Sasa borealis extract exerts an antidiabetic effect via activation of the AMP-activated protein kinase

  • Leaf of Sasa borealis, a species of bamboo, has been reported to exhibit anti-hyperglycemic effect. However, its antidiabetic mechanism is not fully understood. In this study, we examined whether an extract of S. borealis activates AMP-activated protein kinase (AMPK) and exerts anti-hyperglycemic effects. Treatment with the S. borealis extract increased insulin signaling and phosphorylation of AMPK and stimulated the expression of its downstream targets, including $PPAR{\alpha}$, ACO, and CPT-1 in C2C12 cells and $PPAR{\alpha}$ in HepG2 cells. However, inhibition of AMPK activation attenuated insulin signaling and prevented the stimulation of AMPK target genes. The S. borealis extract increased glucose uptake in C2C12 cells and suppressed expression of the gluconeogenic gene, PEPCK in HepG2 cells. The extract significantly reduced blood glucose and triglyceride levels in STZ-induced diabetic mice. The extract enhanced AMPK phosphorylation and increased Glut-4 expression in the skeletal muscle of the mice. These findings demonstrated that the S. borealis extract exerts its anti-hyperglycemic effect through activation of AMPK and enhancement of insulin signaling.

Some Properties and Optimal Culture Conditions of Cyclodextrin Glucanotransferase of Bacillus sp. S-6 Isolated from Kimchi (김치에서 분리한 Bacillus sp. S-6의 Cyclodextrin Glucanotransferase의 특성과 최적생산조건)

  • A microorganism capable of producing high level of extracellular cyclodextrin glucanotransferase(EC 2.4.1.19 ; CGTase) was isolated from Kimchi. 2-O-$\alpha$-D-glucopyranosyl L-ascorbic acid(AA-2G) was synthesized by transglycosylation reaction of CGTase using starch as a donor and L-ascorbic acid as an acceptor. The isolated strain S-6 was identified as Bacillus sp. S-6. The maximal CGTase production was observed in a medium containing 0.5% soluble starch, 1% yeast extract, 1% NaCO3, 0.1% K2HPO4, and 0.02% MgSO4 with initial pH 8.0. The strain was cultured at 37$^{\circ}C$ for 40 hr with reciprocal shaking. Using the culture supernatant as crude enzyme, the optimal pH and temperature of the CGTase activity of this strain were 7.0 and 4$0^{\circ}C$. In the effects of pH and temperature on the stability of the enzyme, the enzyme was stable in the range of pH 6.0~10.0 and up to 45$^{\circ}C$, respectively.

Mapping the Polarization of the Radio-Loud Lyman Alpha Nebula B3 J2330+3927

  • $Ly{\alpha}$ nebulae, or "$Ly{\alpha}$ blobs", are extended (~100 kpc), bright (L[$Ly{\alpha}$] ~ 1044 erg/s) clouds of $Ly{\alpha}$-emitting gas. The origin of the $Ly{\alpha}$ emission remains unknown, but recent theoretical work suggests that measuring the polarization could discriminate among powering mechanisms. we will discuss current status of $Ly{\alpha}$ polarization observations at high-redshift and our on-going survey program. We will present the first narrow-band, imaging polarimetry of a $Ly{\alpha}$ blob, B3 J2330+3927 at z=3.09, with an embedded, radio-loud AGN (C. You et al. in prep.). The AGN lies near the blob's $Ly{\alpha}$ emission peak and its radio lobes align roughly with the blob's semi-major axis. With the SPOL polarimeter on the MMT telescope, we map the polarization in a grid of circular apertures of radius 0.6" (4.4 kpc), detecting a significant (>$2{\sigma}$) polarization fraction P% in 10 apertures and achieving strong upper-limits (as low as 2%) elsewhere. The degree of the polarization map increases from P% ~ 5% at ~5 kpc from the blob center to ~20% at the outer part (~30 kpc). The detections are distributed asymmetrically, roughly along the blob's major axis. The polarization angles (${\Theta}$) are mostly perpendicular to this axis. These results are consistent with the picture that $Ly{\alpha}$ photons produced at the AGN (or the host galaxy) are resonantly scattered away from the center. Higher polarization fraction on the radio jet suggests that the gas is more optically thin along the jet than the off-axis region.

Rheological Properties of the Solutions of Incompatible Polymer Blends

  • A blend polymeric system composed of poly(methyl methacrylate) (PMMA or PM) and polystyrene (PS) dissolved in chloroform was rheologically studied. The viscosities ${\eta}_{bl}$ of the blend system with various blending ratios ${\chi}$ changing from zero (pure PS solution) to unity (pure PMMA solution) were measured at $25{\circ}C$ as a function of shear rates ${\dot{s}}$ by using a Couette type viscometer. ${\eta}_{bl}$ at a given ${\dot{s}}$ decreased exponentially with ${\chi}$ reaching asymptotic constant value of ${\eta}_{bl}$ ; ${\eta}_{bl}$ at a given ${\chi}$ is greater at a smaller ${\dot{s}}$. These results are explained by using Ree-Erying's theory of viscosity, ${\eta}_{bl}=(x_1{\beta}_1/{\alpha}_1)_{b}_1+ (x_2{\beta}_2/{\alpha}_2)_{bl}[sinh^{-1}{\beta}_2(bl) {\dot{s}}]/{\beta}_2(bl){\dot{s}}$. The Gibbs activation energy ${\Delta}G_i^\neq$(i = 2 for non-Newtonian units) entering into the intrinsic relaxation time ${\beta}$ is represented by a linear combination ${\Delta}G_i^\neq(bl) ={\chi}{\Delta}G_i^{\neq}_{iPM}+(1-{\chi}){\Delta}G_i^{\neq}_{iPS}$;the intrinsic shear modulus$[[\alpha}_i]^{-1}$ is also represented by $[{\alpha}_i(bl)]^{-1}={\chi}[{\alpha}_{iPM}]^{-1}+(1-{\chi})[{\alpha}_{iPS}]^{-1}$ and the fraction of area on a shear surface occupied by the ith flow units $x_i(bl)$ is similarly represented, i.e., $x_i(bl) = {\chi}x_{iPM}+(1-{\chi})x_{iPS}$. By using these ideas the Ree-Eyring equation was rewritten which explained the experimental results satisfactorily.

  • Versions of Tannaka duality in operator algebraic context have been obtained in [6], [8] etc. Suppose .sigma.is an automorphism of a von Neumann algebra M, on which there is an action .alpha. of a compact group G such that .sigma. vertical bar $M^{\alpha}$=id, where $M^{\tau}$is the fixed point algebra under the action .alpha.. Then it is shown that if there is an action .tau. of a group H which commutes with .alpha., and which is ergodic in the sense that the fixed point algebra $M^{\tau}$ is trivial, then there exists g.mem.G such that .sigma.=.alpha.(g).   • The effects of linear free energy relationship (LFER) on the imidoyl proton (H${\alpha}$)-substituent chemical shift (SCS) in case of varying para-substituted C-phenyl group in N-benzylideneaniline derivatives were studied. The H${\alpha}$-SCS values and LFER parameters such as ${\sigma}$,${\sigma}^+$, ${\sigma}_I$,${\sigma}_R, F and R were applied to the Hammett, Okamoto-Brown, and Taft, Swain-Lupton's dual substituents parameter (DSP) equations. The results were: (1) the blending coefficient values, ${\lambda}$ = 2.8∼3.2, it's means that the resonance effect (R) was larger than inductive effect (I) and field effect (F), and (2) the values of percent resonance and percent field effects were %R = 66.6 and %F = 33.4, respectively, yielding the ratio of resonance effect (R) to field effect (F) of 2 : 1.

  • The Ag-Pd-Cu alloys containing a small amount of Au is commonly used for dental purposes, because this alloy is cheaper than Au-base alloys for clinical use. However, the most important characteristic of this alloy is age-hardenability, which is not exhibited by other Ag-base dental alloys. The specimens used were Ag-20Pd-20Cu ternary alloy and Au addition alloy. These alloys were melted and casted by induction electic furace and centrifugal casting machine in Ar atmoshpere. These specimens were solution treated for 2hr at $800^{\circ}C$ and were then quenched into iced water, and aged at $350{\sim}550^{\circ}C$ Age-hardening characteristics of the small Au-containing Ag-pPd-Cu dental alloys were investigated by means of hardness testing, X-ray diffraction and electron microscope observations, electrical resistance, differential scanning calorimetric, emergy dispersed spectra and electron probe microanalysis. Principal results are as follows : Hardening occured in two stages, I. e., stage I in low temperature and stage II in high temperature regions, during continuous aging. The case of hardening in stage I was due to the formation of the Llo type face centered tetragonal PdCu-ordered phase in the grain interior and hardening in stage I was affedted by the Cu concentration. In stage II, decomposition of the $\alpha$ solid solution to a PdCu ordered phase(L1o type) and an Agrich ${\alpha}2$ phase occurred and a discontiunous precipitation occurred at the grain boundary. Form the electron microscope study, it was concluded that the cause of age-hardening in this alloy is the precipitation of the PdCu ordered phase, which has AuCu I type face-centered tetragonal structure. Precipitation procedure was ${\alpha}\to{\alpha}+{\alpha}2+PdCu\to{\alpha}1+{\alpha}2+PdCu$ at Pd/Cu = 1 Ag-Pd-Cu alloy is more effective dental alloy as ageing treatment and is suitable to isothermal ageing at $450^{\circ}C$.

Fabrication, Microstructures and High-Strain-Rate Properties of TiC-Reinforced Titanium Matrix Composites

  • TiC ceramic particulate-reinforced titanium matrix composites were fabricated and the resultant densification, microstructure, and static and dynamic mechanical properties were studied. Comparing Ti with TiH₂powders as host materials for TiC ceramic reinforcement by pressureless vacuum sintering, TiH₂-started composites showed better sinterability and resistance to both elastic and plastic deformation than Ti-started ones. When TiH₂and TiH₂-45 vol.%TiC samples were hot pressed, TiH₂matrices transformed to alpha prime Ti and alpha Ti phase, respectively. It is interpreted that the diffusion of an alpha stabilizer carbon from TiC into the matrix is one of the plausible reasons far such a microstructural difference. The 0.2% offset yield strengths of the hot pressed TiH₂and TiH₂-45 vol.%TiC samples were 1008 and 1446 MPa, respectively, in a static compressive mode (strain rate of 1×$10^{-3}$/s). Dynamic compressive strengths of the samples were 1600 and 2060 MPa, respectively, at a strain rate of 4×10³/s.

  • Crystals of tolazamide, $C_{14}H_{21}N_3O_3S$, are triclinic space group $P{\bar{1}}$ with cell dimensions of a = 6.355 (2), b = 9.223 (2), c = 13.510 (3) A, ${\alpha}\;=\;101.04\;(8),\;{\beta}=92.80(5),\;{\gamma}\;=\;85.72\;(6)^{\circ}$ and Z = 2. Intensities were collected on an automated four-circle diffractometer using graphite-monochromated Cu K ${\alpha}$ radiations. The structure was solved by direct method and refined by full-matrix least-squares to an R factor of 0.058 for 1184 observed reflections. The molecules are dimerized by the $N-H{\cdots}O$ hydrogen bonds. There are only van der Waals interactions between these molecular dimers.

  • The cyclodextrin glucanotransferase (CGTase) gene from Bacillus stearothermophilus NO2 was expressed in Saccharomyces cerevisiae 2805 under the adhl promoter. The CGTase was purified from S. cerevisiae 2805/pVT-CGTS. The purified enzyme exhibited a optima of activity around pH 7.0 and $65^{\circ}C$. Thermal stability of the enzyme was increased fairly as compared with the CGTase of B. stearothermophilus NO2. The conversion yield of cyclodextrin (CD) and the production ratio of $\alpha$-, $\beta$,-, ${\gamma}$-CD from starch were showed similarly aspect to the CGTase of B. stearothermophilus NO2.
