Clonorchis sinensis is a Group-I bio-carcinogen, associated with cholangiocarcinoma (CCA). The hamster is the only experimental model of C. sinensis-mediated CCA, but we oblige another animal model. The present study intended to develop a C. sinensis (Cs) mediated CCA model using C3H/He mice, co-stimulated with N-nitrosodimethylamine (NDMA) and dicyclanil (DC). The mice were divided into 8 groups with different combinations of Cs, NDMA, and DC. Six months later the mice were sacrificed and subjected to gross and histopathological examination. The body weights were significantly reduced among the groups treated with 2 or more agents (eg. Cs+NDMA, Cs+DC, NDMA+DC, and Cs+NDMA+DC). In contrast, liver weight percentages to body weight were increased in above groups by 4.1% to 4.7%. A Change of the spleen weight was observed only in Cs+NDMA group. Though C. sinensis infection is evident from hyperplastic changes, only 1 worm was recovered. Two mice, 1 from Cs and the other from Cs+DC group, showed mass forming lesions; 1 ($281.2mm^3$) from the Cs group was a hepatocellular adenoma and the other ($280.6mm^3$) from the Cs+DC group was a cystic mass (peliosis). Higher prevalence of gray-white nodules was observed in Cs group (42.9%) followed by Cs+NDMA+DC group (21.4%). The mice of the Cs+NDMA+DC group showed hyper-proliferation of the bile duct with fibrotic changes. No characteristic change for CCA was recognized in any of the groups. In conclusion, C3H/He mice produce no CCA but extensive fibrosis when they are challenged by Cs, NDMA, and DC together.

Purpose: Disturbed cell cycle regulatory proteins are key events underlying the development and/or progression of human malignancies. The aim of this study is to evaluate the protein expression status involved in G1/S cell cycle in human soft tissue sarcoma. Materials and Methods: We simultaneously evaluated the expression of Cyclin D1, Cyclin E, CDK4, CDK2, p16, p27, Rb, E2F1, p53 and Ki-67 by immunohistochemistry in 43 cases of soft tissue sarcoma Results: The Cyclin D1, Cyclin E, CDK4, CDK2, E2F1, and p53 were expressed in 25 (58.1%), 18 (41.9%), 13 (30.2%), 33 (76.7%), 20 (46.5%), and 18 cases (41.9%). The p16, p27, and Rb expressions were decreased in 26 (60.5%), 22 (51.2%) and 19 cases (44.2%). All of the cases showed alterations of more than one out of the above proteins. The increased Cyclin E expression and Ki-67 labeling index (LI) were significantly associated with histologic grade. The Cyclin E and E2F-1 expressions were increased in relapsed cases and the CDK4 expression was increased in cases of metastasis. Conclusion: Alterations of G1/S cell cycle regulatory proteins may play an important role in the tumoriogensis of soft tissue sarcomas. Our results suggest that increased expressions of Cyclin E, E2F1, and CDK4 were associated with tumor relapse or metastasis and could be considered as parameters of prognosis of soft tissue sarcoma.

Objectives: To analyze whether a concept similar to Korean 'Hwabyung' exists in China. We investigated the status of clinical studies conducted in China for relevant diseases and trends of the traditional Chinese medicinal (TCM) treatment adopted in clinical studies. Methods: To explore the concept of a condition similar to Korean Hwabyung in China, we searched for the existence of concept and pattern that were parallel or similar to those of Hwabyung in TCM text-books and diagnostic guidelines. We searched and analyzed clinical studies of TCM treatment for depression syndrome similar to Hwabyung from CNKI using terms 'depression' and 'qi stagnation transforming into fire'. Using extracted data, characteristics of clinical research, herbal medicine, and acupuncture treatment used in the clinical research and their effects were systematically reviewed. Results: Symptoms of 'qi stagnation transforming into fire' were most similar to those of Hwabyung. Nine articles were selected from a total 258 articles. Most of them used DSM-IV or CCMD-3 for depression diagnosis. They applied 'diagnostic and efficacy guidelines for TCM diseases and syndromes' for pattern diagnosis of 'qi stagnation transforming into fire'. Danzhixiaoyao-san and Jiaweixiaoyao-san were found to be effective when they were used alone or in combination with antidepressants. Acupuncture treatment also showed remarkable effect on LR3, HT7, LI4, PC6, GV20 when it was used alone or in combination with antidepressants. However, careful interpretation is required because a small number of studies are included. Conclusions: 'Qi stagnation transforming into fire' seemed to have symptoms similar to Hwabyung. However, further research is needed to determine its diseases and pattern types compared to Hwabyung. According to included studies of 'depression with Qi stagnation transforming into fire', herbal medicine and acupuncture treatment tended to be effective in relieving depressive symptoms. However, more discussion is required for future application of herbal medicine and acupuncture for treating Hwabyung.
The ginsenoside-hydrolyzing β-glucosidase gene (bgy2) was cloned from Lactobacillus brevis. We expressed this gene in Escherichia coli BL21(DE3), isolated the resulting protein, and then utilized the enzyme for the biotransformation of ginsenosides. The bgy2 gene contains 2,223 bp, and encodes a protein of 741 amino acids that is a member of glycosyl hydrolase family 3. β-Glucosidase (Bgy2) cleaved the outer glucose moieties of ginsenosides at the C-20 position, and the inner glucose at the C-3 position. Under optimal conditions (pH 7.0, 30℃), we used 0.1 mg/ml Bgy2 in 20 mM sodium phosphate buffer (PBS) for enzymatic studies. In these conditions, 1.0 mg/ml ginsenoside Rb1 and ginsenoside F2 were converted into 0.59 mg/ml ginsenoside Rd and 0.72mg/ml compound K, with molar conversion productivities of 69% and 91%, respectively. In pharmaceutical and commercial industries, this recombinant Bgy2 would be suitable for producting ginsenoside Rd and compound K.

Physical, chemical and biological surveys on Nakdong River estuary were made from October, 1985 to September, 1986. 1. For all the seasons except summer, the sea surface temperature in northwest area of the line which linked from Seo Island to Jisim Island was I-2$^{\circ}$C lower than in southeast area, but in summer the sea surface temperature in northwest area was I-2$^{\circ}$C higher than in southeast area. On the contrary, bottom temperature of coastal area was 1$^{\circ}$C higher than that of oceanic area except winter. 2. Surface salinity in summer and fall has minimum value of 1. 9%0 and maximum value of 32.9%0. This large variation was caused by the runoff of the Nakdong River. Effect of less saline Nakdong River water in northwest area of the line which linked from Seo Island to Jisim Island was greater than in southeast area. A strong current rip always formed near this line. 3. The yellowish-green colored zone was observed in the Nakdong River estuary throughout the year by influence of river discharge. The characteristics of the water quality in the zone have shown that the water color was grade 7 by the Forel water-color meter, transparency was less than 4 m, and concentration of suspended solids was more than 5 mg/1. This water body was in the state of eutrophication in terms of chlorophyll-a and inorganic nitrogen concentration during summer season. 4. During the study period, total 276 taxa were identified. Most of them were diatoms and dinoflagellates which consisted of 97.5%. The component ratio of the above two groups was 84.4% and 13.1 % respectively. Diatoms were plentiful in December and dinoflagellates in July. Dominant species were Nitzschia seriata in October, Thalassiosira rotula in December, Skeletoncma costatum in April, and Nitzschia longissima in July. 5. A total of 47 zooplankton taxa was identified from the samples collected. Copepods were numerically the most important components of zooplankton communities in the study area. The domir:ant copepod species were Paracalanus parvus, Acartia clausi and Temora turbinata. Noctiluca scintillans was the next important component. The other zooplankton with minor abundance were Cladocera. Sagitta spp., Cnidaria, Mysidacea, Lucifer spp. and Amphipoda. 6. During the study period, fishes from 47 families and 87 species were sampled in th~ study area. The four most abundant fish species were Rep~mucen//'s valencicnnei, Leiognathus n~cha!is, Amblychaeturicllthys hexanema and Sardirel/a zun:zsi. The fish species of the secondary importance in abundance were Cynoglossus joyneri, Sillago sihama, Engra~lis japonicus, Encdrias nebulos'l, Acanthogobius flavimanus, Trichiurus lepturus, LiParis tanai/ai, Cynagloss//'s interruptus, Aj)ogon line:z!us, Thrissz la 1l.'1la!ensis, and Limanda yokohamac.

The effect of rare earth(RE) at three difference levels of nitrogen on the growth and nitrate content of chicory (Cichorium intybus L.) were studied in terms of leaf size, fresh and dry weight increase, and chlorophyll and nitrate content grown in plastic pots within glasshouse. Leaf size was enlarged as the level of nitrogen applied was high, and leaf within, length, and thickness of leaf were increased by RE treatment showing significant difference at high nitrogen(N+1) plot. As the level of nitrogen applied was high, as fresh and dry weight per plant was increased significantly(p<0.05), and sprinkling cabbage with RE increased fresh and dry matter yield to 2∼12, 4∼6.2% more, respectively. Dry matter content of Chinese cabbage was increased by RE sprinkling. At all levels of nitrogen fertilized chlorophyll contents were increased by RE treatment. As the level of nitrogen was high, as the content of chlorophyll was highly increased by RE. Nitrate was accumulated more at high level of nitrogen application, but nitrate was decreased by RE application, 40% decrease at very high nitrogen plot(N+2). RE stimulated the growth of chicory with high chlorophyll content, and showed the possibility producing high quality agricultural products low in nitrate content.
Fifty-four PIC barrows were used to evaluate the effects of lower dietary lysine content and energy level on carcass characteristics and meat quality in slaughter pigs. Pigs were allotted to one of three treatments by body weight with six replicate pens in each treatment. The dietary treatments for body weights of 20-50 kg, 50-80 kg and 80-90 kg were as follows, respectively: control diet (digestible energy 14.22 MJ/kg, lysine/DE 0.67 g/MJ, 0.53 g/MJ and 0.42 g/MJ); a low lysine group (digestible energy 14.22 MJ/kg, lysine/DE 0.49, 0.38 and 0.30 g/MJ); and a low lysine-low energy group or low nutrient group (digestible energy 13.11 MJ/kg, lysine/DE 0.49, 0.38 and 0.30 g/MJ). The daily weight gain, daily feed intake and feed efficiency were calculated in the overall growth period (nearly 12 weeks). Meanwhile, carcass characteristics and meat quality were evaluated at 60 and 90 kg body weight respectively. During the overall growth trial, lowering dietary lysine and nutrient level both decreased weight gain (p<0.05) and feed efficiency (p<0.01). At 60 kg body weight, decreasing dietary lysine and nutrient level noticeably decreased dressing percentage (p<0.01) and back fat depth at last rib of PIC pigs (p<0.01), but enhanced marbling scores (p<0.10), intramuscular fat content (p<0.10) and water loss rate (p<0.01) of the longissimus dorsi muscle. At 90 kg body weight, lean percentage (p<0.01) was evidently reduced by both lowering lysine content and nutrient level in the diet. However, the shoulder back fat depth (p<0.05) and marbling scores of the loin eye muscle (p<0.05) were increased; Lowering dietary nutrient level could improve back fat depth of 10th rib (p<0.01) and last rib (p<0.01), intramuscular fat content (p<0.10), redness (p<0.01) and water loss rate of the loin eye muscle (p<0.05), but decrease loin area (p<0.05). Finally, when comparing the 60 kg and 90 kg slaughter weights, it was found that the shoulder back fat depth (p<0.01, p<0.10), 6th-7th rib (p<0.01, p<0.01), 10th-rib (p<0.01, p<0.01) and last rib back fat depth (p<0.01, p<0.01) of the low lysine and low nutrient group were all obviously increased comparing with the control group. Taken together, the results showed that decreasing dietary lysine content and nutrient level increased intramuscular fat content and water loss rate of longissimus dorsi muscle; On the other hand, both lowering dietary lysine and nutrient level markedly compensated to increase back fat deposition in the later finishing period (body weight from 60 to 90 kg) in contrast to the control group.
Objective: To develop healthier comminuted meat products to meet consumer demand, the gel properties, rheological properties, microstructure and water distribution of pork meat batters formulated with various amounts of bamboo shoot dietary fiber (BSDF) were investigated. Methods: Different levels of BSDF (0% to 4%) were added to pork batters, and the pH, color, water-holding capacity, texture and rheological properties of pork batters were determined. Then, pork batters were analyzed for their microstructure and water distribution using scanning electron microscopy (SEM) and low-field nuclear magnetic resonance (LF-NMR). Results: Compared with the control, BSDF addition into meat batters showed a significant reduction in L*-value and a significant increase in b*-value (p<0.05). BSDF addition of up to 4% reduced the pH value of pork batters by approximately 0.15 units; however, the cooking loss and expressible water loss decreased significantly (p<0.05) with the increased addition of BSDF. The hardness and gel strength were noticeably enhanced (p<0.05) as the content of BSDF increased. The rheological results showed that BSDF added into pork batters produced higher storage modulus (G') and loss modulus (G") values. The SEM images suggested that the addition of BSDF could promote pork batters to form a more uniform and compact microstructure. The proportion of immobilized water increased significantly (p<0.05), while the population of free water was decreased (p<0.05), indicating that BSDF improved the water-holding capability of pork batters by decreasing the fraction of free water. Conclusion: BSDF could improve the gel properties, rheological properties and water distribution of pork meat batters and decrease the proportion of free water, suggesting that BSDF has great potential as an effective binder in comminuted meat products.
A novel alkaline protease from Streptomyces sp. M30, SapHM, was purified by ammonium sulfate precipitation, hydrophobic interaction chromatography, and DEAE-Sepharose chromatography, with a yield of 15.5% and a specific activity of 29,070 U/mg. Tryptic fragments of the purified SapHM were obtained by electrospray ionization quadrupole time-of-flight mass spectrometry. Nucleotide sequence analysis revealed that the gene sapHM contained 1,179 bp, corresponding to 392 amino acids with conserved Asp156, His187, and Ser339 residues of alkaline protease. The first 24 amino acid residues were predicted to be a signal peptide, and the molecular mass of the mature peptide was 37.1 kDa based on amino acid sequences and mass spectrometry. Pure SapHM was optimally active at 80℃ in 50 mM glycine-NaOH buffer (pH 9.0), and was broadly stable at 0-50℃ and pH 4.0-9.0. The protease relative activity was increased in the presence of Ni2+, Mn2+, and Cu2+ to 112%, 113%, and 147% of control, respectively. Pure SapHM was also activated by dimethylformamide, dimethyl sulfoxide, Tween 80, and urea. The activity of the purified enzyme was completely inhibited by phenylmethylsulfonyl fluoride, indicating that it is a serine-type protease. The Km and Vmax values were estimated to be 35.7 mg/ml, and 5 × 104 U/mg for casein. Substrate specificity analysis showed that SapH was active on casein, bovine serum albumin, and bovine serum fibrin.
FoxO1, FoxO3a and FoxO4 belong to the FoxO gene family, which play important roles in the PI3K/PKB pathway. In this study, we cloned the porcine FoxO1, FoxO3a and FoxO4 sequences and assigned them to SSC11p11-15, SSC1p13 and SSC xq13 using somatic cell hybrid panel (SCHP) and radiation hybrid panel (IMpRH). RT-PCR results showed that these three genes are expressed in multiple tissues. Sequencing of PCR products from different breeds identified a synonymous T/C polymorphism in exon 2 of FoxO3a. This FoxO3a single nucleotide polymorphism (SNP) can be detected by AvaII restriction enzyme. The allele frequencies of this SNP were investigated in Dahuabai, Meishan, Tongcheng, Yushan, Large White, and Duroc pigs. Association of the genotypes with growth and carcass traits showed that different genotypes of FoxO3a were associated with carcass length and backfat thickness between 6th and 7th ribs (BTR) and drip loss (p<0.05).
