The secretory efficiency of recombinant xylanase xynB from yeast Pichia pastoris between the ${\alpha}$-factor preprosequence and a classical mammalian signal peptide derived from bovine ${\beta}$-casein was compared. The results showed that although the bovine ${\beta}$-casein signal peptide could direct high-level secretion of recombinant xylanase, it was relatively less efficient than the ${\alpha}$-factor preprosequence. In contrast, the bovine ${\beta}$-casein signal peptide caused remarkably more recombinant xylanase trapped intracellularly. Real-time RT-PCR analysis indicated that the difference in the secretory level between the two signal sequences was not due to the difference in the transcriptional efficiency.

Vitexin, a natural flavonoid compound identified as apigenin-8-C-${\beta}$-D-glucopyranoside, has been reported to exhibit antioxidative and anti-inflammatory properties. In this study, we investigated its effect on hypoxiainducible factor-$1{\alpha}$ (HIF-$1{\alpha}$) in rat pheochromacytoma (PC12), human osteosarcoma (HOS) and human hepatoma (HepG2) cells. Vitexin inhibited HIF-$1{\alpha}$ in PC12 cells, but not in HOS or HepG2 cells. In addition, it diminished the mRNA levels of hypoxia-inducible genes such as vascular endothelial growth factor (VEGF), smad3, aldolase A, enolase 1, and collagen type III in the PC12 cells. We found that vitexin inhibited the migration of PC12 cells as well as their invasion rates, and it also inhibited tube formation by human umbilical vein endothelium cells (HUVECs). Interestingly, vitexin inhibited the hypoxia-induced activation of c-jun N-terminal kinase (JNK), but not of extracellular-signal regulated protein kinase (ERK), implying that it acts in part via the JNK pathway. Overall, these results suggest the potential use of vitexin as a treatment for diseases such as cancer.

This study evaluated the effect of a combination of acetaminophen and vitamin C (CAV) on reducing serum cortisol and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) concentrations in piglets vaccinated with foot-and-mouth disease (FMD) vaccine. Piglets were vaccinated with FMD vaccine and treated with CAV at concentrations of 0.0, 0.5, 1.0, and 2.0 kg/ton feed (P-CON, AD-1, AD-2, and AD-3, groups, respectively) for 5 days post-vaccination. Cortisol and $TNF-{\alpha}$ levels at 5 days post-treatment in the AD-1-3 groups were significantly lower than that in the P-CON group (p < 0.05). There were no significant differences between AD-2 and AD-3 groups and non-vaccinated, non-CAV-treated piglets.

In this study, $Pb_{0.88}(La_{\alpha}Nd_{1-\alpha})_{0.08}(Mn_{1/3}Sb_{2/3})_{0.02}Ti_{0.98}O_3$ system ceramics with La molar ratio $\alpha$ variation were manufactured for 24 MHz class resonator application. Electromechanical coupling factor, mechanical quality factor and dynamic range of $3^{rd}$ overtone thickness vibration mode were measured as the variations of La and Nd molar ratio. Mechanical quality factor and dynamic range at $\alpha$ = 0.6 composition ceramics showed the highest value of 2691 and 52.37 dB, respectively. The temperature coefficient of resonant frequency measured from $-20^{\circ} to 80^{\circ}$ showed an excellent value of $5ppm/^{\circ}C$ at $\alpha$=1 composition ceramics.

Purpose: The purpose of this study was to evaluate the reliability and validity of the Korean version of the Successful Aging Inventory (SAI-K) to determine its suitability for use with older Korean adults. Methods: Exploratory and confirmatory factor analyses were utilized to assess the factor structure and the construct validity of the SAI-K. First- and second-order Confirmatory Factor Analysis (CFA) were conducted to identify the most adequate model. Cronbach's alpha was used to test the reliability. Results: Using a second-order CFA, a four-factor structure was validated (${\chi}^2$=122.82, p<.001, GFI=.92, AGFI=.88, SRMR=.06 RMSEA=.07, 90% CI=.05-.09, CFI=.93). The four-factor SAI-K showed reliable internal consistency with a Cronbach's alpha for the total scale of .86. Conclusion: The four-factor, 13-item SAI-K showed satisfactory reliability and validity and, thus, has the potential to be an appropriate instrument for measuring successful aging in older Korean adults.

Pro-inflammatory cytokine인 종양괴사인자-$\alpha$ (TNF-$\alpha$)는 B형 간염바이러스(HBV) 감염에 대한 면역반응의 중요한 매개인자이다. TNF-$\alpha$의 생산은 전사수준에서 주로 조절되며, TNF-$\alpha$ promoter 다형성은 TNF-$\alpha$ 유전자 발현을 변화시키기 때문에, TNF-$\alpha$ promoter 다형성이 만성 HBV 감염의 병태생리에 영향을 미칠 가능성이 높다. 그러므로, 본 연구에서는 TNF-$\alpha$ promoter 다형성과 만성 HBV 감염의 연관성을 조사하였다. 본 연구는 만성 HBV 감염환자 181명, HBV 감염이 자연적으로 치유된 201명, 그리고 건강한 대조군 170명을 대상으로 하였다. TNF-$\alpha$ promoter의 -308G/A와 -238G/A 다형성은 PCR-RFLP법을 통하여 분석하였다. 분석결과, 환자군에서 -308과 -238 유전자형의 분포는 자연치유군 및 건강대조군에서의 분포와 통계적인 차이를 보이지 않았다(p>0.05). 또한, 그룹 간에 allele frequency도 통계적 차이가 없었다(p>0.05). 따라서, 본 연구의 결과는 한국인에서 TNF-$\alpha$ -308과 -238 promoter 다형성은 만성 B형 간염 바이러스 감염의 발생과 연관성이 없음을 시사한다.

The estrogen receptor (ER) is activated and degraded by estrogen. We have examined ER downregulation and activation under hypoxia mimetic conditions. Cobalt chloride induced ER downregulation at 24 h of treatment. This degradation involved hypoxia-inducible factor-1$\alpha$ (HIF-1$\alpha$) as examined by using a constitutively active form of HIF-1$\alpha$, HIF-1$\alpha$/VP16, constructed by replacing the transactivation domain of HIF-1$\alpha$ with that of VP16. Western blot analysis revealed that E2-induced ER downregulation was observed within ${\~}6h$, whereas HIF-1$\alpha$/VP16-induced ER degradation was observed within 12${\~}$20h. HIF-1$\alpha$/VP16 activated the transcription of estrogen-responsive reporter gene in the absence of estrogen. These results suggest that ER downregulation and activation under hypoxia maybe mediated in part by a HIP-1$\alpha$ expression.

The effect of Ligustrum Lucidum(LL) on the production of nitric oxide (NO) and superoxide by murine peritoneal macrophages were investigated. Stimulation of the cells with LL in the presence or absence of interferon-r(IFN-r) resulted in the increased accumulation of nitrite in the medium. To further examine the mechanism of LL induced. NO Synthesis, we evaluated the secretion of tumor necrosis $factor-{\alpha}(TNF-{\alpha})$ by LL in murine macrophages. Treatment of LL increased the secretion of bioactive $TNF-{\alpha}$ in cultured medium. In addition, LL induced NO production was decreased by the treatment of anti-murine $TNF-{\alpha}$. neutralizing antibodies, indicating that LL induced superoxide production was decreased by the treatment of anti-murine $TNF-{\alpha}$ neutralizing antibodies. These data suggested that LL induced superoxide production was related to $TNF-{\alpha}$ secretion. In conclusion, our results indicates that LL may enhance innate immune response and be applied as a immunoregulating drug improving phagocytosis.

Extract of HLT inhibit A23187, PMA-induced IL-8, tumor necrosis factor-$\alpha$ (INF-$\alpha$) production in HMC-1 cells. HLT down-regulated LPS-induced IL-8 production and A23187, PMA-induced IL-8, TNF-$\alpha$ production, which may be provide a clinical basis for anti-inflammatory properities of HLT.
