Degradation of Raffinose Oligosaccharides in Soymilk by Immobilized ${\alpha}$-Galactosidase of Aspergillus oryzae

  • [ ${\alpha}$ ]-Galactosidase was immobilized in a mixture of k-carrageenan and locust bean gum. The properties of the free and immobilized enzyme were then determined. The optimum pH for both the soluble and immobilized enzyme was 4.8. The optimum temperature for the soluble enzymes was $50^{\circ}C$, whereas that for the immobilized enzyme was $55^{\circ}C$. The immobilized enzyme was used in batch, repeated batch, and continuous modes to degrade the raffinose-family sugars present in soymilk. Two hours of incubation with the free and immobilized ${\alpha}$-galactosidases resulted in an 80% and 68% reduction in the raffinose oligo saccharides in the soymilk, respectively. In the repeated batch, a 73% reduction was obtained in the fourth cycle. A fluidized bed reactor was also designed to treat soymilk continuously and the performance of the immobilized ${\alpha}$-galactosidase tested at different flow rates, resulting in a 90% reduction of raffinose-family oligosaccharides in the soymilk at a flow rate 40 ml/h. Therefore, the present study demonstrated that immobilized ${\alpha}$-galactosidase in a continuous mode is efficient for reducing the oligosaccharides present in soymilk, which may be of considerable interest for industrial application.

Analysis of Vitamin E in Agricultural Processed Foods in Korea (국내 농산가공식품의 비타민 E 함량 분석)

  • Accurate food composition data are essential for calculation of nutrient intake of a population based on its consumption statistics. In the Korean food composition database, there is a lack of reliable analytical data for tocopherols and tocotrienols. Therefore, this study was conducted to provide information on contents on vitamin E in agricultural processed foods in Korea. Tocopherols and tocotrienols were determined by the saponification extraction method followed by high performance liquid chromatography. Analytical method validation parameters were calculated to ensure the method's validity. Samples were obtained in the years of 2013 and 2014 from the Rural Development Administration. The samples included 34 grains and grain products, 14 snacks, 25 fruits, 5 oils, and 11 sources and spices. All vitamin E isomers were quantitated, and the results were expressed as ${\alpha}$-tocopherol equivalent (${\alpha}-TE$). ${\alpha}-TE$ values of cereal and cereal products, snacks, fruits, oils and sauces and spices ranged from 0.03 to 17.53, 1.01 to 12.84, 0.01 to 1.52, 1.09 to 8.15, and 0.01 to $27.53{\alpha}-TE/100g$, respectively. Accuracy was close to 100% (n=3). Repeatability and reproducibility were 2.04% and 4.69%, respectively. Our study provides reliable data on the tocopherol and tocotrienol contents of agricultural and processed foods in Korea.

Quercetin Inhibits ${\alpha}3{\beta}4$ Nicotinic Acetylcholine Receptor-Mediated Ion Currents Expressed in Xenopus Oocytes

  • Quercetin mainly exists in the skin of colored fruits and vegetables as one of flavonoids. Recent studies show that quercetin, like other flavonoids, has diverse pharmacological actions. However, relatively little is known about quercetin effects in the regulations of ligand-gated ion channels. In the previous reports, we have shown that quercetin regulates subsets of homomeric ligand-gated ion channels such as glycine, 5-$HT_{3A}$ and ${\alpha}7$ nicotinic acetylcholine receptors. In the present study, we examined quercetin effects on heteromeric neuronal ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor channel activity expressed in Xenopus oocytes after injection of cRNA encoding bovine neuronal ${\alpha}3$ and ${\beta}4$ subunits. Treatment with acetylcholine elicited an inward peak current ($I_{ACh}$) in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor. Co-treatment with quercetin and acetylcholine inhibited $I_{ACh}$ in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptors. The inhibition of $I_{ACh}$ by quercetin was reversible and concentration-dependent. The half-inhibitory concentration ($IC_{50}$) of quercetin was $14.9{\pm}0.8\;{\mu}M$ in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor. The inhibition of $I_{ACh}$ by quercetin was voltage-independent and non-competitive. These results indicate that quercetin might regulate ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor and this regulation might be one of the pharmacological actions of quercetin in nervous systems.

Genotypic Difference in Tocopherol and Tocotrienol Contents of Rice Bran (벼 품종별 미강의 Tocopherol과 Tocotrienol 함량 평가)

  • The rice bran has been known to contain tocopherols and tocotrienols carrying antioxidant and cholesterol-lowing activities. The content of 8 isomers of vitamin E : $\alpha\textrm{-}$, $\beta\textrm{-}$, $\gamma\textrm{-}$, $\delta\textrm{-}$tocoperols (T) and tocotrienols ($\textrm{T}_3$) were extracted from 18 major rice varieties and quantified with an HPLC. Tested varieties exhibited T, $\textrm{T}_3$ and total vitamin E ($\textrm{TT}_3$) contents ranging 9.1-14.8, 22.4-37.1, 34.9-46.5 mg/100g with averages of 11.1, 28.0, 39.2 mg/100g, respectively. Among tested varieties, Seojinbyeo and Hwasungbyeo showed high T contents and Andabyeo, Damakum were high in $\textrm{T}_3$, and Andabyeo and Seojinbyeo were high in total $\textrm{TT}_3$ contents. Regardless of varieties, the average 8 isomer contents (in mg/100g) were in descending order of $\gamma\textrm{-}$$\textrm{T}_3$(17.9) >$\alpha\textrm{-}$$\textrm{T}_3$(8.8) >$\alpha\textrm{-}$T (7.8) >$\gamma\textrm{-}$T(2.6) >$\delta\textrm{-}$$\textrm{T}_3$ (0.9)>$\beta\textrm{-}$T (0.7)>$\beta\textrm{-}$$\textrm{T}_3$ (0.4)>$\delta\textrm{-}$T (0.1). In most varieties, $\gamma\textrm{-}$$\textrm{T}_3$, a strong antioxidant and anticancer compound, consisted 64% of total tocotrienol and 46% of total vitamin E in vice bran.

Purification and Biochemical Characterization of Carboxylesterase-III from Plodia interpunctella Hubner (화랑곡나방(Plodia interpunctella Hubner) Carboxylesterase-III의 정제 및 생화학적 특성)

  • Purification and biochemical experiments on the carboxylesterases-III (CE-III) from the indian meal moth, Plodia interpunctella (Hubner) were carried out to understand their enzymemological characteristics. The CE-III from the fifth instar larvae was purified by means of ammonium sulfate fractionation, gel permeation choromatography and ion exchange choromatography. The optimal temperature for the reaction of the CE-III on the 4 substrates ($\alpha$-Na, $\alpha$-Nb, $\beta$-Na and $\beta$-Nb) was confirmed at 4$0^{\circ}C$. The optimal pH for the reactions on the substrates $\alpha$-Na and $\alpha$-Nb was 7.5. But the optimal pH on the substrate $\beta$-Na and $\beta$-Nb was 8.0. The optimal substrate concentration for the reactions of the CE-III was 3.16 X 10$^{-3}$ M in $\alpha$-Na and $\beta$-Nb. On the substrate $\beta$-Na and $\alpha$-Nb, the optimal substrate concentration was 1.0 X 10$^{-3}$ M for CE-III. The $V_{max}$ and $K_{m}$ values of the carboxylesterases were varied by the substrates as followings: the $V_{max}$ of CE-III was 45.9 for $\alpha$-Na, 52.6 for $\beta$-Na, 36.4 for $\alpha$-Nb, and 83.3 ($\mu$ mol/min/mg protein) for $\beta$-Nb. The $K_{m}$ of CE-III was 1.43 X 10$^{-4}$ M for $\alpha$-Na, 3.57 x 10$^{-5}$ M for $\beta$-Na, 9.17 X 10$^{-5}$ M for $\alpha$-Nb, and 7.14 X 10$^{-5}$ M for $\beta$ -Nb, respectively. The CE-III seemed to have somewhat high thermostability considering that the temperature for effective denaturation on activity was about 5$0^{\circ}C$ ~ 6$0^{\circ}C$.EX>.EX>.

Effect of Thrombin on the TNF-$\alpha$ Induced IL-6 Production in HUVECs (혈관내피세포에서 트롬빈이 TNF-$\alpha$에 의해 유도되는 IL-6에 미치는 영향)

  • Here, we evaluated the effect of thrombin on the interleukin-6 production induced by tumor-necrosis-factor-$\alpha$ in endothelial cells. It is well known that tumor-necrosis-factor-$\alpha$ mediates inflammatory responses by activation of nuclear factor-kappa-B in endothelial cells. Here, we showed that lower concentration of thrombin decreased the production of interleukin-6 induced by tumor-necrosis-factor-$\alpha$ and this inhibitory effect of thrombin on interleukin-6 production was mediated by interacting with protease-activated-receptor-1. In addition, phosphoinositide-3-kinase was also involved the anti-inflammatory responses by lower concentration of thrombin in endothelial cells. These results suggested that lower concentration of thrombin mediated anti-inflammatory responses by interacting with protease-activated-receptor-1 on the cell membrane and phosphoinositide-3-kinase in the cell. These findings will provide the important evidence in the development of new medicine for the treatment of severe sepsis and inflammatory diseases and good clue for understanding unknown mechanisms by which thrombin showed the pro-inflammatory or anti-inflammatory activities in endothelial cells.

  • Effect of interfacial electrical conditions on adhesion of ${\alpha}-Fe_2O_3$ particles to PET fabric and the removal of ${\alpha}-Fe_2O_3$ particles from PET fabric, were investigated as functions of pH, electrolyte and ionic strength. The ${\zeta}$ potential of PET fiber and ${\alpha}-Fe_2O_3$ particles in the electrolyte solution were measured by streaming potential and microelectrophoresis methods respectively. The potential energy of interaction between ${\alpha}-Fe_2O_3$ particles and PET fabric were calculated by using the heterocoagulation theory for a sphere-plate model. The negative ${\zeta}$ potential of ${\alpha}-Fe_2O_3$ particle and PET fiber increased with pH, and then decreased certain pH and isoelectric points of ${\alpha}-Fe_2O_3$ particles and PET fiber were pH 6.5 and pH 3.5, respectively. The negative ${\zeta}$ potential of ${\alpha}-Fe_2O_3$ particle and PET fiber affected by electrolytes, were relatively high with polyanion electrolytes in solutions and were low with neutral salts. However, at surfactant solution, ${\zeta}$ potential was levelled off. The influence of the ionic strength on the ${\zeta}$ potential of ${\alpha}-Fe_2O_3$ particle was small but the negative ${\zeta}$ potential of PET fiber increased with the ionic strength. In the presence of anionic surfactant, the ${\zeta}$ potential of ${\alpha}-Fe_2O_3$ particle and PET fiber increased regardless of solution conditions. The interaction energy between ${\alpha}-Fe_2O_3$ particle and PET fabric increased with pH. The interaction energy was relatively high with polyanion electrolytes in solution, and the influence of ionic strength on the interaction energy was small, and the effective thickness of electrical double layer increased with decreasing the ionic strength.

The Effect of Feeding $\alpha$-Tocopherol and Squid Liver Oil on the Development of Smoked Chicken using Old Laying Hens (산란노계를 이용한 훈제닭 개발에 있어서의 $\alpha$-토코페롤과 오징어간유의 급여 효과)

  • This study was performed to investigate the development of smoked chicken using old laying hens. Seventy two-weeks-old, spent laying hens were fed commercial feed (control) supplemented with 100 IU ${\alpha}$-tocopherol plus 10% squid liver oil (treatment) for 15 days and slaughtered. Smoked chickens were manufactured with spent laying hens in this study, Moisture and crude ash contents in smoked chicken of treatments were higher (P<0.05) than those of control group due to the feeding 100 IU ${\alpha}$-tocopherol plus 10% squid liver oil. No differences were observed in fatty acid and amino acid composition between control and treatments. In sensory evaluation, the springiness of smoked chicken was evaluated optimum for 32% consumer. The elastic, blend, specific flavor and smell of the smoked chicken of the treatment were not different from those of the control. However, 46% of tested panel answered that the springiness was higher in the treated-group due to the feeding 100 IU ${\alpha}$-tocopherol plus 10% squid liver oil. These results indicated that smoked chickens would be developed with spent laying hen after feeding 100 IU ${\alpha}$-tocopherol plus 10% squid liver oil and sensory evaluation.

Studies on the Farrowing Induction of Sow with Prostaglandin $F_2\alpha$ I. Effect of Administration Method on the Farrowing Induclion (Prostaglandin $F_2\alpha$의 투여에 의한 돼지의 분만유기에 관한 연구 I. 투여방법이 분만유기에 미치는 영향)

  • This experiment was carried out to clarify the practical use of farrowing induction with PGF2$\alpha$ in swine production. Total 320 heads of pregnant sow were used and the effects of administration time, dosage, administration method of PGF2$\alpha$ and parity of sow on the farrowing induction were investigated. The results obtained in this experiment were summarized as following: 1. Total 228 heads out of 240 head which were treated with PGF2$\alpha$ in order to farrow artificially were farrowed normally. the rate of farrowing induction was 95%. 2. Several farrowing symptoms such as restlessness, bloody eye, udder swelling, milk discharge and nest building for piglets were observed in a few minutes after administration of PGF2$\alpha$. 3. Artifical parturition of sow treated with PGF2 at the 113rd, 112nd, 111st, 110th and 109th day of pregnancy were induced 17.0$\pm$7.3, 24.2$\pm$7.0, 31.3$\pm$14.7, 43.0$\pm$12.2 and 62.2$\pm$15.7 hours after administration, and the duration of pregnancy was significantly (P<0.01) shortened by PGF2$\alpha$ anministration. 4. Minimum amount of PGF2$\alpha$ per head for effective farrowing inductin was considered to be more than 5mg, and the more the amount, the shorter time needed for farrowing induction was resulted. 5. Effective farrowing induction was obtained from the sow treated with PGF2$\alpha$ via vulvomucous rather than from the sow treated via intromuscular, and intervals from PGF2$\alpha$ administration to farrowing in 4th-6th farrowed sows were shorter than those of 1st-3rd farrowed sows but no significant difference was observed.

Studies on the Changes of Steroid Hormone Levels during the Reproductive Stage in Korean Native Goats I.   • The study was conducted to find out the changes of progesterone and 20$\alpha$-dihydroprogesterone (20$\alpha$-OHP) levels during the oestrous cycle in Korean native goats. 4 cyclic goats were offered for this experiment. Blood samples were taken from jugular vein on day 0, 1, 3, 5, 7, 9, 11, 13, 15, 17 and 19 during the oestrous cycle, then the next oestrous day. The serum levels of progesterone and 20$\alpha$-OHP were measured by radioimmunoassay. The progesterone concentration in seurm of the cyclic goats was 0.29$\pm$0.06ng/ml on the first day of oestrous (day 0), increased to 5.29$\pm$0.73ng/ml on day 9, reached to a peak level of 5.73$\pm$0.61ng/ml on day 13, and thereafter decreased to 0.35$\pm$0.30ng/ml on day 1. The serum level of 20$\alpha$-OHP during the oestrous cycle was 0.42$\pm$0.33ng/ml on day 0 and then decreased to 0.28$\pm$0.01ng/ml on day 5. This basal level was maintained until day 13, increased gradually, and reached a peak level of 0.62$\pm$0.05ng/ml on day 19. From the above results, it was suggested that the enzyme 20$\alpha$-hydroxysteroid dehydrogenase(20$\alpha$-HSD) catalyzing the conversion of progesterone to a biologically inactive steroid, 20$\alpha$-OHP, should be active in the luteal cells during the oestrous cycle in Korean native goats.
