Chlorella의 생리적, 생화학적 제활성에 미치는${\gamma}$-선의 영향

  • The sensitivities of Chlorella ellipsoidea to ${\gamma}$-ray from Cobalt-60 were determined by measuring the photosynthetic and respiratory activities and the changes in phosphate contents in various fractions of the irradited cells, which were further grown in a standard medium after irradiation, were compared to those of non-irradiated normal cells. The photosynthetic and repiratory activities of the cells were almost inversely proportional to the dose of ${\gamma}$-ray irradiated and the photosynthetic activity was more sensitive than the respiratory activity of the cells. The most sensitive to ${\gamma}$-ray was growth activity, followed by photosynthesis, exogenous and endogenous respirations of the cells in decreasing order. Chlorella cells were so resistant to ${\gamma}$-ray comapred with other organisms that about 280,000 r dose of ${\gamma}$-ray irradiaton was necessary to reduce as much as half the subsequent photosynthetic activity. When the irradiated algae were further cultured in a standard medium, the phosphate contents in the fraction of DNA, RNA and phosphoprotein decreased considerably compared with those of non-irradiated normal cells, while the phosphate contents in the fraction of polyphosphates increased than those of control. Therefore, it was deduced that ${\gamma}$-ray inhibited the synthesis of DNA from polyphosphates, that the growth of Chlorella cells were consequently retarded.

Effect of Gamma Ray on Germination, Growth and Antioxidant Activity of Senna tora (감마선 조사가 결명자의 생육과 항산화 활성에 미치는 영향)

  • Background: Senna tora is a flowering plant in the legume family Fabaceae. Its seeds are roasted and consumed as tea in Asia, to reduce inflammation in the liver and improve eyesight. Thus, it has been considered as an important medicinal crops in Asia. However, breeding trials to improve its genetic properties are rare. Mutation breeding by gamma ray is known to be an effective and highly successful approach for the generation of agronomically useful cultivars. Here we analyzed the effects of several dosages of gamma ray on the biological conditions of Senna tora seeds. Methods and Results: The germination rate and growth patterns of Senna tora were examined following irradiation with gamma ray at 100, 200, 300 and 400 Gy. The total phenolic compound contents and antioxidant activities of Senna tora were analyzed. Germination increased at 100 and 200 Gy in the M1 and M2 generations compared with that of the control (M0). The total phenolic compound contents and antioxidant activity of the seeds significantly decreased as the radiation dosage increased above 100 Gy in the M1 generation. Conclusions: Senna tora, irradiated with gamma ray at dosages 100, 200, 300, and 400 Gy, showed maximum germination rate at 200 Gy in the M2 generation. Plant height and leaf size gradually decreased with increasing gamma ray intensity in the M2 generation. The total phenolic compound contents decreased significantly at 400 Gy, and the related antioxidant activity was also decreased as the radiation dosage increased.

  • The HyperGam program was developed for the analysis of complex HPGe ${\gamma}$-ray spectra. The previous version of HyperGam was mainly limited to the analysis of ${\gamma}$-ray peaks and the manual logging of the result. In this study, it is specifically developed into a tool for the isotopic analysis of spectra. The newly developed features include nuclide identification and activity determination. An algorithm for nuclide identification was developed to identify the peaks in the spectrum by considering the yield, efficiency, energy and peak area for the ${\gamma}$-ray lines emitted from the radionuclide. The detailed performance of nuclide identification and activity determination was accessed using the IAEA 2002 set of test spectra. By analyzing the test spectra, the numbers of radionuclides identified truly (true hit), falsely (false hit) or missed (misses) were counted and compared with the results from the IAEA 2002 tests. The determined activities of the radionuclides were also compared for four test spectra of several samples. The result of the performance test is promising in comparison with those of the well-known software packages for ${\gamma}$-ray spectrum analysis.

  • Odyssey, one of the NASA's Mars exploration program and SELENE (Kaguya), a Japanese lunar orbiting spacecraft have a payload of Gamma-Ray Spectrometer (GRS) for analyzing radioactive chemical elements of the atmosphere and the surface. In these days, gamma-ray spectroscopy with a High-Purity Germanium (HPGe) detector has been widely used for the activity measurements of natural radionuclides contained in the soil of the Earth. The energy spectra obtained by the HPGe detectors have been generally analyzed by means of the Window Analysis (WA) method. In this method, activity concentrations are determined by using the net counts of energy window around individual peaks. Meanwhile, an alternative method, the so-called Full Spectrum Analysis (FSA) method uses count numbers not only from full-absorption peaks but from the contributions of Compton scattering due to gamma-rays. Consequently, while it takes a substantial time to obtain a statistically significant result in the WA method, the FSA method requires a much shorter time to reach the same level of the statistical significance. This study shows the validation results of FSA method. We have compared the concentration of radioactivity of $^{40}K$, $^{232}Th$ and $^{238}U$ in the soil measured by the WA method and the FSA method, respectively. The gamma-ray spectrum of reference materials (RGU and RGTh, KCl) and soil samples were measured by the 120% HPGe detector with cosmic muon veto detector. According to the comparison result of activity concentrations between the FSA and the WA, we could conclude that FSA method is validated against the WA method. This study implies that the FSA method can be used in a harsh measurement environment, such as the gamma-ray measurement in the Moon, in which the level of statistical significance is usually required in a much shorter data acquisition time than the WA method.

The Comparison of Color and Physiological Properties of Hizikia fusiformis Cooking Juice Ethanol Extract Irradiated with Gamma Ray and Electron Beam (감마선 및 전자선 조사에 따른 톳 자숙액 에탄올 추출물의 색상 및 생물학적 활성 변화 비교)

  • This study was carried out to assess the effect of radiation on the changes of Hizikia fusiformis cooking juice ethanol extract and to compare the effect of gamma ray and electron beam. On the applying radiation, the dark color of cooking juice became changed with higher brightness and lower redness and yellowness. But, there was no difference between gamma ray radiation and electron beam radiation. 1,1-Diphenyl-2-picryl-hydrazyl radical scavenging activity and tyrosinase inhibitory activity of cooking juice were shown to be increased by radiation independent on the radiation source types. The reason for the increased biological activities was caused by higher content of total phenolic compounds. The results could be applied to investigate the effect of radiation source on the color and antioxidant activity of biomaterials, and it was thought that irradiation could be an promising method for enhancing the biological activity of biomaterials.

Comparison of Irradiation Effect of Different Radiation Types on Decontamination of Microorganisms in Red Pepper Powder (고춧가루 오염 미생물의 제어에서 방사선종별 조사 효과)

  • This study investigated the reduction of microbial population and sensory properties in red pepper powders irradiated by gamma ray, electron beam, and X-ray. Populations of total aerobic bacteria and yeast & molds in red pepper powders were decreased by irradiation treatment in a dose-dependent manner. Gamma ray, electron beam, and X-ray at doses above 8 kGy caused 100% inhibition on growth of aerobic bacteria in red pepper powders. Inhibitory activity of X-ray on sterilization of red pepper powders was significantly equal to or higher compared to gamma ray and electron beam. Color and off flavor in red pepper powders were no significant difference among the control and samples irradiated with gamma ray, electron beam, and X-ray. As a result, the gamma ray, electron beam, and X-ray irradiation can be used to sterilize the microbial growth in red pepper powders without quality loss.

The Effect of Red Ginseng Extract on Superoxide Dismutase Activity in the Kidney of Gamma-ray Irradiated Mice (홍삼 투여가 방사선에 조사된 생쥐 신장의 Superoxide Dismutase에 미치는 영향)

  • This study was prepared to observe the change of enzyme activities in kidney treated with red ginseng extract in the gamma ray irradiated mice. Determine the activity of SOD, peroxidase, catalase in the kidney a period of 1 day, 2 day, 3 day, 4 day, 5 day after a saline injection or injection of red ginseng extract or gamma ray irradiatied group into four classify. The activity SOD and catalase showed a tendency to increase and recovery at the early state but pay no regard. Wherease, the activity of peroxide restored and increased pay regard. A physiological saline injection group after gamma ray irradiation showed a tendency to diminish after remakable increase of activity of SOD, peroxidase and catalase than control group. Injection group of red ginseng extract after gamma ray irradiation observed rapid recovery on activity of SOD, peroxidase, catalase than a saline injection group. Experimental result suggested that injection of red ginseng extract after irradiation have the recovery effect on the changed of activity of SOD, peroxidase and catalase against radiation injury.

Determination of Gross-${\beta}$ and ${\gamma}$-Ray Activity Concentrations of Human Tooth (치아의 전베타 농도 및 감마선 방사능 평가)

  • The ${\gamma}$-ray concentration and gross-${\beta}$ activity by age group were measured in the teeth of males and females of the domestic residents. They were divided into 7 age groups from 10s to the age of 70s. The gross-${\beta}$ activity concentration was measured by using the Tennelec XLB measuring instrument filled with P10 gas (argon 90%, methane 10%). The ${\gamma}$-ray was measured through the ${\gamma}$-ray spectroscopic analytical method by using the high purity germanium (HPGe) radiation detector. The range of gross-${\beta}$ activity concentration was measured 0.089 to 0.32 Bq/kg in females and 0.13 to 0.26 Bq/kg in males. From the ${\gamma}$-ray spectroscopic analysis of the teeth, the natural radioactive isotopes of $^{40}K$, $^{208}Tl$, $^{228}Ac$ and $^{234}Th$ were detected and their measured ${\gamma}$-ray activity concentrations were found to be 20.7, 21.9, 3.88 and 5.24 Bq/kg, respectively.

On respiration rate and dehydrogenase activity of yeast cells irradiated by gamma ray from cobalt-60. 1. Oxygen uptake rate of the gamma irraiated yeast cells had been measured with Warburg's manometer, and the $O_{2}$-uptake was compared with those of normal cells. The rate of endogetious respiration increases in its $O_2$-uptake at 150, 000 rentgen dose, and at higher rentoen doses it was decreased. Exogenous respiration begin to decrease in its O_2$-uptake at 5, 000r. doses of irradiation, further decrease with increasing of doses unproportionally. 2. It appears that plasma-membrane and nuclear membrane of yeast cells have changed and denatured by gamma-irradiation, as exogenous respiration of glucose had been decreased at a dose of 200, 000r's irradiation. 3. The activity of glucose, alcoholic, lactic, succinic and glutamic deliydrogenase (G.D.H., A.D.H., L.D.H., S.D.11., and GL.D.H.) in the gamma irradaited cells had been assayed by T.T.C.(Triphenyl tetrazolium chloride) method and spectrophotometry, the obtained results were compared with those of normal cells. 4. At a dose of and 10, 000 rentgens' irradiation of gamma ray, the activty of each debydrogenase (G.D.H., A.D.H., L.D.H., ) shows a sharp and highest peak in optical absorbalicy, but each abtivity of S.D.H and Gl.D.H shows its' maximum peak at a dose of 30, 000r. 5. The curve of each dehydrogenase activity was found to be rhythmical according to dose-rate of gamma irradiation. 6. Comparing with activity of debydrogenase each other, the maximum peak in optical absorbency can be arranged according to order as follows; glucose > alcoholoic > lactic > glutamic > succinic, this order is identical to the order of breakdown utility in respiration of normal yeast cells. 7. The activity of dehydrogenase experimented exhibit a resistance against gamma irradiation at lethal dose of cells, and the activity of dehydrogenase are found to be much resistant than those of respiratory system. We may assume that the membrane substrate of mitochondria or cytoplasm had been destructed by gamma-irradiation much more than that of dehydronase system.

Analysis for the secondary gamma-ray emission for glasses irradiated with various doses of fast neutron: Case study borate and silicate glasses

  • Are borate and silicate glasses suitable for working as shieling materials against fast neutrons? To correctly answer the above question, some silicate, and borate-based glasses were fabricated and irradiated with various doses of fast neutrons varied between 1.73 and 12.10 MGy. The color and hardness of the fabricated glasses were affected by the fast neutron fluence where the transparent glasses turned colored as well as the hardness of the fabricated glasses was decreased. The gamma-ray spectrometric analysis shows a high activity concentration produced in the barium borate glasses due to the formation of radioisotopes Ba-131 and Ba-133 reaches to 5.92E+05 Bq and 4.25E+03 Bq, respectively for sample Cd-5 Batch 3. Additionally, the gamma-ray spectrometric analysis for the sodium silicate glasses shows low activity concentrations emitted from isotopes formed due to the activation of Y2O3-associated impurities. These activities are low compared to that emitted by barium borate-based glasses.
