A Comparative Study on the Antimicrobial Activities of the Seeds of Prunus Species (한국산 Prunus 속 종자의 항균성과 청산배당체에 관한 연구)

  • HPLC analysis of three Prunus species, Armeniacae Semen. Persicae Semen, and Mume Semen, showed that 24. 06 mg/g, 5. 79 mg/g and 3/ 10 mg/g of amygdalin in the MeOH extract and 3. 59 mg/g, 5. 41 mg/g and 13. 48 mg/g of benzaldehyde in the MeOH extract hydrolyzed with ${\beta}-glucosidase$ were contained respectively. The MeOH extract of Mume Semen showed strong antibacterial activities against two species of bacteria (E. coli and B. subtilis) while that of Armeniacae Semen showed mild, and that of Persicae Semen showed no effect. Against A. niger, a strong antifungal activity was observed with Armeniacae Semen and only mild activities with Persicae Semen and Mume Semen. None oh the three inhibited the growth of S. cervisiae. The above results may possibly be suggestive of the correlationships between the contents of amygdalin and benzaldehyde in the test extracts with the antimicrobial potencies.

  • The constituents of Sanguisorba hakusanensis leaves (Rosaceae), of which the roots have been used as an astringent, hemostatics and antiphlogistics, were studied phytoche mically. From water fraction of the MeOH extract, gallic acid 3-O-${\beta}$-D-(6'-O-galloyl)-glucopyranoside(I), quercetin-3-O-${\beta$-D-galactopyranoside(II), quercetin-3-O-${\alpha}$-L-arabinopyranoside(III) and $2{\alpha},\;3{\beta},\;19{\alpha}$, 23-tetrahydroxyurs-12-en-28-oic acid 28-O-${\beta}$-D-glucopyranoside(IV) were isolated by column chromatographic separation using Amberlite XAD-2, ODS-gel and Sephadex LH-20. The structure of these compounds were elucidated by spectroscopic parameters of $^1H$-NMR, $^{13}C$-NMR, $^1H-^1H$ COSY, $^{13}C-^1H$ COSY, EI-Mass, FAB-Mass, IR, UV and by comparison with authentic samples.

IN VITRO DEVELOPMENT OF BOVINE ONE-CELL EMBRYOS FERTILIZED IN VITRO IN SERUM- AND FEEDER CELL-FREE CULTURE SYSTEMS

  • The purpose of this study was to evaluate some factors in the bovine embryonic development from one-cell to blastocyst using modified synthetic oviduct fluid medium (mSOFM), after maturation and in vitro fertilization of the oocytes. The embryonic development to the blastocyst stage was assessed at 7-10 days after in vitro fertilization, and the total cells in the blastocysts were counted by staining nuclei with fluorochrome. Some commercial calf sera (CS) and a superovulated cow serum had different effects on the embryonic development to the blastocyst stage (8.6-21.4%), dependent upon their product lots, although the development might not be affected at least by serum progesterone levels. ${\beta}$-Mercaptoethanol (${\beta}$-ME) supplemented into mSOFM was effective to the embryonic development (27.8%), as well as the co-culture system with cumulus cells (19.5%). In a serum- and feeder cell-free culture using mSOFM containing several growth factors and ${\beta}$-ME instead of CS plus co-cultured cumulus cells, bovine serum albumin (BSA, fraction V), but not polyvinyl alcohol (PVA), was highly effective in embryonic development to the blastocyst stage, almost comparable to CS in the serum-contained culture (CS, BSA and PVA; 27.8, 19.5 and 5.7%, respectively). However, fatty acid free BSA rather reduced the number of developed blastocysts, compared with fraction V BSA (7.3 vs 29.4%). In the serum- and feeder cell-free culture, supplement of glucose to the medium (final 2.0 mM) stimulated the cell proliferation of developing embryos 120 hr after in vitro fertilization. These results indicated that a serum-free medium supplemented with ${\beta}$-ME could successfully support the development of bovine one-cell embryos to the blastocyst stage. Moreover, supplement of glucose and fatty acids to the medium might support preferably the development and cell proliferation of embryos.

  • The fruiting body of Phellinus linteus was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2$O. The repeated silica gel and ODS column chromatographies of the EtOAc fraction led to isolation of four sterols. From the result of spectral data including NMR, MS and IR, the chemical structures of the sterols were determined as ergosta-7,24(28)-dien-3${\beta}$-ol (episterol, 1), 5${\alpha}$,8${\alpha}$-epidioxyergosta-6,9(11),22-trien-3${\beta}$-ol (dehydrop-eroxyergosterol, 2), 5${\alpha}$,8${\alpha}$-epidioxyergosta-6,22-dien-3${\beta}$-ol (ergoterol peroxide, 3), and $3{\beta}$,$5{\alpha}$-dihydroxy-6${\beta}$-methoxyergosta-7,22-diene (6-O-methylcerevisterol, 4). The ergosterols have been first isolated from this mushroom in this study.

Energy Utilization of Growing Chicks in Various Nutritional Conditions

  • For the last two decades, energy utilization of growing chicks has been studied more and more. This paper focuses on the energy utilization estimated by the metabolizable energy (ME) values and the efficiency at which ME is used for growth of chicks under various nutritional environment. Degree of saturation of dietary fats is responsible for nitrogen-corrected apparent metabolizable energy (AMEn) of fats. The effect of dietary fat sources on heat production depends on the kind of unsaturated fatty acids as well as the degree of saturation. Medium chain triglyceride shows lower AME and net energy than long chain triglyceride. Phytase as feed additives increases the AME values of the diet along with improvement of the phosphorous utilization. Ostriches have higher ability to metabolize the energy of fiber-rich foodstuffs than fowls. Their higher ability seems to be associated with fermentation of fiber in the hindgut. Proportions of macronutrients in the diets have influenced not only the gain of body protein and energy but also the oxidative phosphorylation of the chicken liver. Essential amino acids deficiency reduces ME/GE (energy metabolizability) little, if any. Growing chicks respond to a deficiency of single essential amino acids with the reduction of energy retained as protein and increased energy retained as fat. Thus, energy retention is proportional to ME intake despite deficiency, and efficiency of ME utilization is not affected by deficiency of amino acids. Effect of oral administration of clenbuterol, a beta-adrenergic agonist, on the utilization of ME varies with the dose of the agents. Although the heat production related to eating behavior has been estimated less than 5% of ME, tube-feeding diets decreases HI by about 30%.

A New Lupane-Triterpene Glycoside from the Leaves of Acanthopanax gracilistylus

  • A new and two known lupane-triterpene glycosides were isolated from the hot MeOH fraction of the leaves of Acanthopanax gracilistylus W. W. Smith. Based on the physical properties and spectroscopic data, their chemical structures were determined as acankoreoside A (1), acankoreoside D (2), and $3{\alpha}-hydroxy-lup-23-al-20(29)-en-28-oic$ acid $28-O-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}4)-{$beta}-D-glucopyranosyl-(1{\rightarrow}6)-{\beta}-D-glucopyranosyl$ ester (3), respectively. To our best knowledge, compand 3 appears to be novel, which was named as wujiapioside A.

Reducing Power and ${\alpha}-Glucosidase$ Inhibitory profiles of (-)-Catechin and Its glycoside ((-)-Catechin 및 배당체의 환원력 및 ${\alpha}-glucosidase$저해 활성)

  • From the EtOAc fraction of the MeOH extract of Ulmus davidiana, (-)-catechin (1), (-)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and (-)-catechin-7-O-${\beta}$-D-xylopyranoside (3) were isolated and characterized on the basis of $^1H-and\;^{13}C-NMR$, and FABMS spectral data. Compounds 1-3 showed more strong reducing power activities than ${\alpha}-tocopherol$, a positive control.

  • The rhizoma of Polygonatum odoratum (Liliaceae) has been used as the treatment of body fluid deficiency, dryness symptoms and hyper glycemia. To study effects on acute renal failure of P. odorati rhizoma, urinary volume, urinary electrolytes and serum factors associated with renal dysfunction were measured in $HgCl_2-induced$ acute renal failure rats. It was revealed that its ether ex. had significant diuretic effects, regulated hypoelectrolytes$(Na^+,\;K^+,\;Cl^-)$ and inhibited the increase of creatinine, BUN in acute renal failure rats. From the ether ex. which was more effective than MeOH ex., three lipophilic compounds were isolated and elucidated hydrocarbon(l), ${\beta}-sitosteryl$ stearate(2), ${\beta}-sitosterol$(3). The ${\beta}-sitosteryl$ stearate(2) were isolated from Polygonatum sp. at first.

  • For the investigation of medicinal resources in Aconitum species, the studies were carried out to search the less toxic constituents than Aconitum alkaloid in the aerial part of Aconitum jaluense for. album (Ranunculaceae). From the BuOH fraction of MeOH extract, three flavonoid compounds, hyperoside(1), $kaempferol-3-O-{\beta}-_D-galactopyranoside(2)$, $kaempferol-3-O-{\alpha}-_L-rhamnopyranosyl-(1{\rightarrow}6)-{\beta}-_D-glucopyranoside(3)$ were isolated and identified on the basis of their physico-chemical properties, spectroscopic evidences (UV, IR, NMR and Mass etc.) and comparison with authentics, respectively.

Antioxidant compounds from the stem bark of Sorbus commixta

  • The MeOH extract of Sorbus commixta (Rosaceae) exhibited strong DPPH radical scavenging activity, and through activity-guided fractionation two antioxidant compounds were isolated and identified as $catechin-7-O-{\beta}-D-xylopyranoside$ (1) and $catechin-7-O-\;{\beta}-D-apiofuranoside$ (2) by physicochemical and spectrometric methods. To evaluate the antioxidant effect of these compounds, some in vitro tests, such as the DPPH radical scavenging activity test, the superoxide radical scavenging activity test and the lipid peroxidation inhibitory activity test, were performed. Compounds 1 and 2 showed stronger activities than both a-tocopherol and butylated hydroxy anisole (BHA) in each assay.
