Hiwi, a human homologue of the Piwi family, plays an important role in stem cell self-renewal and is overexpressed in various human tumors. This study aimed to determine whether an RNA interference-based strategy to suppress Hiwi expression could inhibit tumor growth in a xenograft mouse model. A rare population of $SSC^{lo}\;Alde^{br}$ cells was isolated and identified as lung cancer stem cells in our previous study. Plasmids containing U6 promoter-driven shRNAs against Hiwi or control plasmids were successfully established. The xenograft tumor model was generated by subcutaneously inoculating with lung cancer stem cell $SSC^{lo}\;Alde^{br}$ cells. After the tumor size reached about 8 mm in diameter, shRNA plasmids were injected into the mice via the tail vein three times a week for two weeks, then xenograft tumor growth was assessed. In nude mice, intravenously delivery of Hiwi shRNA plasmids significantly inhibited tumor growth compared to treatment with control scrambled shRNA plasmids or the vehicle PBS. No mice died during the experiment and no adverse events were observed in mice administered the plasmids. Moreover, delivery of Hiwi shRNA plasmids resulted in a significant suppressed expression of Hiwi and ALDH-1 in xenograft tumor samples, based on immunohistochemical analysis. Thus, shRNA-mediated Hiwi gene silencing in lung cancer stem cells by an effective in vivo gene delivery strategy appeared to be an effective therapeutic approach for lung cancer, and may provide some useful clues for RNAi gene therapy in solid cancers.
The protective effects of the DWP-04 [DDB : selenium yeast: glutathione {31.1 : 6.8 : 62.1 (%, w/w)} against hepatotoxicity by carbon tetrachloride $(CCl_4)$ were studied in rats. The rats were intraperitoneally injected with $CCl_4$ (50% in com oil) at initial dose of 1 ml/kg followed by 0.5 ml/kg 3 times during 1 week. The DWP-04 (50, 100 or 200 mg/kg) or its vehicle was administered everyday before the start of $CCl_4$ injection for two weeks. $CCl_4$ induced hepatocelluar degeneration and necrosis, which led to a great increase in serum aminotransferase, alkaline phosphatase activity and serum lipid levels. It was found by biochemical analysis that $CCl_4$ treatment remarkably increased thiobarbituric acid reactive substances and physphatidylcholine hydroperoxide in hepatic tissues and induced antioxidant enzymes such as catalase and superoxide dismutase (SOD). Liver and serum lipids were significantly lower in rats fed on DWP-04 than in rats induced by $CCl_4$ only-treatment. These results suggested that the DWP-04 could be a promising candidate for the protection of liver injury based on the preventive effects against lipid peroxidation and serum biochemical parameters.

Effects of different photoperiod regimens on the cellular and humoral immunity in broiler chickens were studied(Exp 1). Total one hundred ninety two one-day-old commercial broiler chicks(Cobb$\times$Cobb) were raised between constant lighting(CL) and intermittent lighting (1h light: 3h darkness(IL; 1l; 3D) Body weight, feed intake and feed conversion were measured for seven week. Peripheral blood and splenic lymphocyte activities were tested at 3 and 5 wk of age by performing a mitogen cellproliferation assay with a polyclonal T-cell mitogen, concanavalin A (Con A), and B-cell mitogen, lipopolysaccharide (LPS). To investigate the effect of photoperiod on the humoral immunity, chicks were immunized with sheep red blood cell(SRBC) and iinactivated Newcastle disease virus(NDV) vaccine. Total immunoglobulin G(IgG) concentration was also determined. Diurnal change of melatonin was tested in sera. In experiment 2, 0.1ml melatonin were subcutaneously injected from three to five weeks old if immunomodulation effect of lighting regimen was due to the melatonin or not. Injections of melatonin were made at 0700h and the dosage was 10ng (M2), 100ng(M3), 1$\mu\textrm{g}$(M4) per bird daily, respectively. control were quivalent injections of vehicle(M1). Lymphocyte activities were tested and humoral immunities were examined at 5 weeks of age. Blood melatonin concentration was determined at 0h, 1, h, 2h, and 3h posterior to injection at five weeks old. It was higher in CL chicks than IL chickens during the subsequent period of 3 to 5 wk of age. However, weight gain of chicks raised IL were significantly higher at 6 wk of age than CL(P<0.05). Antibody response to NDV was not affected by both photoperiod regimens and melatonin injection, whereas anti-SRMB titer and IgG concentration were enhanced. Lymphocyte activity of chickens raised under IL was sighificantly higher than those of chickens raised under CL. Melatonin injection also increased lymphocyte activity. When peripheral blood lymphocytes were used, proliferation response to LPS and Con A were significantly increased in M2 and respectively. The results of this experiments suggest that IL improved host immune response and melatonin have immunomodulatory roles.
Recently, many researches regarding the natural products which alternate with chemical products have been done. Among them, boswellia is well known for effect on anti-oxidative effect and inflammation. The aim was the effect boswellia of formalin- induced orofacial and temporomandibular joint (TMJ) pain on experimental animals was investigated. Experiments were carried out using subcutaneous (SC) pain model and TMJ pain model that were induced by the injection of 5% formalin into the right vibrissa pad (SC, $50{\mu}L$) or TMJ ($30{\mu}L$) of rats, respectively. In both models, formalin (5%), formalin after distilled water (vehicle), formalin after boswellia extract (p.o., concentrations of 15, 30 mg/kg) (n=6). The number of scratching on the injected region was scored during the 9 successive periods of 5 min intervals following injection of formalin. Oral administration of boswellia (15, 30 mg / kg) reduced formalin-induced SC orofacial pain behavioral responses. SC orofacial pain behavioral responses was significantly reduced at 20~35 min. In the experimental group injected into temporomandibular joints, the pain response was significantly reduced by $276.2{\pm}8.20$ and $78.3{\pm}4.7$ after oral administration of boswellia (15, 30 mg / kg) at $398.3{\pm}24.8$ times. As a result of the passage of time, the oral administration of boswellia showed a significant effect of reducing the temporomandibular joint pain 30 minutes after the injection of formalin. This study confirmed that oral administration of boswellia modulated the pain behavior in both models. In conclusion, boswellia extract may be a potential therapeutic treatment for orofacial pain.
Gully pot is a part of urban darainage system to butter the runoff water fluxes from road to rivers and to minimize environmental pollution by prevending over the surrounding area. A series of studies have been carried out concerning the physicochemical characteristics of the sewage sludge sediments sttling down in such a gully pot in order to over to evaluate the contamination for heavy metals such as pb, Zn, Cu and Cd. The roadside soil and sewage sludge samples from gully pots were characterized by XRD analyses and sequential extraction : Zn 2595.7$\mu$g/g; Cd9.8$\mu$/g; Cu602.5$\mu$g/g; Pb260.0$\mu$g/g),because of a long-term accumulation of vehicle- and industrial-related pollutants. Mean Zn concentration in Yeouido (3873$\mu$g/g) and Junggu(3262$\mu$g/g)areas are 4-5 times higher than those in Dobonggu area, suggesting that Zn may be derived from automobile traffic (including the rubber of automobile tires). The mean concentrations of Cu and Cr are very significantly high in Junggu and Gurogu areas, possibly due to the industrial activities in these areas. The low Pb levels throughtotut the whole study areas in Seoul can be accounted for the use of unleaded gasoline since 1987. Sequential extraction experiments illustrate that a major part of Zn is bound to FII and FIII, representing about 88% of the total Zn concentration. Fraction IV, related to orgnic matter, is mostly significant for Cu accounting for 60% of the total Cu, and FII is next in importance. The main carriers of Pb are the fractions III, II and FIV, and in that order. The behavior of Cd is quite different from other elements (Zn, Cu, Pb), and most of the Cd is associated with FV. Changes in the physicochemical environments (such as acidification) may result in severe environmental pollution of surface water and rivers with respect to heavy metals (especially Zn and Cu).
Purpose: The purpose of this study is to explain the pivotal role of the travel forecasting process in urban transportation planning. This study emphasizes the use of travel forecasting models to anticipate future traffic. Method: This study examines the methodology used in urban travel demand modeling within transportation planning, specifically focusing on the Urban Transportation Modeling System (UTMS). UTMS is designed to predict various aspects of urban transportation, including quantities, temporal patterns, origin-destination pairs, modal preferences, and optimal routes in metropolitan areas. By analyzing UTMS and its operational framework, this research aims to enhance an understanding of contemporary urban travel demand modeling practices and their implications for transportation planning and urban mobility management. Result: The result of this study provides a nuanced understanding of travel dynamics, emphasizing the influence of variables such as average income, household size, and vehicle ownership on travel patterns. Furthermore, the attraction model highlights specific areas of significance, elucidating the role of retail locations, non-retail areas, and other locales in shaping the observed dynamics of transportation. Conclusion: The study methodically addressed urban travel dynamics in a four-ward area, employing a comprehensive modeling approach involving trip generation, attraction, distribution, modal split, and assignment. The findings, such as the prevalence of motorbikes as the primary mode of transportation and the impact of adjusted traffic patterns on reduced travel times, offer valuable insights for urban planners and policymakers in optimizing transportation networks. These insights can inform strategic decisions to enhance efficiency and sustainability in urban mobility planning.

The AlSi10Mg alloy has garnered significant attention for its application in laser powder bed fusion (L-PBF), due to its lightweight properties and good printability using L-PBF. However, the low production speed of the L-PBF process is the main bottleneck in the industrial commercialization of L-PBF AlSi10Mg alloy parts. Furthermore, while L-PBF AlSi10Mg alloy exhibits excellent mechanical properties, the properties are often over-specified compared to the target properties of parts traditionally fabricated by casting. To accelerate production speed in L-PBF, this study investigated the effects of process parameters on the build rate and mechanical properties of the AlSi10Mg alloy. Guidelines are proposed for high-speed additive manufacturing of the AlSi10Mg alloy for use in automotive parts. The results show a significant increase in the build rate, exceeding the conventional build rate by a factor of 3.6 times or more, while the L-PBF AlSi10Mg alloy met the specifications for automotive prototype parts. This strategy can be expected to offer significant cost advantages while maintaining acceptable mechanical properties of topology-optimized parts used in the automobile industry.
Soil-steel structures have been used for the underpass, or drainage systems in the road embankment. This type of structures sustain external load using the correlations with the steel wall and engineered backfill materials. Buried flexible conduits made of corrugated steel plates for the coastal road was tested under vehicle loading to investigate the effects of live load. Testing conduits was a circular structure with a diameter of 6.25m. Live-load tests were conducted on two sections, one of which an attempt was made to reinforce the soil cover with the two layers of geo-gird. Hoop fiber strains of corrugated plate, normal earth pressures exerted outside the structure, and deformations of structure were instrumented during the tests. This paper describes the measured static and dynamic load responses of structure. Wall thrust by vehicle loads increased mainly at the crown and shoulder part of the conduit. However additional bending moment by vehicle loads was neglectable. The effectiveness of geogrid-reinforced soil cover on reducing hoop thrust is also discussed based on the measurements in two sections of the structure. The maximum thrusts at the section with geogrid-reinforced soil cover was 85-92% of those with un-reinforced soil cover in the static load tests of the circular structure; this confirms the beneficial effect of soil cover reinforcement on reducing the hoop thrust. However, it was revealed that the two layers of geogrid had no effect on reducing the overburden pressure at the crown level of structure. The obtained values of DLA decrease approximately in proportion to the increase in soil cover from 0.9m to 1.5m. These values are about 1.2-1.4 times higher than those specified in CHBDC.

Three types of traffic signal systems are two-color signal systems that flash red and green and are mainly used on crosswalks, next, three-color equalization systems mainly used at T-shaped intersections with red and yellow lights and a green arrow, and third, four-color intersections that generally have red, yellow and green colored lights and a green arrow. In what is known as the "dilemma zone" area, a driver collects information that influences his/her decision whether to stop, speed, tail, interrupt, or violate a traffic light, depending on the intersection width, vehicle speed, cognitive response time and reference yellow signal time. This study examined the impact of changes in the length of the dilemma zone areas based on changes in yellow signal times, the speed of the intersection passages, and signal lamps. Downward adjustments of 50km/h and 60km/h affected yellow signal time. The yellow signal time increased by 0.1 to 2.3[s] due to this effect and the dilemma zone area increased by 1.22 to 26[m]. The driver of the dilemma zone could quickly decide to reduce the time remaining of the straight (3color, 4color) green signal to reduce the potential of a traffic accident at the intersection traffic. Safe entry of red (LED palm) and left-turn signals for entering flashed at the intersection and operated at midnight.
Air pollution levels of gases and aerosol particles inside the Jeongneung and Hongjimun tunnels of the Naebu express way in Seoul were investigated through on-road measurement using a mobile emission laboratory (MEL) on February 8, 2011. The concentrations of $NO_x$, $CO_2$, number concentration of particles ranging 21-560 nm, and surface area of particles deposited on a human lung almost linearly increased with increasing distance from the tunnel entrance, and decreased rapidly before the tunnel exit. This trend was observed regardless of tunnel length and driving directions, which thought to be caused by semi-transverse ventilation facilities of the tunnels. The concentration increments per 1-m distance for $NO_x$, $CO_2$, deposited particle surface area, and number of particles ranging 21-560 nm were 0.61~0.80 ppb, 0.16~0.21 ppm, $0.20{\sim}0.29{\mu}m^2/cm^3$, and 117~192 particles/$cm^3$, respectively. Average pollution levels inside the two tunnels for $CO_2$, deposited particle surface area, and number of particles >5.6 nm ranged 681~748 ppm, $246{\sim}381{\mu}m^2/cm^3$, and $2.4{\sim}6.7{\times}10^5$ particles/$cm^3$, respectively. In case of $NO_x$, the maximum concentration exceeded 1 ppm. These pollution levels inside the tunnels are much higher than those at urban background sites. This result can be utilized as basic data to evaluate the effectiveness of present ventilation system for reducing the pollution level caused by vehicles inside the tunnels.
