Biochemical Characterization of Heterologously Expressed Chitinase 1 (Chi1) from an Inky Cap, Coprinellus congregatus (이형 재조합한 먹물버섯 Coprinellus congregatus Chitinase 1 (Chi1)의 발현과 생화학적 특성 분석)

  • Fungal cell walls consist of various glucans and chitin. Fungi produce chitinases for their growth and development. The inky cap, Coprinellus congregatus, produces at least two different chitinases during its life cycle. Chitinase 1 (chi1) is expresses throughout its life cycle while chitinase 2 (chi2) is expressed at the mushroom autolysing phase. The cloned cDNA of chi1 is successfully expressed as a fusion protein with c-myc in Pichia pastoris, and purified by the affinity chromatography. The optimum pH and temperature of Chi1 was pH 8.0 and $35^{\circ}C$, respectively when 4-nitrophenyl N,N',N"-triacetyl-${\beta}$-D-chitotrioside was used as the substrate. The $K_m$ value and $V_{max}$ for the substrate was 0.780 mM and 0.10 OD $min^{-1}unit^{-1}$, respectively. The addition of purified Chi1 resulted in total growth inhibition against several plant pathogenic fungi such as Alternaria alternata, Fusarium graminearum and Trichoderma harzianum at the concentration of 60 ${\mu}g/ml$.

A Study on the Compatibility of Polycarbonate/Polyamide 6 Blends (Polycarbonate/Polyamide 6 블랜드의 상용성에 관한 연구)

  • A series of polycarbonate(PC)/polyamide 6(PA6) blends were prepared by three different blending methods to investigate their compatibility. From the DSC results, all of these blends have two Tg's in their own Tg regions, and there was no significant depression of the melting point and the crystallization temperature of PA6. With respect to the microstructure of the blends by SEM, the phase separation occurred at very low blend compositions, PC/PA6=95/5 and 5/95, already. In addition, a method is proposed to determine the Flory-Huggins polymer-polymer interaction parameter(${\chi}_{12}$) in polymer blend systems by using the experimentally determined Tg's. The values of ${\chi}_{12}$ obtained were 0.0381, 0.0411, 0.0418, for solution casting, solution precipitation, and extrusion blending methods, respectively. These values were higher than the critical value of ${\chi}_{12}$,($({\chi}_{12})_c$, 0.0271). Therefore it was concluded that the PC/PA6 blend system have little compatibility.

Comparing Feature Selection Methods in Spam Mail Filtering

  • In this work, we compared several feature selection methods in the field of spam mail filtering. The proposed fuzzy inference method outperforms information gain and chi squared test methods as a feature selection method in terms of error rate. In the case of junk mails, since the mail body has little text information, it provides insufficient hints to distinguish spam mails from legitimate ones. To address this problem, we follow hyperlinks contained in the email body, fetch contents of a remote web page, and extract hints from both original email body and fetched web pages. A two-phase approach is applied to filter spam mails in which definite hint is used first, and then less definite textual information is used. In our experiment, the proposed two-phase method achieved an improvement of recall by 32.4% on the average over the $1^{st}$ phase or the $2^{nd}$ phase only works.

  • Glass particle distribution in a stirred solid/liquid systems was investigated using computational fluid dynamics(CFD). The numerical results were compared to experimental data from the available literature which investigated the local dispersed phase volume fraction by means of an endoscope technique. Eulerian multi-phase model and applications considered high loading of solid particle was used to investigate the influence of the particle concentration and mixing tank size on the solid distribution. A good agreement was obtained between the experimental data and simulation results. The results showed different solid particle distribution in an agitator by particle concentration and mixer size.

  • In-situ observations of nano-scale behavior of nanomaterials are very important to understand onthe nano-scale phenomena associated with phase change, atomic movement, electrical or optical properties, and even reactions which take place in gas or liquid phases. We have developed on the in-situ experimental technologies of nano-materials (nano-cluster, nanowire, carbon nanotube, and graphene, et al.) and their interactions (percolation of metal nanoclusters, inter-diffusion, metal contacts and phase changes in nanowire devices, formation of solid nano-pores, melting behavior of isolated nano-metal in a nano-cup, et al.) by nano-discovery membrane platform [1-4]. Between two microelectrodes on a silicon nitride membrane platform, electrical percolations of metal nano-clusters are observed with nano-structures of deposited clusters. Their in-situ monitoring can make percolation devices of different conductance, nanoclusters based memory devices, and surface plasmonic enhancement devices, et al. As basic evidence on the phase change memory, phase change behaviors of nanowire devices are observed at a nano-scale.

  • The main goal of this paper is to develop a robust visual tracking algorithm with particle filtering. Visual Tracking with particle filter technique is not easy task due to cluttered environment, illumination changes. To deal with these problems, we develop an efficient observation model for target tracking with particle filter. We develop a robust phase correlation combined with motion information based observation model for particle filter framework. Phase correlation provides straight-forward estimation of rigid translational motion between two images, which is based on the well-known Fourier shift property. Phase correlation has the advantage that it is not affected by any intensity or contrast differences between two images. On the other hand, motion cue is also very well known technique and widely used due to its simplicity. Therefore, we apply the phase correlation integrated with motion information in particle filter framework for robust tracking. In experimental results, we show that tracking with multiple cues based model provides more reliable performance than single cue.

  • This paper introduces a novel approach to multi-material topology optimization (MTO) targeting in-plane bi-directional functionally graded (IBFG) non-uniform thickness Reissner-Mindlin plates, employing an alternative active phase approach. The mathematical formulation integrates a first shear deformation theory (FSDT) to address compliance minimization as the objective function. Through an alternating active-phase algorithm in conjunction with the block Gauss-Seidel method, the study transforms a multi-phase topology optimization challenge with multi-volume fraction constraints into multiple binary phase sub-problems, each with a single volume fraction constraint. The investigation focuses on IBFG materials that incorporate adequate local bulk and shear moduli to enhance the precision of material interactions. Furthermore, the well-established mixed interpolation of tensorial components 4-node elements (MITC4) is harnessed to tackle shear-locking issues inherent in thin plate models. The study meticulously presents detailed mathematical formulations for IBFG plates in the MTO framework, underscored by numerous numerical examples demonstrating the method's efficiency and reliability.

Study of Wettability Effect on Pressure Drop and Flow Pattern of Two-Phase Flow in Rectangular Microchannel (사각 마이크로채널 내의 2 상유동 압력강하와 유동양식에 대한 젖음성의 영향에 대한 연구)

  • Wettability is a critical parameter in micro-scale two-phase system. Several previous results indicate that wettability has influential affect on two-phase flow pattern in a microchannel. However, previous studies conducted using circular microtube, which was made by conventional fabrication techniques. Although most applications for micro thermal hydraulic system has used a rectangular microchannel, data for the rectangular microchannel is totally lack. In this study, a hydrophilic rectangular microchannel was fabricated using a photosensitive glass. And a hydrophobic rectangular microchannel was prepared using silanization of glass surfaces with OTS (octa-dethyl-trichloro-siliane). Experiments of two-phase flow in the hydrophilic and the hydrophobic rectangular microchannels were conducted using water and nitrogen gas. Visualization of twophase flow pattern was carried out using a high-speed camera and a long distance microscope. Visualization results show that the wettability was important for two-phase flow pattern in rectangular microchannel. In addition, two-phase frictional pressure drop was highly related with flow patterns. Finally, Two-phase frictional pressure drop was analyzed with flow patterns.

Design of Broadband Corrugated Waveguide Polarizer (광대역 커러게이트 도파관 편파기 설계)

  • In this paper, the design theory of the corrugated polarizer using square waveguide is proposed. In order to analysis the characteristic of square waveguide discontinuity as well as achievement of the low VSWR, we apply the modified $TE^\chi_mn$ mode matching method and the corrugated exponential function to the polarizer design. The results show a broadband characteristic that phase shift angle is nearly appeared $90^{\circ}$ in the range of 11.7-15.8GHz. Especially, we could be designed the Ku-band dual polarizer for the satellite broadcasting transponder with 300MHz bandwidth. Its phase shift angle and maximum VSWR, axial ratio are $90^{\circ},\pm1^{\circ}$, 1.03, and 1.0001 in the range of 11.7-12.0GHz and 14.5-14.8GHz, respectively.
