Comparision of the Activity and Characteristics of Lactate Dehydrogenase Isolated from Different Parts of Soybean Seedling (발아초기의 콩 부위별 Lactate Dehydrogenase 활성변화 및 효소성질 비교)

  • The change of lactate dehydrogenase(LDH) activity and the possibility of the existence of LDH. isozyme were examined with different parts of soybean sprout. The enzyme activity was little changed in cotyledons throughout the early stagy of germination. However, hypocotyls and roots showed the continuous decline of the enzyme activity since the radicle emerged from seeds. It was found that LDH from hypocotyls. and roots was unstable as compared with LDH from cotyledons, even at low temperature. The enzyme from hypocotyls and roots was not purified with a good yield when the purification procedure developed for LDH from cotyledons. was employed. LDH from hypocotyls and roots has the Rm value of 0.29, and 0.25 from cotyledons. The apparent Km value for LDH from cotyledons was 0.45mM with sodium pyruvate, while crude homogenate of hypocotyls or roots showed biphasic phenomenon with two Km values 0.014 and 0.45mM. The results indicate the possibility that crude homogenate of hypocotyls or roots may contain a different LDH isozyme from the LDH of soybean reported previously.

Somatic Hybrids by Electro-Protoplast Fusion between N. tabacum and N. glutinosa (담배(N. tabacum)와 N. glutinosa 종간 원형질체 융합식물의 생성)

  • Protoplasts, isolated from leaf of N. tabacum NR-/SR+ and N. glottnosa were electrofused and divided with a plating efficiency of 30∼35% in AAPI 9M medium. Green callus lines were selected in protoplast-derived colonies on MSNO3 selection medium with 1.2mg/ml streptomycin sulfate on the basis of nitrate reductase proficiency and streptomycin resistance. Four putative hybrid plant lines regenerated from the green callus lines had intermediate morphology between that of parents with respect to floral shape, corolla length and ovate leaf blade. Zymograms of leaf peroxidase and esterase from these putative hybrid plant lines showed isozyme profiles derived from both parents and also, they exhibited additional and lost bands. Cytological analysis of two putative hybrid plant lines gave chromosome counts of 2n=66 in L22 and 2n=54 in L44 which were less than the expected number of N. tabacum(2n=48) and N. glutinosa(2n=24).

  • Several metabolic modulators on the generation of carbon monoxide (CO)from dichloromethane (DCM) was examined in adult female rats. It has been known that DCM is converted to CO by cytochrome P-450 or to carbon dioxide $(CO_2)$ by glutathione-dependent metabolic reaction. In rats treated with DCM (3 mmol/kg, ip) only, the carboxyhemoglobin (COHb) level reached a peak of approximately 10% 2 or 3 hr following the treatment. Disulfiram (300 mg/kg, ip) or allylsulfide (200 mg/kg, po), both known as a selective inhibitior for cytochrome P-450 2E1, blocked the increase in COHb concentratlons almost completely suggesting that the metabolic conversion of DCM to CO is mediated by the activity of this specific type of isozyme. YH439 (125 or 250 mg/kg, po), a potential hepatoprotective agent, decreased the COHb elevation as well indicating that this chemical is a potent inhibitor for 2E1. In rats treated with pyrazine (200 mg/kg, ip) 18 hr prior to DCM the peak COHb concentration was decreased by approximately 3 or 4%. However, pretreatment of rats with pyrazine either 24 or 48 hr before DCM increased the peak COHb concentration significantly compared to the rats treated with DCM only. The results in the present study strongly suggest that the generation of CO from DCM depends on the 2E1 activity and that the pharmacological and/or toxicological action of YH439 or pyrazine in animals or human is associated with its effect on this isozyme.

  • Interspecific hybrids between the ASpergillus spp., A. awamori, A. usamii and A. oryzae, were obtained by nuclear transfer technique. Nuclei isolated from an auxotrophic mutant strain were transferred into the protoplasts of a recipient strain of different species. The frequency of interspecific hybrid formation by nuclear transfer was $2*10^{-5}$ $-7*10^{-4}$ In contrast, no interspecific hybrid was isolated by protoplast fusion. Among the hybrids tested, 10 strains showed increased activity of some or all components of cellulases, xylanases and amylase up to more than two times. Isozyme pattern of the hybrids were analyzed by polyacrylamide gel electrophoresis and isoelectric focusing followed by activity staining, which showed that some of the hybrids have isozyme patterns unidentical to either of the two parents. By measuring the DNA contents and the sizes ofthe conidia, the karyotypes of the hybrids were estimated to be aneuploid near to haploid, diploid or triploid. It was concluded that the unclear transfer technique is much more efficient in the formation of interspecific hybrids than protoplast fusion and is very useful for the improvement of Aspergillus strains.

Genotypic and Geographical Variations of $\beta$amylase Isozyme in Soybean Land Races by Isoelectric Focusing (IEF)

  • The experiment was carried out to study the variations and geographical distribution of $\beta$-amylase isozyme by isoelectric focusing (IEF) within Korean, Chinese and Japanese soybean land races. In pH 3-10 gel of IEF, the amylase of soybean accessions was separated into low pI group isozymes (TEX>${$Sp_1$}^b$) and high pI group isozymes(${$Sp_1$}^a$). In pH4-6.5 gel, isoelectric points were at 5.07, 5.15, 5.25, 5.40, and 5.94, and h, j, and k bands also were found. The distribution of Sp$_1$$^{a}$ allele (high pI type) was 29.3% in soybean accessions from Korea, 10.1 % in those from China, and 6.9% in Japanese accessions. The percentage of ${$Sp_1$}^a$) allele was the highest in soybean accessions from Kyungsang province (35 %) in Korea, then central China (32 %) in China, and Honshu (10%) in Japan.

  • 우리나라 여러 딸기 재배지에서 전형적인 시들음 증상을 나타내는 이병식물에서 분리한 32개의 Fusarium oxysporum 균주들을 vegetative compatibility와 전기영동에 의한 동위효소상의 차이에 의해 분류하고 이들과 병원성과의 관계를 알아보았다. Nitrate nonutilizing(nit) mutant를 이용하여 vegetative compatibility group으로 분류해 본 결과 크게 A, B, C, D의 4개 그룹으로 나눌 수 있었으며 A그룹에는 15개 균주, B그룹에는 7개 균주, C와 D그룹에는 각각 2개 균주, 그리고 single VCG인 6개 균주가 존재하였다. 전기영동에 의한 esterase, catalase, acid phosphatase, leucin-aminopeptidase(LAP)의 동위효소상을 비교하여 본 결과 I, II, III, IV의 4개 그룹으로 나눌 수 있었으 I 그룹에는 18개 균주, II그룹에는 2개 균주, III그룹에는 6개 균주, IV그룹에는 6개 균주로 분류할 수 있었다. 또한, VCG의 A와 D그룹에 속하는 모든 균주들의 동위효소상의 I 그룹에 속하였으며 VCG B그룹의 7개 균주들 중에서 5개 균주가 동위효소상의 IV그룹, 7개 균주들 중에서 5개 균주가 동위효소상의 IV그룹, 나머지 2개 균주는 I 그룹과 III 그룹에 속하였으며 C그룹의 균주는 III과 IV 그룹에 속하였고, single VCGs들은 III 그룹에 4개 균주, II 그룹에 2개 균주가 속하므로 VCG와 동위효소상 간에는 밀접한 관계가 있음을 알 수 있었다. 4개 딸기 품종에 대해 병원성을 비교하여 본 결과 VCG A에서 선발된 2개 균주들은 보교조생에 대해 가장 병원성이 강하였으며 정보나 여홍에 대해서도 강한 병원성을 가지고 있었다. 반면에 B 그룹에서 선발된 2개 균주들은 4개 품종에 대해 병원성이 약하거나 거의 없었으며 C 그룹에서 선발된 1개 균주는 여홍에 대해 상대적으로 가장 강한 병원성을 가지고 있었다.

Development of screening systems for modulators on phospholipase-mediated signal transduction

  • Many agonists have been known to activate the hydrolysis of membrane phospholipids through the bindings with corresponding receptors on the various cells. Diacylglycerol and inositol 1,4,5-trisphosphate(IP3) generated by the action of phosphoinositide-specific phospholipase C (PI-PLC) are well known second messengers for the activation of protein kinase C and the mobilization of Ca2+ in many cells. Three types of PI-PLC isozyme (${\alpha}$,${\gamma}$, and $\delta$) and several subtrpes for each type have been identified from mammalian sources by purification of enzymes and cloning of their cDNAs. Each type PI-PLC isozyme is coupled to different receptors and mediators, for example, ${\beta}$-types are coupled to the seven-transmembrane-receptors via Gq family of G-proteins and ${\beta}$-types directly to the receptor tyrosine kinases. Specific modulators for the signaling pathway through each type of PI-PLC should be very useful as potential potential candidates for lend substances in developing novel drugs. To establish the sensitive and convenient screening systems for searching modulators on PI-PLC mediated signaling, two kinds of approaches have been tried. (1) Establishment of in vitro assay condition for each type of PI-PLC isozyme: Overexpression by using vaccinia virus and purification of each isozyme was carried out for the preparation of large amounts of enaymes. Optimum and sensitive assay condition for the measurements of PI-ELC activities were established. (2) Development of the cell lines in which each type of PI-PLC is permanently overexpressed: A fibroblast cell line (3T3${\gamma}$1-7) in which PI-PLC-${\gamma}$1 was overexpressed by using pZip-neo expression vector was developed and used for the measurement of PDGF-induced IP3 formation. The responses for IP3 formed in 3T3${\gamma}$1-7 cells by the treatment of PDGF is 8 times more sensitive than those in control cells. 3T3${\gamma}$l-7 cell is useful for the screening of the inhibitors on the PDGF-induced cellular responses from large number of samples in a small volume(50 ${\mu}$l) and short time(5-15 min). Using these systems, we screened hundreds of herb-extracts for the inhibition of PDGF-induced IP3 formation and selected several extracts that showed the inhibition as the candidates for isolation and characterization of active substances. The determination of the acting point of selected extracts or fractions in the PDGF signaling pathway has been analyzing.

  • ;ABSTRACT-The effects of gastric ulcer induced by restraint and water-immersion stress on gastric carcinogenesis in Wistar male rats treated with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) were examined. Rats of group 1 were 2iven stress for 8 hours before they were received MNNG (100 mg/l) in drinking water for 20 weeks. Rats of group 2 were received MNNG first for 2 weeks and then were given stress once a week from 3rd to 12th weeks, with simultaneous MNNG adminitration and followed by MNNG only until 20th weeks. Rats of group 3 were received MNNG only as a positive control and rats of group 4 were not treated with carcinogen. All groups were sacrificed in 20 weeks. Sections of the pyloric mucosa were stained by avidin-biotin peroxidase complex (ABC) immuno-histochemical method. PAPG (pepsinogen isozyme 1 altered pyloric gland), body weight change, gross lesions and histopathological changes were examined. The results obtained from these studies were summarized as follows: 1. The mean body weight gains of the rats fed with carcinogens (group 1, 2, 3) were significantly lower than that of group 4 (control group, without carcinogen. p<0.05). However, the differences of the mean body weight of rats treated with carcinogen were not significant. 2. Stress treatment (group 1 and 2) increased the appearance of the numbers of PAPG (Pepsinogen 1 Altered Pyloric Gland) induced by carcinogen significantly compared with that of group 3 (carcinogen only, p<0.01). 3. The incidence rate of mucosal hyperplasia in pylorus was significantly increased in group 2 compared with group 3 (p<0.05).0.05).

  • Inheritance of four isozymes, GOT, ACP, MDH and ADH, in Populus alba ${\times}$ P. glandulosa was investigated with starch gel electrophoresis. All four isozymes showed bands. For GOT, six or seven loci were postulated and observed segregation of hybrids at five variable loci was in agreement with expected segregation. Two loci were postulated in ACP; one locus showed no variation but the other locus showed variation. As one additional band was found in P. alba ${\times}$ P. alba (italy), hybrids from P. alba ${\times}$ P. alba ${\times}$ P. glandulosa showed more variation than hybrids from P. alba ${\times}$ P. glandulosa. One monomorphic locus and two variable loci were postulated in MDH. For ADH, both parents were turned out as homozygotes but for different alleles and thus all progenies were heterozygotes. ADH in hybrids seems to be a dieter enzyme as it showed on additional band between two parental bands. There were no variation in band betweens of four enzymes among the clones of P alba and P. glandulosa, respectively.

The Role of G protein in the Activation of Phospholipase C from Bovine Brain (소의 뇌조직 Phospholipase C의 활성화에 미치는 G-단백질의 역할)

  • The objective of the present study was to identify the characteristics of phospholipase C (PLC) isozymes purified from bovine brain and to investigate their interrelationship with G protein. The purified PLC isozymes ${\beta}$, ${\gamma}$ and ${\delta}$ were obtained and the characteristics of PLC activity on various concentrations of free $Ca^{2+}$ were observed. The activity of PLC was increased with increasing $Ca^{2+}$ concentration and the activity PLC ${\delta}$ was increased higher in the presence of phosphatidyl choline(PC) than in the abscence of PC. For vesicle formation as the structure of cell membrane, cholic acid and deoxycholic acid as detergent on phosphatidylinositol bisphosphate($PIP_2$) substrate containing PC were used, and then the activity of PLC isozymes were increased with increasing concentration of cholate, from 0.2% to 1% and were increased slightly in deoxycholate. In the $PIP_2$ containing phospholipid and glycolipid as brain extract, the activity of PLC isozymes were checked in 0.2%-1% cholic acid. The activities of PLC isoyzmes were continuously increased up to 1% cholic acid. The quantitation of PLC isozymes from several bovine organs by radioimmunoassay was made. Brain was the most sufficient organ in terms of amount of PLC ${\beta}$and ${\delta}$. A large amount of PLC ${\delta}$ was existed in adrenal gland. The binding capacity of GTPrS and G protein was observed and other observations of the binding effect of GTPrS-G protein and PLC monoclonal Ab-Protein A from tissue homogenate with PLC were made. From the observation the binding capacity was revealed the range of 0.11%-1.49%. The effects of each type of G protein on the percent activity of purified PLC isozymes were observed. From the observation, activities of isozymes were increased in $Go{\alpha}$ & Gmix, and the activities of PLC ${\beta}$ and ${\delta}$ were increased in $G{\beta}{\gamma}$ and $Gi{\alpha}$. Activities of PLC ${\beta}$ and ${\gamma}$ were decreased in $Gt{\alpha}$ but PLC ${\delta}$ increased.
