  • Acute trauma is a common cause of mortality in individuals aged < 40 years. As organ preservation has become important in treating trauma patients, the treatment is shifting from surgical management to non-operative management. A multidisciplinary team approach, including interventional radiology (IR), is essential for the optimal management of trauma patients, as IR plays an important role in injury evaluation and management. IR also contributes significantly to achieving the best clinical outcomes in critically ill trauma patients. This pictorial essay aims to present and summarize various interventional treatments in trauma patients requiring critical care.

Inhibition of Tumoral VISTA to Overcome TKI Resistance via Downregulation of the AKT/mTOR and JAK2/STAT5 Pathways in Chronic Myeloid Leukemia

  • Tyrosine kinase inhibitors (TKIs) have revolutionized the treatment landscape for chronic myeloid leukemia (CML). However, TKI resistance poses a significant challenge, leading to treatment failure and disease progression. Resistance mechanisms include both BCR::ABL1-dependent and BCR::ABL1-independent pathways. The mechanisms underlying BCR::ABL1 independence remain incompletely understood, with CML cells potentially activating alternative signaling pathways, including the AKT/mTOR and JAK2/STAT5 pathways, to compensate for the loss of BCR::ABL1 kinase activity. This study explored tumoral VISTA (encoded by VSIR) as a contributing factor to TKI resistance in CML patients and identified elevated tumoral VISTA levels as a marker of resistance and poor survival. Through in vitro and in vivo analyses, we demonstrated that VSIR knockdown and the application of NSC-622608, a novel VISTA inhibitor, significantly impeded CML cell proliferation and induced apoptosis by attenuating the AKT/mTOR and JAK2/STAT5 pathways, which are crucial for CML cell survival independent of BCR::ABL1 kinase activity. Moreover, VSIR overexpression promoted TKI resistance in CML cells. Importantly, the synergistic effect of NSC-622608 with TKIs offers a potent therapeutic avenue against both imatinib-sensitive and imatinib-resistant CML cells, including those harboring the challenging T315I mutation. Our findings highlight the role of tumoral VISTA in mediating TKI resistance in CML, suggesting that inhibition of VISTA, particularly in combination with TKIs, is an innovative approach to enhancing treatment outcomes in CML patients, irrespective of BCR::ABL1 mutation status. This study not only identified a new pathway contributing to TKI resistance but also revealed the possibility of targeting tumoral VISTA as a means of overcoming this significant clinical challenge.

Changes in Acidity and Distributions of the Vancomycin-Resistant Lactic Acid Bacteria in the Kimchi Fermented at Different Temperatures (발효 온도에 따른 김치의 산도 변화와 Vancomycin 내성 젖산균의 분포)

  • Chinese cabbage ('Baechu') Kimchi was fermented at the three different temperatures right after it was prepared. Samples were taken everyday for measuring bacterial populations, pH, and titratable acidity through the whole periods of fermentation up to 50 days. pH values and developed acidity were significantly affected by the fermenting temperatures of 4, 10, and $20^{\circ}C$, suggesting that different bacterial flora has been established by the temperatures exposed. The modified MRS agar containing vancomycin (300 $\mu$g/mL) was used for isolating the vancomycin-resistant LAB strains and 127 isolates were finally obtained. Of the LAB isolates, 13 isolates were subjected to the identification experiments based on the biochemical characteristics and the molecular-typing approach, an ITS-PCR, whether they belong to the genus Leuconostoc or not. The data obtained from API 50 CHL kit resulted that six isolates were identified as the members of Leuconostoc and six as Lactobacillus brevis strains except for a single isolate YKI 30-0401, which was not able to be identified because its biochemical traits were not matched to the database of API 50 CHL kit. It was noted that some isolates were distinct in a couple of some biochemical characteristics compared with those of the reference Leuconostoc species. To overcome the limitations experienced in the commercial identification products above, an ITS-PCR experiment was also conducted for the isolates, resulting that eight isolates belong to Leu. mesenteroides ssp. mesenteroides or dextranicum with a single band of 564 bp, and four to L. brevis strains. The ITS-PCR profiles clearly differentiated the closely-related LAB isolates for which same results were obtained by the biochemical method. This molecular approach, however, failed to produce the amplicons for the YKI 20-1003, leaving the strain unidentified. Judging from the identification data obtained in the Kimchi fermented at $4^{\circ}C$ or $10^{\circ}C$, Leuconostoc spp. including Leu. mesenteroides/dextranicum were likely predominant species in the earlier stage and L. brevis occurred at the high level through the whole period. By contrast, L. brevis, as one of the major flora, possibly lead the fermentation from the beginning in the Kimchi fermented at $20^{\circ}C$.}C$.

  • Donguisusaebowon, Kogchigo are wrritten by Dong-Mu. Dokhaengpyun of the Kogchigo has philosophy of Dong-Mu about Gi-In. In order to understand constitutional medicine of Dong-Mu, it is necessary to study about Bi-Bak-Tam-Ra'Ja(four types of man) in Dokhaengpyun. The results are summarized as follows : 1. In Dokhaengpyu, it is defined that In-Eui-Ye-JI - the nature of human - as In-Ja, Eui-Ja, Ye-Ja, Ji-Ja. This attribute is composed of Chung-Sin-Ip-Yong. We can know that this concept, that is Chung-Sin-Yeum-Hae'Ja, is relative to the Bi-Bak-Tam-Ra'Ja. 2. We can think and infer the relation of Sabujisim of Myeng-Ja and Bi-Bak-Tam-Ra'Ja from Bi-Bak-Tam-Ra'Bu that is attribute of In-Eui-Ye-ji'Ja. Bi-Bak-Tam-Ra'Ja can approach the behavior of Chung-Sin-Yeum-Hae'Ja, if they heard the attitude of YouHaHye and BaelYi which they have attribute of In-Eui-Ye-Ji'Ja. 3. They explain Tthat Bi-Bak-Tam-Ra-Jisim is quoted from SaBuJiSim of Dahak. Thinking that Bi-Bak-Tam-Ra-Jisim of Dokaengpyun and SaBujiSim of YuRiak, we know that there is no rule. So, it is difficult that we can infer the Simyok and make some pattern. 4. The relationship of Bi-Bak-Tam-Ra'Ja and In-Eui-Ye-Ji are different from the relationship of Dokhaegpyun, Sadanron, In-Eui-Ye-Ji of YuRiak and Bi-Bak-Tam-Ra'Ja. If we regard the In-Eui-Ye-Ji Sadan as the nature of human, many attritude can be possible and it may be different that apply a Sadan to the proper time and environment. Therefore, we have to be accepted change, as applying method than fixing idea.

Integrated Rotary Genetic Analysis Microsystem for Influenza A Virus Detection

  • A variety of influenza A viruses from animal hosts are continuously prevalent throughout the world which cause human epidemics resulting millions of human infections and enormous industrial and economic damages. Thus, early diagnosis of such pathogen is of paramount importance for biomedical examination and public healthcare screening. To approach this issue, here we propose a fully integrated Rotary genetic analysis system, called Rotary Genetic Analyzer, for on-site detection of influenza A viruses with high speed. The Rotary Genetic Analyzer is made up of four parts including a disposable microchip, a servo motor for precise and high rate spinning of the chip, thermal blocks for temperature control, and a miniaturized optical fluorescence detector as shown Fig. 1. A thermal block made from duralumin is integrated with a film heater at the bottom and a resistance temperature detector (RTD) in the middle. For the efficient performance of RT-PCR, three thermal blocks are placed on the Rotary stage and the temperature of each block is corresponded to the thermal cycling, namely $95^{\circ}C$ (denature), $58^{\circ}C$ (annealing), and $72^{\circ}C$ (extension). Rotary RT-PCR was performed to amplify the target gene which was monitored by an optical fluorescent detector above the extension block. A disposable microdevice (10 cm diameter) consists of a solid-phase extraction based sample pretreatment unit, bead chamber, and 4 ${\mu}L$ of the PCR chamber as shown Fig. 2. The microchip is fabricated using a patterned polycarbonate (PC) sheet with 1 mm thickness and a PC film with 130 ${\mu}m$ thickness, which layers are thermally bonded at $138^{\circ}C$ using acetone vapour. Silicatreated microglass beads with 150~212 ${\mu}L$ diameter are introduced into the sample pretreatment chambers and held in place by weir structure for construction of solid-phase extraction system. Fig. 3 shows strobed images of sequential loading of three samples. Three samples were loaded into the reservoir simultaneously (Fig. 3A), then the influenza A H3N2 viral RNA sample was loaded at 5000 RPM for 10 sec (Fig. 3B). Washing buffer was followed at 5000 RPM for 5 min (Fig. 3C), and angular frequency was decreased to 100 RPM for siphon priming of PCR cocktail to the channel as shown in Figure 3D. Finally the PCR cocktail was loaded to the bead chamber at 2000 RPM for 10 sec, and then RPM was increased up to 5000 RPM for 1 min to obtain the as much as PCR cocktail containing the RNA template (Fig. 3E). In this system, the wastes from RNA samples and washing buffer were transported to the waste chamber, which is fully filled to the chamber with precise optimization. Then, the PCR cocktail was able to transport to the PCR chamber. Fig. 3F shows the final image of the sample pretreatment. PCR cocktail containing RNA template is successfully isolated from waste. To detect the influenza A H3N2 virus, the purified RNA with PCR cocktail in the PCR chamber was amplified by using performed the RNA capture on the proposed microdevice. The fluorescence images were described in Figure 4A at the 0, 40 cycles. The fluorescence signal (40 cycle) was drastically increased confirming the influenza A H3N2 virus. The real-time profiles were successfully obtained using the optical fluorescence detector as shown in Figure 4B. The Rotary PCR and off-chip PCR were compared with same amount of influenza A H3N2 virus. The Ct value of Rotary PCR was smaller than the off-chip PCR without contamination. The whole process of the sample pretreatment and RT-PCR could be accomplished in 30 min on the fully integrated Rotary Genetic Analyzer system. We have demonstrated a fully integrated and portable Rotary Genetic Analyzer for detection of the gene expression of influenza A virus, which has 'Sample-in-answer-out' capability including sample pretreatment, rotary amplification, and optical detection. Target gene amplification was real-time monitored using the integrated Rotary Genetic Analyzer system.

A Comparative Analysis of TOD Planning Elements of Major Urban Railway Station Areas in Seoul (환승역세권 평가를 통한 TOD 계획 요소의 비교분석)

  • Transit Oriented Development(TOD) has been the dominant paradigm for urban development. The traditional approach to the development of urban rail station areas demonstrates apparent weaknesses when it comes to compact and mixed-use types of development for these stations. Thus, the objective of this study is to derive evaluation indicators with which to assess urban rail station areas using TOD planning elements. A focus group interview was conducted to explore TOD planning elements. In order to examine the level of TOD to be achieved, an AHP technique was utilized to analyze the importance of several variables. Z-scores and Re-scores were applied to derive the values for the stations. The Variables related to the land use type, mixed use, floor area, and public transit were found to be the major contributing factors to the achievement of TOD in urban rail station areas. The results of this study show that the level of TOD achievement varies according to the major land use, density, accessibility, and mixed-use planning elements.

  • 인터넷을 기반으로 한 정보통신의 급속한 발전이라는 기업환경의 변화에 적응하기 위해서 기업은 점차 모든 경영시스템을 인터넷을 기반으로 하도록 변화시키고 있을 뿐만 아니라, 기업 조직 또한 전세계를 기반으로한 글로벌 기업 형태로 변화하고 있다. 이러한 급속한 경영환경의 변화로 인해서 기업 내에서는 종전과는 다른 형태의 부서간 상호의사결정조정 과정이 필요하게 되었다. 일반 기업들을 대상으로 한 상호의사결정의 지원과정에 대해서는 기존에 많은 연구들이 있었으나 글로벌기업과 같은 네트워크 형태의 새로운 형태의 기업에 있어서의 상호의사결정과정을 지원할 수 있는 의사결정지원시스템에 대해서는 단순한 그룹의사결정지원시스템 또는 분산의사결정지원시스템과 같은 연구들이 주를 이루고 있다. 따라서 본 연구에서는 인터넷 특히, 웹을 기반으로 한 기업의 글로벌경영 및 분산 경영에서 비롯되는 부서간 상호의사결정이라는 문제를 효율적으로 지원할 수 있는 기업의 글로벌경영 및 분산 경영에서 비롯되는 부서간 상호의사결정이라는 문제를 효율적으로 지원할 수 있는 메커니즘을 제시하고 이에 기반한 프로토타입 형태의 시스템을 구현하여 성능을 검증하고자 한다. 특히, 기업 내에서 가장 대표적으로 상호의사결정지원이 필요한 생산과 마케팅 부서를 대상으로 상호의사결정지원 메커니즘을 개발하고 실험을 진행하였다. 그 결과 글로벌 기업내의 생산과 마케팅 부서간 상호의사결정을 효율적으로 지원 할 수 있는 상호조정 메카니즘인 개선된 PROMISE(PROduction and Marketing Interface Support Environment)를 기반으로 한 웹 분산의사결정지원시스템 (Web-DSS : Web-Decision Support Systems)을 제안하는 바이다.자대상 벤처기업의 선정을 위한 전문가시스템을 구축중이다.의 밀도를 비재무적 지표변수로 산정하여 로지스틱회귀 분석과 인공신경망 기법으로 검증하였다. 로지스틱회귀분석 결과에서는 재무적 지표변수 모형의 전체적 예측적중률이 87.50%인 반면에 재무/비재무적 지표모형은 90.18%로서 비재무적 지표변수 사용에 대한 개선의 효과가 나타났다. 표본기업들을 훈련과 시험용으로 구분하여 분석한 결과는 전체적으로 재무/비재무적 지표를 고려한 인공신경망기법의 예측적중률이 높은 것으로 나타났다. 즉, 로지스틱회귀 분석의 재무적 지표모형은 훈련, 시험용이 84.45%, 85.10%인 반면, 재무/비재무적 지표모형은 84.45%, 85.08%로서 거의 동일한 예측적중률을 가졌으나 인공신경망기법 분석에서는 재무적 지표모형이 92.23%, 85.10%인 반면, 재무/비재무적 지표모형에서는 91.12%, 88.06%로서 향상된 예측적중률을 나타내었다.ting LMS according to increasing the step-size parameter $\mu$ in the experimentally computed. learning curve. Synthesis of Organized $TiO_2$ Electrodes Using Graft Copolymer and Their Applications to Dye-Sensitized Solar Cells (가지형 공중합체를 이용한 나노구조 $TiO_2$ 제조 및 염료감응 태양전지 응용)

  • The morphology of mesoporous $TiO_2$ films plays an important role in the operation of a DSSC. For example, the energy conversion efficiency of DSSCs with well-organized mesoporous $TiO_2$ films is much higher than those with traditional films possessing a random morphology. In previous research, well-organized mesoporous $TiO_2$ films have mainly been synthesized using an amphiphilic block copolymer, e.g., a poly(ethylene oxide) (PEO)-based template. A graft copolymer is more attractive than a block copolymer due to its low cost and the ease with which it can be synthesized. In this work, we provide the first report on the successful synthesis of well-organized mesoporous $TiO_2$ films templated by an organized graft copolymer as a structure directing agent. Well-organized mesoporous $TiO_2$ films with excellent channel connectivities were developed via the sol gel processusing an organized PVC-g-POEM graft copolymer synthesized by one-pot ATRP. The careful adjustment of copolymer composition and solvent affinity using a THF/$H_2O$/HCl mixture was used to systematically vary the material structure. The influence of the material structure on solar cell performance was then investigated. A solid-state DSSC employing both the graft copolymer templated organized 700 nm-thick $TiO_2$ films and graft copolymer electrolytes exhibited a solar conversion efficiency of 2.2% at 100 $mW/cm^2$. This value was approximately two-fold higher than that attained from a DSSC employing a random mesoporous $TiO_2$ film. The solar cell performance was maximized at 4.6% when the film thickness was increased to $2.5{\mu}m$. We believe that this graft copolymer-directed approach introduces a new and simple route toward the synthesis of well-organized metal oxide films as an alternative to a conventional block copolymer-based template.

Effect of Hydrogen Sulfide Removal by Biofilter Seeded with Pseudomonas putida B2 (Pseudomonas putida B2가 접종된 Biofilter의 황화수소제거 효과)

  • A beterotrophic Pseudomonas putida B2 was used to treat of hydrogen sulfide containing gas. The experimental approach involved operating two indentical bench-scale biofilters with media consisting of a mixture of peatmoss, perlite and granular activated carbon(GAC). One column was seeded with Pseudomonas putida B2 and the other was left unseeded. The biofilter was operated for 16 days under EBRT for 20-40 sec, at a temperature of 25-30$^{\circ}C$ and a hydrogen sulfide concentration of 40-190 ppm. The biofilter inocculated with P.putida B2 exhibited high hydrogen sulfide removal efficiency, average of 95%, at a gydrogen sulfide concentration of 40-190 ppm (flow rate 3.6 L/min). However, at a shock loading of 190 ppm the biofiter showed a removal efficiency of 78.9% and the control only showed a removal efficiency of 31.6%. The critical load of this biofilter was 14.83 g/㎥hr, and the critical load of the control column was 4.93 g/㎥hr. These results suggest that P. putida B2 has the potential to be used as a $H_2S$ removal agent in a biofilter.

Non-Enzymatic Browning Reaction of the Dried File-Fish (건조(乾燥) 말쥐치의 비효소적(非酵素的) 갈변(褐變))

  • A kinetical approach for the non-enzymatic browning reaction of the dried file-fish was studied. The reaction rates revealed a tend to increase with increasing water activity and showed the maximum at $0.75\;a_w$ The activation energies obtained from the Arrhenius plot ranged 12.5 to 16.5 Kcal/mole. From these energies of activation, the $Q_{10}$ values at $45^{\circ}C$ showed 1.9 to 2.3 and both activation energy and $Q_{10}$ values were reduced with increase in $a_w$ Shelf-lives, the time to reach an 0.15 O.D./g solid at which severe brown color change could be de ectable, decreased rapidly as the temperature and water activity increase. A storage study under a square-wave fluctuating temperature condition (at 35 and $55^{\circ}C$ periodically with 7 days interval), the rate constants at all water activities used in the experiments were higher than those at $45^{\circ}C$, the mean temperature of the cycle which affects other kinetic parameters including activation energies, $Q_{10}$ values and finally the shelf-lives. The data obtained from the fluctuating temperaure storage study will be used in prediction of shelf-life. The shelf lives assessed at $25^{\circ}C$ from the accelerated shelf-life tests ranged from 179 daysat $0.75\;a_w$ to 302 days at $0.44\;a_w$.
