Effect of Cytokines and bFGF on the Osteoclast Differentiation Induced by $1\;{\alpha},25-(OH)_2D_3$ in Primary Murine Bone Marrow Cultures

  • Bone is a complex tissue in which resorption and formation continue throughout life. The bone tissue contains various types of cells, of which the bone forming osteoblasts and bone resorbing osteoclasts are mainly responsible for bone remodeling. Periodontal disease represents example of abnormal bone remodeling. Osteoclasts are multinucleated cells present only in bone. It is believed that osteoclast progenitors are hematopoietic origin, and they are recruited from hematopoietic tissues such as bone marrow and circulating blood to bone. Cells present in the osteoclast microenvironment include marrow stromal cells, osteoblasts, macrophages, T-lymphocytes, and marrow cells. These cells produce cytokines that can affect osteoclast formation. In vitro model systems using bone marrow cultures have demonstrated that $IL-l{\beta},\;IL-3,\;TNF-{\alpha},$ bFGF can stimulate the formation of osteoclasts. In contrast, IL-4 inhibits osteoclast formation. Knowledge of cytokines and bFGF that affect osteoclast formation and their capacity to modulate the bone-resorbing process should provide critical insights into normal calcium homeostasis and disorders of bone turnover such as periodontal disease, osteoporosis and Paget's disease.

Antioxidant Properties and Quantification of Phenolic Compounds from Safflower (Carthamus tinctorius L.) Seeds

  • The antioxidant properties of twelve phenolic compounds, including matairesinol 4'-O-$\beta$-D-glucoside, 8'-hydroxyarctigenin 4'-O-$\beta$-D-glucoside, matairesinol, 8'-hydroxyarctigenin, N-feruloylserotonin 5-O-$\beta$-D-glucoside, N-(p-coumaroyl)-serotonin-5-O-$\beta$-D-glucoside, N-feruloylserotonin, N-(p-coumaroyl)serotonin, luteolin 7-O-$\beta$-D-glucoside, luteolin, acacetin 7-O-$\beta$-glucuronide, and acacetin, isolated from defatted safflower (Carthamus tinctorius L.) seeds were evaluated with regard to the DPPH, superoxide and hydroxyl radicals. Additionally, levels of phenolic compounds were determined by HPLC in two cultivars of safflower seeds. Among them, four serotonin derivatives showed potent DPPH ($IC_{50}=10.83-21.75\;{\mu}M$) and hydroxyl ($IC_{50}=75.93-374.63\;{\mu}M$) radical scavenging activities, and their activities were significantly stronger than that of ${\alpha}-tocopherol$. Four flavonoids ($IC_{50}=170.65-275.83\;{\mu}M$) and four lignans ($IC_{50}=114.22-406.10\;{\mu}M$) exhibited significant superoxide and hydroxyl radical scavenging activities, respectively, whereas these compounds contained less activity toward the DPPH and hydroxyl radicals than serotonin derivatives. The levels of serotonin derivatives, lignans and flavonoids in safflower seeds of two cultivars ranged from 49.30 to 260.40, 3.72 to 158.90, and 11.72 to 214.97 mg% (dry base), respectively. Of the two cultivars, 'Cheongsu' had somewthat higher concentrations of phenolic compounds than 'Uisan'. These results suggest that phenolic compounds in safflower seeds may playa role as protective phytochemical antioxidants against reactive oxygen-mediated pathological diseases.

Antifungal Activity of the Crude Extract from Quintinia acutifolia on the Dermatophytic fungus

  • The crude extract of Quintinia acutifolia Kirk inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (3 mm inhibition zone at $150\;{\mu}g/disc$) and the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (3 mm inhibition zone at $150\;{\mu}g/disc$), and cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_{50}$ $50,000\;{\mu}g/mL$ at $150\;{\mu}g/disc$). However, Candida albicans (ATCC 14053) did not observed the antimicrobial activity and the cytotoxic activity to BSC monkey kidney cells ($({\alpha})$ 5 mg/mL, $150\;{\mu}g/disc$).

한국의 임상과 자연환경에서 분리된 Cryptococcus neoformans의 혈청형과 효소생성능 (Serotype and Enzymatic Profile of Crypfococcus neoformans Isolates from Clinical and Environmental Sources in Korea)

  • 한국의 임상검체와 자연환경에서 분리된 Cryptococcus neoformans 58주에 대한 혈청형과 세포외효소 생성능에 관한 셍물학적 특성을 조사하였다. 환자로부터 분리된 임상균주 51주 중 48주는 혈청형 A (94.1%) 였으며 2주는 혈청형 B (3.92%),그리고 나머지 1주는 혈청형 D (1.96%)였다. 자연환경에서 분리된 7주는 비둘기 분변에서 분리된 것들이며 모두 혈정형 A였다. 모든 균주는 proteinase와 phnospholipase를 생성하였고, 또한 API-ZYM system을 이용한 19종류의 효소생성능 시험에서는 alkaline phosphatase, esterase (C4), esterase lipase (C8), leucine arylamidase, acid phosphatase, naphthol-AS-BI-phosphohydrolase, $\alpha$-glucosidase, 그리고 $\beta$-glucosidase를 생성하였으나, N-acetyl-$\beta$-glucosarninidase는 39주 (67.2%)에서만 생성하였다. 혈청형 B로 동정된 2주와 혈청형 A로 동정된 균주중 1주는$\beta$-glucuronidase를 생성하였다. 본 연구에 사용된 총21종류의 효소 생성능 시험을 기초로 하여 생물형을 구분하였는데, 모두 4가지의 유형을 나타내었고, 또한 임상과 환경균주에서 혈정형과 생물형 특성간의 유의한 상관성를 나타내었다.

독일가문비(Picea abies Karsten) 잎 추출성분의 항산화 활성 (Antioxidant Activity of the Extractives from the Needles of Picea abies Karsten)

  • 독일가문비(Picea ahies Karsten) 잎을 채취하여 건조시킨 후 분쇄하여 1.5 kg을 acetone-$H_2O$(7:3, v/v)로 추출하고 핵산, 메틸렌클로라이드, 에틸아세테이트 및 물 가용부로 분획하여 동결건조 시켰다. 에틸아세테이트 및 물 가용부를 Sephadex LH-20으로 충진한 칼럼에서 MeOH와 EtOH-n-hexane 혼합액을 용출용매로 사용하여 칼럼크로마토그래피를 실시하였다. 단리된 화합물들은 셀룰로오스 박층크로마토그래피(TLC)로 확인한 후 $^1H$-, $^{13}C$-NMR 등의 스펙트럼을 사용하여 정확한 구조를 규명하였고 FAB 및 El-MS로써 분자량을 측정하였다. 독일가문비 잎의 에틸아세테이트 가용부에는 (+)-catechin(화합물 I), (-)-epicatechin(화합물 II), kaempferol-3-O-${\beta}$-D-glucopyranoside(화합물 III), 4-hydroxyacetophenone(화합물 IV)가 분리되었으며 물 가용부에서는 (+)-catechin(화합물 I)과 protocatechuic acid(화합물 V)가 분리되었다. 각 분획물 및 단리된 화합물들은 DPPH 라디칼 소거법을 이용하여 항산화 시험을 실시하였으며, 분획물중에는 에틸아세테이트 가용부, 그리고 화합물중에는 (T)-catechin과 (-)-epicatechin에서 기준물질과 유사한 우수한 항산화 효능이 있는 것으로 나타났다.

Isolation and Structural Determination of Free Radical Scavenging Compounds from Korean Fermented Red Pepper Paste (Kochujang)

  • Sixteen antioxidative active compounds isolated from the EtOAc layer of MeOH extracts of kochujang, Korean fermented red pepper paste, were structurally elucidated as fumaric acid, methyl succinate, succinic acid furan-2-yl ester methyl ester (gochujangate, a novel compound), 2-hydroxy-3-phenylpropanoic acid, 3,4-dihydroxybenzoic acid, 2,3-dihydroxybenzoic acid, 2,4-dihydroxybenzoic acid, 6,7-dihydroxy-2H-chromen-2-one (esculetin), caffeic acid, cis-p-coumaric acid, trans-p-coumaric acid, daidzin, genistin, apigenin 7-O-$\beta$-D-apiofuranosyl($1{\rightarrow}2$)-$\beta$-D-glucopyranoside, apigenin 7-O-$\beta$-Dglucopyranoside, and quercetin 3-O-$\alpha$-L-rhamnopyranoside by mass spectrometry (MS) and nuclear magnetic resonance (NMR) experiments. These compounds were analyzed for the first time as antioxidants from kochujang.

구강악안면 영역의 치성 감염 환자에 대한 세균학적 연구 (A STUDY OF MICROORGANISMS IN ORAL & MAXILLOFACIAL INFECTED PATIENTS)

  • Oral & maxillofacial infections are most commonly odontogenic in origin. Although such infections are usually self-limiting, they may occasionally spread deeply into fascial spaces or planes far from the initial site of involvement. If early diagnosis and appropriate therapy is delayed, complications such as mediastinal extension, retropharyngeal spread and airway obstruction could happen to the patients. For the study of the microbiology, we have retrospectively analysed the oral & maxillofacial infected patients in the Dept. of Oral & Maxillofacial Surgery. In-Ha University Hospital from 1997 September to 2000 April. The results were as follows 1. The male patients were more common than female, with male 61.9% and female 38.1%. 2. Dental originated infections were most common cause with the incidence of 62%. 3. Most common fascial space involved was buccal space 42cases(37.2%) followed by submandibular space 13cases(11.5%), infraorbital space 13cases(11.5%), masseteric space 11cases(9.7%), periapical abscess 11cases(9.7%). 4. The causative organisms isolated from the pus culture were Gram Positive Bacterial species, which were 46cases(31.9%) of Streptococcus viridans, 16cases(8.6%) of ${\alpha}$ and ${\beta}-hemolytic$ streptococcus, 4cases(3.1%) of Strep.-group D non enterococci, 7cases(5.1%) of Staphylococcus Coa. neg., 5cases(3.9%) of Staphylococcus aureus, 3cases(2.3%) of Enterococcus faecalis, 1case(0.8%) of Bacillus species, 1case(0.8%) of Peptostreptococcus, 1case(0.8%) of Clostridium and Gram negative bacterial species, which were 4cases(3.1%) of Acinetobacter baumannii, 2cases(1.6%) of Pseudomonas aeruginosa, 2cases(1.6%) of Burkholderia cepacia, 1case(0.8%) of Neisseria species, 1case(0.8%) of Klebsiella pneumoniae, 1case(0.8%) of Klebsiella oxytoca, 1case(0.8%) of Escherichia coli. 5. In drug sensitivity test, high resistant tendency was found in Penicillin system(Penicillin G 83.3%, Ampicillin 60%) and Aminoglycosides (Gentamycin 50%, Tobramycin 45.5%), but tertiary Cephalosporin system(Cefoperazone 9.1%, Ceftazidime 18.2%), and glycopeptides system (Teicoplanin 0%, Vancomycin 0%) showed lower resistancy.

복강경 보조 위 전절제술-연속된 77예의 경험 (Laparoscopy Assisted Total Gastrectomy with Lymph Node Dissection-77 Consecutive Cases)

  • 목적: 조기 위암 환자에 대한 복강경 보조 원위부 위절제술은 삶의 질 향상을 위한 치료의 하나로 점차 많이 시행되고 있다. 하지만, 복강경 보조 위 전절제술은 위아전절제술에 비해 술기상의 복잡성 등의 이유로 그 빈도가 높지 않다. 이에 저자 등은 본 교실에서 시행한 복강경 보조위 전절제술의 초기 경험을 분석하여 술기상의 가능성과 안정성 및 수술 결과를 평가하고자 한다. 대상 및 방법: 연세대학교 의과대학 외과학교실에서 조기 위암으로 2003년 7월부터 2007년 6월까지 한 명의 술자에 의해 수술 전 조기위암으로 진단받고 복강경 보조 위 전절제술과 D1+$\beta$ 림프절 절제술 및 Roux-en-Y 재건술을 시행받은 77명의 환자를 대상으로 임상병리학적 특징, 수술 후 경과, 합병증을 후향적으로 분석하였다. 결과: 환자들의 평균 나이는 61세였고 남자와 여자의 수는 각각 49명, 28명이었다. 평균 수술시간은 210분(범위 100-400)이었고 각 수술 시간은 수술 경험이 많아짐에 따라서 점 차 감소하였다. 수술 후 13예(16.9%)에서 합병증이 발생하였고 사망한 예는 없었다. 장관 운동은 평균 수술 후 3.2일에 회복되었고, 수술 후 연식을 시작한 시기는 4.4일, 수술 후 평균 재원일은 10일이었다. 위의 절제연은 근위부 원위부 각각 평균 3.1 cm, 10.7 cm로 충분한 절제연을 확보하였으나 한 예에서 원위부 절제연에 현미경적 암세포의 침윤이 있었다. 림프절 절제 범위는 D1+$\alpha$가 2예(2.6%), D1+$\beta$가 55예(71.4%), D2가 20예(26.0%)이었고 평균 절제된 림프절 수는 41.8개이었다. 결론: 복강경 보조 위 전절제술의 초기 경험을 분석하였을 때 조기위암의 치료에 있어 효과적이고 안전한 술식임을 확인하였다. 그 적용 범위의 확대를 위해서는 앞으로 복강경 보조 위 전절제술과 개복 수술의 전향적인 비교 연구와 장기 생존율 등 종양학적인 치료결과에 대한 연구가 필요하다.

Low Density Lipoprotein-oxidation Inhibitory Phytochemicals from the Fruits of Rhus parviflora

  • Fruits of Rhus parviflora were extracted with 80% aqueous methanol (MeOH), and the concentrated extract was partitioned using ethyl acetate (EtOAc), n-butanol (n-BuOH), and $H_2O$, successively. Purification of EtOAc fraction led to isolation of fifteen polyphenols of which structures were identified by spectroscopic methods including 2D-NMR. Most compounds apart from compound 10 inhibited low density lipoproteinoxidation within $IC_{50}$ value of $10{\mu}M$. Among compounds, taxifolin (2), quercetin 3-O-${\alpha}$-L-rhamnopyranoside (13), agathisflavone (5) sulfuretin (4), and aureusidin (3) showed $IC_{50}$ values 0.9, 0.8, 5.8, 2.9, and $2.4{\mu}M$ which were of highly significant in comparison positive control butylated hydroxytoluene with $IC_{50}$ value of $2.1{\mu}M$. The results indicate fruits of R. parviflora as a source of antihypercholesterolemic compounds.

Lysophosphatidic acid increases mesangial cell proliferation in models of diabetic nephropathy via Rac1/MAPK/KLF5 signaling

  • Mesangial cell proliferation has been identified as a major factor contributing to glomerulosclerosis, which is a typical symptom of diabetic nephropathy (DN). Lysophosphatidic acid (LPA) levels are increased in the glomerulus of the kidney in diabetic mice. LPA is a critical regulator that induces mesangial cell proliferation; however, its effect and molecular mechanisms remain unknown. The proportion of ${\alpha}-SMA^+/PCNA^+$ cells was increased in the kidney cortex of db/db mice compared with control mice. Treatment with LPA concomitantly increased the proliferation of mouse mesangial cells (SV40 MES13) and the expression of cyclin D1 and CDK4. On the other hand, the expression of $p27^{Kip1}$ was decreased. The expression of $Kr{\ddot{u}}ppel$-like factor 5 (KLF5) was upregulated in the kidney cortex of db/db mice and LPA-treated SV40 MES13 cells. RNAi-mediated silencing of KLF5 reversed these effects and inhibited the proliferation of LPA-treated cells. Mitogen-activated protein kinases (MAPKs) were activated, and the expression of early growth response 1 (Egr1) was subsequently increased in LPA-treated SV40 MES13 cells and the kidney cortex of db/db mice. Moreover, LPA significantly increased the activity of the Ras-related C3 botulinum toxin substrate (Rac1) GTPase in SV40 MES13 cells, and the dominant-negative form of Rac1 partially inhibited the phosphorylation of p38 and upregulation of Egr1 and KLF5 induced by LPA. LPA-induced hyperproliferation was attenuated by the inhibition of Rac1 activity. Based on these results, the Rac1/MAPK/KLF5 signaling pathway was one of the mechanisms by which LPA induced mesangial cell proliferation in DN models.
