Protective Effects of the Water Extracts of Herbal Medicine on BNL cl.2 Cells (수종의 생약 물 추출물의 BNL cl.2 Cells 보호효과)

  • The protective effects of the water extracts of nine kinds of medicinal herbs, which have been reputed to having the hepatoprotective activity in Chinese herbal medicine, on BNL cl.2 cells using a MTT assay were investigated. Five extracts including Coriolus versica, Curcuma longa, Phellinus linteus, Po-lygonum aviculare, and Salvia miltiorrhiza showed the protective effects on BNL cl.2 cells damaged by $CCl_4$ with $ED_{50}$ values of less than $100\;{\mu}g/ml$. Silymarin had been used as a positive control.

  • The fluorescence quenching of 5-methyl-3-phenyl-2-[s-oxadiazol-2'-thionen5'-yl] indole by carbon tetrachloride ($CCl_4$) and aniline in different solvents viz., dioxane, benzene, toluene, methanol, propanol has been carried out at room temperature to understand the role of quenching mechanisms. The Stern-Volmer plots have been found to be linear. As probability of quenching per encounter 'p' is less than unity, and the activation energy for quenching 'E$_{a}$' is greater than the activation energy of diffusion 'E$_{d}$', it is inferred that the fluorescence of quenching mechanism is not due to material diffusion alone.e.e.

  • 이성분 용액에 대한 천이상태 이론의 적용 방법을 재고찰하여 이론적 보충을 하여주었으며, 이 새로운 방법에 따라서 $C_6H_{12}-CCl_4$계의 열역학적 성질들을 계산하였다. 또한 전의 방법에 따라서 계산된 $C_6H_6-C_6H_{12}$$C_6H_6-CCl_4$계의 열역학적 성질들도 재계산하였다. 이 방법에 따라서 계산된 열역학적 성질들을 측정치와 대단히 잘 맞는다.

  • Liver and serum xanthine oxidase(XOD) activity were determined in rats treated with DL-ethionine. Concomitantly, the enzyme activity was compared with that of rats treated with CCl$_{4}$, actinomycin D and those fed a protein depleted diet. The activity of XOD in liver was inhibited by injection of ethionine to the rats. But, there were no differences in activity of serum XOD between control group and ethionine-treated rats. And the pattern of changes in enzyme activity of serum and liver in ethionine-treated rats, was similar with actinomycin D treated rats or those fed a protein depleted diet. On the other hand, the activity of XOD was rather elevated both in serum and liver by injection of CCl$_{4}$ to rats.

  • Water fractions prepared from Plantaginis Semen and Plantaginis Folium exhibited both significant liver-protective activities against $CCl_4$ intoxication. Aucubin, an iridoid glycoside. isolated from both leaves and seeds of Plantago asiatica(Plantaginaceae) also showed potent liver protective activities against the animal model of hepatitis induced by $CCl_4$ intoxication.

  • The reactions between organic halides$(CCl_4,\;C_6H_5Br,\;C_6H_5Cl,\;C_6H_5I)$ and amines $(C_6H_5NH_2,\;R_2NH,\;R_3N,\;(CH_2)_5NH,\;pyridine)$ were studied under mixed u.v. irradiation. The modes of reactions were examined by means of gas chromatography and product-reactant ratio determination. The reaction of $CCl_4$ with amines give chloroform and hexachloroethanes, and the reaction of aromatic halides with amines gave biphenyl and benzene. In each series of reaction there obtained mainly corresponding amine hydrohalides, but no amination products. The reactivity was in the order of the basicity of amines and of the reactivity of organic hahides, except in the case of cyclic tertiary amine. The result was interpreted as a non-chain photodecomposition process. A competitive proton abstraction reaction path via the formation of a change transfer complex was proposed as the reaction mechanism.

독성물질 대사효소 조절기전에 관한 연구

  • 약물, hormone, 독성물질등의 대사능과 발암 가능성등이 간장 장해시 및 ketosis시에 달라지는 원인과 기전, 독성물질 대사효소의 변동과 그 작용기전을 규명하고자, 대표적인 간장장해 물질인 carbon tetrachloride를 rat에 투여하여 간장 장해를 일으키고, 당뇨병, starvation, high-fat diet처리하여 ketosls상태를 만든 후에, specific cytochrome P45O polyclonal antibodies와 cDNA probes를 사용하여, enzyme activitieg, Western immunoblot analysis와 mRNA Northern blot analysis 등을 실험하여, 간장 장해와 ketosis시 cytochrome P45O의 변동과 그 작용기전, regulation을 규명하고자 하였다. 실험 결과, $CCl_4$투여후 P450IIE enzyme (aniline hydroxylase) 활성이 시간 의존적으로 급격히 떨어졌고, P450IIE protein양이 똑같은 방식으로 감소되었으나 mRNA level은 변화가 없었다. $CCl_4$에 의해서 P450IIE는 protein의 특이적인 파괴에 의한 post-translational reduction됨을 알 수 있었다. 반면에 당뇨병, starvation, high-fat diet등 ketosis시에는 P450IIE 효소활성이 2-3배 증가되었고, P450IIE protein양도 같은 수준으로 증가되었으며, mRNA도 증가 되었다. Ketosis시에는 P450IIE가 pretranslational activation됨을 알 수 있었다.

Hepatoprotective and antioxidant activity of the methanolic extract of Mucuna pruriens Bak. roots

  • The methanolic extract of natural roots of Mucuna pruriens was screened for hepatoprotective and antioxidant effects in carbon tetrachloride ($CCl_4$) induced hepatotoxicity in Albino rats. The degree of protection was measured by estimating biochemical parameters like serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, total protein and level of serum bilirubin (both total and direct). The extract also reduced $CCl_4$ induced lipid peroxidation both in-vitro and in-vivo. Hepatoprotective activity of methanolic extract at dose 300 mg/kg body weight, i.p., was comparable with standard drug Silymarin (25 mg/kg, i.p.). Furthermore, histopathological experiments were also carried out to support the study.

  • The effects of various fractions from the whole plant of Pelvetia siliquosa Tseng et Chang (Fucaceae) on the $CCl_4$-induced hepatotoxicity as well as on streptozotocin (STZ)-induced diabetes in rats were investigated. The ether fraction exhibited a potent rat lens aldose reductase (RLAR) inhibition in vitro and showed a significant inhibition of not only serum glucose concentrations but also sorbitol accumulations in the lens, red blood cells and sciatic nerves in the STZ-induced diabetic rats. When administered orally in Sprague-Dawley rats, $H_{2}O$ fraction was found to cause a significant inhibition of the rise in the serum transaminase activities in $CCl_4$-intoxicated rats. These results suggested that this plant might possess constituents with hepatoprotective, anti-diabetic effects and those effects on diabetic complications.

  • Hepatic fibrosis/cirrhosis is characterized by increased production and deposition of collagen, noncollagenous glycaproteins, and proteoglycans, which mainly compose the extracellular matrix (ECM). Recently, activations of macrophages, myofibroblasts and mast cells are thaught ta be associated with the accumulation of ECM. In order to investigate the kinetics of macrophages, myofibroblasts and mast cells and the relationship between these cells and the accumulation of ECM in carban tetrachloride (CCl$_4$)-injected rat liver, we induced liver cirrhosis of rat by an injection of CCl$_4$ for 14 weeks.
