Red pepper seed water extract inhibits preadipocyte differentiation and induces mature adipocyte apoptosis in 3T3-L1 cells

  • BACKGROUND/OBJECTIVES: Reducing the number of adipocytes by inducing apoptosis of mature adipocytes as well as suppressing differentiation of preadipocytes plays an important role in preventing obesity. This study examines the anti-adipogenic and pro-apoptotic effect of red pepper seed water extract (RPS) prepared at $4^{\circ}C$ (RPS4) in 3T3-L1 cells. MATERIALS/METHODS: Effect of RPS4 or its fractions on lipid accumulation was determined in 3T3-L1 cells using oil red O (ORO) staining. The expressions of AMP-activated protein kinase (AMPK) and adipogenic associated proteins [peroxisome proliferator-activated receptor-${\gamma}$ ($PPAR-{\gamma}$), CCAAT/enhancer-binding proteins ${\alpha}$ (C/EBP ${\alpha}$), sterol regulatory element binding protein-1c (SREBP-1c), fatty acid synthase (FAS), and acetyl-CoA carboxylase (ACC)] were measured in 3T3-L1 cells treated with RPS4. Apoptosis and the expression of Akt and Bcl-2 family proteins [B-cell lymphoma 2 (Bcl-2), Bcl-2-associated death promoter (Bad), Bcl-2 like protein 4 (Bax), Bal-2 homologous antagonist/killer (Bak)] were measured in mature 3T3-L1 cells treated with RPS4. RESULTS: Treatment of RPS4 ($0-75{\mu}g/mL$) or its fractions ($0-50{\mu}g/mL$) for 24 h did not have an apparent cytotoxicity on pre and mature 3T3-L1 cells. RPS4 significantly suppressed differentiation and cellular lipid accumulation by increasing the phosphorylation of AMPK and reducing the expression of $PPAR-{\gamma}$, C/EBP ${\alpha}$, SREBP-1c, FAS, and ACC. In addition, all fractions except ethyl acetate fraction significantly suppressed cellular lipid accumulation. RPS4 induced the apoptosis of mature adipocytes by hypophosphorylating Akt, increasing the expression of the pro-apoptotic proteins, Bak, Bax, and Bad, and reducing the expression of the anti-apoptotic proteins, Bcl-2 and p-Bad. CONCLUSIONS: These finding suggest that RPS4 can reduce the numbers as well as the size of adipocytes and might useful for preventing and treating obesity.

Clinical Implications According to Diagnostic Methods of Human Epidermal Growth Factor Receptor 2 Positivity in Breast Cancer: A Retrospective Study

  • Purpose: According to American Society of Clinical Oncology/College of American Pathologists guidelines, breast cancer is human epidermal growth factor receptor 2 (HER2) positive if there is HER2 protein overexpression at a 3+ level on immunohistochemistry (IHC 3+) or gene amplification (more than six copies per nucleus) on fluorescence in situ hybridization (FISH+). However, there have been few reports on whether outcomes differ based on diagnosis by these two techniques. In this study, we compared outcomes based on the two methods in patients with HER2-positive breast cancer. Methods: This study was a retrospective analysis of HER2-positive breast cancer in 18,304 patients, including 14,652 IHC 3+ patients and 3,652 FISH+ patients from the Korean Breast Cancer Society Registry. We compared breast cancer-specific survival and overall survival based on IHC 3+ and FISH+ status with or without trastuzumab. Results: Breast cancer-specific survival was significantly different between the IHC 3+ and FISH+ groups, with 5-year cumulative survival rates of 95.0% for IHC 3+ and 98.5% for FISH+ patients who did not receive trastuzumab (p=0.001) in Kaplan-Meier methods. However, there were no significant differences in breast cancer-specific survival and overall survival between IHC 3+ and FISH+ groups regardless of trastuzumab treatment in Cox proportional hazards models. Conclusion: The survival outcomes were not affected by the different two diagnostic methods of HER2-positive breast cancer. Further research to evaluate differences in prognosis and other characteristics according to the diagnostic methods of HER2 positivity is needed in the future.

Comparative evaluation of supplemental zilpaterol hydrochloride sources on growth performance, dietary energetics and carcass characteristics of finishing lambs

  • Objective: We compare the effects of three different approved sources of supplemental zilpaterol on growth-performance responses and carcass characteristics of finishing lambs. Methods: Twenty four Pelibuey${\times}$Katahdin lambs ($46.75{\pm}2.43kg$) were used in a 33-day feeding trial. Lambs were fed a dry rolled corn-based finishing diet. Treatments consisted of the non-supplemental basal diet (Control) versus the basal diet supplemented with 125 mg zilpaterol/kg of diet (as fed basis) from three commercial sources marketed in Mexico: Zilmax (ZIL), Grofactor, and Zipamix. Results: Compared to controls, zilpaterol (ZH) supplementation did not affect dry matter intake (DMI), but increased carcass adjusted daily weight gain (ADG, 36.7%), gain efficiency (34.2%), and dietary net energy (26.0%), and decreased (23.4%) the ratio of observed:expected DMI. Compared to controls, supplemental ZH increased hot carcass weight (6.4%), dressing percentage (3.2%), m. longissimus thoracis (LM) area (15.6%), and shoulder muscle:fat ratio (28.7%), but decreased kidney-pelvic-heart fat, and fat thickness. Supplemental ZH increased 10.9% and 14.3% whole cut weight of loin and leg, respectively, and the proportion (as percentage of cold carcass weight) of leg (4.3%). These increases were reflected in greater forequarter and hindquarter weights. Lambs fed ZH increased (4.6%) empty body weight (EBW) and reduced (14.7%) liver/spleen weight (as g/kg EBW). Likewise, ZH supplementation tended (p = 0.08) to lower (8.9%) visceral fat. Growth performance, energetic efficiency, hot carcass weight, dressing percentage, LM area and whole cuts were not different across supplemental ZH sources. However, compared with non-supplemented controls, only ZIL appreciably decreased carcass fat distribution, including fat thickness, percentage kidney pelvic and heart fat, shoulder fat, and visceral fat. Conclusion: Supplemental ZH increases ADG, gain efficiency, carcass dressing percentage, and LM area. The magnitude of these responses was similar among ZH sources. Nevertheless, compared with non-supplemented controls, only ZIL appreciably decreases carcass fat. The basis for this is uncertain, but indicative that some practical differences in zilpaterol bio-equivalency may exist across commercial sources tested.

운동과 영양 조절이 남성 연령별 체조성 및 비만에 미치는 영향 (Effects of Exercise and Nutrition on Male Body Composition and Obesity)

  • 본 연구는 운동과 영양 조절을 통한 남성 체 구성을 조사하였다. 훈련을 수행하는데 문제가 없는 건강한 40명을 임의로 선정하여 20명에게는 운동과 영양조절 그룹으로 선정하였고, 나머지 20명에게는 운동만 6개월간 수행하도록 하였다. 실험조건 동안 운동강도를 정확하게 측정하기 위해 최대산소 소모량($VO_2$ max)을 측정하였다. 운동과 영양조절일 실시한 20대 남자와 운동만 수행한 남자는 6개월 후 체중이 2.3 kg과 0.4 kg이 감소하였다. 0~3개월 간 실시한 결과에서 체중 측정값에 p값은 0.697이였으며, 3~6개월 측정값의 p값은 0.535로 나타났고, 0~6개월 측정 값의 p값은 0.617로 나타났다. 20대의 골격근은 약간 증가하였다. 30대의 체중은 6개월 후 운동과 영양조절 그룹이 4.0 kg, 운동만 수행한 그룹에서 1.3 kg이 감소하였다. 비록 체중의 감소는 골격근의 증가와 연관성이 있지만 유의성은 없었다. 체중의 변화는 지질 감소와 수분(세포 내 및 세포 간 수분) 감소와 연관이 있었다. 30대를 제외한 20대, 40대, 50대의 체중 감소는 있었지만 유의성은 없었다.

쪽잎의 생리활성 평가 (Evaluation of the Bioactivity of Polygonium tinctorium Leaf: Potential Clinical Uses)

  • 본 연구에서는 천연 염재, 약용 및 식용으로 사용되고 있는 쪽잎의 고부가가치화를 위해, 쪽잎의 열수 추출물 및 ethanol 추출물을 조제하고, 이의 항산화, 항균, 항당뇨, 항응고, 혈소판 응집저해 활성을 평가하였다. 쪽잎의 열수 추출효율은 27.6%로 ethanol 추출효율보다 약 2배 높았으며, 열수 추출물의 TP 함량은 51.2 mg/g으로 ethanol 추출물보다 높았으며, 특이하게 총당 함량은 ethanol 추출물(297.8 mg/g)에서 열수 추출물보다 1.2배 높게 나타났다, 반면 TF 및 환원당 함량은 열수 및 ethanol 추출물에서 유사하게 나타났다(p<0.05). 항균 활성 평가 결과, ethanol 추출물은 L. monocytogenes, S. epidermidis, S. aureus 및 B. subtilis의 그람 양성균에 대해서만 강력한 항균력을 나타내었으며, 열수 추출물은 항균 활성이 나타나지 않았다. 또한 ethanol 추출물이 열수 추출물보다 강력한 38.8%의 DPPH 소거능, 84.5%의 ABTS 소거능과 환원력($OD_{700}=0.357$)을 나타내었으나, nitrite 소거능은 열수 추출물이 효과적임을 확인하였다. 한편 쪽잎 ethanol 추출물은 5 mg/ml 농도에서 무첨가구에 비해 TT는 1.86배, PT는 1.79배, aPTT는 1.65배 연장시켜 매우 강력한 항응고 활성을 나타내었으나, 빠르게 혈소판 응집을 촉진하는 것을 확인하였다. 반면 열수 추출물은 강력한 혈소판 응집저해를 나타내었다. 현재까지 쪽잎의 항혈전 활성은 보고된 바 없으며, 이러한 쪽잎의 강력한 항산화 및 항혈전 활성은 쪽잎을 이용한 항혈전 소재 개발이 가능함을 제시하고 있다.

Combination Therapy with a PI3K/mTOR Dual Inhibitor and Chloroquine Enhances Synergistic Apoptotic Cell Death in Epstein-Barr Virus-Infected Gastric Cancer Cells

  • The phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin (PI3K/AKT/mTOR) signaling pathway is a promising target for gastric cancer (GC) treatment; however the efficacy of PI3K/mTOR dual inhibitors in GC has not yet been maximized. Additionally, the effect of autophagy regulation by PI3K/mTOR dual inhibitors has not been clearly elucidated in GC treatment. We aimed to show that our newly developed PI3K/mTOR dual inhibitor, CMG002, when combined with an autophagy inhibitor, chloroquine (CQ), potently induces effective cancer cell death in Epstein-Barr virus (EBV)-associated gastric cancer (EBVaGC) cells, where both the PI3K/AKT/mTOR and autophagy pathways play important roles in disease pathogenesis. EBV- and mock-infected AGS and NUGC3 GC cell lines were treated with CMG002 +/- CQ. PI3K/AKT/mTOR signaling pathway mediators, cellular apoptosis and autophagy markers were confirmed by Western blot assay. Cell viability was assessed using the Cell Counting Kit-8 (CCK-8) assay. CMG002 effectively blocked the PI3K/AKT/mTOR pathway by markedly decreasing phosphorylation of AKT and its downstream mediator S6. CMG002 induced G0/G1 cell cycle arrest and enhanced apoptotic cell death in AGS and NUGC3 cells, particularly EBV-infected cells compared with mock-infected cells, as confirmed by flow cytometric analyses and TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) assays. The combination of CMG002 plus CQ synergistically increased apoptotic cell death in EBV-infected GC cell lines when compared with CMG002 alone (P < 0.05). Our results suggest that the new PI3K/mTOR dual inhibitor, CMG002, when used in combination with the autophagy inhibitor, CQ, provides enhanced therapeutic efficacy against EBVaGC.

울산 해안의 최종간빙기 최온난기 추정 해성단구 (The estimation of the marine terrace of the Last Interglacial culmination stage(MIS 5e) in the Sanhari of Ulsan coast,southeastern Korea)

  • The formation age and depositional environment of the marine terrace I of the estimated paleoshoreline altitude of 18m in Sanhari of Ulsan coast, southeastern Korea were investigated on the basis of examination of lithofacies and stratigraphy of terrace deposits. Marine deposits of the terrace is composed of rounded boulders(70cm in diameter) and rounded pebbles(1.0cm in diameter) which overlay them. The above rounded boulders which lie on the paleo-shore platform are considered to have been formed by wave abrasion in the same period that the paleo-shore platform was developed. The rounded pebbles which lie on the rounded boulder layer are considered to have been deposited in gravel beach and berm environment, judging from the laminae developed in this layer. The paleo-shore platform and marine rounded gravel layer of the terrace are assumed to have been formed in the large transgression period of the Last Interglacial culmination stage(MIS 5e), judging from the comparision of the formation age of 125ka B.P. of Juckcheon terrace I in the adjacent Pohang coast which was dated by amino acid dating. The terrestrial deposit of this terrace was largely composed of angular and subangular gravel mixed with marine rounded pebble which has been carried away mainly from the deposit of previous marine terraces and redeposited in this terrace. The lowest peat layer of terrastrial deposit was considered to have been deposited during the period from the late MIS 5e which is the estimated finishing time of deposition of the above marine gravels to the early stage of following regression period(MIS 5d) in which the sea level was still high. The sediments of angular and subangular gravel deposit which lie on this peat layer were assumed to have been deposited during the period from the early stage of the first regression period(MIS 5d) of the Last Interglacial to the Last Glacial. The lower part of the angular gravel layer is composed of the deposits of the fluvial and colluvial sediments, whereas most of the upper and middle part of the layer is mainly composed of angular gravels of colluvial sediments formed in the cold environment.

중금속 흡착능 Leuconostoc mesenteroides CJNU 0705 균주를 활용한 피부 건강기능성 소재 개발 (Development of Skin Health Promoting Materials Using Leuconostoc mesenteroides)

  • Leu. mesenteroides CJNU 0705 균주는 모유에서 분리되었으며 16S rRNA 유전자 염기서열 분석과 자당(2%)이 포함된 tryptic soy agar plate상에서의 덱스트란 생성능을 통하여 동정하였다. Leu. mesenteroides CJNU 0705 균주는 미세먼지 속에 존재하는 피부 유해성 중금속인 납과 카드뮴에 대한 흡착능이 있으며 이러한 성질은 사균 상태에서도 확인되었다. 또한 Leu. mesenteroides CJNU 0705 균주는 여드름 원인균인 P. acnes 균주에 대하여 항균활성을 나타내었다. 마지막으로 피부 건강기능소재로 잘 알려진 녹차, 당근, 개똥쑥, 파슬리, 브로콜리 및 옥수수수염 추출물을 기본 배지(A)로 하여 2% 자당(B) 혹은 2% 자당과 3% 효모추출물(C)을 각각의 추출물에 첨가한 후 Leu. mesenteroides CJNU 0705 균주를 접종 및 배양하여 덱스트란 생산량을 확인한 결과 C 배지가 최적배지로 확인되었으며 덱스트란의 생산은 주사전자현미경 사진으로도 관찰되었다. 결론적으로 모유 유래의 Leu. mesenteroides CJNU 0705 균주나 이를 이용한 천연발효추출물은 피부 건강기능소재로 활용될 수 있을 것이다.

2011년 3월 일본 동북지방 태평양 연안 지진재해시의 쓰나미 재해와 지각변동 (Tsunami Disasters and Tectonic Movements along the Coastal Areas of Northeast Japan Derived from Mega-Earthquake in March, 2011)

  • 2011년 3월 일본 동북지방의 태평양 연안 해저에서 발생한 거대 지진(M 9.0)에 의한 쓰나미 재해와 지각변동에 대하여 조사하였다. 센다이 해안 평야의 경우 쓰나미에 의한 침수 거리는 현 해안으로부터 4.0km에 달하였다. 쓰나미 사질 퇴적물은 쓰나미 침수범위 중의 바다쪽 60~75%의 구간, 니질 퇴적물은 육지쪽 25~40%의 구간에 분포한다. 이 분포 비율은 센다이 해안평야의 충적층 속에 묻혀있는 과거 거대 쓰나미 때의 퇴적물 분포로부터 당시의 내륙쪽 침수한계를 추정하는데 활용될 수 있을 것으로 보인다. 센다이 해안평야 지역에서는 표고가 낮고 저평한 지형조건이, 그리고 리아스식 해안인 산리쿠 해안지역에서는 좁은 만입부에서의 쓰나미 증고 현상이 쓰나미 재해를 크게 증대시킨 요인으로 작용하였다. 이시노마키, 오가와, 오가치, 오나가와 등 동북 일본 해안지역의 여러 곳에서 이번의 대지진으로 인한 지각변동으로 0.5~1.0m정도의 지반침강이 일어났음을 확인하였다.

고령의 만성 C형 간염 환자에서 Daclatasvir와 Asunaprevir 병용 요법의 유효성 및 안전성 평가 (Efficacy and Safety of Daclatasvir and Asunaprevir Combination Therapy in Elderly Chronic Hepatitis C Patients)

  • Background : The prevalence of chronic hepatitis C virus (HCV) tends to be higher in the elderly. Pegylated interferon and ribavirin therapy (Peg-IFN/RBV) was recommended as the first-line treatment in the past decades, but this regimen showed unsatisfactory results in terms of safety and efficacy especially in elderly patients. Recently, it was demonstrated that dual therapy with daclatasvir and asunaprevir was well tolerated and led to high sustained virological response (SVR) rates, irrespective of age. We conducted a study to evaluate the efficacy and safety of daclatasvir plus asunaprevir by involving elderly patients aged above 65 years. Methods : We retrospectively analyzed clinical data from chronic hepatitis C virus (HCV) genotype 1b patients treated with daclatasvir plus asunaprevir from September 2015 to December 2016 at Seoul St. Mary's hospital. The patients were divided into two groups as elderly patients (older than 65 years) and non-elderly patients (younger than 65 years) and compared the efficacy and safety. Results : A total of 112 patients were treated with daclatasvir plus asunaprevir for chronic hepatitis C. Among them, 101 patients completed the whole treatment, and in 88 patients the amount of HCV RNA was measured after 12 weeks of treatment. There was no significant difference in SVR at 12 weeks between both the groups (p=0.68). Typically, 91.4%(32/35) of elderly patients and 94.3%(50/53) of non-elderly patients achieved SVR12. Common adverse events included elevation in transaminase level, headache, and gastrointestinal disorders. There was no statistical difference in the symptoms between the two groups. Conclusions : The combination therapy with daclatasvir plus asunaprevir exhibited similar rates of SVR12 in HCV elderly patients without leading to further adverse events compared to non-elderly patients. Therefore, it is proposed that daclatasvir plus asunaprevir therapy could be considered as an effective and safe treatment, even in patients aged over 65 years.
