천연 올레인산 기반 폴리아미드계 TPEs 단량체 합성 (Synthesis of Monomers for Polyamide-type TPEs from Oleic Acid)

  • 식물성 오일로부터 추출한 물질을 기반으로 하여 열가소성 탄성체(thermoplastic elastomer, TPE) 중에서도 뛰어난 물성을 가진 것으로 잘 알려진 폴리아미드계 TPE(polyamide-type TPE, TPAE)의 단량체 합성공정을 개발하였다. 대다수의 식물성 오일에 함유되어 있는 불포화 지방산으로 잘 알려진 올레인산을 사용하여 TPAE의 단량체 6종($C_9$, $C_{10}$, $C_{11}$ ${\omega}$-amino acid 와 ${\alpha}$, ${\omega}$-dicarboxylic acid)을 좋은 수율로 합성하였다.

Identification of ${\omega}$-Aminotransferase from Caulobacter crescentus and Sitedirected Mutagenesis to Broaden Substrate Specificity

  • A putative ${\omega}$-aminotransferase gene, cc3143 (aptA), from Caulobacter crescentus was screened by bioinformatical tools and overexpressed in E. coli, and the substrate specificity of the ${\omega}$-aminotransferase was investigated. AptA showed high activity for short-chain ${\beta}$-amino acids. It showed the highest activity for 3-amino-n-butyric acid. It showed higher activity toward aromatic amines than aliphatic amines. The 3D model of the ${\omega}$-aminotransferase was constructed by homology modeling using a dialkylglycine decarboxylase (PDB ID: 1DGE) as a template. Then, the ${\omega}$-aminotransferase was rationally redesigned to increase the activity for 3-amino-3-phenylpropionic acid. The mutants N285A and V227G increased the relative activity for 3-amino-3-phenylpropionic acid to 3-amino-n-butyric acid by 11-fold and 3-fold, respectively, over that of wild type.

  • ${\omega}$-아미노산인 ${\beta}$-alanine과 ${\gamma}$-aminobutyric acid의 해리상수를 전도도법을 이용하여 온도($20{\sim}40^{\circ}C$)와 압력($1{\sim}2500$ bar)을 변화시키면서 측정하였다. 각각 아미노산의 두 가지 해리상수는 온도가 증가하면 증가하지만 압력이 증가하면 $K_1$은 증가하지만 $K_2$는 감소하였다. 해리상수를 이용하여 여러 가지 열역학적 성질들을 계산하고 이들로부터 해리반응의 특성을 알아보았다. ${\omega}$-아미노산의 해리상수를 해리기와 치환기간의 거리의 상관관계로 알아보았다. 반응특성을 알아보기 위해 Hammett 법칙을 적용하여 반응의 치환기 상수와 압력에 따른 반응상수를 계산하였다. ${\omega}$-아미노산의 특성을 치환기 효과로 설명하고 반응유형을 반응상수로서 알아보았다.

타우린 복용이 정상 성인여성의 혈장 총지방산 및 인지질 지방산패턴에 미치는 영향 (Effects of Oral Taurine Supplementation on Plasma Total and Phospholipid Fatty Acid Patterns in Healthy Female Adults)

  • Effect of oral taurine supplementation on plasma total and phospholidpid -fatty acid profiles and their metabolism were evaluated in healthy female adults. Among twenty five female volunteers(23.6$\pm$0.3 years old ) participated in the taruine supplementation program(6g taurine /day), twenty four subjects succesfully completed the 2 week program , and only nine subjects continued to take taurine for another 2 weeks. Levels of plasma fatty acids and taruine were measured by gas-liquid chromatobraphy and an automated amino acid analyzer based on ion exchange chromatography, respectively. Plasma taurine concentration s of the subjects were 108. 7$\pm$3.4 , 184.2$\pm$8.2 and 235.9$\pm$77.0$\mu$emol/L at 0 , 2 and 4 weeks of taurine supplementation. Fatty acid compositions and elongation and desaturation indices of polyunsaturated fatty acids (PUFA) in plasma total lipids were not influenced by oral taurine supplementation. However, fatty acid compositions and their metabolism in plasma phospholipids were significantly affected by taurine supplementation in female adults. Compared to the values for 0 week, the percentage of saturated fatty acids (SFA) in plasma phospholipid was significantly lowered at 2 weeks, but elevated at 4 weeks of taurine supplementation. In contrast , the percentage of phospholipid PUFA significantly increased at 2 weeks and decreased at 4 weeks of taurine supplementation from to the values for 0 weeks. Foru weeks of oral taurine supplementation signifinatly elevated the eongation index(20 : 4$\omega$6 ⇒22 : 4 $\omega$6, p<0.01), and decreased the desaturation index (20 : 3 $\omega$6 ⇒20 : 4 $\omega$6 , p<0.01) of $\omega$6 fatty acids in plasma phospholipids. Plasma taurine concentration was positively correlated with the percentage of 14 : 0 fatty acids and the enlongation index o f$\omega$3 fatty acids(20 : 5 $\omega$3 ⇒22 : 5 $\omega$3), and thenegatively correlated with the percentage of 20 : 0 in plasma phospholipids. These results indicate that oral taurine supplementation for 4 weeks signidicantly elelvated the percentage of SFA, and lowered the percentage of PUFA in plasma phospholipids with no influence on plasm total fatty aicd composition in healthy female adults.

Arabidopsis thaliana로부터 지방산 불포화효소 유전자의 분석 (Characterization of a fad3 cDNA Encoding Microsomal Fatty Acid Desaturase from Arabidopsis thaliana)

  • Linoleic acid를 linolenic acid로 전환시키는 지방산불포화효소의 유전자(fad3)를 유채의 fad3 DNA probe를 이용한 plaque hybridization방법으로 $\lambda$ZAPII Arabidopsis thaliana cDNA expression library로부터 분리하였으며 1.8 kb-EcoRI DNA조각을 지니는 lambda clone을 pGEM7으로 subcloning하여 염기서열을 분석하였다. 그 결과로부터의 아미노산서열분석에 의하면 fad3 유전자는 open reading frame이 386개의 아미노산으로 이루어졌으며 44,075 Da의 분자량이 예측되고 있다. 엽록체의 $\omega$-3 지방산불포화효소(fad7)와 endoplasmic reticulum의 지방산불포화효소(fad2)와의 비교시 각각 70%와 58%의 유사성이 나타났다. 특히 82-151의 아미노산서열과 276-333의 아미노산지역은 보존성이 높았으며 이는 불포화지방산효소의 기능에 필수적인 지역으로 보여진다. 한편 대장균을 이용한 IPTG에 의한 Fad3단백질의 유도생성은 대장균의 생육에 독성효과를 나타내는 것으로 나타났다.

Divergent Process for C10, C11 and C12 ω-Amino Acid and α,ω-Dicarboxylic Acid Monomers of Polyamides from Castor Oil as a Renewable Resource

  • Polyamides have great potentials for diverse applications and the present production of their monomers mostly relies on resources from fossil fuel. Starting from undecylenic acid, a natural resource, we have developed both divergent and efficient processes for $C_{10}$, $C_{11}$ and $C_{12}$ ${\omega}$-amino acid and ${\alpha},{\omega}$-dicarboxylic acid monomers of the polyamides.

영양평가 전산프로그램을 이용한 시판김밥 및 김밥균형식단의 영양균형성 평가 - 지방산, 아미노산 및 항산화 비타민 균형을 중심으로 - (Evaluation on Nutritional Balance of Market-Kimbab and Nutritionally Adjusted Kimbab Menu by the Self-Developed Computer Program)

  • This research was designed to develop a computer program and evaluate the nutritional balances especially the balance of fatty acids, amino acids and antioxidant vitamins for convenience foods. The Korean convenience food, Kimbab purchased from markets was evaluated by using the self-developed computer program. Contents of calories, protein and calcium were lower$(1/3^{\circ}{\neq}1/2)$ than the recommended levels of Korean adult woman, and the carbohydrate/ protein/ fat(CPF) energy ratio was 70: 13: 17. The mean P/ M/ S ratio was 2.2/ 1.4/ 1 and that of ${\omega}6/\;{\omega}3$ fatty acids was 17.9/ 1, which was higher than the desirable ratio of $4{\sim}8/\;1$. Average essential amino acid balance of market-Kimbab samples was within the desirable range even though the absolute amount of protein was lower than the recommended level. Contents of antioxidant vitamins (A, C &E) were lower than recommended levels. Two kinds of nutritionally adjusted Kimbab menu were established by self-developed computer program. Some of major changes was adding food ingredients such as tuna fish and perilla leaf cooked with sesame oil and soybean oil to increase ${\omega}3$ series fatty acids. Some fruits and milk were also added to the menu. The adjusted CPF ratios was 63: 15: 22 and the new values for P/ M/ S and ${\omega}6/\;{\omega}3$ fatty acids ratios were 1.0/ 1.2/ 1/0 and 6.1/ 1 respectively. In sensory evaluation of two kinds of adjusted Kimbab, the taste and overall estimation scores were higher than unadjusted Kimbab. The computer program developed in this study might be used as a tool for the evaluation of nutritional balance of other convenience foods and menu planning.

제주도 용암 해수로 대량배양한 미세조류 멜로시라(Melosira nummuloides)의 성분 특성 (Compositional Characteristics of the Microalga Melosira nummuloides Mass-cultured Using Jeju Lava Seawater)

  • In this study, we analyzed the compositional characteristics of Melosira nummuloides mass-cultured using Jeju lava seawater. M. nummuloides showed the highest growth rate when cultured for 14 days at 17-20℃ and 15,000 lx. Proximate composition of raw-material (RM) and freeze-dried M. nummuloides (FM) showed high ash content (65-72%), while ethanol-extracted M. nummuloides (EM) had low ash and high lipid contents. The predominantly occurring mineral, Si, was 334 g/kg in RM and 269 g/kg in FM, but EM contained only 1.97 g/kg. The ratio of essential amino acids was similar in RM (38.93%) and FM (36.89%) lower in EM (17.83%), but branched chain amino acids required for muscle metabolism was high (63.40%). The polyunsaturated fatty acids of EM (34.74%) was 11% more than that in RM (23.81%), and the ratio of omega-6 to omega-3 fatty acids was 1:3.6. Furthermore, the concentration of total chlorophyll was higher by 5.15 times (62.32 mg/g) and that of fucoxanthin by 7.06 times (11.02 mg/g) in EM than in RM (12.11, 1.56 mg/g, respectively). The mass cultivation and filtration of M. nummuloides using lava seawater has high prospects of commercialization as a competitive bio silica, cosmetic and healthy functional food material.
