Feasible monitoring of the inhibitory effects of free NH3 on NO2- oxidation

  • This study investigated nitrite ($NO_2{^-}$) accumulation due to FA (Free Ammonia: $NH_3$) inhibition in an anaerobic-aerobic-anoxic (AOA) process reactor to mainly treat wastewater containing 302-610 mg/L of $NH_3/NH_4{^+}-N$. Based on an experimental operation focusing on the nitrification, it was observed that $NO_2{^-}$ was accumulated in the aerobic nitrification zone as pH increased, due to inhibition of $NO_2{^-}$ conversion to $NO_3{^-}$ by FA. This result implied FA inhibition to NOB ($NO_2{^-}$-Oxidizing Bacteria) for converting $NO_2{^-}$ to $NO_3{^-}$. The objective of this study is to develop a feasible monitoring procedure for early detection of the FA inhibition toward $NO_2{^-}$ accumulation and poor nitrification. Thus, in order to rapidly assess FA concentrations, an $NH_3$ probe was utilized to measure $NH_3$ concentrations together with applying a simple model prediction using the measured $NH_4{^+}$ concentrations, the Henry's law constant of $NH_3$ and measured pH. The predictive model $NH_3$ levels were verified by a good correlation (89%) with the corresponding measured data, but the model prediction underestimated FA concentrations at less than 7.4 and a little overestimated at pH above 7.5. Interestingly, accumulated $NO_2{^-}$ levels were roughly correlated with FA levels that were observed at delayed time points. This reflects the detected FA levels can be good indicators of $NO_2{^-}$ levels with some delayed time. $NO_2{^-}$ accumulation started at measured FA concentrations of higher than approximately 3 mg/L and ceased below that FA level.

  • In order to study the influence of nitrate reduction to ionic balance in tissue of tobacco plant, differneces in amounts of those cations and anions were determined and these balances were compared with contents of organic acids and activities of nitrate reductase, while they were fertilized with different nitrogen sources ($NO_3-N$, $NH_4-N$, $NO_3+NH_4-N$) in water culture. The results of studies are summerized as follows; 1. Total uptake of inorganic cations was the highest in nitrate-fed plants, whereas that of inorganic anions showed the highest level in the plants grown with the mixture ($NO_3+NH_4$). The amounts of inorganic cations and anions were comparable in two treatments containing $NH_4-N$, but in plants treated with nitrate only had much higher level of inorganic cations than others. 2. Deficiency in the amount of inorganic anions in nitrate-fed plants was balanced with organic acids, dominantly with malic acid among them. But another two $NH_4-N$ fed plant sustained equilibrium between inorganic cations and anions. 3. Reduction of nitrate was raised in tissues of nitrate-fed plants. By the results of nitrate reduction, cations maintained equilibrium with nitrate ion were let loose. The replacement of inorganic anions with organic anions could be a compensation process for the loss of uptaken nitrate ions which must be reduced to be incorporated into organic N compounds.

  • The elution mechanism of rare earth elements in cation exchange resin which was substituted with $NH_4^+,\;Zn^{2+}\;or\;Al^{3+}$ as a retaining ion had been investigated. Rare earths or rare earths-EDTA complex solution was loaded on the top of resin bed and eluted with 0.0269M EDTA solution. When the rare earth-EDTA complex was adsorbed on the $Zn^{2+}\;or\;Al^{3+}$ resin form, retaining ion was complexed with EDTA and liberated rare earths was adsorbed in the resin again. Adsorbed rare earths in resin phase could be eluted by the complexation reaction with EDTA eluent. On $NH_4^+$ resin form, the rare earth-EDTA complex which had negative charge could not adsorbed on the cation exchange resin because the complexation reaction between $NH_4^+$ and EDTA was impossible. So the elution time was much shorter than in $Zn^{2+}\;or\;Al^{3+}$ resin form. When the rare earths solution was loaded on the $Zn^{2+},\;Al^{3+}$ resin form bed, rare earths was adsorbed in the resin and the retaining ion was liberated. Adsorbed rare earths in resin bed was exchanged by EDTA eluent forming rare earths-EDTA complex, and eluted through these processes. On $NH_4^+$ resin form, rare earths loaded was adsorbed by exchange reaction with $NH_4^+$. As the EDTA eluent was added, rare earths was liberated from resin forming negatively charged rare earth-EDTA complex and eluted without any exchange reaction. So the elution time was greatly shortened and there was no metallic ion except rare earths in effluent. When the $Zn^{2+}\;and\;Al^{3+}$ was used as retaining ion, the pH of efflent was decreased seriousely because the $H^+$ liberated from EDTA molecule.

  • Ion exchange equilibria in bulk and rhizosphere soil collected from peach seedlings were studied to find exchange equations that could be used in chromatographic models dealing with movement and distribution of fertilizer ammonium and exchangeable cations in soil profiles. Soil samples were equilibrated with mixtures of $NH_4Cl$, KCI, and $CaCl_2$ solutions and then extracted with $Sr(NO_3)_2$ solution to determine exchangeable cation compositions at equilibrium. Exchange data were fitted to Vanselow's, Gapon's, and Kerr's equations, but those formulations did not adequately describe the equilibria. An empirical equation of the form : ${\frac{\alpha_i^m}{a_j^n}}=K{\frac{(iX)^{mPi}}{(jX)^{nPj}}}$ which has an exponent on each of the exchangeable cation concentrations could describe the equilibria very well over the range of treatments. In this equation ${\alpha}^i$ and ${\alpha}^j$ are activities of cation i and j with valences m and n respectively. (iX) and (jX) are concentrations of exchangeable cations. Mole or equivalent fractions can be considered as the exchangeable ion concentration unit. Arbitrary constants $P_i$ and $P_j$, and distribution coefficient K can be found with multiple regression for the logarithmic form of the equation.

  • As the concentration of nitrogen in the sewage flowing into the sewage treatment plant increases due to urbanization and industrialization, the degree of adverse effects such as eutrophication and toxicity to the aquatic ecosystem is also increasing. In order to treat sewage containing high concentration of nitrogen, various studies on the biological nitrogen removal process are being conducted. Existing biological nitrogen removal processes require significant costs for supplying oxygen and supplementing external carbon sources. In this respect, as a high-level nitrogen removal process with economic improvement is required, an anaerobic ammonium oxidation process (ANAMMOX), which is more efficient and economical than the existing nitrification and denitrification processes, has been proposed. The purpose of this study is to confirm the stability of the ANAMMOX process in the water treatment process and to derive the ratio of ammonia nitrogen (NH4+) to nitrite nitrogen (NO2-) for the implementation of the mainstream ANAMMOX process. A laboratory-scale Mainstream ANAMMOX reactor was operated by applying the ratio calculated based on the substrate ratio suggested in the previous study. In the initial range, the removal efficiency of NH4+ was 58~86%, and the average removal efficiency was 70%. In the advanced range, the removal efficiency of NH4+ was 94~99%, and the average removal efficiency was 95%. As a result of the study, as the NH4+/NO2- ratio increased, the stability of the mainstream ANAMMOX process was secured, and it was confirmed that the NH4+ removal efficiency and the total nitrogen (TN) removal efficiency increased. As a result, the results of this study are expected to be used as basic data in the application of the ANAMMOX process in the mainstream.

  • This study was focussed to compare the concentrations of the volatile oil components of flue-cured tobacco grown by two types of nitrogen-ammonium and nitrate-under vinyl mulching. The results were summarized as follows; 1. The content of nicotine was lower and reducing sugars degraded from starch was higher content in $NO_3-N$ application than $NH_4-N$. 2. The concentrations of furfural, benzyl alcohol, and phenethyl alcohol in the cured leaf were not different between $NH_4-N$ and $NO_3-N$ treatments. 3. Some of the important flavour components-linalool, solanone, damascenone, neophytadiene, and oxyolanone + megastigmatriene-one - had much higher concentrations in $NO_3-N$ than $NH_4-N$ application. 4. In conclusion, nitrate nitrogen was more favorable to decrease nicotine and to increase some important flavour components of cured leaf than ammonium nitrogen.

Annealing effects in $N_2$ and $NH_3/N_2$ ambient are estimated with dry $O_2$ and $NH_4$ oxidation($NH_3$ added in dry $O_2$ oxidation) method. C;em'rated defects in dry $O_2$ oxidation are lengthened according to oxidation time. but any defects in $NH_3$ oxidation are not found. Dry oxidation, after $NH_3$ oxidation as an initial oxidation. lias the defect -removing effect at the interface of Si -$SiO_2$. After dry or $NH_3$. oxidation. the annealmg 7.5% $NH_3/N_2$ ambient brings out gettering effect of OSF. The annealing in 7.5% $NH_3/N_2$ ambient for NI L oxidation method decreaSE,s $NH_3$ length of OSF about 20 % compared with dry oxidation method. Tlw feature of OSF is pit type, the gettering is directed to (011) plane for (111) plane. and OSFs are etched following to 110) directIon.

  • Microorganisms capable of accumulating poly-p-hydroxybutyric acid(PHB) were isolated from soil by enrichment culture technique. Among them, the strain designated as FL-027 had high PHB productivity and was identified as Alcaligenes. The optimal medium composition for cell growth was 8.0 $g/\ell$ of fructose and 3.0 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 5.04 at pH 7.0 and $30^{\circ}C$. To investigate the optimal conditions for the PHB accumulation, we divided the process into two stages; the first stage for the growth of the cell in nutrient-rich medium and the second stage for the PHB accumulation in nutrient-deficiency medium. The optimal conditions for PHB accumulation were 8.0 $g/\ell$ of fructose and 0.25 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 60 at pH 6.5 and $30^{\circ}C$. PHB accumulation was stimulated by deficiency of nutrients such as $NH_4^+$, $Ca^{2+}$, $SO_4^{2+}$ in medium. Among them. $NH_4^+$ deficiency was chosen because of its effectiveness. We found the inhibition of cell growth by fructose in batch culture. In order to keep the fructose concentration at an optimal level, intermittent feeding fed-batch culture was employed, and the cell concentration as high as 10.83 $g/\ell$ whose PHB content was responsible for 43% of the dry cell weight. The purified PHB was identified as homopolymer of 3-hydroxybutyric acid by using IR and $^1H-NMR$.

Effect of NH4F Flux on the Characteristics of Barium Strontium Silicate Phosphor Particles (NH4F 융제가 바륨 스트론튬 실리케이트계 형광체의 특성에 미치는 영향)

  • [ $Ba_{1.5}Sr_{0.5}SiO_4:Eu$ ] phosphor particles with high photoluminescence intensity under long wavelength ultraviolet were prepared by spray pyrolysis. We investigated the effect of $NH_4F$ flux added into starting solution on the morphology and photoluminescence intensity of $Ba_{1.5}Sr_{0.5}SiO_4:Eu$ phosphor prepared by spray pyrolysis. $Ba_{1.5}Sr_{0.5}SiO_4:Eu$ phosphor particles prepared from starting solution containing $NH_4F$ flux had the maximum photoluminescence intensity at the post-treatment temperature of $1200^{\circ}C$ and the maximum photoluminescence intensity of $Ba_{1.5}Sr_{0.5}SiO_4:Eu$ phosphor particles prepared from the starting solution containing $NH_4F$ flux was $137\%$ of that of the phosphor particles prepared from the starting solution without flux material. $Ba_{1.5}Sr_{0.5}SiO_4:Eu$ phosphor particles prepared from starting solution containing $NH_4F$ flux had larger size and more aggregated morphology than those prepared from starting solution without flux material. The photoluminescence intensity of $Ba_{1.5}Sr_{0.5}SiO_4:Eu$ phosphor particles prepared from starting solution containing $NH_4F$ flux above $3wt.\%$ had high photoluminescence intensities. The addition amount of $NH_4F$ flux showing the maximum photoluminescence intensity was $12wt.\%$. The optimum amount of $NH_4F$ flux was $5wt.\%$ when we considered the morphological and photoluminescence characteristics of $Ba_{1.5}Sr_{0.5}SiO_4:Eu$ ohosphor particles prepared by spray pyrolysis.

Purification of Hydrogenase from Thiocapsa roseopersicina : Effect of Ammonium Sulfate Precipitation and Heat-Treatment (홍색 유황세균 Thiocapsa roseopersicina가 생산하는 수소생산 효소의 정제 : Ammonium sulfate 침전 및 열처리의 효과)

  • Effect of $(NH_4)_2SO_4$ precipitation and heat-treatment on hydrogenase which was extracted from the cytoplasmic fraction of the phototrophic purple sulfur bacterium Thiocapsa roseopersicina NCIB 8347 was studied. Crude enzyme extract was prepared by centrifugation($28,000{\times}g$, $400,000{\times}g$) after sonication of cells grown under photosynthetic condition for 96 hrs. Various conditions of $(NH_4)_2SO_4$ precipitation and heat-treatment were examined and the effect of protein concentration was analyzed by SDS-electrophoresis between the treatments. Optimum conditions for $(NH_4)_2SO_4$ precipitation and heat-treatment for evolution hydrogenase activity were 40-60% saturation and $60^{\circ}C$ for 20 min, respectively, which exhibited the specific hydrogenase activity of 0.78 U/mg-protein. Specific hydrogenase activity was decreased to 31.6% when the heat-treatment at $60^{\circ}C$ increased from 20 min to 5 hrs.
