Soil Respiration in Pinus rigida and Larix leptolepis Plantations (리기다소나무와 낙엽송(落葉松) 인공조림지내(人工造林地內) 토양발생(土壤發生) 이산화탄소(二酸化炭素)에 관한 연구(硏究))

  • Soil respiration was measured every two weeks from May through November 1995 using the soda lime method in 40-Year-old Pinus rigida and Larix leptolepis plantations on a similar soil in Yangpyeong, Kyonggi Province. Treatments included control and no-roots(plots trenched and root regrowth into plots prevented). Root respiration was evaluated by comparing no-roots sub-plots to control plots. Mean soil respiration showed highly significant species effects(p<0.01) and was highest at the Pinus rigida control plot($0.38g/m^2/hr$) and lowest at the Larix leptolepis no-roots plot($0.31g/m^2/hr$). High soil respiration in Pinus rigida may be related to aboveground litter production. The annual $CO_2$ fluxes ranged from 23 to 27t/ha/yr. We found significant correlations between temperatures(air : $R^2$=0.53, soil : $R^2$=0.55) and soil respiration(p<0.01), but no significant correlations between soil moisture and soil respiration(p>0.1). Root respiration was 3% of total soil respiration. We might underestimate rapt respiration because of shallow trenches and $CO_2$measurements right after trenching. Factors controlling soil respiration including belowground litterfall(especially fine roots) inputs, litter quality should be well understood to predict soil carbon fluxes and relative contributions to total soil respiration in forest ecosystems.

Commercial Scale Evaluation of Practical Extruded Pellet Feed for the Olive Flounder (Paralichthys olivaceus) (넙치 실용배합사료개발을 위한 현장적용시험)

  • This study was conducted to evaluate the effects of extruded pellet (EP) diets, as compared to a raw fish moist pellet (MP) diet for olive flounder, Paralichthys olivaceus, grown in commercial-scale aquaculture for 1 year. Four diets with duplication per diet were formulated for this experiment: two experimental EP diets (EP1 and EP2), one commercial EP diet (CEP), and a raw fish MP diet (MP). The MP diet consisted of 80% frozen horse mackerel and 20% commercial binder meal. Fish weighing $30.1{\pm}0.1 g$ ($mean{\pm}SD$) were distributed randomly to each aquarium as a group of 2,600 fish. Weight gain (WG) and feed efficiency ratio (FER) of fish fed EP2 and MP were higher (P<0.05) than those of fish fed CEP, while those of fish fed EP1 did not differ (P>0.05) from those fed EP2 and MP. However, fish fed the MP diet had a higher survival rate than fish fed the other diets. Fish fed EP2 had higher serum, phospholipids and total protein levels, and lower levels of serum glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT), and total cholesterol than fish fed MP (P<0.05). Dorsal muscle and liver proteins and lipid of fish fed EP1 were higher (P<0.05) than those of fish fed CEP, while those of fish fed EP1 and MP did not differ from those of fish fed EP2 (P>0.05). These results strongly suggest that EP1 could be developed to replace MP for grow-out stage production of olive flounder without adverse effects on growth performance.

Prediction of Digestible and Metabolizable Energy Content of Rice Bran Fed to Growing Pigs

  • Two experiments were conducted to determine the digestible energy (DE) and metabolizable energy (ME) content of 19 rice bran samples and to develop prediction equations for DE and ME based on their chemical composition. The 19 rice bran samples came from different rice varieties, processing methods and regions. The basal diet was formulated using corn and soybean meal (74.43% corn and 22.91% soybean meal and 2.66% vitamins and minerals). The 19 experimental diets based on a mixture of corn, soybean meal and 29.2% of each source of rice bran, respectively. In Exp. 1, 108 growing barrows ($32.1{\pm}4.2kg$) were allotted to 1 of 18 treatments according to a completely randomized design with 6 pigs per treatment. The treatment 1 was the control group which was fed with basal diet. The treatments 2 to 18 were fed with experimental diets. In Exp. 2, two additional rice bran samples were measured to verify the prediction equations developed in Exp. 1. A control diet and two rice bran diets were fed to 18 growing barrows ($34.6{\pm}3.5kg$). The control and experimental diets formulations were the same as diets in Exp. 1. The results showed that the DE ranged from 14.48 to 16.85 (mean 15.84) MJ/kg of dry matter while the ME ranged from 12.49 to 15.84 (mean 14.31) MJ/kg of dry matter. The predicted values of DE and ME of the two additional samples in Exp. 2 were very close to the measured values.

Effect of Early Pregnant Heifer as Donor on the Ovum Pick-Up Derived Oocyte Aspiration and Embryo Production (초기 임신우의 공란우 활용이 초음파 유도 난자 채취 및 수정란 생산에 미치는 영향)

  • This study was carried out to evaluate the effect of early pregnant cow as donor for Ovum Pick-Up (OPU) derived oocyte aspiration and embryo production in Holstein heifers. Four non-pregnant and 2 pregnant Holstein heifers were used as donor and then carried out total 17 OPU session for 10 weeks (2 times per week). Recovered cumulus-oocyte-complexes (COCs) were classified into 4 grade by oocyte cytoplasm and cumulus cells and matured in vitro in TCM-199 supplemented with 10% FBS, 10 mg/ml FSH and 1 mg/ml estradiol in 5% $CO_2$ and over 99% humidity for 24 h. After 24 h co-incubation with post-thaw sperm, the presumed zygotes were cultured in CR1aa medium with 4 mg/ml BSA for 3 days and then changed CR1aa medium with 10% of FBS for another 3~4 days. The Mean number of aspirated follicles and collected oocytes in the early stage pregnant and non-pregnant heifers were $13.0{\pm}4.3$ and $10.6{\pm}3.9$, $5.4{\pm}3.4$ and $7.7{\pm}3.6$ per session, respectively. Rate of collected oocyte from aspirated follicles were 59.2% and 50.5%, respectively. The average number of good quality oocytes (Grade I and II) in the early stage pregnant and non-pregnant heifers was $3.7{\pm}2.7$ and $4.9{\pm}2.6$ (Mean${\pm}$SD). Cleavage and blastocyst developmental rates in Grade I and II were 22.2% and 25.5%, and then $1.7{\pm}0.9$ and $1.4{\pm}1.1$ blastocyst per session, respectively. In conclusion, OPU technology can be used in early stage pregnant and non-pregnant heifers without any problem and so applied OPU derived embryo production to maximize the ability of genetically valuable females.

Liquid Boar Sperm Quality during Storage and In vitro Fertilization and Culture of Pig Oocytes

  • The percentages of sperm motility and normal acrosome on the liquid boar semen diluted and preserved at $4^{\circ}C$ with lactose hydrate, egg yolk and N-acetyl-D-glucosamine (LEN) diluent were significant differences according to preservation day and incubation time, respectively. The sperm motility steadily declined from 96.9% at 0.5 h incubation to 78.8% at 6 h incubation at 1 day of preservation. However, the sperm motility rapidly declined after 4 day of preservation during incubation. The normal acrosome steadily declined from 93.3% at 0.5 h incubation to 73.8% at 6 h incubation at 1 day of preservation. However, the normal acrosome rapidly declined after 3 day of preservation during incubation. The rates of sperm penetration and polyspermy were higher in 5 and $10{\times}10^6$ sperm/ml than in 0.2 and $1{\times}10^6$ sperm/ml. Mean numbers of sperm in penetrated oocyte were highest in $10{\times}10^6$ sperm/ml compared with other sperm concentrations. The rates of blastocysts from the cleaved oocytes (2-4 cell stage) were highest in $1{\times}10^6$sperm/ml compared with other sperm concentrations. In conclusion, we found out that liquid boar sperm stored at $4^{\circ}C$ could be used for in vitro fertilization of pig oocytes matured in vitro. Also, we recommend $1{\times}10^6$sperm/ml concentration for in vitro fertilization of pig oocytes.

The Effects of Milking Time and Dairy Processes on Melatonin Contents in Milk (착유 시간 및 유처리 공정이 우유 내 멜라토닌 함량에 미치는 영향)

  • Melatonin is one of the functional hormones in the milk and found in all mammalian species. The primary motivation for the use of melatonin as a supplement is as a natural aid to better sleep. Melatonin contents in milk vary according to the naturally occurring factors such as seasons and milking times. As a number of studies indicate melatonin supplementation helps to reduce the age-related decline in hormone production and overall enhancement of health, the more detailed studies are required to know the nature of milk derived hormones and to clarify them as a functional value adaptation. The objectives of this study were to examine the change of melatonin content in bovine milk during different milking times at the two dairy farms and during dairy process including homogenization and pasteurization. Commercial dairy products including night time milking products(night milk) marketed in Japan were also investigated. Melatonin content was determined by radioimmunoassay using 125I. Individual milk was collected from lactating Holstein cows at Kyong-Ki province in Korea. At farm A, the melatonin contents milking at 4 a.m. was higher than those of 7 p.m. and 12 p.m.($6.90{\pm}3.55,\;2.01{\pm}1.47$ and $0.16{\pm}0.04pg/mL$, respectively). At farm B, the mean melatonin contents milked from 24:00 AM to 05:00 AM have shown the highest contents ($4.65{\pm}$0.72pg/mL), and milking samples of the rest time had very little or non-detection of melatonin contents. Melatonin contents of commercial night milk samples were about 5 times higher than those of other common milk products. These results suggested that melatonin might be heat-resistant and commercial dairy process might not affect its contents in the final products.

The Study on Marginal discrepancy of Manufactured Coping Crown according to Production method and materials by using CAD/CAM System (CAD/CAM System을 활용하여 제작된 Coping Crown의 제작방식 및 재료에 따른 변연 적합도에 관한 연구)

  • The purpose of this study was to compare the marginal discrepancy of coping crowns according to manufacturing method and materials by using the CAD/CAM system to compare the optimum manufacturing method and materials. The coping crowns were fabricated by using manufacturing methods and materials, and marginal fit was measured using the replica technique. For comparative analysis of manufacturing methods and materials, analysis of variance was performed. One-way ANOVA was performed to compare and analyze different mean values. For the milling method of the alloy, 114.6 was obtained for the MM group, 111.4 for the MS group, 67.2 for the MSC group, and 50.9 for the MSS group, respectively, using the milling/sintering method. In the milling/sintering method of ceramic, 35.6 and 36.3, respectively, were obtained for the SLME group and SLMR group, respectively. However,there was a significant difference according to the material. The milling/sintering method of the ceramic material gave the lowest value, and it was verified by the optimal method. However, marginal discrepancy of coping crowns fabricated using the CAD/CAM system is applicable to the clinic since it corresponds to acceptable numerical values in clinical practice.

Effects of Dietary Probiotics as an Alternative to Antibiotics on Growth Performance, Biochemical Characteristics and Immune Response in Weaning Pigs (이유자돈에 있어서 복합 생균제(MR-1)의 사료 내 첨가가 성장 능력 및 생화학적 조성, 면역 반 응에 미치는 영향)

  • We evaluated the growth performance, biochemical characteristics, and immune responses in weaning pigs given a diet containing MR-1 (0.2%/feed) or antibiotics (0.1%/feed) for 45 days. In vitro study showed that MR-1 has antibacterial activity against a variety of strains of pathogenic bacteria, especially a strain of cattle-derived Escherichia coli K99 (E. coli K99) by agar diffusion assay. In the in vivo model, 0.2% MR-1-given group clearly ameliorated the weight gain and feed efficiency in the growth performance of weaning pigs compared to the basal diet group (p<0.05). Additionally, 0.2% MR-1 induced an elevation in the levels of mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) and showed a similar pattern ($TNF{\alpha}$ and $IFN{\gamma}$ production) to the antibiotic treated pigs. Taken together, we suggest that 0.2% MR-1 makes probiotics an alternative to antibiotics in weaning pigs.

Effect of Peanut Seed Orientation on Germination, Seedling Biomass, and Morphology in an Oak Tree Sawdust Cultivation System

  • We performed seed germination tests to investigate the effects of seed sowing orientation on germination viability on peanut (Arachis hypogaea L.) sprouts. Specifically, we assessed the influence of seed sowing orientation on germination rate, seedling weight, and seedling length, as well as the seedling vigor index. The seeds were sown in oak tree sawdust at 3.0 cm depth. Four seed orientations were tested: vertical with the hypocotyl end down, vertical with the hypocotyl end up, horizontal with the hypocotyl end down, and horizontal with the hypocotyl end up. The mean seed germination percentages of the four seed orientations were significantly different (p < 0.01) and ranged from 25 to 91.7%. The vertical orientation with hypocotyl-end-down and hypocotyl-end-up orientations showed the highest (91.7%) and lowest (25%) germination rates, respectively. The vertical orientation with the hypocotyl end down produced the heaviest (4.9 g) seedlings and the longest hypocotyls (4.65 cm). This orientation also produced the longest true leaf + epycotyl (2.15 cm) and had the highest seedling vigor index (197.1). The seedlings had a straight growth pattern, whereas seedlings from seeds sown with the hypocotyl up had an awkward plumular hook shape. Taken together, to produce peanut sprouts, we recommend placing the seeds vertically with the hypocotyl end down because this orientation leads to a high germination rate, high biomass production, and high overall seedling quality.

Estimation of co-variance components, genetic parameters, and genetic trends of reproductive traits in community-based breeding program of Bonga sheep in Ethiopia

  • Objective: The objectives of the study were to evaluate reproductive performance and selection response through genetic trend of community-based breeding programs (CBBPs) of Bonga sheep. Methods: Reproduction traits data were collected between 2012 and 2018 from Bonga sheep CBBPs. Phenotypic performance was analyzed using the general linear model procedures of Statistical Analysis System. Genetic parameters were estimated by univariate animal model for age at first lambing (AFL) and repeatability models for lambing interval (LI), litter size (LS), and annual reproductive rate (ARR) traits using restricted maximum likelihood method of WOMBAT. For correlations bivariate animal model was used. Best model was chosen based on likelihood ratio test. The genetic trends were estimated by the weighted regression of the average breeding value of the animals on the year of birth/lambing. Results: The overall least squares mean±standard error of AFL, LI, LS, and ARR were 375±12.5, 284±9.9, 1.45±0.010, and 2.31±0.050, respectively. Direct heritability estimates for AFL, LI, LS, and ARR were 0.07±0.190, 0.06±0.120, 0.18±0.070, and 0.25±0.203, respectively. The low heritability for both AFL and LI showed that these traits respond little to selection programs but rather highly depend on animal management options. The annual genetic gains were -0.0281 days, -0.016 days, -0.0002 lambs and 0.0003 lambs for AFL, LI, LS, and ARR, respectively. Conclusion: Implications of the result to future improvement programs were improving management of animals, conservation of prolific flocks and out scaling the CBBP to get better results.
