Factors Affecting Color Loss in Plum Nectar (자두 Nectar의 퇴색(褪色)에 미치는 영향인자(影響因子))

  • The effects of various factors on the color stability of plum nectar were studied. The data showed that pH and temperature were the most important factors affecting the rate of color loss. The concentration of ascorbic acid affected the rate of color loss in plum nectar. Although the color loss could be influenced by the presence of sugars and their degradation products such as 5-hydroxymethyl-2-furfural(HMF), HMF did not accumulate in sufficient amounts during the storage in the plum nectar to affect appreciably the degradation rate. The pigment was very unstable in the fermented juice and its. spectrophotometric spectrum was changed differently from that of natural plum color material. Materials present in plum nectar itself apparently have an appreciable effect on the rate of color loss.

In vitro evaluation of nano zinc oxide (nZnO) on mitigation of gaseous emissions

  • Background: Enteric methane ($CH_4$) accounts for about 70% of total $CH_4$ emissions from the ruminant animals. Researchers are exploring ways to mitigate enteric $CH_4$ emissions from ruminants. Recently, nano zinc oxide (nZnO) has shown potential in reducing $CH_4$ and hydrogen sulfide ($H_2S$) production from the liquid manure under anaerobic storage conditions. Four different levels of nZnO and two types of feed were mixed with rumen fluid to investigate the efficacy of nZnO in mitigating gaseous production. Methods: All experiments with four replicates were conducted in batches in 250 mL glass bottles paired with the ANKOM$^{RF}$ wireless gas production monitoring system. Gas production was monitored continuously for 72 h at a constant temperature of $39{\pm}1^{\circ}C$ in a water bath. Headspace gas samples were collected using gas-tight syringes from the Tedlar bags connected to the glass bottles and analyzed for greenhouse gases ($CH_4$ and carbon dioxide-$CO_2$) and $H_2S$ concentrations. $CH_4$ and $CO_2$ gas concentrations were analyzed using an SRI-8610 Gas Chromatograph and $H_2S$ concentrations were measured using a Jerome 631X meter. At the same time, substrate (i.e. mixed rumen fluid+ NP treatment+ feed composite) samples were collected from the glass bottles at the beginning and at the end of an experiment for bacterial counts, and volatile fatty acids (VFAs) analysis. Results: Compared to the control treatment the $H_2S$ and GHGs concentration reduction after 72 h of the tested nZnO levels varied between 4.89 to 53.65%. Additionally, 0.47 to 22.21% microbial population reduction was observed from the applied nZnO treatments. Application of nZnO at a rate of $1000{\mu}g\;g^{-1}$ have exhibited the highest amount of concentration reductions for all three gases and microbial population. Conclusion: Results suggest that both 500 and $1000{\mu}g\;g^{-1}$ nZnO application levels have the potential to reduce GHG and $H_2S$ concentrations.

Studies on the Ginseng Tea using Spore-forming Lactic Acid Bacteria (유포자성 유산균을 이용한 인삼차 개발에 관한 연구)

  • In order to develop ginseng tea powder with spore forming lactic acid bacteria Lactobacillus sporogenes was used. In the jar fermentor experiment under optimal culture conditions, the number of spore of L. sporogenes reached about $20{\times}10^8\;CFU/mL$ and sporulation rate was 97%. Granulated ginseng tea was made from glucose 7 kg, lactose 2 kg, ginseng extract 1 kg and spores 5 g $(5200{\times}10^8\;CFU/g)$. In the treatment of artificial gastric juice (pH 3.0) for 4 h and artificial bile for 8 h, the survival rate of spores in the granulated ginseng tea was 55.4% and 90.0% respectively. The spores survived 77.6% after incubation for 20 min in boiled water. Its storage stability was about 75% for 12 months at room temperature.

  • Electron micrographs of muscle of plaice killed instantly by spiking at the head clearly showed the A-band, I-band, Z-line and M-line in muscle strips, whereas these bands could not be distinguished from each other in electrically stimulated plaice muscle strips. As the electrical stimulation time increased, the continuity of Z-line disrupted rapidly. Electron microscopic observation showed that sarcoplasmic reticulum (SR) in unstimulated plaice had natural triad structures between Z-line. However these structures were disrupted in the electrically stimulated sample. These structures were clearly observed after storage for 10 hrs at $5^{\circ}C$ from all the samples. In association with the $Ca^{2+}$ translocation, $Ca^{2+}$-pyroantimonate deposits were observed at the inner part of SR immediately after spiked the plaice, However, $Ca^{2+}$-pyroantimonate deposits in electrically stimulated plaices were observed in the muscle strips and this phenomenon was clearly observed when electrical stimulation was prolonged.

  • 2018년 5월 전라북도 남원군에서 재배하여 수확 후 $0^{\circ}C$ 저장고에서 예냉을 마친 산마늘을 사용하였으며 외 포장 상자는 골판지로 구성된 통기공이 없는 골판지상자 1kg용을 사용하였다. 골판지상자와 함께 내부 포장으로 사용하기 위한 내포장 필름은 $20{\mu}m$ 두께의 HDPE으로 된 필름 봉지, 흡습종이는 파라핀왁스 코팅 종이($32{\times}35cm$, 태영산업)를 이용하여 조합 처리하고 국내에서 개발한 기능성 필름2종으로 포장처리는 총 4가지로 (1) PE(관행) (2) PE+흡습지 (3) 기능성필름1($30{\times}58cm$, 기능성물질 1%+40,000OTR, 제조사: 신영산업사) (4) 기능성필름2($30{\times}58cm$, Key fresh, 제조사: 씨앤케이프로팩(주)) 각 처리당 1kg 씩의 산마늘 잎을 포장하였다. 실험은 포장 처리 후 상온상태로 전북 완주군 실험실까지 수송하였으며 실험실 도착 후 상온 실험실에 두고 2일과 6일 후 중량감소율, 색도, 엽록소, 손실률 및 종합선도 등 품질특성 변화를 조사하였고 각 포장방법별 온도와 상대습도 변화를 보기 위하여 산마늘 잎이 포장된 상자 내부 중앙 부분에 디지털온습도기록계(SP-2000-20R)를 고정한 후 측정하였다. 대조구로 사용한 PE필름 단용으로 포장한 산마늘 잎은 상온저장 6일 경과 후 중량 소율이 1.7%, PE필름+신선지 처리는 1.0%, 기능성필름1과 기능성필름2는 거의 중량감소가 없어 1% 미만의 변화를 보였다. 산마늘 잎의 SPAD 값에 의한 엽록소 수치와 색도를 측정한 명도($L^*$)와 Hue angle 값의 변화는 기능성필름1과 기능성필름2가 유사하게 높게 유지되었고 반면에 PE필름은 저장 2일 후 부터 값이 감소하기 시작하여 6일 후 크게 감소하였다. 저장 6일 후 기능성필름1과 기능성필름2가 외관품위에서 높은 평가를 받아 상품성을 유지하고 있었으나 PE필름 처리와 PE필름+흡습지 처리는 상품성을 상실한 상태였다. 이러한 경향은 건전율에 있어서도 유사하였으며 기능성필름1(다공성 나노 물질이 혼입되어 있으며 산소투과율을 $40,000cc/m2{\cdot}day{\cdot}atm$으로 조성한 필름)이 상온저장 6일 후 상품성을 유지하는 건전율이 83%로 가장 높았다.

  • This study was conducted to determine the effects of gamma and e-beam irradiation on the quality of minced pork and pork patties. Each sample was irradiated at 5 to 20kGy, and its quality characteristics were then evaluated during storage at 30. The results of the total bacterial populations in the minced-pork and pork patty samples showed that the antimicrobial effect of gamma irradiation was superior to that of e-beam irradiation. The 2-thiobarbituric acid reactive substances (TBARS) value of all the samples significantly increased (p<0.05) as the irradiation dose and storage period increased. In addition, the gamma-irradiated (GI) samples had higher (p<0.05) TBARS values than the e-beam-irradiated (EI) samples. The volatile basic nitrogen contents of the GI samples were lower (p<0.05) than those of the EI samples. The color values, such as the $L^*$(brightness), $a^*$(redness), and $b^*$(yellowness) of the minced pork and pork patties, were increased (p<0.05) by irradiation. The hardness and sensory properties, such as the color, chewiness, taste, and overall acceptability of the pork patties, were decreased when the irradiation dose increased, and the hardness and sensory scores of the GI samples were lower than those of the EI samples.

Dietary Effects of an Animal Fat with High Free- Fatty Acid Content on Feed Utilization and Meat Quality in Broilers (고산가 동물성 지방의 첨가가 육계의 사료이용성과 육질에 미치는 영향)

  • Present experiment was undertaken to compare the effects of two dietary fats whose free fatty acid content was quite different on performances and on meat quality of broilers. Yellow grease or animal fat (trade name) containing 15 or 38.6% free fatty acid, respectively, was added to the broiler starter and finisher diets at levels of 3.0 and 5,0%, respectively. A total of 108, day old, male Maniker(Chunho) broiler chicks was alloted to 3 dietary treatments with 3 replications per treatment and 12 chicks per replication Though control diet was not supplemented with fat, the three dietary groups were made isocaloric and isonitrogenous. All the chicks were ad libitum fed the test diets for 8 weeks. Feed intake and body weight were measured every other week. Shank color was measured at the end of feeding trial by Roche Color Fan. To evaluate meat quality, 7 chicks of mean body weight were selected from each treatment group after the trial. Measurements were made for abdominal fat content, organoleptic scores for thigh and breast, and for contents of total lipids, free fatty acids, iodine values and peroxide values of the breasts. During the period from 0 to 4 weeks of age, the broilers fed the diets added with fats performed the same as those fed the control diet. However, the body weight gam (25%), feed intake (10.8%) and feed efficiency (11.3%) of chicks fed the fat-supplemented diets, during the finisher period (5-8 weeks), appeared significantly improved compared to those of control group (p<0.05). During overall period of 8 weeks, body weight gain, feed intake, and feed efficiency of chicks fed the diets added with fats were, on the average, 16.2. 8.8 and 6.8%, respectively, better than, those of control Monwhile the performance between the chicks fed diets added with the two different fat sources appeared to be of the same tendency. Though the shank pigmentation was not statistically different among the treatments, the chicks fed the fat-supple ented diets tended to have slightly less pigments than the control. Organoleptic scores of thigh or breast of chicks fed the animal fat diet were of the same range as those of the other two groups. The contents of total lipids, free fatty acids, iodine values and peroxide values of breasts from broilers fed the various diets appeared to be in the same ranges among the treatments. The values for control, yellow grease and animal fat groups were 7.77, 6.66 and 6.32% for total lipids, 9.23, 9.7 and 9.31mg oleic acid/g fat for free fatty acids, 65.36, 63.89 and 59.25g/ 100g fat for iodine values, and 9.62, 10.46 and 8.79 meq/kg fat for peroxide values, respect vely. Changes of free fatty acids contents of breast during a storage for 10 da s at 4$^{\circ}C$ were also not different among the dietary groups. From the observations n. this experiment, it seems possible to conclude that the animal fat containing 38.6% free fatty acid can be used as efficiently as yellow grease in broiler diets without any adverse effects on meat quality.

Effect of Chitosan Treatment on the Quality of Dried Lavers (건조김 제조시 키토산처리가 품질에 미치는 영향)

  • Chitosan was used to treat lavers during the production of dried lavers. In the procedure of washing the harvested lavers, the lavers were treated with various solutions of 0.1 and 0.01% of 30 cp chitosan and of 0.005 and 0.04% of 90 cp chitosan. Then treated lavers were processed as usual for the production of dried lavers. Significant differences between chitosan-treated and non-treated lavers were observed in flavor, in total microbial counts, and in the number of coliform microorganisms. Off-flavor of dried lavers was reduced significantly by treating with chitosan. Total microbial count was $3.7{\times}10^{4}$ cells/g in the non-treated dried lavers, but they were $5.7{\times}10^{3}$ cells/g and $2.1{\times}10^{3}$ cells/g, respectively, in the dried lavers treated with 0.01% and 0.1% of 30 cp chitosan. The number of coliform microorganisms was 240 cells/g in non-treated dried lavers. However, coliform microorganisms were not detected in the dried lavers treated with 0.1% of 30 cp chitosan. When the dried lavers were used to make Kimbab (rice rolled with laver), the number of total microorganisms increased very slowly during storage time at $20^{\circ}C$ in the Kimbab rolled with chitosan-treated dried lavers compared to that in the Kimbab rolled with non-treated dried lavers.

  • File fish Navodon modestus was dehydrated in cabinet type hot-sir drier at $48-50^{\circ}C$ for 11 hours and also yellowfin puffer Fugu xanthopterum was dried in open air at $26-28^{\circ}C$ for 30 hours. Nucleotides and their related compounds were collected by extraction with cold perchloric acid and their amounts were determined by anion exchange column chromatography. The contents of ADP, IMP, ATP and hypoxanthine in fresh file fish muscle were 22.9, 12.1, 4.9, and 3.2 ${\mu}mole/g,$ dry wt. respectively. AMP and inosine were 0.9 ${\mu}mole/g,$ dry wt. equally. In fresh yellowfin puffer muscle, the contents of ADP, ATP, AMP, inosine and hypoxanthine were 25.6, 2.4, 1.6, 0.3, 0.6, and 0.4 ${\mu}mole/g,$ dry wt. respectively. In the case of file fish, ADP and ATP tended to degrade rapidly during hot-air dehydration. The contents of IMP were decreased slightly while AMP and inosine were increased. And another case of yellowfin puffer, ADP also tended to degrade rapidly during sun drying while AMP, IMP, inosine and hypoxanthine were increased. Especially, in both case of file fish and yellowfin puffer, inosine was increased twenty five and thirty five times during drying respectively.

Estimation of storability for Korean apples (Malus domestica) using Md-ACS1 and Md-ACO1 DNA marker (Md-ACS1 및 Md-ACO1 분자표지를 이용한 국내육성사과의 저장성 예측)

  • Apple (Malus domestica) is a climacteric fruit because of its high respiration and ethylene production. Ethylene affects the fruit by decreasing its quality and storability. Md-ACS1 and Md-ACO1 genes are involved in ethylene biosynthesis in apple; the Md-ACS1-2 and Md-ACO1-1 alleles are associated with low ethylene production. We conducted an analysis to study Md-ACS1 and Md-ACO1, and to examine ethylene production and softening rate of fruit at room temperature ($20^{\circ}C$) storage in 'Fuji (FJ)', 'Golden Supreme (GS)', and 5 cultivars of Korean apples ('RubyS (RS)', 'Hongro (HR)', 'Arisoo (AS)', 'Summer King (SK)', 'Greenball (GB)'). The result showed that an increase in the number of the alleles (ACS1-2, ACO1-1) decreased the ethylene production and softening rate. The presence of ACS1-1/1, ACO1-1/2 was confirmed in GS and the highest ethylene production and softening rate was observed. Ethylene production and softening rate of SK and GB expressing ACS1-1/2, ACO1-1/2 were higher than that of HR and AS, expressing ACS1-2/2, ACO1-1/2, but lower than GS. FJ with ACS1-2/2, ACO1-1/1 showed the lowest ethylene production and softening rate among all cultivars except RS. The Md-ACS1 and Md-ACO1 DNA markers could potentially be used to estimate storability and applied in marker assisted selection the improve the efficiency of apple breeding.
