Combined Effect of Radiation and $7{\beta}$-Hydroxycholeslerol on Human Cervical Cancer Cells in vitro

  • [ $7{\beta}$ ]-Hydroxycholesterol (cholest-5-ene-3, 7-diol, $7{\beta}$-OHC) showed the cytotoxicity on human cervical carcinoma cells (HeLa), $10{\mu}M$ of 50% inhibitory concentration. We evaluated $7{\beta}$-OHC as the possibility of radiation sensitizer. The combination effect of $7{\beta}-OHC\;and\;{\gamma}$-irradiation was measured using colony forming assay and flow cytometer with propidium iodide and $DiOC_6$ stained cells, respectively. The combined treatment of $7{\beta}-OHC\;and\;{\gamma}$-irradiation did not show significant enhancing effects on HeLa cells.

U-937 세포에서 방사선 및 $H_2$O_2$에 의한 ceruloplasmin의 mRNA 유전자 발현 (Ceruloplasmin Gene Expression in U-937 Cells exposed to ${\gamma}$-Irradiation and $H_2$O_2)

  • In human U-937 cell exposed to ${\gamma}$-irradiation and $H_2O$$_2$, the level of mRNA efrpression in ceruloplasmin gene was measured by using comparative RT.PCR (reverse transcriptase-polymerase chain reaction). At the normal growth condition, the level of ceruloplasmin transcript was estimated as 8.2% and 0.0068% of hprt (hypoxantine phosphoribosyl transferase) transcript and of $\beta$-actin transcript, respectively. In U-937 cells exposed to a dose of 100 rad ${\gamma}$-irradiation, the level of ceruloplasmin transcript was increased about 2.7 and 1.6 fold compared to un-treated cell by using compensation with the levels of hprt and $\beta$-actin transcript. By contrast, the expression of ceruloplasmin gene in U-937 cells exposed to $H_2O$$_2$(50 $\mu$M, 24 h), was shown no significant difference compared to un-treated cell. These results indicated that the expression system of ceruloplasmin gene may react only some specific oxygen species, such as reactive oxygen species induced by ${\gamma}$-irradiation.

  • The purpose of this study was to determine the effects of laser Irradiation on Human plasma $\beta-endorphin$ levels, by treating with low level helium-neon (He-Ne) and Infrared(lR) laser. The Laser was fixed frequency of 2400Hz by continuous scanning and irradiating time was 8 minutes each point. Blood samples were taken at before, after, after 15min's treatment and Plasma $\beta-endorphin$ was measured by radioimmunoassay. The samples for this study were 6 normal subjects(3male, 3female). The data were analyzed by paired t-test, one-way ANOVA and simple regression. The results of this study were as follows : 1. The human plasma $\beta-endorphin$ levels were noted as significant increase in after-treatment $(22.84{\pm}10.63pg/ml)$ as compared with before-treatment $(16.96{\pm}9.23pg/ml)$ and significant increase in after 15min's $(27.27{\pm}8.81pg/ml)$ as compared with after-treatment (p<0.05). 2. There were no significant changes in plasma g-endorphin levels between male and female. 3. The human plasma $\beta-endorphin$ levels were high associated in between session reliability (p<0.05).

Effects of Whole Body Irradiation on Morphine, DAMGO, DPDPE, U50,488H and $\beta$-endorphin-Induced Antinociception

  • Opioid receptors have been pharmacologically classified as ${\mu}$, ${\delta}$, ${\kappa}$ and ${\varepsilon}$. We have recently reported that the antinociceptive effect of morphine (a ${\mu}$-opioid receptor agonist), but not that of ${\beta}$-endorphin (a novel ${\mu}/{\varepsilon}$-opioid receptor agonist), is attenuated by whole body irradiation (WBI). It is unclear at present whether WBI has differential effects on the antinociceptive effects of ${\mu}-$, ${\delta}-$, ${\kappa}-$ and ${\varepsilon}$-opioid receptor agonists. In our current experiments, male ICR mice were exposed to WBI (5Gy) from a $^{60}Co$ gamma-source and the antinociceptive effects of opioid receptor agonists were assessed two hours later using the hot water ($52^{\circ}C$) tail-immersion test. Morphine and $D-Ala^2$, $N-Me-Phe^4$, Gly-olenkephalin (DAMGO), [$D-Pen^2-D-Pen^5$] enkephalin (DPDPE), trans-3,4-Dichloro-N-methyl-N-[2-(1-pyrrolidinyl)-cyclohexyl]-benzeneacetamide (U50,488H), and ${\beta}$-endorphin were tested as agonists for ${\mu}$, ${\delta}$, ${\kappa}$, and ${\varepsilon}$-opioid receptors, respectively. WBI significantly attenuated the antinociceptive effects of morphine and DAMGO, but increased those of ${\beta}$-endorphin. The antinociceptive effects of DPDPE and U50,488H were not affected by WBI. In addition, to more preciously understand the differential effects of WBI on ${\mu}-$ and ${\varepsilon}$-opioid receptor agonists, we assessed pretreatment effects of ${\beta}$-funaltrexamine (${\beta}$-FNA, a ${\mu}$-opioid receptor antagonist) or ${\beta}$-$endorphin_{1-27}$ (${\beta}$-$EP_{1-27}$, an ${\varepsilon}$-opioid receptor antagonist), and found that pretreatment with ${\beta}$-FNA significantly attenuated the antinociceptive effects of morphine and ${\beta}$-endorphin by WBI. ${\beta}$-$EP_{1-27}$ significantly reversed the attenuation of morphine by WBI and significantly attenuated the increased effects of ${\beta}$-endorphin by WBI. The results demonstrate differential sensitivities of opioid receptors to WBI, especially for ${\mu}-$ and ${\varepsilon}$-opioid receptors.

Polytetrafluoroethylene의 열자격전류 특성에 미치는 $^{60}Co\;{\gamma}-$선 조사효과 (Effects of $^{60}Co\;{\gamma}-Ray$ Irradiation on TSC Characteristics of Polytetrafluoroethylene)

  • The effects of $^{60}Co\;{\gamma}-Ray$ irradiation on TSC characteristics of TFE type polytetrafluoroethylene(PTFE) were investigated. The $\alpha$ peak on TSC curve of PTFE was decreased, while the $\beta$ peak was reversely increased by the increasing of $\gamma$-irradiation doses up to 80 krad(0.8 kGy). Moreover, $\alpha$ and $\beta$ peaks on TSC curve of the irradiated PTFE are changed with the increasing of the forming temperature and forming electric field. In annealing the irradiated PTFE specimens at room temperature in air, it was shown a continuous recovery of TSC characteristics with time, which is assumed the traped charge carriers are liberated from the shallow traps and undergoes a recombination process during room temperature decay.

오디에서 열처리와 자외선 조사가 Anthocyanin-Polyphenol Copigment Complex의 안전성에 미치는 효과 (Effects of Temperature and UV Irradiation on Stability of Anthocyanin-Polyphenol Copigment Complex in Mulberry Fruits)

  • Anthocyanin and polyphenolic compounds present in fruits of mulberry (Morus alba L.) were determined and the influence of temperature and UV irradiation on stability of the anthocyanin-copigment complex were investigated. The copigmentation substance selected in non-anthocyanin fraction from mulberry for the study included: phenolic acid (hydroxybenzoic acid) and flavonoid (quercetin-3-O-$\beta$-D-glucopyranoside). The copigmentation effect increased with the copigment content. UV irradiation had a stronger degradation effect on the copigmentation complex than heating at $80^{\circ}C$. The non-anthocyanin fraction of mulberry and isolated flavonoid (quercetin-3-O-$\beta$-D-glucopyranoside) from mulberry fruit predominated over other copigment substances.

견의 결정성에 관한 연구(II) -견 피브로인의 산 처리에 의한 결정화와 황변억제효과에 대하여- (A Study on the Crystallinity in Silk Fibers(II) -Crystallization in Silk Fibroin by Acid Treatment and Its Photoyellowing Reduction Effect-)

  • In order to investigate the crystallization in silk fibroin induced by acid treatment and its photo-yellowing reduction effect. silk fibroin were immersed in PH 3.5~4.0 sulfuric acid and 0.01N hydro-chloric acid at 2$0^{\circ}C$, and the changes of crystallinity were measured by infrared spectroscopy, X-ray diffraction. Yellowness index caused by UV irradiation were also observed. The results are as follows : 1. ${\beta}$-form transition occurs when the silk fibroin are immersed at 2$0^{\circ}C$ in pH 3.5~4.0 sulfuric acid and 0.01N hydro-chloric acid. Formation of new hydrogenbond in molecules on the amorphous region induced ${\beta}$-form structure, and increased crystallinity. 2. Crystallization owing to $\beta$-form transition reduced the initial yellowness index by ultraviolet irradiation.

  • This study deals with the radiation protection effect of the pretreatment of $\beta$-carotene and combination with selenium on the DNA damage in mice after whole body ${\gamma}$-irradiation. This was obtained the radioprotective effect by evaluation of DNA damage levels in mice spleen and blood after irradiation. Six-week-old ICR male mice were administrated with $\beta$-carotene and combination with selenium orally once a day for 5 days and then irradiated with 8.0 Gy of $\gamma$-ray at a dose rate of 1.0 Gy/min. Development of an easy-to-handle murine model for the characterization of radiation-induced gross and molecular changes in skin

  • Background Radiation-induced skin injury is a dose-limiting complication of radiotherapy. To investigate this problem and to develop a framework for making decisions on treatment and dose prescription, a murine model of radiation-induced skin injury was developed. Methods The dorsal skin of the mice was isolated, and irradiation was applied at single doses of 15, 30, and 50 Gy. The mice were followed for 12 weeks with serial photography and laser Doppler analysis. Sequential skin biopsy samples were obtained and subjected to a histological analysis, immunostaining against transforming growth factor beta (TGF-${\beta}$), and Western blotting with Wnt-3 and ${\beta}$-catenin. Increases in the levels of TGF-${\beta}$, Wnt, and ${\beta}$-catenin were detected after irradiation. Results All tested radiation doses caused progressive dermal thickening and fibrosis. The cause of this process, however, may not be radiation alone, as the natural course of wound healing may elicit a similar response. The latent appearance of molecular and histological markers that induce fibrosis in the 15 Gy group without causing apparent gross skin injuries indicates that 15 Gy is an appropriate dose for characterizing the effects of chronic irradiation alone. Thus, this model best mimics the patterns of injury that occur in human subjects. Conclusions This animal model can be used to elucidate the gross and molecular changes that occur in radiation-induced skin injury and provides an effective platform for studying this adverse effect without complicating the process of wound healing.

감마선 조사된 우유단백질에 대한 우유 알러지 환자의 IgE 결합능의 변화 (Changes of Binding Ability of Milk-Hypersensitive Patients질 IgE to Gamma-Irradiated Milk Proteins)

  • This study was carried out to evaluate the application of food irradiation technology as a method for reducing milk allergies. Bovine $\alpha$-casein, $\beta$-casein, $textsc{k}$-casein, $\alpha$-lactalbumin(ALA), $\beta$-lactoglobulin (BLG) and serum albumin (BSA) were used as model allergens of milk proteins and the proten solution (2.0 mg/mL) with 0.01 M phosphate buffered saline (pH 7.4) was irradiated at 3, 5 and 10 kGy. Using milk-hypersensitive patients IgE (MHP-IgE), the changes of binding ability to irradiated proteins were observed by competitive indirect enzyme-linked immunosorbent assay (Ci-ELISA). Affinity of MHP-IgE to milk proteins was higher in ALA and BLG than that of other proteins. Standard curve to each non-irradiated protein could be made with MHP-IgE for quantifying milk allergens. Binding abilities of MHP-IgE to the irradiated proteins, however, decreased with different slopes of the standard curves. Sensitivity of gamma irradiation was higher in ALA and BLG than of other proteins. These results indicated that irradiation technology can be used to reduce the milk hypersensitivity.
