정상과 설사증 송아지에서 혈중 retinol과 $\alpha$-tocopherol의 비교 (The Comparison of Blood Retinol and $\alpha$-Tocopherol Concentrations between Diarrhea and Normal Calves)

  • The purpose of this study is to examine and compare the concentrations of blood retinol and $\alpha$-tocopherol in normal and diarrhea calves. The subjects of the experiments are from three groups. Each group consists of 5 calves(diarrhea, treatment and control group). The CBC, serum chemistry, serum retinol and a-tocopherol concentrations were estimated in each group. Blood retinol concentration was $13.3{\pm}7.0{\mu}g/100ml$ in diarrhea group, $31.5{\pm}6.9{\mu}g/100ml$ in treatment group and $28.1{\pm}11.8{\mu}g/100ml$ in the control group. The blood concentration in diarrhea group was significantly lower than that of the control group in the case of retinol(p<0.05). However, there were no significance between the treatment poop and the control group. The $\alpha$-tocopherol concentration in blood was $266.0{\pm}127.6{\mu}g/100ml$ in diarrhea group, $432.2{\pm}172.7{\mu}g/100ml$ in the treated group and $579.3{\pm}145.8{\mu}g/100ml$ in the control group. In the case of $\alpha$-tocopherol, the laboratory group were significantly lower than the control group(p<0.05), except for the treatment group. As in retinol concentration there were no significance between treatment group and the control group. In the test of CBC, PCV was significantly lower in the group with diarrhea than the control group(p<0.05). Fibrinogen concentrations in diarrhea calves were significantly higher than the treatment and control group. In conclusion, the blood retinol and $\alpha$-tocopherol concentration in diarrhea calves are lower than normal calves. Medication of retinol and $\alpha$-tocopherol on calves with diarrhea is recommended.

에탄올을 장기간 섭취한 흰쥐의 간기능에 미치는 아스코르빈산 및 알파토코페롤 투여의 영향 (Effects of Ascorbate and $\alpha$-Tocopherol Administration on Liver Function in Chronically Ethanol-Treated Rats)

  • 본 연구는 장기간의 에탄올을 섭취시킨 흰쥐에 있어서 최근에 개발된 CL-HPLC를 이용해 측정한 과산화물 및 혈액 생화학치의 변화를 조사하고, 또한 이들 수치에 미치는 아스코르빈산, 알파토코페롤 및 아스코르빈산 알파토코페롤 병합투여의 영향을 알아보기 위하여 실시하였다. 그 결과, 에탄올 투여군은 대조군에 비하여 체중은 감소하였으나, 상대적 간장중량, GPT의 수치가 증가하였으며, 특히 과산화물가가 유의하게(p<0.001) 증가하였다. 반면에 에탄올 투여군에 3주동안 아스코르빈산, 알파토코페롤 및 아스코르빈산.알파토코페롤 병합 투여를 한 결과, 알파토코페롤 투여군 및 아스코르빈산.알파토코페롤 병합투여군에서 상대적 간장중량, 콜레스테롤함량, GPT의 수치 및 과산화물가가 회복되었다. 이 결과는 식이중의 알파토코페롤 및 아스코르빈산.알파토코페롤 투여가 에탄올로 유도된 간장의 과산화지질의 형성을 방지해, 간기능을 회복시키는데 효과적이라는 것을 시사한다 하겠다.

  • Effects of tocopherols and $\beta$-carotene on the oxidation of the solid model system of a free fatty acid mixture (64.5% of linolic acid ; 26.4% of oleic acid ; 5.0% of palmitic acid) with tocopherols and $\beta$-carotene were studied. $\alpha$-tocopherol revealed an antioxidant activity at the concentration below 0.05%, however, it showed a prooxidant activity when the concentration was higher than 0.05%. The antioxidant activity of ${\gamma}$ -tocopherol was not affected by the concentrations in the range of 0.01~0.10% in the model and ${\gamma}$-tocopherol showed higher antioxidant activity than that of $\alpha$-tocopherol. It seemed that $\alpha$-tocopherol was unstable compared to ${\gamma}$-tocopherol during oxidation. $\beta$-carotene showed a weak antioxidative activity at the initial stage of this system while $\beta$-carotene showed a prooxidant activity in the presence of tocopherol. $\beta$ -carotene was highly susceptible to autoxidative degradation during oxidation.

  • Safflower is an important medicinal plant that has been used in China, Korea and Japan for thousands of years. The red and yellow pigments obtained from the petals of safflower can invigorate blood, release stagnation and promote menstruation. In addition, these pigments are used safely in processed foods and soft drinks as naturally harmless rotor additives. On the other hand, the seed of safflower contains 30-40% oil with higher level of mono- and poly-unsaturated fatfy acid profiles and elevated levels of $\alpha$-tocopherol. In this paper, we describe advances in the production of pigments and $\alpha$-tocopherol by cell culture in safflower.

  • Induction of micronuclei by diesel emission particle extract (DEPE) in mouse bone marrow cells was suppressed by pre-treatment with ascorbic acid, ${\alpha}-tocopherol$ or glutathione (GSH). These compounds were given orally to mice at the dose of 1, 10 or 100 mg/kg/day for 5 consecutive days. The dose of DEPE (200 mg/kg) was administered intraperitoneally once to mice with the 5th administration of test compounds. Ascorbic acid, ${\alpha}-tocopherol$ and GSH all showed the dose-dependent suppression on DEPE-induced micronuclei.

  • Rice, the staple food in Korea, is deficient to some extent in protein, lipid and vitamins. This study was carried out in order to investigate the effects of dietary supplementation to the rice diet of cellulose, ginseng, and ${\alpha}$-tocopherol on lead toxicity in rats. Using male rats fed the rice diet with the distilled drinking water containing 750mg of lead as nitrate per liter, for 11 weeks, organ weights, hemoglobin levels, serum glutamic pyruvic transaminase activity and accumulation of lead in liver, blood and kidney were observed. Supplementation of cellulose, ginseng and ${\alpha}$-tocopherol to the lead groups showed the protective effect significantly in the weight of liver but no influence in hemoglobin levels. Ginseng especially decreased the serum glutamic pyruvic transaminase activity to normal level. The three supplemented diets reduced the lead accumulation in kidney and blood, but not in liver.

  • The antioxidant effects of aqueous green tea extracts with $\alpha$-tocopherol and lecithin dissolved in lard were investigated on the lipid metabolism in serum and liver of Sprague-Dawley male rats. Elderly male rats were fed for 8 weeks by different experimental diets containing 10% lard(w/w), 0.5% aqueous green tea extracts, 0.05% $\alpha$-tocopherol and 1.0% lecithin. By the addition of the mixture of the antioxidants, the level of total cholesterol. HDL-cholesterol and triglyceride in serum and liver were not changed. The activities of glutathione peroxidase were significantly increased in the case of addition of the antioxdant mixtures but the activities of superoxide dismutase and glutathione S-transferase were not affected.

대두유의 자동산화 및 가열산화에 미치는 정향추출물의 항산화 효과 (Effects of Clove Extracts on the Autoxidation and Thermal Oxidation of Soybean Oil)

  • This study was conducted to investigate the antioxidant activities of clove extracts in water, methanol and ether. The clove extracts, BHA and ${\alpha}$-tocopherol were added to each oil at a level of 200 ppm. The activities of the substrate oils and controls were tested under autoxidation and thermal oxidation conditions. The degree of the effects of the antioxidant activities under autoxidation condition were in the following order; ether extract 〉 methanol extract 〉BHA 〉 ${\alpha}$-tocopherol 〉 water extract = control group. The induction periods of the control, water, methanol and ether extracts, and BHA and ${\alpha}$-tocopherol were 9.5, 9.6, 10.7 11.8, 10.4 and 9.7 days, respectively. Under thermal oxidation condition, the methanol extract showed stronger antioxidant activity than those of the water and ether extracts. The antioxidant activity of the methanol extract was attributed to ${\alpha}$-tocopherol and BHA.

Liquid chromatographic determination of α-, β-, γ-, and δ-tocopherol in sesame oils of different origin

  • The contents of ${\alpha}$-, ${\beta}$-, ${\gamma}$-, and $\delta$-tocopherols in sesame oils were determined by high performance liquid chromatography with UV detection. ${\alpha}$-Tocopherol contents ranged from 10.28 to 19.79 mg/g oil, ${\beta}$-tocopherol contents from 8.22 to 20.10 mg/g. However, both ${\gamma}$- and $\delta$-tocopherol were less than 1.49 mg/g or not detected. ${\gamma}$-Tocopherol was not detected from both unroasted white and black sesame seed oils. Significantly higher level of tocopherol in sesame oil than other oils is an evidence of the reason why it is highly stable and prevents oxidation. The tocopherol composition for twenty sesame oils was classified by using principal component analysis.

  • 친수성 아스코르빈산(L-ascorbic acid)과 소수성 토코페롤(${\alpha}$-tocopherol)을 모두 함유하는 ethosome 베시클을 제조하면서 이들의 특성을 살펴보았다. Ethosome에 포집되는 아스코르빈산 수용액 농도가 증가할수록 베시클 입자 크기는 작아졌다. 토코페롤은 HPC에 혼합되어 ethosome 베시클 액정막의 구성 성분이 되는데 토코페롤 혼합에 의해 베시클 입자크기와 아스코르빈산 수용액의 포집효율이 다소 증가하였다. 그러나 혼합비율이 25 wt% 이상이 되면 베시클 막이 불완전한 액정구조로 변하면서 입자크기는 크게 증가하고 포집효율은 크게 감소하였다. 아스코르빈산과 토코페롤이 함께 ethosome에 포집되어 있을 때 항산화 효능이 고온($40^{\circ}C$)에서도 5주간 안정하게 유지됨을 확인하였다. UV 조사 실험에서도 아스코르빈산은 수용액 상태로 있을 때에 비해 ethosome에 포집되어 있을 때 안정성이 향상되었다.
