Quality Evaluation and Components of Euphoria longana (용안육의 품질평가 및 성분연구)

  • The fruit of Euphoria longana Lamarck (Sapindaceae) is a sweet food which is available in China and South Asia. Its pulp or flesh (Longan Arillus) is used as a tonic, and for the treatment of amnesia, insomnia, various palpitations due to fright, etc. It is traditionally said to fortify the heart and stabilize the spirit. Four compounds were isolated from the pulp of E. longana. The structures of these compounds were determinded as ${\beta}-sitosterol$, daucosterol, uracil and adenosine by means of spectroscopic methods.

  • The methanol extract of Euphorbia milii exhibited strong inhibitory effect on platelet aggregation in the cource of our search for anti-platelet component from succulent plants. Two components, components 1 and 2 were isolated from this plant. 1 was the mixture of 72% of 1-octacosanol (1a) and 28% of 1-triacontanol (1b), and 2 was identified as ${\beta}-sitosterol$. 2 ($IC_{50}$: $195\;{\mu}M$, and $170\;{\mu}M$ respectively) was about two fold stronger than ASA ($IC_{50}$: $420\;{\mu}M$ and $340\;{\mu}M$ respectively) on both collagen and U46619 induced aggregation, while the effect of 1 to platelets was negligible.

  • Ten compounds were isolated from the stem barks of Acanthopanax gracilistylus WW Smith (AGS) by steam distillation, they were p-menta-1,5,8-triene, n-butyl isobutylphthalate, p-mentha-1,5-diene-8-ol, 8-hydroxy-p-cymene, myrtenol, trans-(+)-carveol, 1,3-di-tert-butylbenzene, 4-methyl-2,6-di-butylphenol, valencene and verbenone, respectively, characterized by GS-Mass spectra. And we have also extracted and isolated from the MeOH extracts of the stem barks of AGS, two compounds were obtained. On the basis of chemical and spectral evidence, they were syringin(l), ${\beta}-sitosterol(2)$.

  • Five compounds were isolated from the fruits of Evodia daniellii $H_{EMSLEY}$. Their structures were identified as 2-oxotridecanyl acetate, bergapten, limonin, squalene, ${\beta}-sitosterol$ on the basis of spectral data. Among these compounds, 2-oxotridecanyl acetate and squalene were isolated in this plant for the first time.

  • The rate of growth and production of bioactive substances from Rehmannia glutinosa Liboschitz (Scrophulariaceae) were studied with the variation on the constituents of the culture media. The best growth was observed from MS basal medium containing 3.0 ppm NAA and 2.0 ppm kinetin. Carbohydrates (fructose, glucose and sucrose), phytosterols(${\beta}-sitosterol$, campesterol and stigmasterol) and carotenoid like substances were identified by GC-MS and TLC from the callus mass. However, catalpol was not detected from both solid and cell suspension cultures containing geraniol. Callus cultured Rehmannia glutinosa in the MS basal medium containing 0.1 ppm NAA and 0.1 ppm kinetin become differentiated to root.

  • The photochemical study of Viscum album (Loranthaceae) led to the isolation of twelve compounds, lupeol (1), betulonic acid (2), betulinic acid (3), terminic acid (4), ursolic acid (5), $\beta$-sitosterol (6), $\alpha$- spinasterol (7), oleanolic acid (8) , 5-hydroxy-1- (4′-hydoxyphenyl) -7- (4"-hydroxyphenyl) -hepta-1- en-3-on (9), 2′-hydroxy -4′, 6′- dimethoxychalcone -4-O-glycoside (10) ,2′-hydroxy-4′, 6′-dimethoxychalcone -4-O- [apiosal(1longrightarrow12)]glucoside (11) and syringin (12). Their structures were established by chemical and spectroscopic methods. The cytotoxicity of the isolated compounds was evaluated by sulforhodamine B assay against five cultured human tumor cell lines.

  • Three non-alkaloidal constituents, (-)-loliolide as an iridoid, friedelin as a triterpenoid, and ${\beta}-sitosterol$ as a steroid, were isolated by fractional extraction and silica gel column chromatography from the herb of Commelina communis L. (Commelinaceae). The structures of the native components were elucidated on the basis of physical and spectral (UV, IR, PMR, CMR, and mass spectra) data.

  • To elucidate the composition of phytosterols in the fruit body of Pholiota spp. 6 species(P. adiposa, P. aurivella, P. highlandensis, P. nameko, P. squarrosa and Pholiota sp.) were analyzed with Gas chromatography(GC). Pholiota spp. were contained campesterol, stigmasterol, and $\beta$-sitosterol as the major phytosterols.

  • From the dried roots of Caragana chamlagu Lamarck which have been known to be effective against neuralgia, arthritis, migraine, and sputum, etc. in the folk medicines of Korea, a colorless sterol-like substance, C$_{29}H_{50}O$, m.p. $115-117^{\circ}$, was isolated from the non-saponified fraction of methanolic extract. It was confirmed to be a mixture of four sterols, namely cholesterol, brassicasterol, campesterol, and $\beta$-sitosterol by physico-chemical properties, ir, nmr, mass spectra and gas liquid chromatographic method with the comparison of the authentic samples. In mass fragments of four sterols, furthermore, the same ions of m/e 273, 255, 229 and 213 were assigned to fragments of stigmastane skeletons of the unsaturated phytosterol, and the characteristic ions of each sterol were observed 386, 371, 368, 353 in cholesterol, 398, 383, 380, 365, 355, 337 in brassicasterol, 400, 385, 382, 367 in campesterol and 414, 399, 396, 381 in $\beta$-sitosterol, respectively.

  • It is well known that inflammation is associated with neurodegenerative disorders, including Alzheimer' disease, Parkinson's disease and ischemia. Nitric oxide (NO), a pro-inflammatory mediator, is produced by inducible NO synthase (iNOS) in microglia as well as macrophages and appears to account for neurodegeneration. In this study, we aimed to isolate NO inhibitors from Pyrus pyrifolia by activity guided purification. As a result, we identified daucosterol and ${\beta}$-sitosterol, which have not been isolated from this plant before. This article also describes NO inhibitory activities of the methanol extract of Pyrus pyrifolia fruit and the isolated compounds from this, which are lupeol, betulin, betulinic acid, ${\beta}$-sitosterol and daucosterol, in LPS-activated RAW 264.7 and BV2 cell lines. Western blot analysis was performed to clarify the underlying mechanism of NO inhibition in the two cell lines.
