GLB1-related disorders: GM1 gangliosidosis and Morquio B disease

  • GLB1-related disorders comprise two phenotypically unique disorders: GM1 gangliosidosis and Morquio B disease. These autosomal recessive disorders are caused by b-galactosidase deficiency. A hallmark of GM1 gangliosidosis is central nervous system degeneration where ganglioside synthesis is highest. The accumulation of keratan sulfate is the suspected cause of the bone findings in Morquio B disease. GM1 gangliosidosis is clinically characterized by a neurodegenerative disorder associated with dysostosis multiplex, while Morquio B disease is characterized by severe skeletal manifestations and the preservation of intelligence. Morquio B disease and GM1 gangliosidosis may be on a continuum of skeletal involvement. There is currently no effective treatment for GLB1-related disorders. Recently, multiple interventions have been developed and there are several ongoing clinical trials.

Nucleus-Selective Expression of Laccase Genes in the Dikaryotic Strain of Lentinula edodes

  • In mating of Lentinula edodes, dikaryotic strains generated from certain monokaryotic strains such as the B2 used in this study tend to show better quality of fruiting bodies regardless of the mated monokaryotic strains. Unlike B2, dikaryotic strains generated from B16 generally show low yields, with deformed or underdeveloped fruiting bodies. This indicates that the two nuclei in the cytoplasm do not contribute equally to the physiology of dikaryotic L. edodes, suggesting an expression bias in the allelic genes of the two nuclei. To understand the role of each nucleus in dikaryotic strains, we investigated single nucleotide polymorphisms (SNPs) in laccase genes of monokaryotic strains to reveal nuclear origin of the expressed mRNAs in dikaryotic strain. We performed reverse transcription PCR (RT-PCR) analysis using total RNAs extracted from dikaryotic strains (A5B2, A18B2, and A2B16) as well as from compatible monokaryotic strains (A5, A18, and B2 for A5B2 and A18B2; A2 and B16 for A2B16). RT-PCR results revealed that Lcc1, Lcc2, Lcc4, Lcc7, and Lcc10 were the mainly expressed laccase genes in the L. edodes genome. To determine the nuclear origin of these laccase genes, the genomic DNA sequences in monokaryotic strains were analyzed, thereby revealing five SNPs in Lcc4 and two in Lcc7. Subsequent sequence analysis of laccase mRNAs expressed in dikaryotic strains revealed that these were almost exclusively expressed from B2-originated nuclei in A5B2 and A18B2 whereas B16 nucleus did not contribute to laccase expression in A2B16 strain. This suggests that B2 nucleus dominates the expression of allelic genes, thereby governing the physiology of dikaryons.

미세구조 제어를 통한 (Nd,Dy)-Fe-B 소결자석의 보자력 증가 (The Coercivity Enhancement of (Nd,Dy)-Fe-B Sintered Magnet by Microstructure Control)

  • Sintered Nd-Fe-B magnets are widely used in many fields such as motors, generators, actuators, microwaves and so on due to their excellent magnetic properties. Many researchers have shown that the Nd-rich phase was essentially important for high magnet properties. In this study, we focused on controlling of the Nd-rich phase to enhance magnetic properties by the cyclic sintering process. Nd-Fe-B based sintered magnets were prepared by isothermal sintering and cyclic sintering processes. Magnetic properties and microstructure of the magnets were investigated. The coercivity was enhanced from 21.2 kOe to 23.27 kOe after 10 cycles of the sintering. The Nd-rich phase was effectively penetrated into the grain boundary between the $Nd_2Fe_{14}B$ grains by the cyclic sintering.

Heterologous Regulation of BCG hsp65 Promoter by M.leprae 18 kDa Transcription Repression Responsive Element

  • Among a number of antigens characterized in M leprae, an etiological agent of Leprosy, the 18 kDa antigen, is unique to M leprae. We have previously determined a sequence specific element in the 18 kDa gene of M leprae, which confers transcriptional repression. In this report, we have examined if the element could be applied to genes other than the 18 kDa gene of M leprae. To identify the roles of the regulatory sequence in heterologous promoter, we have constructed pB3 vector series, which contains BCG hsp65 promoter and the M leprae 18 kDa transcription repression responsive element in tandem using LacZ gene as a reporter gene. Cloning of hsp65 promoters of M bovis BCG or M smegmatis in front of LacZ gene resulted in normal $\beta$­galactosidase activity as expected. However, when the sequence element was placed between the promoter and the LacZ gene, $\beta$-galactosidase activity was reduced 10-fold less. Also we have examined with pB3(-) vector, that harbors the transcription repression responsive element in a reversed orientation, the $\beta$-galactosidase activity was found to be similar to pB3(+) vector. Thus, these results further confirm that M leprae 18 kDa transcription repression responsive element could regulate BCG hsp65 heterologous promoter and that the element could act as an operator for the transcription of mycobacteria.

고추역병을 방제하는 PGPR균주 Bacillus subtilis AH18의 항진균성 Cellulase 유전자의 Cloning 및 효소 특성 조사 (Cloning and Characterization of a Cellulase Gene from a Plant Growth Promoting Rhizobacterium, Bacillus subtilis AH18 against Phytophthora Blight Disease in Red-Pepper)

  • 식물생육을 촉진하고 고추역병균을 방제하는 다기능 PGPR 균주 Bacillus subtilis AH18 항진균성 cellulase 유전자를 PCR을 이용해 pUC18과 재조합 후 E. coli DH5$\alpha$에 cloning하여 E. coli내에 발현시켰으며, 그 형질전환 균주를 E. coli DH5$\alpha$(pCM 41)이라 명명하였고, 발현된 cellulase를 ce/H라 하였다. E. coli DH5$\alpha$(pCM 41)의 inset 부위는 B. subtilis AH18의 1,582 bp 유전자를 포함하며 cellulase의 유전자는 1,524 bp로 508개의 amino acid가 암호화된 것으로 추정되었고, CMC를 함유한 SDS-PAGE의 방법으로 약 55 kDa의 분자량을 확인하였다. B. subtilis AH18이 가지는 ce/H는 3종의 대표적인 Bacillus spp.들의 cellulase 유전자의 DNA와 아미노산 배열이 98% 이상 유사하였으며, CMC(carboxymethyl-cellulose) 뿐만 아니라, 불용성 섬유소인 Avicel, filter paper(Whatman No. 1) 특히 고추역병균인 Phytophthora capsici의 건조 cell wall도 분해하였다. 또한 colH의 cellulase는 $50^{\circ}C$에서 효소활성이 가장 높았으며, 최적 pH는 pH 6.0이었다. 그리고 $AgNO_3$ 또는 $CoCl_2$ 첨가시 활성이 1.7배, 2배 정도 증가하였고 $HgC1_2$ 첨가시는 활성이 20%까지 떨어졌다. 또한 여러 화학 저해제들 중 Sodium azide 또는 Hydroxy urea는 효소 활성을 증가시켰으며, CDTA 또는 EDIA는 섬유소분해능을 감소시켰다. 이들의 결과는 고추역병균 P. capsici의 생육을 억제하는 B. subtilis AM18의 진균세포벽 용해성 cellula의 효소학적 특성을 구명한 것이라고 할 수 있다.

0.18 um CMOS 공정을 이용한 UWB 단일 입력-차동 출력 이득 제어 저잡음 증폭기 설계 (A Design of Ultra Wide Band Single-to-Differential Gain Controlled Low Noise Amplifier Using 0.18 um CMOS)

  • 본 논문에서는 CMOS 0.18 um 공정을 이용하여 UWB(Ultra Wide Band) 시스템의 낮은 대역 $3.1{\sim}4.8GHz$에서 사용할 수 있는 단일 입력-차동 출력 이득 제어 저잡음 증폭기를 설계하였다. 측정 결과는 높은 이득 모드에서 차동 출력 전력 이득은 각각 $14.1{\sim}15.8dB,\;13.3{\sim}15dB$로, 입력 반사 계수는 -10dB 이하로, lIIP3는 -19.3dBm, 잡음 지수는 $4.85{\sim}5.09dB$로 측정되었으며, 이때 전원 전압 1.8V에서 사용 전력은 19.8 mW를 사용하였다. 낮은 이득 모드에서 차동 출력 전력 이득은 각각 $-6.1{\sim}-4.2dB,\;-7.6{\sim}-5.6dB$로, 입력 반사 계수는 -10dB 이하로, IIP3는 -1.45 dBm, 잡음 지수는 $8.8{\sim}10.3dB$로 측정되었으며, 이때 전원 전압 1.8V에서 사용 전력은 5.4mW를 사용하였다.

Sexual Behaviors, Estrus Detection and Conception of Heifers Synchronized by Progesterone Intravaginal Device(Prid) and Synchromate-B

  • Seventy four Holstein heifers were randomly assigned over three trials to PRID-7+PG-6 and Synchromate B-9 regimens to synchronize estrus cycle for embryo transfer. Sexual behaviors; moounting, standing, orientation, chin-resting, sniffing, licking, rubbing and butting, vaginal swelling and mucus discharge were observed between 06-08, 12-14 and 18-20 h on 1st day and 00-02, 06-08, 12-14 and 18-20 h on 2nd day after removal of hormones. Synchromate-B treatment (81.6%) showed higher synchronized estrus rate than PRID treatment (77.8%) during observation period. Standing estrus was observed within 74 h after PG injection in PRID and within 52 h after removal of implant in Synchromate-B. About 68% of heifers in PRID and 74% of heifers in Synchromate-B showed standing estrus between 0-14 h on 2nd day after removal of the hormones. Synchromate-B resulted in a tighter synchrony of standing estrus than PRID. Incidence of average mounting and standing per head during observation period was 22.3 and 16.6 in PRID and 28.1 and 13.6 in Synchromate-B. The PRID showed peak in active mounting at 18-20 h on 1st day, however, the Synchromate-B showed at 0-2 h on 2nd day after removal of hormone. Active standing was shown between 18 h on 1st day to 20h on 2nd day in PRID, however, between 0-14 h on 2nd day after removal of hormone is Synchromate-B. There was slight difference in pattern of active mounting and standing during estrus between PRID and Synchromate-B. Conception rate of synchronized heifers transferred with fresh and frozen embryos by non-surgical and surgical methods was higher in synchromate-B (62.5%) than in PRID (38.5%). Chin-resting showed highest incidence among 6 sexual behavioral components in the both treatments. Synchromate-B showed higher incidence of chin-resting (16.6) than PRID (10.7). Synchromate-B group showed also higher incidence of orientation, sniffing and butting than PRID group. Synchromate-B resulted in more active sexual behaviors than PRID. The pattern of incidence of chin-resting, licking and butting was almost symmetrical in PRID with their peak values at 6-8 h on 2nd day, however in Synchromate-B chin-resting and sniffing was symmetrical with their peak values at 12-14 h on 2nd day after removal of hormone. There was tendency to increase vaginal swelling according to time passage of synchronized estrus in the both treatments. Incidence of mucus discharge in Synchromate-B was slightly higher than in PRID. Twenty to 40% was false negative in conception rate by tall painting before re-estrus day as judged by rectal palpation.
