Quantitative Evaluation of the First Order Creatine-Kinase Reaction Rate Constant in in vivo Shunted Ovine Heart Treated with Oxandrolone Using Magnetization Transfer 31P Magnetic Resonance Spectroscopy (MT-31P-MRS) and 1 H/31P Double-Tuned Surface Coil: a Preliminary Study

  • Purpose: Children born with single ventricle physiology demonstrate poor growth rate and suffer from malnutrition, which lead to increased morbidity and mortality in this population. We assume that an anabolic steroid, oxandrolone, will promote growth in these infants by improving myocardial energy utilization. The purpose of this paper is to study the efficacy of oxandrolone on myocardial energy consumption in these infants. Materials and Methods: We modeled single ventricle physiology in a lamb by prenatally shunting the aorta to the pulmonary artery and then postnatally, we monitored cardiac energy utilization by quantitatively measuring the first order reaction rate constant, $k_f$ of the creatine-kinase reaction in the heart using magnetization transfer $^{31}P$ magnetic resonance spectroscopy, home built $^1H/^{31}P$ transmit/receive double tuned coil, and transmit/receive switch. We also performed cine MRI to study the structure and dynamic function of the myocardium and the left ventricular chamber. The spectroscopy data were processed using home-developed python software, while cine data were analyzed using Argus software. Results: We quantitatively measured both the first order reaction rate constant and ejection fraction in the control, shunted, and the oxandrolone-treated lambs. Both $k_f$ and ejection fraction were found to be more significantly reduced in the shunted lambs compared to the control lambs, and they are increased in oxandrolone-treated lambs. Conclusion: Some improvement was observed in both the first order reaction rate constant and ejection fraction for the lamb treated with oxandrolone in our preliminary study.

인($^{31}$P) 자기공명분광법을 사용하여 사립체 근질병환자와 정상인과의 대사물질 비교조사 (Metabolic Abnormalities in Patients with Mitochondrial Myopathy Evaluated by In Vivo $^{31}$P Magnetic Resonance Spectroscopy)

  • 목적 : 인($^{31}P$) 자기공명분광법을 사용하여 사립체 근병(mitochondria myopathy) 환자의 대퇴부 근조직의 대사물질의 변화를 정상인과 비교조사하였다. 대상 및 방법 : 사립체 근병환자 10명과 정상인 10명을 대상으로 1.5T MRI/MRS 장비를 사용하여 인($^{31}P$) 자기공명분광법을 적용하였다. 오른쪽 대퇴부위의 근조직에 $4{\;}{\times}{\;}4{\;}{\times}4{\;}cm^{3}$ 의 관심부위 (volume of interest ; VOI)를 선정하여 image selected in vivo spectroscopy (ISIS)를 저용하였다. 인대사불질의 정\ulcorner분석은 Marquart algorithm을 사용하였다. 결과 : 사립체 근병환자의 특징은 정상인과 비교하여 Pe/PCr 대사비율이 상당히 증가하고 (P=0.003), ATP/PCr 대사비율은 상당히 감소하였다(p=0.004). 특히 ATP 중 ${\beta}-ATP/PCr$ 비율의 변화가 가장 심하게 나타났다. 환자군과 정상군의 pH 차이는 통계학적으로 큰 의의는 없었다. 결론 : 인($^{31}P$) 자기 공명분광법은 사립체 근병환자의 대퇴부 근조직의 ATP/PCr 과 Pi/PCr 대사비율을 토대로 유용한 임상 평가 자료를 제공하고, 따라서 근대사물질의 질병을 이해하는데 도움을 줄 것으로 사료된다.

Human in-vivo 31P MR Spectroscopy of Benign and Malignant Breast Tumors

  • Objective: To assess the potential clinical utility of in-vivo 31P magnetic resonance spectroscopy (MRS) in patients with various malignant and benign breast lesions. Materials and Methods: Seventeen patients with untreated primary malignant breast lesions (group I), eight patients with untreated benign breast lesions (group II) and seven normal breasts (group III) were included in this study. In-vivo 31P MRS was performed using a 1.5 Tesla MR scanner. Because of the characteristics of the coil, the volume of the tumor had to exceed 12 cc (3×2×2 cm), with a superoinferior diameter at least 3 cm. Mean and standard deviations of each metabolite were calculated and metabolite ratios, such as PME/PCr, PDE/PCr, T-ATP/PCr and PCr/T-ATP were calculated and statistically analyzed. Results: Significant differences in PME were noted between groups I and III (p=0.0213), and between groups II and III (p=0.0213). The metabolite ratios which showed significant differences were PME/PCr (between groups II and III) (p=0.0201), PDE/PCr (between groups I and III, and between groups II and III) (p=0.0172), T-ATP/PCr (between groups II and III) (p=0.0287), and PCr/T-ATP (between groups II and III) (p=0.0287). There were no significant parameters between groups I and II. Conclusion: In-vivo 31P MRS is not helpful for establishing a differential diagnosis between benign and malignant breast lesions, at least with relatively large lesions greater than 3 cm in one or more dimensions.

Localized In Vivo $^{31}P$ NMR Studies on Rabbit Skeletal Muscle Tissue from Premortem to Postmortem Period

  • Localized in vivo 31P NMR spectroscopy was applied to evaluate the postmortem catabolism of high energy phosphates in rabbit skeletal muscle tissue. In the premortem processes all of the important high energy phosphate metabolites were characterized, and particularly phosphocreatine (PCr) resonance signal was the strongest. In the immediate phases of the postmortem processes the signal intensities of PCr, phosphomonoesters (PME), phosphodiesters(PDE), $\alpha$-, $\beta$- and ${\gamma}$-adenosine triphosphate (ATP) resonance began to decrease while the signal intensity of inorganic phosphorus (Pi) resonance began to increase. The present study suggests that localized in vivo 31P NMR spectroscopy may provide more precise biochemical information of the early postmortem period based on the metabolic alterations of phosphate. The unique ability of localized in vivo 31P NMR spectroscopy to offer noninvasive information about tissue biochemistry in animals as well as human may have an impact on thanatochronology and medicolegal science.

The Status of Guanine Nucleotides in Taxol-Stabilized Microtubules Probed by 31P CPMAS NMR Spectroscopy

  • Rapid exchange and hydrolysis of the tubulin-bound guanine nucleotides have been known to govern the dynamics of microtubules. However, the instability and low concentration have made it difficult for the microtubule-bound GTP to be observed directly. In this study, we circumvent these problems by lyophilization and using cross-polarization techniques. $^{31}P$ NMR signals were detected from the tubulin-bound GTP in microtubules for the first time. Analysis of the $^{31}P$ CPMAS NMR spectrum indicates that GTP hydrolysis was delayed by the presence of taxol.

Characterization of the Catalytic Heteropoly Compounds using Solid-state NMR

  • Heteropoly compounds, H3PMo12O40, CsxH3-xPMo12O40, and vanadium containing heteropoly compound were characterized by Solid-state broad line 1H MAS NMR, 31P MAS NMR, and High Speed MAS 51V NMR spectroscopy of quadrupolar nuclei. The effects of calcination, dehydration, and the number of protons on the structure of heteropoly compounds were studied. The results of this study demonstrate that these Solid-state NMR techniques are very useful tools to study heteropoly compounds.

  • [ $^{31}P$ ] powder pattern spectra were measured to investigate the aspects of the interaction between the MLV (Multilamellar vesicle) and PMAP-23, a membrane of cathelicidin family and then CSAs(chemical shift anisotropy) were calculated to indentify the extent of perturbation of phospholipid mobility by the peptides. We found that acidic phospholipid interacts strongly with PMAP-23, and the analogues which modified to increase the amphipathic property showed that larger change of CSA. The analogue which introduced positive charge showed the same effects with amphipathic property.

Hard Tissue Analysis of NMR after Fluoride Administration

  • Fluoride (F) is an important element for the mineralization of body tissues. The purpose of this study was to administer fluoride prenatally to rats to evaluate its beneficial concentration for rat bone using microstructural analysis, to analyze its effect on the bone structure, and to evaluate the effect of its transfer through rat placenta. Fourier transform infrared spectroscopy (FTIR) and nuclear magnetic resonance spectrometry (NMR) were performed. The $^{19}F$ NMR and $^{31}P$ NMR signals suggested the existence of fluoride ions in the apatite lattice because the signals were caused by the fluoride ions that were coupled to the phosphate atoms and were affected in the phosphate phases other than the element phases in the apatite. Consequently, if it was not affected too much, the desirable concentration of prenatal fluoride treatment could have a helpful effect on the bone crystal structure through placental fluoride transfer.
