  • Ion effects on $11{\alpha}-hydroxylation$ of progesterone and $5{\alpha}-reduction\;of\;11{\alpha}-hydroxyprogesterone$ by Rhizopus nigricans were investigated. Metal ions such as $Cu^{2+},\;Cd^{2+},\;Co^{2+},\;Mn^{2+},\;Zn^{2+},\;Fe^{2+},\;Mg^{2+},\;Fe^{3+}\;and\;Na^+$ reduced the $11{\alpha}-hydroxylation$ activity, while $K^+$ stimulated the same reaction. Enzyme activity for the $5{\alpha}-reduction$ of $11{\alpha}-hydroxyprogesterone$ was increased in the presence of $Fe^{2+},\;Mn^{2+},\;Mg^{2+},\;Co^{2+},\;Zn^{2+},\;Fe^{3+},\;K^+\;and\;Na^+$, whereas it was decreased in the presence of $Cd^{2+}\;and\;Cu^{2+}$. Potassium ion of $10^{-3}\;M\;$ of concentration was found to be effective for the promotion of $11{\alpha}-hydroxylation$. On the other hand, cadmium ion of $10^{-4}\;M$ was proved to suppress the $5{\alpha}-reduction$ reaction. Progesterone is reported to be transformed into $11{\alpha}-hydroxyprogesterone$ which, in turn, is converted further into $11{\alpha}-hydroxy-allopregnane-3$, 20-dione by R. nigricans. From this point of view, the highest yield of $11{\alpha}-hydroxyprogesterone$ could be obtained when potassium ion of $10^{-3}\;M$ was given initially followed by addition of cadmium ion of $10^{-4}\;M$ to limit conversion of 11{\alpha}-hydroxyprogesterone into $11{\alpha}-hydroxy-allopregnane-\;3$, 20-dione.

  • To decompose carbon dioxide, magnetite was synthesized with 0.2M-FeSO4$.$7H2O and 0.5 M-NaOH by coprecipitation. The deoxidized magnetite was prepared from the magnetite by hydrogen reduction for 1, 1.5, 2 hr. The degree of hydrogen reduction and the decomposition rate of carbon dioxide were investigated with hydrogen reduction time. The crystal structure of the magnetite was identified spinel structute by the X-ray powder diffractions. After magnetite was reduced by hydrogen, magnetite reduced by hydrogen become new phae(${\alpha}$-Fe2O3, ${\alpha}$-Fe) and spinel type simultaneously. After decomposing of carbon dioxide at 350$^{\circ}C$, new phse(${\alpha}$-Fe2O3, ${\alpha}$-Fe) were removed and the spinel type only existed. The specific surface area of the synthesized magnetite was 46.69㎡/g. With the increase of the hydrogen reduction time, the grain size, the hydrogen reduction degree and the decomposition rate of carbon dioxide was increased.

  • The asymmetric reduction of representative prochiral ${\alpha},{\beta}$-acetylenic ketones with a new chiral borohydride reducing agent, potassium 9-0-(1,2: 5,6-Di-O-isopropylidene-${\alpha}$ -D-glucofuranosyl)-9-boratabicyclo[3.3.1]nonane, 1, in THF at $-78^{\circ}C$ was studied. Structurally different acetylenic ketones such as internal ${\alpha},{\beta}$-acetylenic ketones $RC {\equiv} CCOCH_3$ and terminal ${\alpha},{\beta}$-acetylenic ketones $HC {\equiv} CCOR$ were chosen. Thus, the reduction of internal ${\alpha},{\beta}$-acetylenic ketones yields the corresponding propargyl alcohols, such as 67% ee for 3-hexyn-2-one, 75% ee for 5-methyl-3-hexyn-2-one, 86% ee for 5,5-dimethyl-3-hexyn-2-one, 74% ee for 3-nonyn-2-one and 61% ee for 4-phenyl-3-butyn-2-one. Terminal ${\alpha},{\beta}$-acetylenic ketones, such as 3-butyn-2-one, 1-pentyn-3-one, 4-methyl-1-pentyn-3-one and 1-octyn-3-one, are reduced to the corresponding alcohols with 59% ee, 17% ee, 44% ee and 12% ee of optical induction respectively. With one exception (4-methyl-1-pentyn-3-one), all propargyl alcohols obtained are enriched in R-enantiomers.

  • Five subcellular fractions were obtained by successive centrifugation from the liver of rats within 6 hours of life and characterized by comparing marker compound or marker enzyms. After incubating $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one with the each fraction, the steroids were analyzed by TLC, GLC and GC-MS. A $6{\alpha}$-hydroxylase which hydroxylizes the tetra-hydrogenated compound of progesterone, $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one, was localized in the crude plasma membrane fraction, but not in the microsome fraction. The maximum 6α-hydroxylation was observed at pH 7.0. While this 6α-steroid hydroxylase was not able to hydroxlyze the progesterone, the $3{\alpha}$-isomer was hydroxylized at the $6{\alpha}$-position.

  • Rate of 11.alpha.-hydroxylation of progesterone with Rhizopus nigricans was accelerated by glucose. Glucose seemed to play an important role in the formation of cofactor because its effects on the reaction were almost same as those of electron acceptors such as NADPH and $NaIO_{4}$. Rate of glucose consumption appeared to increase in proportion as the rate of hydroxylation reaction, which enhanced with increase in the glucose concentration to level off at 0.5 g/l for mycelia and at 20 g/l for spores. However, for mycelia immobilized in polyacrylamide gel, externally added glucose did not affect the reaction rate at all because of the glucose accumulated in the gel during the cultivation period. 5.alpha.-Reduction of 11.alpha.-hydroxyprogesternoe required much higher concentration of glucose than 11.alpha.-hydroxylation of progesternoe so that high yield of 11.alpha.-hydroxyprogesternoe can be obtained by repressing the activity of 5.alpha.-reductase at low concentration of glucose.

  • In this study, Matsuda formula used to evaluate the loads acting on the pillar was investigated and load reduction factor(${\alpha}$) was evaluated by numerical analysis to better apply for the design. From the results, normal stress was concentrated to one side due to excavation of preceding tunnel after construction of pillar. And 86.5% of maximum normal stress was revealed partly unequally when the ground was poor. By numerical analysis, $14{\sim}83%$ of total loads calculated by Matsuda formula decreased and then, from these results, load reduction factor(${\alpha}$) was estimated. From now on, stability and economic aspects could be guaranteed by applying the load reduction factor(${\alpha}$).

  • Thirteen analogs of tridecapeptide ${\alpha}$-factor (WHWLQLKPGQPMY) of Saccharomyces cerevisiae with C- or N-terminal Trp extension and isosteric replacement by Aib at position 8 and 11, Trp at position 13, D-Ala at position 9, and Orn and Glu at position 6 were synthesized and assayed for their biological activity. Receptor binding assay was carried out using our newly developed spectrophotometric method with detector peptide 14. C- or N-terminal extended analogs, ${\alpha}$-factor-$[Trp]_n$ (n =1-5) 1-5 and $[N-Trp]_1$-${\alpha}$-factor 6, were all less active than native ${\alpha}$-factor and gradual decreases in both activity and receptor affinity were observed with greater Trp extension. Trp-substituted analog at position 13, $[Trp^{13}]{\alpha}$-factor 7, exhibited about 2-fold reductions in both activity and receptor affinity. Aib-substituted analogs, $[Aib^8]{\alpha}$-factor 8 and $[Aib^{11}]{\alpha}$-factor 9, showed 5- to 10-fold reduction in activity as well as 3-fold reduction in receptor affinity compared to native ${\alpha}$-factor. $[Orn^6]{\alpha}$-factor 10 demonstrated strong potency with a 7.0-fold increase in halo activity as well as 1.8-fold increase in receptor affinity compared to native ${\alpha}$-factor. For two double substituted analogs, [$Glu^6,{\small{D}}-Ala^9$]${\alpha}$-factor 12 showed the slightly decreased potency in halo activity compared to analog 10, whereas [$Orn^6,{\small{D}}-Ala^9$]${\alpha}$-factor 11 exhibited 15-fold higher halo activity as well as nearly 3-fold higher receptor affinity compared to native ${\alpha}$-factor.

  • Rhizopus nigricans produces 11.alpha.-hydroxyprogesternoe with a unidentified byproduct, which is hardly separated. Results of chromatography, IR and NMR spectroscopy identified the byproduct to be 11.alpha.-hydroxy-allopregnane-3,20-dione. R. nigricans was found to transform progesternoe into a monoform intermediate, 11.alpha.-hydroxyprogesterone, from which 11.alpha.-hydroxy-allopregnane-3,20-dione and 6.betha., 11.alpha. - dihydroxyprogesterone were formed respectively by 5.alpha.-reduction and 6.betha.-hydroxylation.

  • The change of thermal expansion and mechanical behaviors by cold working has been investigated in Fe-29%Ni-17%Co low thermal expansion Kovar alloy. Fe-29%Ni-17%Co alloy was cold rolled gradually and prepared to plates having reduction ratio of 0%, 20%, 40%, 60%, and 80%. Annealing effect on the properties was also studied. Thermal expansion was measured from $25^{\circ}C$ to $600^{\circ}C$ with a heating rate of $5^{\circ}C$/min by using vacuum differential dilatometer. It was found that thermal expansion coefficient ($\alpha_{30{\sim}400}$) slightly decreased (reduction ration of 20%) and then remarkably increased (above reduction ration of 40%) with increasing reduction ratio of cold rolling. Thermal expansion coefficient ($\alpha_{30{\sim}400}$) was sharply decreased after annealing heat-treatment. Yield and tensile strengths were continuously increased and elongation was decreased by cold roiling. Microstructural observation and X-ray diffraction analysis results showed that the $\alpha$ phase significantly increased as the reduction ratio increased. The slight decrease of thermal expansion coefficient bellow reduction ration of 20% could be explained by the destroying short-range ordering and the decreasing of grain size. The significant increase of thermal expansion coefficient with cold rolling mainly attributed to the appearance of $\alpha$ phase. The correlation between the microstructural cause and invar phenomena for the low thermal expansion behavior was also discussed.
