감마선에 의한 감귤 궤양병균의 불활성화와 감귤 궤양병 감염에 미치는 영향 (Inactivation of Xanthomonas citri subsp. citri and Effect on Infection of Citrus Canker by Gamma Irradiation)

  • 감귤 궤양병은 Xanthomonas citri subsp. citri에 의해 발생하는 병으로서 많은 나라에서 이 병에 대해 검역을 실시하고 있다. 최근 methyl bromide의 검역 시 사용이 제한됨에 따라 감마선을 이용한 농수축산물에 대한 살균 방안이 대두되고 있다. 본 연구에서는 현탁액 형태의 감귤 궤양병균과 생과실 과피에 존재하는 감귤 궤양병균의 밀도를 감소시킬 수 있는 감마선의 선량을 알아보았다. 미생물의 밀도를 90% 감소시키는 선량인 감귤 궤양병균의 $D_{10}$ value는 현탁액과 감귤 생과실 과피에서 각각 55와 28 Gy였다. 또한 감마선을 처리한 감귤 궤양병균 현탁액을 감귤 잎에 접종하였더니 병 발생이 억제되었다. 본 연구를 통하여 감마선 처리에 의해 감귤 생과실에 존재하는 감귤 궤양병균을 박멸할 수 있다고 판단되며 본 연구 결과는 감마선을 검역에 활용하는데 가치가 있다고 생각된다.

  • 생쥐의 난소에 미치는 방사선의 조직학적 및 기능적 영향을 조사하기 위하여 2.88Gy 및 7.2Gy의 방사선(Co-60)을 전신조사하였다. $7{\mu}m$ 두께의 조직절편을 제작하여 조직학적 관찰을 하였고, 방사면역측정 법을 이용하여 난소마쇄현탁액내 프로게스테론, 테스토스테론 및 에스트라디올을 정량하였다. 방사선조사로 유강소난포 및 무강소난포가 공히 높은 폐쇄율을 보였고, 난포내 프로게스테론의 농도가 증가되었으며 테스토스테론 및 에스트라디올의 농도는 감소되었다. 즉, 방사선을 조사함으로써 난소내 난포세포인 협막세포에 영향이 미친 결과 $3{\beta}-HSD(3{\beta}-hydroxysteroid\;dehydrogenase) $ 및 isomerase의 활성이 억제 혹은 둔화되어 테스토스테론 및 에스트라디올의 합성이 저하되었고 그 결과 난포의 폐쇄가 가속화되었다고 사료된다.

  • In uranium enrichment plants and nuclear fuel fabrication facilities, exact measurement of fissile isotope enrichment of uranium is required for material accounting in international safeguards inspection as well as process quality control. The purpose of this study was to develop a simple measurement system which can portably be used at nuclear fuel fabrication plants especially dealing with low enriched uranium. For this purpose, a small size CZT (CdZnTe) detector was used, and the detector performance in low uranium gamma/X -rays energy range was investigated by use of various enriched uranium oxide samples. New enrichment measurement technique and analysis method for low enriched uranium oxide, so-called, 'semi-peak ratio technique' was developed. The newly developed method was considered as an alternative technique for the low enrichment and would be useful to account nuclear material in safeguarding activity at nuclear fuel fabrication facility.

Protective Activity Against Oxidative Stress of Plants Indigenous to Korea

  • We have screened the cytoprotective effect against $H_2O_2$ and $\gamma-ray$ radiation induced oxidative stress from 32 Korean plants. Betula ermani var.saitoana (caulis, leaves), Rosa wichuraiana (caulis), Sorbus commixta (caulis), Weigela florida (leaves), Cirsium rhinoceros (whole plant), and Viburnum erosum (caulis) were found to scavenge 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of six plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by $\gamma-ray$ radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Betula ermani var. saitoana, Rosa wichuraiana, Sorbus commixta, Weigela florida, Cirsium rhinoceros, and Vibumum erosum protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

The status of NORMs in natural environment adjacent to the Rooppur nuclear power plant of Bangladesh

  • The Rooppur Nuclear Power Plant (RNPP), the first nuclear power plant in Bangladesh with a capacity of 2.4 GWe, is under construction on the bank of the river Padma, at Rooppur in Bangladesh. Measurement of background radioactivity in the natural environment adjacent to RNPP finds great importance for future perspectives. Soil and sediment samples collected from upstream and downstream positions of the Padma River (adjacent to RNPP) were collected and analyzed by HPGe gamma-ray spectrometry for primordial radionuclides. The average activity concentrations (in Bqkg-1) of 226Ra, 232Th and 40K radionuclides in soil samples were found to be 44.99 ± 3.89, 66.28 ± 6.55 and 553 ± 82.17 respectively. Respective values in sediment samples were found to be 44.59 ± 4.58, 67.64 ± 7.93, 782 ± 108. Relevant radiation hazard indices and dosimetric parameters were calculated and compared with the world average data recommended by US-EPA. Analytical results show non-negligible radiation hazards to the surrounding populace. Measured data will be useful to monitor any change of background radioactivity in the surrounding environment of RNPP following its operation for the generation of nuclear energy.

방사선조사(放射線照査)에 의한 Prostaglandin E2 및 자연살해세포(自然殺害細胞) 활성도(活性度)의 변화(變化) (Relationships between Radiation-induced Prostaglandin E2 and Natural Killer Cell Activity in Mice)

  • The number of splenic lymphocyte, serum prostaglandin $E_2$ level and natural killer cell activity were assayed after single whole body irradiation of a sublethal dose of $^{60}Co-{\gamma}$ ray to C57BL/6J mice. With a view to knowing the relationships between radiation induced prostaglandin $E_2$ level and the normal natural killer cell activity after natural killer cell-target cell conjugation, The change of normal natural killer cell activity were measured by administration of prostaglandin $E_2$ containing serum from irradiated mice. The results were summarized as follows; 1. The total number of splenic lymphocyte was significantly decreased by irradiation and the number was not affected by indometacin, prostaglandin synthesis inhibitor, treatment. 2. Serum prostaglandin $E_2$ level was increased in irradiated mice, but indometacin treated mice group showed low level of prostaglandin $E_2$. 3. In the case of irradiated mice, natural killer cell activity was not shown any difference between irradiated group and indometacin combined group. But when natural killer cell-target cell conjugations were exposed to the serum of each group during cytotoxic activity assay, whereas the normal natural killer cell activity was significantly decreased by treatment of serum from irradiated mice, the activity was not changed by treatment of indometacin pretreated mice serum. This result indicated that the prostaglandin $E_2$ induced by the radiation inhibited the post-target binding cytolytic process of natural killer activity.

방사선 조사에 의한 감자 발아 억제시 생화학적 효과 (Biochemical Effect on Potato Tubers Irradiated by Gamma-Ray at Sprout-Inhibition Dose)

  • 방사선 조사에 의한 발아 억제기작을 살펴보기 위하여 $4^{\circ}C$ 에서 저온 처리 한 감자에 0.12 kGy 감마선을 조사한 후 5주 동안 $20{\pm}2^{\circ}C$, 상대습도 $70{\sim}90%$에서 저장하였다. 이 기간 중 발아에 관여된다고 생각되는 ${\alpha}-amylase$, peroxidase, indole acetic acid (IAA) oxidase, IAA synthesizing enzyme의 활성 변화를 추적하였다. 시판 ${\alpha}-amylase$와 peroxidase의 방사선 감수성은 0.94와 0.36 kGy의 감마선 조사하여 $D_{37}$ 값을 얻었다. 감마선 조사를 행한 감자의 IAA oxidase 역가는 조사후 급격히 증가됐고 IAA 합성 효소의 역가는 조사직후 조금 증가하였으나 저장 기간에 따라 크게 감소하였다. 형태적인 변화에서 조사된 감자에서는 성장점의 고사와 변형된 눈이 관찰되었고 IAA 흑은 gibberellin 등의 처리에 의해 이들 자체로는 회복되지 않았고 그 주위로 새로운 눈의 성장이 관찰되었다. 결론으로 방사선 조사에 의해 야기된 감자의 호흡은 탄수화물이 ${\alpha}-amylase$등의 효소에 의한 분해로 이루어져 ATP를 공급하며 뒤이은 peroxidase 역가의 증가와 같은 효소의 합성은 방사선에 의한 손상을 치유하려는 과정에 속한다 할 수 있다. 또한 눈의 생장점에서의 고사 현상은 손상에 대한 방어 작용의 일종이라 해석되며 이로인한 발아 억제 현상을 페놀 대사와 관련지어 추측할 수 있었고 증가된 IAA oxidase와 IAA 합성계의 파괴에 의한 IAA의 고갈 역시 방사선 조사에 의한 발아 억제 현상의 원인이 됨을 알 수 있었다.

A New Composition of Nanosized Silica-Silver for Control of Various Plant Diseases

  • The present study addressed the efficacy of nanosized silica-silver for controlling plant pathogenic microorganisms. The nanosized silica-silver consisted of nano-silver combined with silica molecules and water soluble polymer, prepared by exposing a solution including silver salt, silicate and water soluble polymer to radioactive rays. The nanosized silica-silver showed antifungal activity against the tested phytopathogenic fungi at 3.0 ppm with varied degrees. In contrast, a number of beneficial bacteria or plant pathogenic bacteria were not significantly affected at 10 ppm level but completely inhibited by 100 ppm of nanosized silicasilver. Among the tested plant pathogenic fungi, the new product effectively controlled powdery mildews of pumpkin at 0.3 ppm in both field and greenhouse tests. The pathogens disappeared from the infected leaves 3 days after spray and the plants remained healthy thereafter. Our results suggested that the product developed in this study was effective in controlling various plant fungal diseases.

방사선조사가 골무기질함량에 미치는 영양에 관한 실험적 연구 (AN EXPERIMENTAL STUDY ON THE CHANGE OF BONE MINERAL METABOLISM AFTER IRRADIATION)

  • Irradiation is widely used for the treatment of malignant diseases, and possibly cause the osteoporosis. The bone densitometry and bone scintigraphy are valuable when used to monitor the patients longitudinally to access the progression of osteoporosis and risk of osteoradionecrosis. To evaluate the osteoporosis after irradiation of cobalt-60 gamma ray on the lumbar spines of New Zealand white rabbits, bone densitometry by dual photon absortiometry and bone scintigraphy were performed weekly. The decrease of bone density began at the first week after irradiation, and were in the nadir at 4-6th week. The osteoblastic activity measured by bone scintigraphy decreased in the first week, and was in the nadir at 4-6th week. The severity of these changes were related to the radiation dose. In conclusion, the osteoporosis before presentation of the osteoradionecrosis can be developed at low dose irradiation and confirmed by bone densitometry, bone scanning, and histopathology.
