Effects of Gamma Irradiation on Antioxidant Properties and Physical Characteristics of Sargassum siliquastrum Water Extract (꽈배기 모자반 물 추출물의 항산화능과 물리적 특성에 대한 감마선 조사의 영향)

  • In this study, the antioxidant properties and physical characteristics of irradiated Sargassum siliquastrum water extract were evaluated. Samples were irradiated with $Co^{60}{\gamma}$-ray at doses ranging from 3 to 20 kGy. They were then analyzed to investigate antioxidant properties, including total phenolic compound content and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. In addition, physical properties such as viscosity and color were evaluated. The results demonstrated that total phenolic compound content and DPPH radical scavenging activity were significantly improved (p<0.05) by irradiation. In terms of physical properties, viscosity and color were reduced in the irradiated extracts as compared to non-irradiated samples. In conclusion, gamma irradiation improved the antioxidant properties of Sargassum siliquastrum water extract, improving its original weak point as a natural antioxidant when applied in the food industry.

Comparison of Irradiated Food with Electron Beam and Gamma-ray by PSL and TL Methods (광자극발광법과 열발광법 분석을 통한 전자선과 감마선 조사식품의 비교 연구)

  • This study was conducted to determine the PSL and TL properties of foods irradiated with electron beam and gamma-ray. 5 kinds of food including cereal, pulse, fish powder, dried vegetable and tea were irradiated at 0 to 10 kGy by electron beam accelerator or $^{60}Co$ gamma-ray irradiator. The PSL analysis showed negative results for most of the non-irradiated samples. Non-irradiated shrimp powder showed intermediate result. Irradiated samples gave negative or intermediate or positive value which presented the limitation of PSL technique. In TL analysis, there were TL glow curves at around $300^{\circ}C$ with low intensity on non-irradiated samples. Maximum peak in the range of $150-250^{\circ}C$ was appeared on irradiated samples. TL ratio obtained by re-irradiation with 1 kGy was less than 0.1 on non-irradiated samples and higher than 0.1 on irradiated samples. Therefore, in PSL measurement, electron-beam irradiated samples could obtain more clear results. TL analysis showed obvious difference between non-irradiated and irradiated samples. But the identification was impossible for the sample of rice and lemon tea. Because of it's low contents of mineral.

  • The gaseous mixtures of iso-propanol of 10 torr with carbon dioxide of 0∼1990 torr were irradiated with Co-60 gamma radiation up to 50 kGy, and the products such as carbon monoxide, acetone, methane, ethane, C$_3$H$_a$, C$_4$H$_b$ and tert-butanol were analyzed by gas chromatography. G(CO) value of 4 was obtained from the gas mixtures containing more than 490 torr of carbon dioxide. The production rates of the organic products and the decomposition rate of iso-propanol increased linearly with the pressure of carbon dioxide, and G(-iso-Propanol) and G(Acetone) values increased by 4 and 2, respectively, with each 10 torr increment of carbon dioxide pressure. The mechanisms of the radiolytic decomposition of iso-propanol and the production of carbon monoxide and organic materials are discussed on the basis of the experimental results of the present study.

Characteristics of Low Molecular Weight Alginate Film Prepared with ${\gamma}$-irradiation (방사선 처리에 의해 제조한 저분자 알긴산 필름의 특성)

  • To modify the physical properties of alginate film, the gamma ray irradiation was performed, and alginate film properties such as elongation strength (ES), elongation rate (ER), water vapor permeability(WVP), and water solubility (WS) were measured. The film made from 1.75% alginate solution (w/w) with the addition of 2% CaCl$_2$ solution suggested a good application for a film preparation in that elongation strength(ES) increased 5 fold by CaCl$_2$ treatment and water vapor permeability was decreased by 17${\sim}$22%. A solid form of alginates were irradiated with Co$^{60}$ gamma rays in the dose range of 2 to 10 kGy to make a low molecular weight alginate film, and then the intrinsic viscosity and the molecular weight of alginates were measured. Increasing the dose range of 2 to 10 kGy produced lower intrinsic viscosity and lower molecular weight, causing the decrease in the ES and WS. However, ER increased four times compared to that of without gamma ray dose, which is an indication of the different properties of algiante film. WVP of the films did not show any significant change at different doses.

  • In this study, we studied the gamma-radiation effect of fiber Bragg gratings (FBGs) on the high temperature annealing condition after grating inscription using a KrF UV laser (248 nm). The FBGs were fabricated in a different annealing temperature using the same commercial Ge-doped silica core fiber (SMF-28e) and exposed to gamma-radiation up to a dose of 31 kGy at the dose rate of 115 Gy/min. The high temperature annealing procedure for grating stabilization was applied to change the radiation sensitivity of the FBGs. According to the experimental data and analysis results, the gratings that were stabilized at different temperatures at 100, 150 and $200^{\circ}C$ have clearly shown that exposure to higher temperatures increases their radiation sensitivity. The radiation-induced Bragg wavelength shift (BWS) was shown a difference of up to about a factor of two depending on the annealing temperature conditions of the gratings.

  • Optical fibers are going to be used for telecommunication, image fibers, sensors under irradiation in nuclear power plants and various irradiation facilities. Especially, Temperature detection sensors using Raman light scattering, temperature or strain sensors using fiber gratings, magnet-optical sensors using photo-magnetic effect, are already commercialized. However, When fibers are exposed to ionizing radiation, color centers are formed in fibers which reduces their light transmission, and it is limited in applying under radiation environments. In this study, $Co^{60}$ gamma-ray induced optical attenuation on Ge-doped single mode(SM) fiber has been measured. Gamma-ray is irradiated for 4hours at the dose rate of 0.5kGy/hr, 2kGy/hr, 8kGy/hr. Consequently, gamma-ray induced loss based on radiation effects in Ge-doped SM fiber occur precisely. Furthermore, dose rate effect that the higher dose rate in the same total dose, the more increase loss of optical fiber and annealing effect that the higher the loss after irradiation, the more increase the recovery rate of the loss are observed in the fiber. This results plan to make use of bases in the study of the radiation-hardened optical fiber.

  • This study was undertaken to investigate ultrastructural changes according to the radiosensitivity in the spermatogenic cells and Sertoli cell of the seminiferous tubules in Korean native pheasants. During spermatogenetic period, testes were collected from male adult Korean native pheasant and they were used as experimental and control birds. The experimental group was divided into a single-dose whole body irradiation group (400, 600, 800 and 1,000 rads) and a split-dose whole body irradiation groups(400/2, 600/2, 800/2 and 1,000/2 rads). The experimental birds were sacrificed at 24 and 72 hrs after irradiation and the control pheasants were sacrificed at the same time. Ultrastructural changes of Sertoli cells and spermatogonia were investigated by ultrathin section with electron microscope. The results obtained are summarized as follows; 1. The apoptosis was observed after 72 hrs group of the single-dose irradiation of 400 rads. 2. The cytoplasmic organelles of spermatogonia were severely damaged more than that of sertoli cell in 72 hours group of split-dose irradiation of 800 rads. 3. The cytoplasmic organelles of Sertoli cell were severely damaged except the nuclear membrane of Sertoli cells in 72 hrs group of split-dose irradiation of 1,000 rads.

groES Expression Related to Antifungal Activity of Streptomyces sp. SAR01 (Streptomyces sp. SAR01 균주에서의 항진균 관련 groES의 발현)

  • To analyse proteins and gene related to antifungal activity, SAR01 strain was isolated from a brown seaweed and identified as Streptomyces sp. by FAME(fatty acid methyl ester) analysis. Antifungal activity deficient mutant(SAR535) of Streptomyces sp. SAR01 was induced by gamma radiation$({60}^Co)$. It was found that 6 specific protein spots appeared only in SAR01 by 2-D electrophoresis analysis. Among them, a protein of 10 kDa had homology of 96% with 10 kD chaperonin cpn 10 (GroES) by Basic Local Alignment Search Tool(BLAST, NCBI) analysis. SAR535 transformants into which groES was transferred by electroporation revealed antifungal activity newly similar with SAR01 It suggested that groES be supposed to be related to the antifungal activity of Streptomyces sp. SAR01.

Potential Detection of γ-Irradiated Panicum miliaceum by Viscosity Measurement during Storage (점도 측정법을 이용한 기장(Panicum miliaceum)의 저장 중 감마선 조사 검지 가능성)

  • To elucidate the physicochemical change of Panicm miliaceum powders caused by irradiation and identify detection of irradiation, viscosity, TBA value, Hunter color and total viable count were measured after irradiation of Panicum miliaceum powder. Panicum miliaceum powders from Korea and China were irradiated at 0, 2.5, 5, 7.5, 10, 15 kGy using a $Co^{60}$ irradiator and stored for 9 months at $0^{\circ}C$ and $20^{\circ}C$. All samples resulted in decrease for viscosity and specific parameter values by increasing dosage at 50 rpm after irradiation. These values showed dose-dependent relationship (above $r^2$=0.92) between non-irradiated and irradiated samples during storage. These results suggest that the detection of irradiated Panicum miliaceum at various doses is possible by using viscometric method during storage. Total viable count, TBA value and Hunter color were determined as assistant indices for the samples viscosity measured. Total viable count, TBA values showed dose-dependent relationship (2.5 ~15 kGy). The difference in viscosity along with total viable count, TBA values between non-irradiated samples little or some changed with the lapse of post-irradiation time, but was still distinguishable each other more than 6 months at $0^{\circ}C$ and $20^{\circ}C$ for Panicum miliaceum powders from Korea and China.

Preservation of Fried Fish Meat Paste by Irradiation (감마선 조사에 의한 튀김어묵의 품질보존)

  • Radurization effects on fried fish meat paste with Co-60 gamma irradiation at 0-5 kGy and physicochemical aspects of the stored samples at low ($3{\pm}1^{\circ}C$, LT) and room ($10-20^{\circ}C$, RT) temperatures were investigated. The initial microbial loads of the samples were $2.2{\times}10^3/g$ in total bacterial count, $2.8{\times}10^2/g$ in yeasts & molds, and $1.0{\times}10^2/g$ in coliform group, respectively. An irradiation dose of more than 3 kGy was shown to be effective for the radurization of stored samples and there is no apparent difference between air and vacuum packaging groups. The chemical components related to the quality underwent similar changes in the nonirradiated and irradiated groups; however, as the storage time was prolonged the quality of nonirradiated samples rapidly deteriorated. Textural parameters of the samples were little affected by the applied doses, and sensory evaluations showed that 3 kGy irradiation was the optimum dose level to extend the shelf-life of fried fish meat paste up to 2 times at RT and 3 to 4 times at LT, respectively compared with the nonirradiated control.
