식물유래 천연물, TGR-N1의 잔디 진균병에 대한 방제효과 검정 (Control Activities of Natural Compound, TGR-N1, against Turfgrass Fungal Diseases in vitro and in Field)

  • 이 연구는 식물유래 천연물, TGR-N1의 잔디진균병에 대한 방제효과를 조사하기 위해 수행되었다. In vitro실험에서 TGR-N1은 잔디진균병에 대해 항균효과를 나타내었다. TGR-N1의 500배, 1000배액은 Rhizoctonia solani AG 1-1, Rhizoctonia solani AG2-2, Rhizoctonia cerealis, Sclerotinia homoeocarpa, Colletotrichum graminicola 에 대해 강한 항균력을 나타내었다. 그러나 Pythium spp.에 대해서는 항균효과가 없었다. TGR-N1의 Rhizoctonia solani AG 1-1, Rhizoctonia solani AG2-2에 대한 포장방제 실험결과, TGR-N1 500배액의 방제가는 각각 86.1%와 83.3%의 방제가를 나타내었다. 이 연구 결과 식물유래 천연물 TGR-N1의 항균 활성이 확인되었는 바 천연물 농약으로서의 잠재성을 보여 주었다.

  • 실옥산 사슬과 크라운 에테르가 연결된 유연성이 큰 2가지 새로운 비스-크라운 에테르를 합성하였다. 1,3-Bis(trimethylsiloxy)-1,3-dimethyl-1,3-dipentaoxacyclohexadecamethyl disiloxane(1)는 백금촉매 존재하에서 3-methylene-16-crown-5 (8)과 1,3-bis(trimethylsiloxy)-1,3-dimethyl disiloxane (10)의 반응에 의해 합성하였다. 1,1,3,3,5,5-Hexamethyl-1,5-dipentaoxacyclohexadecamethyl trisiloxane (2)은 백금촉매 존재하에서 crown ether (8)과 1,1,3,3,5,5-hexamethyl trisiloxane (11)의 반응에 의해 합성하였다. 3-Methylene-13-crown-4 (7), 3-methylene-16-crown-5 (8), 및 3-methylene-19-crown-6 (9)은 NaH 존재하에서 triethylene glycol (3), tetraethylene glycol (4), 및 pentaethylene glycol (5)을 각각 3-chloro-2-chloro-methyl-1-propene (6)와 반응시켜서 합성하였다.

Genetic Polymorphism of Interleukin-1A (IL-1A), IL-1B, and IL-1 Receptor Antagonist (IL-1RN) and Prostate Cancer Risk

  • Purpose: We aimed to investigate the associations between polymorphisms of interleukin-1A (IL-1A), IL-1B, and IL-1 receptor antagonist (IL-1RN) and prostate cancer (PCa) risk. Materials and Methods: A comprehensive search for articles of MEDLINE and EMBASE databases and bibliographies of retrieved articles published up to August 3, 2014 was performed. Methodological quality assessment of the trials was based on a standard quality scoring system. The meta-analysis was performed using STATA 12.0. Results: We included 9 studies (1 study for IL-1A, 5 studies for IL-1B, and 3 studies for IL-1RN), and significant association was found between polymorphisms of IL-1B-511 (rs16944) as well as IL-1B-31 (rs1143627) and PCa risk. IL-1B-511 (rs16944) polymorphism was significantly associated with PCa risk in homozygote and recessive models, as well as allele contrast (TT vs CC: OR, 0.74; 95%CI, 0.58-0.94; P=0.012; TT vs TC+CC; OR, 0.79; 95%CI, 0.63-0.98; P=0.033; T vs C: OR, 0.86; 95%CI, 0.77-0.96; P=0.008). The association between IL-1B-31 (rs1143627) polymorphism and PCa risk was weakly significant under a heterozygote model (OR, 1.35; 95%CI, 1.00-1.80; P=0.047). Conclusions: Sequence variants in IL-1B-511 (rs16944) and IL-1B-31 (rs1143627) are significantly associated with PCa risk, which provides additional novel evidence that proinflammatory cytokines and inflammation play an important role in the etiology of PCa.

mRNA Expression and Clinical Significance of ERCC1, BRCA1, RRM1, TYMS and TUBB3 in Postoperative Patients with Non-Small Cell Lung Cancer

  • Background: To explore mRNA expression and clinical significance of ERCC1, BRCA1, RRM1, TYMS and TUBB3 genes in tumor tissue of postoperative patients with non-small cell lung cancer (NSCLC). Materials and Methods: Sixty NSCLC patients undergoing radical operation in our hospital from Nov., 2011 to Jun., 2012 were selected. Plasmid standards of ERCC1, BRCA1, RRM1, TYMS and TUBB3 were established and standard curves were prepared by SYBR fluorescent real-time quantitative PCR analysis. Samples from tumor centers were taken to detect mRNA expression of ERCC1, BRCA1, RRM1, TYMS and TUBB3 genes in cancerous tissue during operation. The total mRNA expression quantities were compared according to different clinical characteristics. Results: The total expression quantities of 5 genotypes from high to low were ERCC1>RRM1>TUBB3>TYMS>BRCA1 in turn. By pairwise comparisons, other differences showed statistical significance (p<0.05 or p<0.01) except for TYMS and TUBB3 (p>0.05); the low expression rates from high to low were ERCC1>TYMS>TUBB3>TUBB3>RRM1>BRCA1 in turn. The expression quantities of BRCA1, RRM1 and TYMS in males, smokers and patients without adenocarcinoma were all significantly higher than that in females, non-smokers and patients with adenocarcinoma, and significant differences were present (p<0.05 or p<0.01). In terms of pathological staging, the expression quantities of BRCA1, RRM1 and TYMS in phases IIa~IIb and IIIa~IIIb had a tendency to be greater than in phases I and IV. Conclusions: Resistance to chemotherapy and sensitivity to targeted therapy differ among patients with NSCLC. Differences in gene expression in different individuals were also revealed. Only according to personalized detection results can individualized therapeutic regimens be worked out, which is a new direction for oncotherapy.

The prevalence of dog erythrocyte antigen 1 in relation to breed in the Daejeon area

  • This study was performed to collect the basic data of DEA 1.1 in four small breed (Maltese, Shih-tzu, Poodle, Yorkshire terrier) and in three large breed (German shepherd, Labrador retriever and Jindo) dogs in the Daejeon area. 105 dogs from 7 breeds (Maltese=20, Shih-tzu=19, Poodle=15, Yorkshire terrier=11, German shepherd=10, Labrador retriever=10, Jindo=20) were selected and tested using the dog blood typing Kit$^{(R)}$ (Korea Animal Blood Bank Inc., South Korea). The prevalence of DEA 1.1 was 83%, that of DEA 1.2 was 17%, and there was no DEA (-) blood type identified in this study. Prevalence according to breeds was Maltese (DEA 1.1, 85%; DEA 1.2, 15%), Shih-tzu (DEA 1.1, 95%; DEA 1.2 5%), Yorkshire terrier (DEA 1.1, 91%; DEA 1.2, 9%), Labrador retriever (DEA 1.1, 90%; DEA 1.2, 10%). One hundred percent of DEA blood type 1.1 was discovered in all of the Poodles and German shepherds, and a higher prevalence of DEA 1.2 was found (DEA 1.1, 40%; DEA 1.2 60%) in Jindo dogs. The prevalence of DEA 1.2 in the Jindo dogs was significantly higher than in other breeds (P<0.01). German shepherds and Labrador retrievers may be more suitable as donor dogs than Jindo dogs in the Daejeon area. Larger scale studies are necessary from more dogs and other areas in South Korea.

Analysis of the Molecular Event of ICAM-1 Interaction with LFA-1 During Leukocyte Adhesion Using a Reconstituted Mammalian Cell Expression Model

  • Ligand-receptor clustering event is the most important step in leukocyte adhesion and spreading on endothelial cells. Intercellular adhesion molecule-1 (ICAM-1) has been shown to enhance leukocyte adhesion, but the molecular event during the process of adhesion is unclear. To visualize the dynamics of ICAM-1 movement during adhesion, we have engineered stable Chinese hamster ovary cell lines expressing ICAM-1 fused to a green fluorescent protein (IC1_GFP/CHO) and examined them under the fluorescence microscopy. The transfection of IC1_GFP alone in these cells was sufficient to support the adhesion of K562 cells that express $\alpha$L$\beta$2 (LFA-1) integrin stimulated by CBR LFA-1/2 mAb. This phenomenon was mediated by ICAM-1-LFA-1 interactions, as an mAb that specifically inhibits ICAM-1-LFA-1 interaction (RRl/l) completely abolished the adhesion of LFA-1* cells to IC1_ GFP/CHO cells. We found that the characteristic of adhesion was followed almost immediately (~10 min) by the rapid accumulation of ICAM-1 on CHO cells at a tight interface between the two cells. Interestingly, ICI_GFP/CHO cells projected plasma membrane and encircled approximately half surface of LFA-1+ cells, as defined by confocal microscopy. This unusual phenomenon was also confirmed on HUVEC after adhesion of LFA-1* cells. Neither cytochalasin D nor 2,3-butanedione 2-monoxime an inhibitor of myosin light chain kinase blocked LFA-1-mediated ICAM-1 clustering, indicating that actin cytoskeleton and myosin-dependent contractility are not necessary for ICAM-1 clustering. Taken together, we suggest that leukocyte adhesion to endothelial cells induces specialized form of ICAM-1 clustering that is distinct from immunological synapse mediated by T cell interaction with antigen presenting cells.

  • CONSTANS (CO) induces the expression of FLOWERING LOCUS T (FT) in the photoperiodic pathway, and thereby regulates the seasonal timing of flowering. CO expression is induced and CO protein is stabilized by FLAVIN-BINDING KELCH REPEAT F-BOX PROTEIN 1 (FKF1) in the late afternoon, while CO is degraded by CONSTITUTIVE PHOTOMORPHOGENIC 1 (COP1) during the night. These regulatory cascades were thought to act independently. In our study, we investigated the relationship between FKF1 and COP1 in the regulation of CO stability in response to ambient light conditions. A genetic analysis revealed that FKF1 acts as a direct upstream negative regulator of COP1, in which cop1 mutation is epistatic to fkf1 mutation in the photoperiodic regulation of flowering. COP1 activity requires the formation of a hetero-tetramer with SUPPRESSOR OF PHYA-105 (SPA1), [$(COP1)_2(SPA1)_2$]. Light-activated FKF1 has an increased binding capacity for COP1, forming a FKF1-COP1 hetero-dimer, and inhibiting COP1 homo-dimerization at its coiled-coil (CC) domain. Mutations in the CC domain result in poor COP1 dimerization and misregulation of photoperiodic floral induction. We propose that FKF1 represses COP1 activity by inhibiting COP1 dimerization in the late afternoon under long-day conditions, resulting in early flowering.

Enhanced Degradation of TNT and RDX by Bio-reduced Iron Bearing Soil Minerals

  • We demonstrated that reductive degradation of 2,4,6-Trinitrotoluene (TNT) and hexahydro-1,3,5-trinitro-1,3,5-triazine (Royal Demolition Explosive, RDX) can be enhanced by bio-reduced iron-bearing soil minerals (IBSMs) using Shewanella putrefaciens CN32 (CN32). The degradation kinetic rate constant of TNT by bio-reduced magnetite was the highest (0.0039 $h^{-1}$), followed by green rust (0.0022 $h^{-1}$), goethite (0.0017 $h^{-1}$), lepidocrocite (0.0016 $h^{-1}$), and hematite (0.0006 $h^{-1}$). The highest rate constant was obtained by bio-reduced lepidocrocite (0.1811 $h^{-1}$) during RDX degradation, followed by magnetite (0.1700 $h^{-1}$), green rust (0.0757 $h^{-1}$), hematite (0.0495 $h^{-1}$), and goethite (0.0394 $h^{-1}$). Significant increase of Fe(II) was observed during the reductive degradation of TNT and RDX by bio-reduced IBSMs. X-ray diffraction and electron microscope analyses were conducted for identification of degradation mechanism of TNT and RDX in this study. 4-amino-dinitrotoluene were detected as products during TNT degradation, while Hexahydro-1-nitroso-3,5-dinitro-1,3,5-triazine, Hexahydro-1,3-dinitroso-5-nitro-1,3,5triazine, and Hexahydro-1,3,5-trinitroso-1,3,5-triazine were observed during RDX degradation.

  • Silica sulfuric acid (SSA)를 이용하여 ethylenediamine (EDA)과 o-phenylenediamine (o-PDA)을 2-(4-methylthio benzenesulfonyl)-1,3-dimethyl/1-methyl-3-phenyl/1,3-diphenyl/1-methyl-3-ethoxypropane-1,3-dione 3a-d과의 헤테로고리화 반응을 통하여 좋은 생리활성을 나타내는 1H-1,4-diazepines 4a-d과 3H-1,5-benzodiazepines 5a-d을 좋은 수율로 합성하였다. 이 반응에서 ${\beta}$-diketones/${\beta}$-ketoesters 3a-d는 4-methylthiobenzenesulfonyl chloride 1과 다양한 ${\beta}$-diketones/${\beta}$-ketoesters 2a-d과의 축합반응으로 합성하였으며, 합성한 4a-d와 5a-d 화합물들에 대해서 fantimicrobial, antifungal 및 anthelmintic 활성을 측정하였다.

1-Methylcyclopropene(1-MCP) 처리가 오이의 신선도 유지에 미치는 영향 (Effect of 1-Methylcyclopropene(1-MCP) on the Postharvest Life and Fruit Quality of Cucumber)

  • 1-MCP(1-methylcyclopropene) 처리에 의한 오이 'Baekdadaki'의 수확 후 품질에 관한 연구를 수행하였다. 1-MCP의 농도별 250ppb, 500ppb, 1,000ppb 처리 후 상온과 저온($10^{\circ}C$)으로 나누어 저장하면서 생리적 특성변화 및 호흡율, 에틸렌 발생율을 조사하였다. 물리화학적 특성변화로 생체중과 경도는 상온, 저온 모두 저장 중 감소하였고 1-MCP 처리중에는 저온의 500ppb 에서 가장 적은 감소율을 보였다. 노화의 지표인 과피 b값은 상온괴 저온 모두 1-MCP 250, 1,000ppb에서 가장 높은 증가율을 보였고 저온저장 15일째에 황화와 함께 과실 무름 현상이 나타났다. 오이의 저온저장 15일 동안 1-MCP 500ppb 처리에서 가장 낮은 부패율을 나타내어 상품성을 상대적으로 높게 유지하였다. 호흡율은 1-MCP 250ppb 처리에서 높은 증가율을 보였고, 에틸렌 발생량은 상온 저장된 1,000ppb 에서 저장 6일 동안 가장 높은 증가율을 보였다. 오이 'Baekdadaki' 품종의 수확 후 품질은 1-MCP 500ppb 농도에서 가장 좋은 품질을 유지하였고 여러 농도의 1-MCP 처리에서 에틸렌 생성이 억제되지는 않았다.
