Effects of Artificial Stomach Fluid and Digestive Enzymes on the Aglycone Isoflavone Contents of Soybean and Black Bean (Rhynchosia Molubilis : Yak-Kong) (대두와 쥐눈이콩의 비배당체 이소플라본 함량에 대한 인공위액과 소화효소 처리효과)

  • Phytoestrogens, especially soy-derived isoflavones, are receiving great scrutiny as a food supplement for preventing hormone dependent disease such as postmenopausal osteoporosis. Their beneficial effects are derived from aglycone form of isoflavones, such as daidzein, genistein or glycitein. In contrast to the common usage of soybean, black bean (Rhynchosia Molubilis : Yak-kong) has been used as a supplement for preventing postmenopausal osteoporosis in oriental medicine. To investigate the effects of the saliva, artificial stomach fluid, and digestive enzymes on the conversion of glycosidic isoflavone to aglycone form, soybean and black bean were extracted with 70% methanol and freeze-dried. The recovery yield of methanol extracts of black bean was 14.1% which was higher than that of soybean, 13.5%. In terms of total isoflavones, we routinely obtained larger amount of isoflavones from black bean than those from soybean. By incubating methanol extracts of soybean and black bean with IN HCI for 180 min, the proportions of aglycones relative to the total isoflavone were significantly increased (32.4% and 52.4%, respectively). In vitro conversion, digestive enzymes ($\beta$-glucosidase and $\alpha$-glucosidase) may hydrolyze glycosidic bond of isoflavone more effectively than saliva or artificial stomach fluid did. It seems to say that the activity of $\beta$-glucosidase was higher than those of $\alpha$-glucosidase. The rate of conversion of glucoside form to aglycone form in black bean and soybean was low in physiological condition (pH) tested, although the enzymatic hydrolysis of glucoside was active. These results demonstrated that the composition of aglycone in food may be the important factors in terms of the bioavailability of isoflavones. (Korean J Nutrition 36(1): 32-39, 2003)

The Effects of Soyumjungjang-tang on DSS-Induced Ulcerative Colitis in Mouse (소염정장탕(消炎整腸湯)이 DSS로 유발(誘發)된 생쥐의 궤양성 대장염에 미치는 영향(影響))

  • Objectives : This study was carried out to investigate the effects of Soyumjungjang-tang(SJT) on the experimental ulcerative colitis induced by dextran sulfate sodium(DSS) in mice. Methods : Ulcerative colitis was induced through supplying 4% DSS solution as the drinking water for 7 days in 6-week-old male ICR mice. The colitic mice were divided into three groups: the sample groups were orally administered SJT in doses of 25mg/kg(S25 group) or 100mg/kg(S100 group) once a day for 10 days, from 3 days before starting drinking the DSS solution, and the control(C) group was administered normal saline instead of SJT. The DSS solution or SJT was not administered to the normal(N) group. The length of colon, histologic finding, the activities of myeloperoxidase(MPO) and alkaline phosphatase(AP), and the expressions of $IL-1{\beta}$, IL-6, COX-2, $NF-{\kappa}B$, and $I{\kappa}B$ in colonic mucosa was checked using immunoblot, ELISA, etc. The activities of chondroitinase, tryptophanase, ${\beta}-glucuronidase$ and ${\beta}-glucosidase$ in stool were also measured. Results : The length of colon shortened, histologic finding deteriorated, the activities of MPO, AP, chondroitinase, tryptophanase, ${\beta}-glucuronidase$ and ${\beta}-glucosidase$, and the expressions of $IL-1{\beta}$, IL-6, COX-2, $NF-{\kappa}B$ increased, and the expression of $I{\kappa}B$ decreased in the C group. All measures, except $NF-{\kappa}B$, were restored in S25 group, but some measures deteriorated more in the S100 group than in the C group. Conclusions : According to the above results, it is supposed that SJT has a potential therapeutic effect on ulcerative colitis.

Optimization of Glycosyl Aesculin Synthesis by Thermotoga neapolitana β-Glucosidase Using Response-surface Methodology (반응표면분석법을 이용한 Thermotoga neapolitana β-glucosidase의 당전이 활성을 통한 glycosyl aesculin 합성 최적화)

  • Glycosyl aesculin, a potent anti-inflammatory agent, was synthesized by transglycosylation reaction, catalyzed by Thermotoga neapolitana ${\beta}-glucosidase$, with aesculin as an acceptor. The key reaction parameters were optimized using response-surface methodology (RSM) and $2{\mu}g$ of the enzyme. As shown by a statistical analysis, a second-order polynomial model fitted well to the data (p<0.05). The response surface curve for the interaction between aesculin and other parameters revealed that the aesculin concentration and reaction time were the primary factors that affected the yield of glycosyl aesculin. Among the tested factors, the optimum values for glycosyl aesculin production were as follows: aesculin concentration of 9.5 g/l, temperature of $84^{\circ}C$, reaction time of 81 min, and pH of 8.2. Under these conditions, 61.7% of glycosyl aesculin was obtained, with a predicted yield of 5.86 g/l. The maximum amount of glycosyl aesculin was 6.02 g/l. Good agreement between the predicted and experimental results confirmed the validity of the RSM. The optimization of reaction conditions by RSM resulted in a 1.6-fold increase in the production of glycosyl aesculin as compared to the yield before optimization. These results indicate that RSM can be effectively used for process optimization in the synthesis of a variety of biologically active glycosides using bacterial glycosidases.

Single-dose Oral Toxicity Study of β-glucosidase 1 (AtBG1) Protein Introduced into Genetically Modified Rapeseed (Brassica napus L.) (GM 유채에 도입된 β-glucosidase 1 (AtBG1)의 단회투여독성시험)

  • Rapeseed (Brassica napus L.) is an oil crop classified as Brassicaceae, and it is widely grown worldwide. To develop a drought-resistant rapeseed, the ${\beta}$-glucosidase 1 (AtBG1) gene was introduced into rapeseed because drought- and salt-resistance phenotypes were observed when the AtBG1 gene was overexpressed in arabidopsis. Newly developed genetically modified crop must be proved to be safe. Safety assessments are based on the historical usage and scientific reports of a crop. In this study, we examined the potential acute oral toxicity of AtBG1 protein expressed in genetically modified (GM) rapeseed and calculated the minimum lethal dose at 6 weeks in both male and female ICR mice. AtBG1 protein was fed at a dose of 2,000 mg/kg body weight in five male and five female mice according to the marginal capacity concentration of OECD, 2,000 mg/15 ml/kg. Mortalities, clinical findings, and body weight changes were monitored for 14 days after dosing, and postmortem necropsy was performed on day 14. This study showed that no deaths occurred in the test group, and AtBG1 protein did not result in variations in common symptoms, body weight, and postmortem findings between the two groups. This showed that the minimum lethal dose of AtBG1 protein expressed in transgenic rapeseed exceed 2,000 mg/kg body weight in both sexes.

Cultural Conditions of Pleurotus ostreatus 201 for the Production of Cellulolytic Enzymes in Synthetic Medium (합성배지에서 저온성 느타리(Pleurotus ostreatus 201)의 섬유소분해효소 생산조건)

  • Cultural conditions and carbon sources affecting the productivity of cellulolytic enzymes of Pleurotus ostreatus 201 were examined in synthetic media. The optimum cultural temperature and initial pH for the production of enzymes were $25^{\circ}C$ and 5.5 in avicelase, and 3$0^{\circ}C$ and 5.0 In CMCase, and 3$0^{\circ}C$ and 6.5 in B-glucosidase. Among carbon sources, cellulose powder was the best for the production of avicelase, and Na-CMC for CMCase, and cellobiose for $\beta$-glucosidase. The optimum concentration of cellulose powder was 1.0% (w/v), and glucose depressed the production of enzymes remarkably.

  • Precisely reliable and quantitative reporters can provide phenotypes that are consistent with research goals in protein expression. Here, we developed an improved reporter mATglu III 5 by directed evolution using a versatile β-glucosidase ATglu derived from Agrobacterium tumefaciens. When expressed in hosts, a vector containing this mutant distinctly showed a colored or fluorescent phenotype, according to the supplemented substrate, without any inducer. Analysis of mATglu III 5 showed it to be fully functional in fusion state with oligomeric proteins, especially under non-induction conditions, thereby offering an alternative to conventional reporters.

Effects of Carbon and Nitrogen Sources in the Production of Cellulolytic Enzymes by Trametes trogii (Trametes trogii에 의한 섬유소 분해효소의 생산에 있어서 탄소원과 질소원의 영향)

  • For the purpose of utilizingcellulosesresources by cellulolytic enzymes of Trametes trogii, its cultural conditions for the production of cellulolytic enzymes in synthetic media were investigated. The optimum conditions for the production of cellulase by T. trogii in synthetic media were $30{\sim}35^{\circ}C,\;pH\;4.0{\sim}6.0,\;and\;11{\sim}15$ day's cultivation. Among the carbon sources, carboxymethyl cellulose was good for the production of avicelase and ${\beta}-glucosidase$, but cellulose was good for the production of CMCase. The optimum concentration of Na-CMC was 3% for the production of all the three cellulolytic enzymes. As the nitrogen source, $0.03{\sim}0.04%$ N as ammonium tartrate was effective for the production of the cellulases.

Increased glucose metabolism and alpha-glucosidase inhibition in Cordyceps militaris water extract-treated HepG2 cells

  • BACKGROUND/OBJECTIVES: Recent living condition improvements, changes in dietary habits, and reductions in physical activity are contributing to an increase in metabolic syndrome symptoms including diabetes and obesity. Through such societal developments, humankind is continuously exposed to metabolic diseases such as diabetes, and the number of the victims is increasing. This study investigated Cordyceps militaris water extract (CMW)-induced glucose uptake in HepG2 cells and the effect of CMW treatment on glucose metabolism. MATERIALS/METHODS: Colorimetric assay kits were used to determine the glucokinase (GK) and pyruvate dehydrogenase (PDH) activities, glucose uptake, and glycogen content. Either RT-PCR or western blot analysis was performed for quantitation of glucose transporter 2 (GLUT2), hepatocyte nuclear factor 1 alpha ($HNF-1{\alpha}$), phosphatidylinositol 3-kinase (PI3k), protein kinase B (Akt), phosphorylated AMP-activated protein kinase (pAMPK), phosphoenolpyruvate carboxykinase, GK, PDH, and glycogen synthase kinase 3 beta ($GSK-3{\beta}$) expression levels. The ${\alpha}-glucosidase$ inhibitory activities of acarbose and CMW were evaluated by absorbance measurement. RESULTS: CMW induced glucose uptake in HepG2 cells by increasing GLUT2 through $HNF-1{\alpha}$ expression stimulation. Glucose in the cells increased the CMW-induced phosphorylation of AMPK. In turn, glycolysis was stimulated, and glyconeogenesis was inhibited. Furthermore, by studying the mechanism of action of PI3k, Akt, and $GSK-3{\beta}$, and measuring glycogen content, the study confirmed that the glucose was stored in the liver as glycogen. Finally, CMW resulted in a higher level of ${\alpha}-glucosidase$ inhibitory activity than that from acarbose. CONCLUSION: CMW induced the uptake of glucose into HepG2 cells, as well, it induced metabolism of the absorbed glucose. It is concluded that CMW is a candidate or potential use in diabetes prevention and treatment.

Impacts of Soil Microbial Populations on Soil Chemical and Biological Properties under Tropical Dry Evergreen Forest, Coromandel Coast, India

  • There are very few studies about soil chemical and biological properties under tropical dry evergreen forest Coromandel Coast, India. The present study was conducted in six tropical dry evergreen forests sites such as Oorani, Puthupet, Vadaagram, Kotthatai, Sendrakillai and Palvathunnan. We measured the quantity of soil chemical, biological properties and selected soil microorganisms for investigating the impacts of soil microbial populations on soil chemical and biological properties. The result showed that total N, P, Ca, S, Fe, Mn, Cu, Co, exchangeable K, Olson P, extractable Ca and phosphobacterial population were higher in the soil from Kothattai forest site. Organic carbon, total Mg, extractable Na, soil respiration, ${\beta}$-glucosidase activity, bacterial population, fungi population and actinomycetes population were higher in the soil from Palvathunn forest site. Total K, $NH_4{^+}$-N, $NO_3{^-}$-N, exchangeable K, extractable Ca, extractable Na, azotobacter population, bacillus population and rhizobacteria population were higher in the soil from Sendrakillai. Beijerinckia population, rhizobacteria and soluble sodium were higher in Puthupet forest soil. Total Si, total Na and exchangeable K were higher in soil from Oorani forest site. Total Mo and exchangeable K were higher in the soil from Vadaagaram forest site. The results showed that organic carbon, total N, $NH_4{^+}$-N, $NO_3{^-}$-N, extractable P, extractable Ca, soil respiration and ${\beta}$-glucosidase were significantly correlated with soil microbial populations. Therefore soil microorganisms are important factor for maintaining soil quality in tropical dry evergreen forest.
