Production of Amylases from Herpetosiphon geysericola (Herpetosiphon geysericola 균주의 Amylase 생성)

  • A thermophilic and cellulolytic bacterium, Herpetosiphon geysericola CUM 317 isolated from the compost, produced ${\alpha}-amylase,\;{\beta}-amylase$, and glucoamylase. Mutual relationships on the production of the three amylases were studied by changing the cultivation conditions. ${\alpha}-Amylase$ and glucoamylase were produced highly after 40 hrs on wheat bran medium at $50^{\circ}C$ and after 30 hrs on liquid medium at $40^{\circ}C$, though ${\beta}-amylase$ was produced best at 10 hrs of initial cultivation phase. The production of the amylases was generally repressed by the addition of carbon sources in liquid medium containing polypeptone. ${\alpha}-Amylase$ production was enhanced relatively by the addition of cupric sulfate in the liquid medium, ${\beta}-amylase$ was enhanced by cadmium sulfate, and glucoamylase was enhanced by calcium chloride.

Flavonoid Analysis from the Leaves of Eucommia ulmoides (두충나무잎의 생리활성 Flavonoid 분석)

  • Three flavonoid compounds, astragalin(1), isoquercitrin(2) and quercetin $3-O-{\beta}-D-xylopyranosyl(1-2)-{\beta}-D-glucopyranoside(3)$ isolated from the leaves of Eucommia ulmoides were identified and quantified by HPLC. All flavonoids were well separated on a ${\mu}-Bondapak\;C_{18}$ column with a mobile phase composed of THF-dioxane-MeOH-HOAc-5% $H_3PO_4-H_2O$(145 : 125 : 50 : 20 : 2 : 658). The contents of compound 1 in the methanol extract and n-butanol fraction were 0.09%(w/w) and 0.46%(w/w), of compound 2 were 0.08%(w/w) and 0.48%(w/w), and of compound 3 were 0.40%(w/w) and 1.22%(w/w), respectively.

The effect of β-sitosterol proliferation and apoptosis in human uterine leiomyoma cells (계혈등(鷄血藤)의 Beta-sitosterol 성분이 자궁근종세포의 증식억제와 세포자멸사의 유도에 미치는 영향)

  • Purpose : ${\beta}$-sitosterol is kind of phytosterols or plant which are structurally similar to cholesterol. This study was aimed to investigate the inhibitory effect of the ${\beta}$-sitosterol on the proliferation of human uterine leiomyoma cells and the expression of gene related the mechanism of cell apoptosis. Methods : We counted the number of death cells treated with indicated time of the ${\beta}$-sitosterol and investigated cell death rate by cell count assay. Furthermore, flow cytometry analysis and DNA fragmentation assay were used to dissect between necrosis and apoptosis. and then we observed the differential gene expression by western blot analysis. Results : 1) The inhibitory effect on the growth of uterine leiomyoma cell treated with the ${\beta}$-sitosterol $16{\mu}M$ was increased in a time dependent. 2) The result of flow cytometry analysis, subG1 phase arrest related cell apoptosis was investigated 16.97% in uterine leiomyoma cell treated with the ${\beta}$-sitosterol $16{\mu}M$ and showed the fashion of proportional time dependent. 3) The gene expression of p27, p21 related cell cycle was increased according to increasing time interval but cyclin E-CDK2 complex was decreased expression. 4) The character of apoptosis, DNA fragmentation was significantly observed on the time dependent. 5) The expression of pro-caspase 3 and PARP were decreased dependent on treatment with time dependent. Conclusion : This study showed that the ${\beta}$-sitosterol have the inhibitory effect on the proliferation of human uterine leiomyoma cell and the effect was related with apoptosis.

Comparison of Volatile Aroma Compounds between Synurus deltoides and Aster scaber Leaves (수리취와 참취 엽의 휘발성 향기성분 비교)

  • This study was investigated to compare the volatile aroma compounds of Synurus deltoides and Aster scaber. The volatile aroma compounds from Synurus deltoides and Aster scaber were extracted by soild-phase microextraction (SPME) methods. S. deltoides had 97 volatile aroma compounds such as including 5-acetyl-1,2-dihydro acenaphtylene (14.63%), ${\beta}$-cubebene (9.31%), caryophyllene (8.97%), ${\beta}$-chamigrene (7.14%), ${\beta}$-selinene (2.71), ${\alpha}$-farnesene (2.47%), ${\alpha}$-bergamotene (2.26%), ${\beta}$-elemene (1.94%), etc. A. scaber had 84 volatile aroma compounds such as (+)-epi-bicyclosesquiphellandrene (10.38%), terpinolen (10.09%), caryophyllene (6.04%), 8-isopropenyl-1,5-dimethyl-1,5-cyclodeca diene (5.42%), ${\alpha}$-himachalene (5.04%), ${\beta}$-thujene (4.37%), ${\beta}$-pinene (4.28%), ${\beta}$-cubebene (3.99%), etc. Conclusively, the main common volatile aroma compounds in S. deltoides and A. scaber leaves were 19 volatile aroma compounds such as caryophyllene, terpinolen, ${\beta}$-cubebene. But the composition and amount of volatile aroma compounds were very different between the two species.

  • The effects of Sn and Nb content on the characteristics of ${\beta}$ to ${\alpha}$ phase transformation for Zr were studied by optical microscopy and transmission electron microscopy. The basketweave structures consisted of fine ${\alpha}$-laths were shown in all the air-cooled specimens. As the Nb content increased, the ${\beta}{\to}{\alpha}$+${\beta}$ transformation temperature decreased thus allowing the width of ${\alpha}$-lath, however, did not change with Sn content. While water-quenched pure Zr and Zr-xSn alloys were found to be mailny slipped martensite, water-quenched Zr-xNb alloys showed predominantly a twinned martensite. The transition of slipped martensite to twinned martensite was contributed to the decrease of Ms temperature.

Influence of antioxidants on β-carotene degradation in nanoemulsions (나노에멀션 내의 베타카로텐 분해에 미치는 산화방지제의 영향)

  • In this study, we examined the effects of carrier oil type (MCT oil: MO, corn oil: CO, palm oil: PO), pH of dispersion solution, and antioxidants on the chemical degradation of ${\beta}$-carotene in oil-in-water nanoemulsions. The pH of the emulsion had a significant influence on the stability of ${\beta}$-carotene, which showed rapid degradation in emulsions at low pH value and relatively higher stability at high pH values. The influence of the carrier oil type on ${\beta}$-carotene stability was assessed. The rate of ${\beta}$-carotene degradation increased in the following order: CO > PO > MO. The effect of antioxidants on ${\beta}$-carotene degradation was monitored during storage at $25^{\circ}C$ for 4 weeks. The rate of ${\beta}$-carotene degradation decreased upon addition of water-soluble (ascorbic acid) or oil-soluble (tocopherol) antioxidants. In general, tocopherol was more effective than ascorbic acid in reducing ${\beta}$-carotene degradation. To utilize this nanoemulsion for producing acidic beverages, adding a higher concentration of antioxidants is required.

The Study on Phase Separation Development by Curing Reaction Rate for Unsaturated Polyester/Polyvinylacetate Semi-IPN (Unsaturated Polyester/Polyvinylacetate Semi-IPN의 경화반응속도에 따른 상분리현상 연구)

  • Morphological changes of unsaturated polyester/polyvinylacetate semi-IPN were studied while the phase separation and the cure reaction occurred in a competing fashion. The light scattering and thermal analysis techniques were used to investigate the phase separation rates and mechanical properties resultantly induced by molecular diffusion of thermoplastic polymer during the curing process of thermosetting polymer. The reaction activation energy was calculated by using Flynn-Wall method and the semi-IPN structure exhibited various phase-separation morphological characteristics. When PVAc composition was 10 wt%, the phase separation was not observed during the curing reaction, but the phase separation occurred in a similar fashion to nucleation and growth(NG) mechanism at room temperature. On the other hand, when PVAc composition was over 11.65 wt%, the phase separation was generated in the middle of the curing process. Consequently, the phase separation seemed to influence the curing reaction rate, which was also supported by the changing activation energy with conversion and PVAc composition. Finally, the total scattered intensity was measured at various temperature, and subsequently the diffusion rates of phase separation R(${\beta}m$) were evaluated.

A Study on the Inclusion Complexation of Octyldimethyl p-aminobengoate with \brta -Cyclodextrin$ (Octyldiinethyl p-aminobenzoate와 \brta -Cyclodextrin$의 포접화합물에 관한 연구)

  • Inclusion complex formation of octyldimethl p-aminobenzoate with $\beta$-cyclodextrin in aqueous solution and in the solid state was studied by the solubility method, spectroscopic(UV, FT-lR) and X -ray diffractometry. The solid complex of octyldimethy p-aminobenzoate with $\beta$-cyclodextrin was obtained in molar ratio of 1 : 2(guest/host). A spatial relationship between host and guest molecule was clearly reflected in the magnitude of the apparent stability constant (K') and in the stoichiometry of the inclusion complex. Furthermore, a typical type Bs phase-solubility diagram was obtained for octyldimethyl p-aminobenzoate and p -cyclodextrin in water at $25^{\circ}C$. The results indicated that the solubility of the guest molecule was higher by the formation of $\beta$-cyclodextrin inclusion complex.

  • Dopamine beta-hydroxylase (DBH) is a neurotransmitter biosynthetic enzyme which catalyzes the conversion of dopamine to norepinephrine. Although structural studies of the mature form of this enzyme have been extensive, the culmination of these finding had been hightly controversial and contradictory. In this study, biochemical approaches were taken to characterize the structure of mature DBH. Soluble bovine DBH was purified from aderenal medulla. Three bands of 69 kDa, 72 kDa and 75 kDa which were physically separable and similar in structure were observed by SDS-PAGE. Furthermore, gas phase sequence analysis revealed that the 72 kDa band consists of two polypeptides which are present at equimolar concentrations and differed in that one had three extra amino acids at the N-terminus. Taken together, the soluble form of DBH exist in at least four forms, three identified by SDS-PAGE, one of which consists of two polypeptides as identified by N-terminal sequence analysis. The significance of these forms and their possible biosynthetic mechanisms are discussed.

  • Biphasic Calcium Phosphate (BCP) consisted of hydroxyapatite (HAp) and $\beta$-tricalcium phosphate (P-TCP) has been prepared by solid state reaction. The size reduction of the resultant BCP agglomerate was carried out by reaction with hot water under atmospheric condition uld also under high pressure using an autoclave. The influence of processing conditions on the change of crystalline phase and composition, relative amount of constituent, specific surface area, and microstructure was investigated by means of XRD, FT-IR, BET method using a nitrogen adsorption and SEM.
