PIV를 이용한 직렬배열에서 2원주 주위의 유동장 특성 연구 (The Characteristic Investigation of the Flowfield around Two Circular Cylinders in the Tandem Arrangement Using the PIV)

  • The Characteristics of the flowfield around two circular cylinders in tandem arrangement was investigated by PIV. Strouhal numbers. velocity vectors and velocity profiles were observed at centre-to-centre space ratios of P/D= 1.5. 2.0 and 2.5, and Reynolds number of $Re=3.0{\times}10^3{\sim}5.0{\times}10^3$. As the results the Strouhal numbers measured in the rear region of the cylinder of wake side were decreased with the space ratios. The flow between two cylinders was almost stagnated and the size of the stagnated region was larger in the close side than in the far side of the front cylinder. The direction of vortex between two cylinders was opposed each other with the small difference(${\alpha}\;{\pm}1.0^{\circ}$) of the attack angle ${\alpha}$.

대두 ${\alpha}-galactosidase$의 정제 및 성질 (Purification and properties of soybean ${\alpha}-galactosidase$)

  • 대두 발아 과정 중의 ${\alpha}-galactosidase$를 추출하여 염석, 이온교환 크로마토그래피 및 겔 여과 등의 방법으로 정제한 후 정제효소의 효소학적 성질을 검토하였다. 대두 ${\alpha}-galactosidase$의 활성은 $25^{\circ}C$에서 120시간 발아시켰을 때 가장 높았으며, 대두 중의 raffinose는 96시간, stachyose는 120시간 발아시켰을 때 완전히 분해되었다. 대두 ${\alpha}-galactosidase$는 황산암모늄염석, DEAE-Cellulose 및 DEAE-Sephadex A-50 이온교환 크로마토그래피, Sephadex G-150 겔 여과 등에 의하여 비활성은 825U/mg protein으로써 6.6배까지 정제되었으며 수율은 2.5%이었고 HPLC와 PAGE에 의하여 순도를 확인하였다. 정제효소의 등전점은 pH 4.8이었고, 분자량은 30,000인 monomer이었으며 정제효소의 최적작용 PH는 6.0, 최적작용온도는 $40^{\circ}C$ 이었고, $60^{\circ}C$에서 10분 처리시 25%의 잔존 활성을 나타내었다. 정제효소는 stachyose보다 raffinose를 쉽게 분해하였으며 PNPG에 대한 Km값은 5.3 mM, 활성화 에너지는 13.02 cal/mole이었다.

  • The purpose of this study was to process extraction and separation for polyglucose porpyran from pophyra yezoensis as high yield. The acidic porphyran solution was extracted with 0.1~1N of ${\alpha},{\beta}$-dichloromaleic acid or ${\alpha}$-chloromaleic acid of 2% solution as organic acid instead of inorganic acid at $60^{\circ}C$ and then porphyran was collection of high yield to acidic solution and it was neutralization treated with 0.1N of $d-Ca(OH)_2$ solution and the mixture was conversion to porphyran salt form and treated with a shell powder of ouster and then added of ethanol as precipitator. It recovery porphyran contained of violet purple laver coloring matter was obtained as a crystalline and used for the next step without future purification to prepare of porphyran laver jam. so, The resulting porphyran and porphyran jam was characterized by it component and physical properties.

방사선조사후 마우스에서의 Cytokine 생산능 및 Listeria monecytogenes에 대한 저항성의 변화 (Effect of lonizing Radiation on the Host Resistance Against Listeria Monocytogenes Infection and the Cytokine Production in Mice)

  • 목적 : 방사선조사가 세포내재성 병원균인 Listeria monocytogenes (LM)의 감염에 미치는 영향과 함께 감염면역과 밀접한 관계가 있는 대식세포에서의 $TNF-\alpha$ 및 Nitric oxide (NO) 생산능의 변화와 비장세포에서의 $IFN-\gamma$ 및 IL-2생산능에 미치는 영향을 알아보고자 본 실험을 시행하였다. 대상 및 방법 : 실험동물로는 BALB/c 마우스를 사용하였으며 Co-60 원격치료기를 이용하여 방사선을 조사하였다. LM감염에 미치는 영향을 관찰하기 위해서 방사선조사 1일후 $10^5$의 LM균을 복강내에 주사하고 1일, 3일, 5일후에 비장조직에서 LM생균수를 측정하였다. 방사선조사가 마우스의 생체내에서와 시험관내에서의 $TNF-\alpha$의 생산능에 미치는 효과를 관찰하기 위해서 각각 LPS로 유도하여 L929/Actinomycin D assay에 의해 $TNF-\alpha$량을 측정하였다. $IFN-\gamma$의 생성능은 방사선조사후 비장을 적출하여 비장세포액을 제조하여 Concavalin A (Con-A)로 자극한 후 정량검사를 하였다. IL-2의 생성능은 $IFN-\gamma$실험에서와 같이 비장세포를 얻어서 Con-A로 자극하여 CTLL-2세포의 성장촉진능력을 관찰함으로써 판정하였다. 방사선조사가 복강내 대식세포에서 생산되는 NO에 미치는 영향도 관찰하였다. 결과 : 방사선 (300cGy)을 조사한 군에 LM을 감염시킨 1일후 비장으로부터 검출되는 생균수는 대조군에 비해 감소하였으나 감염 3일 5일후의 생균수는 대조군에 비하여 오히려 증가하였다. 시험관내 복강대식세포에 방사선 (100-850cGy)을 조사하면 1일후 생산되는 $TNF-\alpha$의 양은 대조군에 비하여 증가하였으나, 방사선조사 (100-600cGy) 5일후 수집된 복강대식세포에 의한 $TNF-\alpha$의 생산은 오히려 감소하였고. 방사선조사 (300cGy)후 LM감염시 5일후 유도된 생체내 $TNF-\alpha$의 생산도 대조군에 비해 감소하였다. 방사선 (300cGy)을 조사한 마우스로부터 적출한 비장세포로부터 생산되는 $IFN-\gamma$와 IL-2의 양은 대조군에 비해 감소하였다. NO의 양은 100cGy 및 300cGy조사시 대조군에 비해 증가하였으며 그 이상 조사량을 증가하면 점차 감소하였다. 결론 : 방사선조사후 세포내재성 병원균인 LM감염에 대한 초기 저항성의 증가와 감염중반이후의 저항성 감소는 대식세포에서의 초기 $TNF-\alpha$ 생산능 증가후 감소, 그리고 T림프구에서의 $IFN-\gamma$및 IL-2 생산능 감소와 관련이 있을 것으로 사료된다.

Characterization of the Functional Properties of Soy Milk Cake Fermented by Bacillus sp.

  • The mucilage production and tyrosine content in soy milk cake (SMC) fermented by Bacillus firmus NA-1, Bacillus subtilis GT-D, and B. subtilis KU-A was improved by fortification with 10% defatted soybean flour. The fibrinolytic activity and consistency of the SMC were drastically increased by solid-state fermentation for 1 day. However, the consistency of the fermented SMC gradually decreased during fermentation for 3 days. Furthermore, the tyrosine content of the freeze-dried powder of SMC fermented by three Bacillus sp. was 9 times higher than that of unfermented SMC. The soybean proteins, including the 7S and 11S subunits, were partially digested during alkaline fermentation, producing lower molecular-weight peptides. The fibrinolytic enzyme produced in SMC fermented by B. firmus NA-l and B. subtilis KU-A exhibited higher thermal stability than that of B. subtilis GT-D fermentation. The powder obtained from B. subtilis GT-D fermentation had an ${\alpha}$-amylase activity and lower consistency compared to those of B. firmus NA-1 and B. subtilis KU-A. In addition, this powder contained 6.3% moisture content, 27% crude protein content and 9 units of fibrinolytic activity and proteolytic activity.

Nitric Oxide Inhibitory Constituents from Fruits of Opuntia humifusa

  • Opuntia humifusa, also called as Cheonnyuncho, is a cactus widely cultivated in southern regions of Korea. It has been known to have diverse biological activities, but most of the studies were performed with the MeOH extracts or solvent-partitioned fractions. Furthermore, the efforts to identify the responsible compounds for the biological activities are very limited. In this study, we tested the inhibitory effect of extracts and solvent-partitioned fractions of O. humifusa against LPS-induced nitric oxide (NO) production in Raw264.7 cells. The butanol fractions of O. humifusa efficiently inhibited the production of NO in Raw264.7 cells, but it was not due to the reduction of cell viability. Bioassay-guided isolation of butanol fractions of O. humifusa allowed the isolation of three flavonoids isorhamnetin 3-O-${\beta}$-$\small{D}$-galactosyl-4'-O-${\beta}$-$\small{D}$-glucoside (1), isorhamnetin 3,4'-di-O-${\beta}$-$\small{D}$-glucoside (2) and isorhamnetin 3-O-${\beta}$-$\small{D}$-(6-O-${\alpha}$-$\small{L}$-rhamnosyl)glucoside (3), and one lignan syringaresinol O-${\beta}$-$\small{D}$-glucopyranoside (4). Among them, isorhamnetin 3-O-${\beta}$-$\small{D}$-galactosyl-4'-O-${\beta}$-$\small{D}$-glucoside (1) and isorhamnetin 3,4'-di-O-${\beta}$-$\small{D}$-glucoside (2) exhibited the moderate inhibitory effects against LPS-induced NO production. This is the first time to report anti-inflammatory effects of these compounds.

황칠, 닥나무, 꾸지뽕 혼합 추출물의 항당뇨 효과 (Antidiabetic Effects of Mixed Extract from Dendropanax morbiferus, Broussonetia kazinoki, and Cudrania tricuspidata)

  • Dengropanax morfiferus (D), Broussonitia kazinoki (B), and Cudriania tricuspidata (E), a widely cultivated species in South Korea, has been used as traditional medicine to treat numerous diseases. In this study, we evaluated the antidiabetic effects in a various signaling mechanisms using mixed extract and major component contents were analyzed by HPLC in the combined extracts from Dengropanax morfiferus, Broussonitia kazinoki, and Cudriania tricuspidata (DBCE). DBCE inhibited ${\alpha}$-glucosidase and ${\alpha}$-amylase activation and showed potent antioxidant effects, which are evaluated using DPPH, ABTS, and SOD assay. Cytokines, which are released by inflammatory cells in pancreatic islets, are involved in the pathogenesis of type 1 diabetes mellitus. DBCE showed the protective effects in RINm5F cells against cytokines-induced damage by suppressing inducible nitric oxide (NO) synthase and COX-2 expression and NO production. Insulin resistance is the primary characteristic of type 2 diabetes. Therefore, the regulatory effect of DBCE on glucose uptake and production are investigated in insulin-responsive human HepG2 cells. DBCE stimulated glucose uptake, prevented Glut2 and phosphor-IRS1 downregulation induced by high glucose (HG, 30 mM). Moreover, DBCE pretreatment diminished glucose levels, PEPCK and G6Pase overexpression provoked by HG. These findings suggest that DBCE might be used for diabetes treatment through alpha-glucosidase or alpha-amylase activity regulation, pancreatic beta cell protection, hepatic glucose sensitivity improvement. Cytokines, which are released by inflammatory cells' infiltrations around the pancreatic islets, are involved in the pathogenesis of type 1 diabetes mellitus.

1-Deoxynojirimycin을 생산하는 Bacillus subtilis S10 배양액의 혈당강하 효과 (Hypoglycemic Effect of Culture Broth of Bacillus subtilis S10 Producing 1-Deoxynojirimycin)

  • 본 연구는 $\alpha$-glucosidase의 강력한 효소저해제인 1-deoxynojirimycin(DNJ)을 고효율로 생산하는 균주의 선발 및 동정, 배양최적화 및 균 배양액의 혈당강하효과 등에 관하여 조사하였다. 먼저 토양으로부터 $\alpha$-glucosidase에 대해 강력한 저해능을 나타내는 9균주를 대상으로 이 중에서 DNJ 생산성이 가장 우수한 한 개 균주를 최종 선발하였고, 이 균주를 16S rDNA 염기서열분석으로 균주동정을 하여 Bacillus subtilis S10이라 명명하였다. 본 균주의 배양액을 이온교환수지법(Amberlyst 15 $H^+$ form, Dowex $1{\times}2$-100 $OH^-$ form, Amberlite CG-50 $NH_3^+$ form)에 의해 DNJ를 정제하고, 정제된 DNJ를 LC-MS로 분석한 결과 표준 DNJ와 동일한 물질임을 확인하였다. DNJ 대량생산을 위한 S10균주의 배양 최적탄소원 및 최적질소원과 이들의 최적농도를 조사한 결과 각각 1% galactose, 1.6% polypeptone임이 밝혀졌으며, 확립된 최적화 조건에서의 DNJ 생산량은 0.75 g/L이었다. 고혈당 유도 마우스를 대상으로 각종 혈당강하제의 효과를 알아보기 위해 혈당치를 비교한 결과 무처리구의 혈당치 $510{\pm}10.9\;mg/dL$에 비해 acarbose 처리구는 $139.4{\pm}33.1\;mg/dL$, 누에분말 처리구는 $209.1{\pm}19.6\;mg/dL$, 그리고 S10 균주 배양액 처리구는 $208.6{\pm}39.8\;mg/dL$로써 무처리구에 비해 각각 72.7%, 59.0%, 그리고 59.1%의 혈당 억제율을 나타냈다. 이상의 연구결과 S10균주 배양액은 누에분말과 같은 수준의 고혈당 억제 및 완화효과가 있음이 확인되었으며 향후 S10균주의 대량생산을 통해 식후혈당조절을 위한 건 강기능식품 개발이 기대된다 하겠다.

Crystal Structure of Byakangelicin ($C_{17}H_{18}O_{7}$)

  • The crystal structure of byakangelicin, one of furanocoumarin aldose reductase inhibitors, was determined by X-ray diffraction method. The crystal is triclinic, with a=8.114(1), b=10.194(1), $c=11.428(1)\AA,{\;}{\alpha}=111.50(1),{\}{\beta}=95.57(1),{\}{\gamma}=112.52(1)^{circ},{\;}D_x=1.41,{\;}D_m=1.39{\;}g/cm^3$, space group P1 and Z=2. The intensity data were collected by ${\omega}-2{\theta}$ scan method with $CuK_{a}$ radiations. The structure was solved by direct method and refined by full matrix least-squares procedure to the final R-value of 0.056. There are two molecules with different conformations in an asymmetric unit. The molecules are kept by two intermolecular O-HO type hydrogen bonds and van der Waal's forces in the crystal. The absolute configuration of the molecules was estimated to S-form by the 'Eta refinement' procedure.

Antioxidant Constituents from the Leaves of Cedrela sinensis A. Juss

  • Phytochemical study on the EtOAc fraction from the MeOH extract of the leaves of Cedrela sinensis led to the isolation of five known phenolic compounds (1-5), whose structures were identified as (+)-catechin (1), $kaempferol-3-0-{\alpha}- L-rhamnopyranoside$ (2), quercetin (3), $quercetin-3-O-{\alpha}-L-rhamnopyranoside$ (4), and $quercetin-3-O-{\beta}-D-glucopyranoside$ (5), respectively, by comparing their spectral $(uv,\;JR,\;IH\;and\;^{13}C-NMR,\;and\;ESI-MS)$ and physicochemical data with those reported in the literature. Among the isolated compounds (1-5), compounds 1 and 3-5 exhibited significant DPPH radical scavenging effects with $IC{_50}$ values ranging from $21.3{\pm}1.4\;to\;38.1{\pm}3.2 {\mu}M$ as well as superoxide anion radical scavenging effects with $IC{_50}$ values ranging from $9.4{\pm}0.7\;to\;21.2{\pm}3.6 {\mu}M$. Furthermore, compounds 1 and 3-5 also exhibited considerable inhibitory effects on LDL peroxidation induced by either $CU^{2+}$ or AAPH with $IC{_50}$ values ranging from $1.4{\pm}0.4\;to\;11.9{\pm}1.4\;{\mu}M$. These results indicated that flavonoids are the major constituents of C. sinensis and considered to be antioxidant principles of this plant.
