Cloning of Cellobiohydrolase Gene (cbhI) in Radiation Induced Mutant of Pleurotus florida

  • The cellobiohydrolase gene (cbhI), a key component of a cellulolytic system, of a mutant PfCM4 (Pleurotus florida), developed through gamma ray radiation mutagenesis, was isolated and cloned. The deduced amino acid sequence was closely related to the glycoside hydrolase family 7 (GH7). The molecular weight of the deduced amino acid sequence of cbhI gene was found to be 22.4 kDa. Though the percent identity was found to be much less (35.61%) between the wild type and mutant, the cellulolytic activity of PfCM4 was 17.24% higher than that of the wild type. This shows that the catalytic domain of the cbhI gene was conserved in the mutant PfCM4.

Radionuclide concentrations in agricultural soil and lifetime cancer risk due to gamma radioactivity in district Swabi, KPK, Pakistan

  • This study focuses on measuring the levels of naturally occurring radioactivity in the soil of Swabi, Khyber Pakhtunkhwa, Pakistan, as well as the associated health hazard. Thirty (30) soil samples were collected from various locations and analyzed for 226Ra, 232Th, and 40K radioactivity levels using a High Purity Germanium detector (HPGe) gamma-ray spectrometer with a photo-peak efficiency of approximately 52.3%. The average values obtained for these radionuclides are 35.6 ± 5.7 Bqkg-1, 47 ± 12.5 Bqkg-1, and 877 ± 153 Bqkg-1, respectively. The level of 232Th is slightly higher and 40K is 2.2 times higher than the internationally recommended limit of 30 Bqkg-1 and 400 Bqkg-1, respectively. Various parameters were calculated based on the results obtained, including Radium Equivalent (Raeq), External Hazard (Hex), Absorbed Dose Rate (D), Annual Gonadal Equivalent Dose (AGDE), Annual Effective Dose Rate, and Excess Lifetime Cancer Risk (ELCR), which are 170.3 ± 24 Bqkg-1, 0.46 ± 0.06 Bqkg-1, 81.4 ± 2.04 nGy h-1, 582 ± 78.08 µSvy-1, 99.8 ± 13.5 µSv Gy-1, and 0.349 ± 0.04, respectively. These values are below the limits recommended by the United Nations Scientific Committee on the Effects of Atomic Radiation (UNSCEAR) in 2002. This study highlights the potential radiation threats associated with natural radioactivity levels in the soil of Swabi and provides valuable information for public health and safety.

  • We investigated the cytoprotective effect of phloroglucinol, which was isolated from Ecklonia cava (brown seaweed), against oxidative stress induced cell damage in Chinese hamster lung fibroblast(V79-4) cells. Phloroglucinol was found to scavenge intracellular reactive oxygen species (ROS) generated by $\gamma-ray$ radiation. In addition, Phloroglucinol inhibited cell damage induced by radiation through scavenging ROS. Phloroglucinol increased the superoxide dismutase and glutathione peroxidase activity, Taken together, the results suggest that phloroglucinol protectes V79-4 cells against oxidative damage by enhancing the cellular antioxidant enzymes activity.

  • It is important to measure and protect from the radiation space dose and induced activity at the high energy medical linear accelerator facilities. These are to consider the additional risk to patients undergoing treatment, machine operators and staff members. Measurements of the space dose distribution and induced radioactivity at the 18 MeV medical linear accelerator facility in the Yonsei Cancer Center. 1. Exposure space dose for 300 rads monitor doses of 18 MeV electron are measured as 50 mR at 1 meter from patients. 2. Exposure space dose for 300 rads monitor doses of 10 MV X-ray are detected as 350 mR at 1 meter from phantom. 3. Induced radioactivity by photonuclear reaction was measured as 0.65 mR/hr from collimater after 30 Gy(3,000 rads) irradiated. 4. Analyzing the decay curves and energy spectrum of induced radioactivity, detected a few materials to be activated by photoneutron reaction, $^{65}Cu({\gamma}{\cdot}n)\;^{64}Cu,\;^{186}W({\gamma}{\cdot}n)\;^{185}W,\;^{181}Ta({\gamma}{\cdot}n)\;^{180}Ta,\;^{199}Au({\gamma}{\cdot}n)\;^{198}Au$.

Gamma Radiation-Induced Changes of Antioxidant Enzymes in Callus Cultures of Cassava(Manihot esculenta Crantz) (감마선에 의한 카사바 (Manihot esculenta Crantz) 배양세포의 항산화효소 활성 변화)

  • The gamma radiation-induced changes of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) in callus cultures of cassava (Manihot esculenta Crantz) selected as a high yield of cell line for SOD were investigated. In normal cultures, the cell growth reached a maximum at 30 days after subculture (DAS), followed by a rapid decrease with further cultures. The SOD and POD specific activities (units/mg protein) showed the highest at the immediately after subculture and subsequently decreased to 20 DAS, and then increased to 30 DAS, whereas the CAT activity showed the lowest at just after subculture, and it continuously increased from 15 DAS to 30 DAS, showing a good correlation with the cell growth. Irradiation of gamma-ray of 50 and 70 Gy on 7 DAS inhibited significantly the cell growth by 50% and 80% at 14 days after treatment (DAT), respectively. In the cells irradiated with 70 Gy, SOD and POD specific activities increased by 4 and 2.5 folds at 14 DAT, respectively, whereas CAT activity was not affected. The results indicate that SOD and POD may be involved in the antioxidative mechanism in relation to oxidative stresses induced by subcultures and by gamma radiation in callus cultures of cassava.

The Flow-rate Measurements in a Multi-phase Flow Pipeline by Using a Clamp-on Sealed Radioisotope Cross Correlation Flowmeter (투과 감마선 계측신호의 Cross correlation 기법 적용에 의한 다중상 유체의 유량측정)

  • The flow rate measurements in a multi-phase flow pipeline were evaluated quantitatively by means of a clamp-on sealed radioisotope based on a cross correlation signal processing technique. The flow rates were calculated by a determination of the transit time between two sealed gamma sources by using a cross correlation function following FFT filtering, then corrected with vapor fraction in the pipeline which was measured by the ${\gamma}$-ray attenuation method. The pipeline model was manufactured by acrylic resin(ID. 8 cm, L=3.5 m, t=10 mm), and the multi-phase flow patterns were realized by an injection of compressed $N_2$ gas. Two sealed gamma sources of $^{137}Cs$ (E=0.662 MeV, ${\Gamma}$ $factor=0.326\;R{\cdot}h^{-1}{\cdot}m^2{\cdot}Ci^{-1}$) of 20 mCi and 17 mCi, and radiation detectors of $2"{\times}2"$ NaI(Tl) scintillation counter (Eberline, SP-3) were used for this study. Under the given conditions(the distance between two sources: 4D(D; inner diameter), N/S ratio: $0.12{\sim}0.15$, sampling time ${\Delta}t$: 4msec), the measured flow rates showed the maximum. relative error of 1.7 % when compared to the real ones through the vapor content corrections($6.1\;%{\sim}9.2\;%$). From a subsequent experiment, it was proven that the closer the distance between the two sealed sources is, the more precise the measured flow rates are. Provided additional studies related to the selection of radioisotopes their activity, and an optimization of the experimental geometry are carried out, it is anticipated that a radioisotope application for flow rate measurements can be used as an important tool for monitoring multi-phase facilities belonging to petrochemical and refinery industries and contributes economically in the light of maintenance and control of them.

Changes of Microbiological and Physicochemical Characteristics of Doenjang Prepared with Low Salt Content and Gamma Irradiation (감마선 조사와 저염함량으로 제조한 된장의 미생물 및 품질특성 변화)

  • Changes of microbiological and physicochemical characteristics of gamma irradiated Doenjang with low salt were studied. Samples were prepared by mixing the Doenjang and the Chungkukjang with 6% and 8% of salinity. The mixture was irradiated by gamma-ray with dose of 0, 5, 10, 20 kGy, and each sample was stored at $25^{\circ}C$ for 8 weeks. The results showed that the Bacillus cell was inactivated by $10^4-10^5\;cells/g$ with dose of 10 kGy, and yeast and Lactobacillus group were nearly eliminated by 10 kGy. The general quality of gamma irradiated low salt Doenjang, such as amino nitrogen, protease activity, and pH, were more stable than that of control during storage periods. The sensory evaluations showed that 8% (salt)-5 kGy and 6% (salt)-10 kGy irradiated samples were more acceptable than market purchased Doenjang. Therefore, it was considered that gamma irradiation was effective for maintaining better quality of low salt Doenjang and suitable radiation dose was 5 kGy in 8% salted, and 10 kGy in 6% salted sample.

Separation and Recovery for the Analysis of Radioiodine in RI Wastes (RI 폐기물 내 방사성요오드 분석을 위한 분리 및 회수)

  • Various kinds of RI wastes are discharged from licensed organizations of radioisotopes les such as hospitals and clinic organizations, educational organizations, research institutions, and public organizations. Radioiodines such as $^{125}I\;and\;^{131}I$ are radioisotopes mainly used in nuclear medicine and industry. A method for the determination of radioiodines in RI wastes has been applied to measure low level activity using acid decomposition method and HPGe gamma ray spectrometer. Prior to analysis of real samples, $^{131}I$ reference solution and 10 g of yellow tissue paper was added to flask in mantle and was heated in 100 mL of 0.4 N $K_2Cr_2O_7$ and 100 mL of 9 M $H_2SO_4$, and then distilled after adding 10 mL of 30% $H_2PO_3$ and 1 mL of 30% $H_2O_2$. The condensed iodine by circulator was extracted into $CCl_4$, then back-extracted into the aqueous phase with 10 mL of 5% $K_2SO_2$ solution. Finally, $^{131}I$ was measured at 364.48 keV using HPGe gamma ray spectrometer after precipitation and filtration. Chemical yield of three steps such as acid decomposition process, chemical separation process, and precipitation and filtration process was more han 94% respectively, MDA(Minimum Detectable Activity) of $^{131}I$ at this analytical condition was 0.6 Bq/g.

Protective Effects of Chitosanoligosaccharide on Gamma Ray-Induced Hepatic Damage in Mice (생쥐에서 키토산올리고당의 감마선 저항 효과)

  • The purpose of this study was to evaluate the effect the radiation-resistance of chitosan on the mice. A healthy male ICR mice were used for experiment. The SOD and MDA activity was measured from the liver of mice at 48 and 72 hours after the irradiation. The ultrastructural changes of the liver by irradiation was observed at 24, 48 and 72 hours after irradiation. The experimental groups were divided into three groups. Group 1 was the control group which was not treated with chitosanoligosaccharide before or after iradiation. Group 2 was the prefeeding group which chitosanoligosaccharide solution was supplied by feeding ad libitum for 30 days before irradiation. Group 3 was the postfeeding group which chitosanoligosaccharide solution was supplied by feeding after irradiation. In all groups 10 mice were used. The results were as follow: The SOD and MDA activity of the prefeeding group was decreased significantly (P<0.01). Control group - The nuclei were condensed. The mitochondria and rough endoplasmic reticulum (rER) were extended and the ribosome was dropped from the rER. Prefeeding group - The nuclei were rounded. The mitochondria was elongated. And the rER attached ribosomes. Postfeeding group - The nuclei were slightly condensed. The mitochondria and the rER were extended and the ribosome was dropped from the rER. It was concluded that the chitosanoligosaccharide was effective in the radiation-protection. So, chitosan would have the potential as the radiation-protection materials.

Inhibition of an Inducible Nitric Oxide Synthase Expression by a Hexane Extract from Perilla frutescens cv. Chookyoupjaso Mutant Induced by Mutagenesis with Gamma-ray (방사선 유도 돌연변이 약용들깨 핵산 추출물의 Inducible Nitric Oxide Synthase 저해활성)

  • In earlier investigations, seeds of Perilla frutescens(L.) Britt. cv. Chookyoupjaso were irradiated with 200 Gy gamma ray to generate mutagenesis. The aim of this study is to investigate the effects of a hexane extract from Perilla frutescens (L.) Britt. cv. Chookyoupjaso mutant 45 on the actions of anti-inflammatory activity on inducible nitric oxide synthase, and an identification of the major active compound. The hexane extract from P. frutescens exhibited activity of inhibition of a NO production ($IC_{50}$, $295.1{\mu}g\;ml^{-1}$). The hexane extract was further divided into subfractions by silica-gel chromatogarphy. Inhibition of the NO production by various fractions was assayed in LPS-stimulated RAW 264.7 cells. Among the seven fractions, the 5th fraction was the most effective ($IC_{50}$, $19.5{\mu}g\;ml^{-1}$). The 5th fraction suppressed the expression of protein of iNOS in LPS-induced RAW 264.7 cells, and GC/MS analyses showed that isoegomaketone is a major bio-active compound in the 5th fraction. The result indicated that isoegomaketone has a good potential to be developed as an anti-inflammation agent.
