  • Manuka oil sometime named New Zealand's tea tree oil is soluble in oil and come from nature. The $\alpha$-pinene extracted from Manuka oil and R-limonene which is one of the component of extracted Citrex from Grapefruit were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion and broth dilution methods were used to measure the antimicrobial activity Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus is similar when the concentration of Manuka oil and $\alpha$-pinene is 10${mu}ell$. However, Antimicrobial activity of Manuka oil for EscherEchta coli, Staphylococcus aureus is better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene is low. Antimicrobial activity of Citrex is superior to that of R-limonene. The proper ratio of Maunka oil and Citrex can Improve the antimicrobial activity. The proper ratio obtained from studies was 75% of Maunka oil and 25% Citrex for Escherichia coli, 25% of Maunka oil and 75% Citrex for Staphylococcus aureus.

  • A study for the evaluation of selected filter media that need further characterization particularily in the context of alpha measurements of radon progeny has been carried out by investigating physical characteristics and using the alpha spectroscopy. Physical characteristics was investigated by electrostatic charging and mechanical strength of filters, and then pressure drop before and after sampling was tested. Alpha spectroscopy was used to analyze the energy spectra from the deposition of radon progeny into filters. The results of the assessment showed that the newer filter types do not have a great advantage over the 'old standard' Millipore type AA. But Metricel DM-800 is recommended for those situations where electrostatic charging is a problem. Also this method will be used more effective for the evaluation of new developing filters in future.

  • Hexagonal M-type barium ferrite $BaFe_{12}O_{19}$ was prepared by a direct solid state reaction between $BaCO_3$ and ${\Alpha}-Fe_2O_3$ powders at temperatures ranging from 800 to 1000 $^{\circ}C$ in a nitrogen or air environment. The effects of NaCl on phase transformation of mixture of $BaCO_3$ and ${\Alpha}-Fe_2O_3$ to $BaFe_{12}O_{19}$ were studied using X-ray diffraction and M$\"{o}$ssbauer spectroscopy. Regardless of the environment for heat treatment or the added NaCl, two phases, BaFe and ${\Alpha}-Fe_2O_3$, coexist for samples heat treated at 800 $^{\circ}C$, but a single phase was observed from samples heat treated at 100$0^{\circ}C$ in both air and nitrogen environments. It was found that the addition of NaCl reduced the amount of $a-Fe_2O_3$ phase.hase.

  • The concentrations of astaxanthin and $\alpha$-tocopherol were measured from the muscle of the rainbow trout(Oncorhynchus mykiss) that had been fed the red yeast(Phaffia rhodozyma) containing 0.2% astaxanthin for 7, 14 and 21 days. The effect of the astaxanthin supplementation for 21 days on peroxidation of liver and muscle lipids of the rainbow trouts was examined. The astaxanthin was found to be accumulated in the rainbow trout muscle when fed for 7 days with astaxanthin supplementation(80mg/kg diet) in the form of the red yeast and the content did not increase further when fed longer up to 21 days. Seven days supplementation of astaxanthin raised the rainbow trout muscle content of the astaxanthin to 17.3$\mu\textrm{g}$/g tissue from 11.8$\mu\textrm{g}$/g tissue in mature control group. Although the hepatic TBARS level was found to be significantly decreased, the astaxanthin supplementation did not alter the $\alpha$-tocopherol and TBARS contents of the rainbow trout muscle.

PHA-Induced Peripheral Blood Cytogenetics and Molecular Anslysis : a Valid Diagnostic and Follow-up Modality For Acute Primyelocytic Leukemia Patients Treated With ATRA and/or Arsenic Tri-oxide

  • Background: Acute promyelocytic leukemia (APML) is characterized by the reciprocal translocation t(15;17) (p22;p12) resulting in the PML-$RAR{\alpha}$ fusion gene. A dual diagnostic and follow up approach was applied including cytogenetic demonstration of the t(15;17) translocation and detection dg PML-$RAR{\alpha}$ chimeric transcripts by molecular means. Purpose: Conventional cytogenetics involving bone marrow is beset with high probability of poor metaphase index and was substituted with phytohemagglutinin (PHA)-induced peripheral blood culture based cytogenetic analysis as a diagnostic & follow up modality in APML patients of Kashmir (North India). Both qualitative (RT-PCR) and quantitative (Q-PCR) tests were simultaneously carried out to authenticte the modified cytogenetics. Materials and Method: Patient samples were subjected to the said techniques to establish their baseline as well as follow-up status. Results: Initial cytogenetics revealed 30 patients (81%) Positive for t(15;17) whereas 7 (19%) had either cryptic translocation or were negative for t(15;17). Two cases had chromosome 16q deletion and no hallmark translocation t(15;17). Q-PCR status for PML-$RAR{\alpha}$ was found to be positive for all patients. All the APML patients were reassessed at the end of consolidation phase and during maintenance phase of chemotherapy where 6 patients had molecular relapse, wherein 4 also demonstrated cytogenetic relapse. Conclusions: It was found that PHA-induced peripheral blood cytogenetics along with molecular analysis could prove a reliable modality in the diagnosis and assessment of follow up response of APML patients.

Effect of Aging Treatment on the Microstructure and Mechanical Properties of Mg-6Al-xZn (x : 1.5, 2.5) Alloys Fabricated by Squeeze Casting (용탕단조된 Mg-6Al-xZn (x=1.5, 2.5) 합금(合金)의 미세조직 및 기계적 성질에 미치는 시효의 영향)

  • This study has investigated the effect of aging treatment on the microstructure and mechanical properties of Mg-6Al-xZn(x = 1.5, 2.5) alloys fabricated by the squeeze casting process. The microstructures of as-squeeze cast were composed of pro-eutectic ${\alpha}$, super saturated ${\alpha}$ and ${\beta}(Mg_{17}Al_{12})$ compound. Aged at both $200^{\circ}C$ and $240^{\circ}C$, Mg-6Al-xZn alloys showed the peak hardness due to the formation of ${\beta}(Mg_{17}Al_{12})$ precipitates. The discontinuous precipitates of the lamella type are predominant at $200^{\circ}C$ aging treatment, while the finely dispersed continuous precipitates were dominant at $240^{\circ}C$ aging treatment. Mg-6Al-xZn alloys fabricated by the squeeze casting process had the better combination of tensile strength and elongation compared to the conventionally cast alloys. As zinc contents increased, the tensile strength was increased by the solid solution strengthening effect of zinc.

  • Guppy fry were treated for the first 40 days of life with 0, 20, 40, 60 & 100$\mu\textrm{g}$ of estradiol per gram of food in order to change the sex of normal males to functional females(genetic male). The present investigation deals with the effects of steroid hormones, such as $\beta$-estradiol and testosterone, on the sex differentiation in guppy and tilapia. The results obtained were summarized as follows. 1. In B (20$\mu\textrm{g}$/g diet) group 17$\beta$-estradiol-treated, 67.8% of male offsprings were produced. 2. In D (60$\mu\textrm{g}$/g diet) group 17$\beta$-treated, 67% of female offsprings were produced. 3. B, D groups of genetic male brooders had significantly different effects (P<0.01) upon sex ratios of their progeny. 4. This strongly indicates that sex direction has been achieved and that the male is the heterogametic sex. 5. The group that produced the highest percentage of male offspring(male percentage of observed number to expected number was 91%) contained only full-sibling male brooders to the sex-reversed female brooders. 6. After 7 months following treatment, the sex-reversed males had ovarian portion in the anterior region and a testicular portion in the posterior region of the same intersexual gonad, respectively. 7. At 7 months after treatment, the ovareis revealed a complete arrest of the ovarian formation, and appearances of spermatogenetic cell cysts among surviving auxocytes. 8. In most of sex-reversed fish, anterior portion of test is was devoid of sperm ducts including the seminal vesicle and vas deferens. 9. The male transferrin showed two strong bands, while the female transferrin showed a single weak band. 10. One of the two bands of male transferrin showed the same mobility with band of female transferrin.

Influence of 2-bromo-α-ergocryptine on Plasma Prolactin, Oestradiol-17β and Progesterone Levels in Domestic Hen

  • This study investigated the effect of 2-bromo-$\alpha$-ergocryptine (anti prolactin agent) on plasma levels of prolactin, oestradiol-17$\beta$ and progesterone in domestic hen during the active period of lay. Fifty healthy female White Leghorn birds were administered with anti prolactin agent (2-bromo-$\alpha$-ergocryptine, Sigma-USA., methane sulphonate salt, $C_{32}H_{40}BrN_5O_5.CH_4SO_3$) subcutaneously @100$\mu$g/kg body weight at weekly intervals from 17th to 36th week of age. Another group of fifty birds as controls were given placebo in place of bromocriptine. The level of prolactin remained lower in treated birds than in the control birds from 19 to 36 weeks of age. Level of prolactin even in the control group was found to decrease during the peak production period. Oestradiol-$17{\beta}$ and progesterone concentration in treated birds were significantly (p<0.01) higher than the controls during the treatment. Egg production, is positively correlated with oestradiol-$17{\beta}$ (r=0.02; r=0.67) and progesterone (r=0.49; r=0.90) in control and treated groups respectively where as prolactin level is positively correlated with egg production in the control birds (r=0.07). Prolactin levels were negatively correlated with egg production (r=-0.55) in treated birds; and oestradiol-$17{\beta}$ (r =-0.71; r=-0.53) and progesterone (r=-0.22; r=-0.27) respectively in control and treated groups. The total number of pause days during the treatment period decreased significantly (p<0.01) in the treated group compared to the control group. The reduction in pause days in treated group resulted in 1.76% increase in egg production over that in control group. The increase in egg laying days and the total egg production were found to be significant (p<0.01). These results indicate that a lower level of prolactin in circulatory blood enhances egg production in the domestic hen.

Analytical Methods for the Isolation of Dehydrotomatine and ${\alpha}$-Tomatine in Tomato Fruits by Use of Alumina Column Chromatography and High-Performance Liquid Chromatography (Alumina Column Chromatography와 HPLC에 의한 토마토의 Dehydrotomatine 및 ${\alpha}$-Tomatine 단리방법 연구)

  • Tomato fruits(Lycoperisicon esculentum) synthesize the glycoalkaloids dehydrotomatine and ${\alpha}$-tomatine, possibly as defense against bacteria, fungi and insects. We developed a new effective method to prepare and purify dehydrotomatine and ${\alpha}$-tomatine that exists in tomato fruits using alumina column chromatography and high performance liquid chromatography (HPLC). The tomato glycoalkaloids(TGA) in tomato was extracted with 2% acetic acid, and then precipitated with ammonium hydroxide(pH=10.5). The dry precipitate substance was applied on alumina column, and then fractionated with water saturated n-butylalcohol. The TGA(Fr. No. 26~36) were collected and dried under reduced pressure. The TGA was performed on a reverse phase HPLC(Inertsil ODS-2, $5\;{\mu}m$), eluted with acetonitrile/20mM $KH_2PO_4$(24:76, v/v) at 208 nm. Two peaks were detected on HPLC, and individual peak was collected by repeating HPLC. Furthermore, to confirm the identity dehydrotomatine and ${\alpha}$-tomatine, each peak isolated was hydrolyzed with 1N HCl into sugar and aglycone tomatidine. The sugars were converted to trimethylsilyl ester derivatives. The nature and molar ratios of sugars were identified by gas-liquid chromatography(GLC) and the aglycone by high-performance liquid chromatography(HPLC). The first peak (Rt=17.5 min) eluted from HPLC was identified as dehydrotomatine, and second peak(Rt=21.0 min) was as ${\alpha}$-tomatine. This technique has been used effectively to prepare and isolate dehydrotomatine and ${\alpha}$-tomatine from tomato fruits.

Modulation of the Tendency Towards Inclusion Body Formation of Recombinant Protein by the Addition of Glucose in the araBAD Promoter System of Escherichia coli

  • We attempted to modulate the overall protein expression rate through the addition of a repressor against the araBAD promoter system of Escherichia coli, in which glucose was used as a repressor. Therefore, 0.5% L-arabinose was initially contained as an inducer in culture medium, and either 2% glucose or 2% glycerol was used as a carbon source, and it was found that the expression of recombinant interferon-${\alpha}$ could be observed at the beginning of the batch culture when glycerol was used as a carbon source. However, when glucose was used, the initiation of recombinant interferon-${\alpha}$ expression was delayed compared with that when glycerol was used. Furthermore, when the addition of 0.5% glucose was carried out once or twice after 0.5% L-arabinose induction during DO-stat fed-batch culture, the distributions of soluble and insoluble recombinant interferon-${\alpha}$ were modulated. When glucose was not added after the induction of L-arabinose, all of the expressed recombinant interferon-${\alpha}$ formed an inclusion body during the later half of culturing. However, when glucose was added after induction, the expressed recombinant interferon-${\alpha}$ did not all form an inclusion body, and about half of the total recombinant interferon-${\alpha}$ was expressed in a soluble form. It was deduced that the addition of glucose after the induction of L-arabinose might lower the cAMP level, and thus, CAP (catabolite activator protein) might not be activated. The transcription rate of recombinant interferon-${\alpha}$ in the araBAD promoter system might be delayed by the partial repression. This inhibition of the transcription rate probably resulted in more soluble interferon-${\alpha}$ expression caused by the reduction of the protein synthesis rate.
