parametric analyses with monthly median values revealed that $14.8{\sim}20.0%$ of water levels were decreased, $24.2{\sim}36.9%$ of electrical conductivities were increased, and $27.4{\sim}32.5%$ of water temperatures were increased at a confidence level of 99%. Highly proportions of increasing or decreasing trends were unexpected and they resulted from the relatively short term of data collection (maximum 6 years). Meanwhile, the investigation of groundwater around the national groundwater monitoring stations showed that the decreasing or increasing trends of water levels, electrical conductivities, themselves, didn't indicate directly groundwater hazards such as groundwater depletion or groundwater contamination. Both the values and variation rates (slopes) of water level, electrical conductivity and temperature in the longer period are considered simultaneously. This study is the first comprehensive work in analyzing trends of groundwater data obtained from the national groundwater monitoring stations. Based on this study, the periodical and regular analysis of groundwater data is essentially required to grasp the overall variational trend of groundwater resources in the country.

Analysis on the Snow Cover Variations at Mt. Kilimanjaro Using Landsat Satellite Images (Landsat 위성영상을 이용한 킬리만자로 만년설 변화 분석)

  • Since the Industrial Revolution, CO2 levels have been increasing with climate change. In this study, Analyze time-series changes in snow cover quantitatively and predict the vanishing point of snow cover statistically using remote sensing. The study area is Mt. Kilimanjaro, Tanzania. 23 image data of Landsat-5 TM and Landsat-7 ETM+, spanning the 27 years from June 1984 to July 2011, were acquired. For this study, first, atmospheric correction was performed on each image using the COST atmospheric correction model. Second, the snow cover area was extracted using the NDSI (Normalized Difference Snow Index) algorithm. Third, the minimum height of snow cover was determined using SRTM DEM. Finally, the vanishing point of snow cover was predicted using the trend line of a linear function. Analysis was divided using a total of 23 images and 17 images during the dry season. Results show that snow cover area decreased by approximately $6.47km^2$ from $9.01km^2$ to $2.54km^2$, equivalent to a 73% reduction. The minimum height of snow cover increased by approximately 290 m, from 4,603 m to 4,893 m. Using the trend line result shows that the snow cover area decreased by approximately $0.342km^2$ in the dry season and $0.421km^2$ overall each year. In contrast, the annual increase in the minimum height of snow cover was approximately 9.848 m in the dry season and 11.251 m overall. Based on this analysis of vanishing point, there will be no snow cover 2020 at 95% confidence interval. This study can be used to monitor global climate change by providing the change in snow cover area and reference data when studying this area or similar areas in future research.

Development of Prediction Model for the Na Content of Leaves of Spring Potatoes Using Hyperspectral Imagery (초분광 영상을 이용한 봄감자의 잎 Na 함량 예측 모델 개발)

  • In this study, the leaf Na content prediction model for spring potato was established using 400-1000 nm hyperspectral sensor to develop the multispectral sensor for the salinity monitoring in reclaimed land. The irrigation conditions were standard, drought, and salinity (2, 4, 8 dS/m), and the irrigation amount was calculated based on the amount of evaporation. The leaves' Na contents were measured 1st and 2nd weeks after starting irrigation in the vegetative, tuber formative, and tuber growing periods, respectively. The reflectance of the leaves was converted from 5 nm to 10 nm, 25 nm, and 50 nm of FWHM (full width at half maximum) based on the 10 nm wavelength intervals. Using the variance importance in projections of partial least square regression(PLSR-VIP), ten band ratios were selected as the variables to predict salinity damage levels with Na content of spring potato leaves. The MLR(Multiple linear regression) models were estimated by removing the band ratios one by one in the order of the lowest weight among the ten band ratios. The performance of models was compared by not only R2, MAPE but also the number of band ratios, optimal FWHM to develop the compact multispectral sensor. It was an advantage to use 25 nm of FWHM to predict the amount of Na in leaves for spring potatoes during the 1st and 2nd weeks vegetative and tuber formative periods and 2 weeks tuber growing periods. The selected bandpass filters were 15 bands and mainly in red and red-edge regions such as 430/440, 490/500, 500/510, 550/560, 570/580, 590/600, 640/650, 650/660, 670/680, 680/690, 690/700, 700/710, 710/720, 720/730, 730/740 nm.

Development of Multi-Reservoir System Operation Rule Curves for Hydropower Maximization in the Nam Ngum River Basin of Lao PDR (라오스 남능강 유역 다중 저수지 시스템의 최적 수력발전 운영규정 곡선 개발)

  • The Lao government is continuously developing hydro-power dams in addition to the existing eight power plants in the Nam Ngum River basin and is expanding the power capacity of the existing power plants to meet the expected increase in electricity demand. Accordingly, the Lao government has requested an update on the existing reservoir operating rule curve in order to run the power plants efficiently. To this end, this study reviewed the current independent operating system as well as the joint operating system in order to maximize the annual power generation produced by a power plant by using CSUDP, general-purpose dynamic programming (DP) software. The appropriate operating regulation curve forms (URC/LRC, MRC) were extracted from the DP results, and the annual power generations were simulated by inputting them as the basic operating data of the reservoir operation set of the HEC-ResSim program. By synthesizing the amount of the annual power generation simulated, the existing operation regulation curve, the operational performance, and the opinion of the field operator, the optimal reservoir operation regulation curves that maximize the annual power generation of the target power plant were developed. Results revealed that a system operating in conjunction with the reservoir produces about 2.5 % more power generation than an independent reservoir due to the synergistic effect of the connection.
