Direct Headspace Sampling 방법을 이용한 당근의 휘발성 Terpenoids 분석에 관한 연구 (Studies on Measuring Volatile Terpenoids in Carrots Using the Direct Headspace Sampling Method)

  • 고품질 당근을 육성하기 위한 연구의 일환으로 당근의 휘발성 terpenoids를 GC 분석으로 측정하였다. 여러가지 휘발성 terpenoids 표준물질을 DHS(direct headspace sampling)방법으로 검출한 결과 ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, ${\alpha}-terpinene$, limonene, ${\gamma}-terpinene$, terpinolene 의 7가지 성분이 명확히 분리되었으나, ${\alpha}-phellandrene$${\beta}-myrcene$으로부터 분리되지 않았다. 또한 terpinen-4-ol, bornyl acetate, ${\alpha}-bisbolol$ 등 비등점이 높은 물질은 검출되지 않았다. 검출된 7가지 terpenoids 성분의 표준곡선은 결정계수$(r^2)$가 전부 0.99 이상으로서 고도로 유의한 표준곡선이었다. DHS 방법을 통한 본실험의 재현성을 측정한 결과, ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, limonene, ${\gamma}-terpinene$, terpinolene, total terpenoids의 변이 계수(C.V.)가 각각 6.8, 6.8, 8.4, 7.1, 3.8, 10.1, 7.1%로 $3.8{\sim}10.1%$의 범위를 나타내었다. 세계적으로 많이 재배되는 당근 65품종에 대한 ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, ${\alpha}-terpinene$, limonene, ${\gamma}-terpinene$, terpinolene, total terpenoids를 측정한 결과, 각각 $0.28{\sim}2.48\;ppm$, $0.35{\sim}1.87\;ppm$, $0.56{\sim}1.51\;ppm$, 0 ppm, $0.59{\sim}1.84\;ppm$, $0.87{\sim}3.33\;ppm$, $5.15{\sim}35.81\;ppm$, $9.07{\sim}42.30\;ppm$으로서 품종에 따라 상당한 차이를 보였다. Total terpenoids 의 경우 65품종 중 5품종(7.7%)이 10 ppm미만이고, 15품종(23.1%)이 $10{\sim}11.99\;ppm$, 14품종(21.5%)이 $12{\sim}13.99\;ppm$, 9품종(13.8%)이 $14{\sim}15.99\;ppm$, 10품종(15.4%)이 $16{\sim}17.99\;ppm$, 4품종(6.2%)이 $18{\sim}19.99\;ppm$, 5품종(7.7%)이 $20{\sim}29.99\;ppm$, 3품종(4.6%)이 30 ppm 이상이었다. 전반적으로 일본에서 육성된 품종이 구미에서 육성되어 보급되고 있는 품종들에 비해 total terpenoids 함량이 낮은 편이었다.

한국산 생강의 휘발성 향기성분 (Volatile Flavor Components of Korean Ginger(Zingiber officinale Roscoe))

  • 국내에서 소비되고 있는 대표적인 향신료 중의 하나인 생강의 essential oil 조성을 분석코저 SDE 방법에 의해서 분리하여 silica gel column chromatography에 의해 탄화수소 화합물 분획과 함산소화합물 분획으로 분리한 다음 GC 및 GC-MS에 의해 성분을 확인하였다. 신선한 생강의 essential oil의 수율은 0.32%로서 탄화수소화합물 분획이 68.1%, 함산소화합물 분획이 31.9%를 차지하였으며, mass spectrum에 의해서 잠정적으로 확인된 47종을 포함하여 101종의 성분이 확인되었다. 그 중 탄화수소화합물 분획에서는 camphene, ${\beta}-phellandrene$, zingiberene, ${\gamma}-bisabolene,\;{\bate}-bisabolene$, ${\beta}-sesquiphellandrene$이 주성분이었으며, 함산소화합물 분획에서는 1,8-cineol, neral, geranyl acetate, citronellol, geranial, ${\alpha}-terpineol+borneol$등이 주성분이었다. 외국산 생강의 essential oil의 조성과 비교한 결과 국내산 생강은 essential oil의 수율은 높은 편이나 sesquiterpene hydrocarbon의 함량이 높은 경향을 보였다.

침엽수 4종 목부 정유의 항염증 효과 평가 (Anti-Inflammatory Effect of Essential Oils Extracted from Wood of Four Coniferous Tree Species)

  • 본 연구는 국내산 편백, 소나무, 잣나무, 일본잎갈나무 목부 정유의 항염증 효과를 평가 하였다. 실험에 사용된 정유는 각 수종의 목부에서 증류법(hydrodistillation)으로 추출되었다. 추출한 정유들을 GC-MS를 통해 성분 분석한 결과, 편백의 경우 ${\alpha}$-cadinol (19.25%), ${\tau}$-muurolol (14.20%) 및 ${\alpha}$-pinene (13.74%)이, 소나무는 ${\alpha}$-pinene (47.16%), longifolene (14.31%), ${\beta}$-phellandrene (11.78%) 및 ${\beta}$-pinene (11.02%)이 주요성분이었으며, 잣나무의 경우 ${\alpha}$-pinene (13.49%)와 longifolene(10.79%), 낙엽송의 경우 geranyl linalool (23.58%) 및 ${\alpha}$-pinene (18.57%)이 주요성분으로 확인되었다. 침엽수 목부 정유들의 항염증 효과는 LPS(Lipopolysaccharide)로 염증반응이 유도된 RBL-2H3 비만세포주에 정유를 처리한 후 IL-4(interleukin-4), IL-13(interleukin-13), ${\beta}$-hexosaminidase의 발현 변화를 조사함으로써 평가하였다. MTT assay를 통한 세포 독성 평가 결과, 침엽수 4종 정유 모두 $10^{-7}{\sim}10^{-5}%$ 농도에서 세포 독성을 보이지 않았다. 염증반응이 유도된 RBL-2H3 세포에 침엽수 4종의 목부 정유를 $10^{-7}%$ 농도로 처리하였을 때, LPS 처리군 대비 침엽수 4종의 목부 처리군에서 IL-4와 IL-13의 상대적 발현량이 큰 폭으로 감소하였으며, ${\beta}$-hexosaminidase 분비가 유의하게 억제되었다. 침엽수 정유 4종의 IL-4 상대적 발현량은 LPS 처리군 대비 소나무 69.6%, 잣나무 63.2%, 편백 55.1% 그리고 낙엽송 45.8% 감소하였다. IL-13의 상대적 발현량은 소나무 57.8%, 잣나무 57.1%, 편백 51.1% 그리고 낙엽송 34.5% 감소하였다. ${\beta}$-Hexosaminidase 분비의 경우 LPS 처리군 대비 편백 38.1%, 소나무 33.0%, 잣나무 27.4% 그리고 낙엽송 9.1% 억제하였다. Il-4, IL-13 및 ${\beta}$-hexosaminidase 분비를 비교한 결과, 침엽수 목부 정유 4종 중 소나무 정유의 활성이 가장 우수하였다. 이러한 결과는 국내 침엽수 4종의 목부 정유는 염증반응을 유발하는 싸이토카인의 발현 억제를 통하여 항염증 효과를 나타냄을 확인할 수 있었다.

소나무의 정상(正常)잎, 피해(被害)잎 및 솔방울의 테르페노이드성분(成分) 분석(分析) (Terpenoid Analysis of the Normal, Damaged Needle and Pinecone in Pinus densiflora)

  • To analyze terpene components, the essentail oil were extracted with steam distillation method from normal needle, damaged needle and pinecone of Pinus densiflora. The extracted essential oil was analyzed by gas chromatography and gas chromatography-mass spectroscopy. The results were summarized as follows; 1. Normal needles involve 43 kinds of terpene components, but damaged needles contained only 29 kinds. The most abundant components of normal and damaged needles were respectable ${\alpha}$-pinene and caryophyllene oxide. ${\alpha}$-pinene content in normal meedles amounted to 15.99 percents and caryophyllene oxide in damaged was 8.15 percents. 2. Pinecone showed 23 kinds of terpene components and among them the most abundant component was ${\beta}$-phellandrene, of which content showed 19.31 percents. 3. In normal needles, excluding ${\alpha}$-pinene, the contents of 8 kinds of other monoterpenes, reached to 48 percents of the total terpenes, 4. In damaged needles, excluding ${\alpha}$-pinene, the contents of 4 kinds of other monoterpenes, reached to 11 percents of the total terpenes. 5. In pinecone, excluding camphene, the contents of 6 kinds of other monoterpenes, reached to 58 percents of the total terpenes.

  • The essential oils of stems, roots, fruits and leaves of Lindera obtusiloba BL. were collected in the winter and summer extracted by simultaneous distillation extraction (SDE) apparatus and analyzed by gas chromatography-mass spectrometry (GC-MS). In present study, 58 kinds of volatile components in the winter stems (WS), 70 in the winter roots (WR), 77 in the summer stems (SS), 78 in the summer roots (SR), 70 in the summer fruits (SF) and 76 in the summer leaves (SL) were identified. The results showed that, the major components were monoterpenes including ${\alpha}$-thujene (1.22~13.80%) camphene (1.56~18.40%), ${\beta}$-mycrene (1.75~9.27%), limonene (5.57~12.83%), ${\beta}$-phellandrene (3.03~7.72%), linalyl acetate (2.29~12.55%), dihydromycrene (0~11.15%), germacrene B (0~7.54%) of which the contents had major fluctuations in different seasons and parts. In general, monoterpenes were the major constituent of SF in L. obtusiloba BL. that have presented possibilities for industrial applications.

Analysis of Aroma Components from Zanthoxylum

  • Zanthoxylum schinifolium and Zanthoxylum piperitum A.P. DC. belong to the Rutaceae family and are perennial, aromatic, and medicinal herbaceous plants. In this study, their aroma compounds were isolated by steam distillation extraction using a Clevenger-type apparatus, and then further analyzed by gas chromatography (GC) and gas chromatograph/mass spectrometry (GC/MS). The yields of the essential oils from Z. schinifolium and Z. piperitum AP. DC. were 2.5 and 2.0%(w/w), respectively, and the color of their oils was quite similar, a pale yellow. From the distilled oil of Z. schinifolium, 60 volatile compounds which make up 87.24% of the total composition were tentatively identified, with monoterpenes predominating. $\beta$-Phellandrene (22.54%), citronellal (16.48%), and geranyl acetate (11.39%) were the predominantly abundant components of Z. schinifolium. In the essential oil of Z. piperitum AP. DC., 60 volatile flavor components constituted 94.78% of the total peak area were tentatively characterized. Limonene (18.04%), geranyl acetate (15.33%), and cryptone (8.52%) were the major volatile flavor compounds of Z. piperitum A.P. DC.

Constituents of the Essential Oil from Eclipta prostrata L.

  • The volatile aroma constituents of Eclipta prostrata L. (leaves, stems, and flowers) were isolated by hydro-distillation extraction method and analyzed by GC/MS. The yield of Eclipta prostrata L. essential oil was 0.1% (v/w), and its color was yellow. Sixty-eight volatile flavor compounds, which make up 71.15% of the total volatile composition of the essential oil were tentatively characterized. It contained 35 hydrocarbons (56.25%) with sesquiterpene predominating, 12 alcohols (3.05%), 8 ketones (3.83%), 9 aldehydes (1.86%), 2 oxides (6.03%), and 2 esters (0.13%). ${\alpha}$-Humulene, 6,9-heptadecadiene, (E)-${\beta}$-farnesene, and ${\alpha}$-phellandrene were the major abundant aroma components in Eclipta prostrata L., aromatic and medicinal plant.

Angelica속 생약의 정유성분에 관한 연구(IV) -강활의 정유성분- (Studies on Essential Oils of Plants of Angelica Genus in Korea (IV) -Essential Oils of Angelicae koreanae Radix-)

  • Essential oil of the root of Angelica koreana Max. (Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain nine monoterpenes such as ${\alpha}-pinene$(7.0%), camphene, ${\beta}-pinene$, myrcene, ${\alpha}-phellandrene$, ${\Delta}-3-carene$, p-cymene, limonene(2.8%), terpinolene and also found to contain m-cresol(11.6%), citronellol, citral b, methylcinnamate, eudesmol and osthol. 2-Hydroxy-5-methylacetophenone and twelve compounds were tentatively identified.

Comparative Analysis of Volatile Flavor Compounds from Zanthoxylum pipperitum A.P. DC

  • The volatile flavor components of Zanthoxylum pipperitum A.P. DC. produced in Korea and China were isolated using a Clevenger-type apparatus by steam distillation extraction, and analyzed by gas chromatography-mass spectrometry (GC/MS). The yields of oils from Korean and Chinese Z. pipperitum A.P. DC. were 2.0 and 1.2% (w/w), respectively. From the two Z. pipperitum A.P. DC. oils, sixty and fifty-four volatile flavor compounds were tentatively identified, and they constituted 94.78 and 87.34% of the total peak area, respectively. Piperitone(p-menth-1-en-3-one) (13.48%) was the most abundant compound in the Chinese Zanthoxylum pipperitum A.P. DC. oil, followed by $\beta$-phellandrene, sabinene, terpinen-4-ol and linalool (each >5%). Whereas, the most abundant compound in the Korean Zanthoxylum pipperitum A.P. DC. oil was limonene (18.04%), followed by geranyl acetate, cryptone, citronellal, cuminal and phellandral (each >5%).

A Multivariate Statistical Approach to Comparison of Essential Oil Composition from Three Mentha Species

  • The chemical composition of essential oils obtained from aerial parts in spearmint, apple mint and chocolate mint, was investigated by gas chromatography/mass spectrometry analyses. (-)-Carvone (33.0%) was quantitatively major compound in spearmint, followed by R-(+)-limonene (11.7%) and ${\beta}$-phellandrene (9.7%); (-)-carvone (37.4%) and germacrene D (11.9%) in apple mint; and (-)-menthol (34.3%), p-menthone (18.4%) and menthofuran (9.8%) in chocolate mint. Hierarchical cluster analysis and principle components analysis showed the clear difference in chemical composition of the three mint oils.
