  • This experiment was performed to evaluate the ultrastructural responses of the absorptive cells in the appendix of the mouse, inoculated with Ehrlich carcinoma cells in the inguinal area, following administration of BCG (Bacillus Calmette-Guerin). Healthy adult ICR mice weighing 25 gm each were divided into normal and experimental groups (experimental control group and BCG treated group). In the experimental groups, each mouse was inoculated with $1{\time}10^7$ Ehrlich carcinoma cells subcutaneously in the inguinal area. From next day after inoculations, 0.5mL of saline or BCG (0.5 mL/25gm B.W.: $0.03{\times}10^8{\sim}0.32{\times}10^8CFU$) were injected subcutaneously to the animals every other day. The day following the last injection, each mouse was sacrificed. Pieces of the tissue were taken from the appendix, prefixed with 2.5% glutaraldehyde-1.5% paraformaldehyde solution, followed by post-fixation with 1% osmium tetroxide solution. The ultrathin sections were stained with uranyl acetate and lead citrate. In the normal control, experimental control and BCG treated mice, general morphology of the absorptive cells of appendix were similar. But myelin figures and intramitochondrial dense granules were more frequently observed in the absorptive cells of BCG treated mice than normal control ones. Above results show that BCG did show slight ultrastructural alterations in the absorptive cell of the appendix. These results that BCG may slightly suppress function of the absorptive cells of the appendix.

  • This study is to identify potential factors to impact on appearance of wildbirds which live in the core forest around urban area in Gangdong-gu, Seoul. As a result of analysis of, studies on biotope showed most of urbanization biotope was biotope of residential areas with high green coverage and biotope of residential and business areas with low green coverage while most of biotope of green and openspace was core green biotope. The research area was divided into several blocks based on biotope types in the urbanization areas excluding green and openspace. As a result of research on wildbirds, total 51 species 3,419 individuals appeared in spring and total 35 species 4,213 individuals appeared in winter. 24 wild bird species were selected as subjects of this study among 31 species seen in urbanization areas, since urban birds, rapacious birds, waterside birds were excluded from the study for the proper consideration. Then this study looked at how many species and individuals of the subjects were observed at each research block in urbanization areas during spring and winter separately. Landuse structure and green structure in each block were examined to see whether these structures affect the number of wild birds observed in urbanization areas of Gangdong-gu. Furthermore, the distance between these blocks and green was assessed. While studying the potential links between the landuse structure and the number of wild birds observed in urbanization areas of Gangdong-gu, block area, green coverage, and building-to-land ratio were believed to affect the number of types and species of wild birds in the research area. In terms of correlation analysis of whether green structure affected the number of wild birds observed in urbanization areas of Gangdong-gu, crown volume of layers, the average green patch area, the average height of canopy layer were found to have an impact not only on the number of types but also species of wild birds in the research area.

  • In this study to evaluate the involvement of EGFR, HER-2/neu and ALCAM (CD166) oncogene products in canine mammary neoplastic lesions, sections of archived paraffin-embedded samples of 49 mammary tumors were analyzed immunohistochemically using antibodies against human EGFR and HER-2/neu and ALCAM. These 49 tumors were divided into 2 groups: 22 benign (19 adenoma, 3 benign mixed tumors) and 27 malignant tumors (2 simple adenocarcinomas, 5 complex adenocarcinomas, 3 solid carcinoma, 5 sclerosing carcinoma, 8 malignant mixed tumors and 4 malignant myoepithelioma). As a result of immunostaining, 31.8% (7/22) of the benign tumors and 29.6% (8/27) of the malignant tumors expressed the HER-2/neu oncogene product, EGFR expression was detected in 27.3% (6/22) of benign tumors and in 22.2% (6/27) of the malignant tumors. ALCAM expression was detected in 40.9% (9/22) of benign tumors and in 7.4% (2/27) of the malignant tumors. These results suggest that some of the biological and morphological characteristics of the tumor are associated with canine mammary gland tumors, as also reported for human breast cancer, the possibility of using anti-HER-2/neu antibodies in the treatment of canine mammary tumors.

  • In order to increase ginsenoside content, to reduce chemical change, to shorten extracting procedure, new methods of extraction and fractionation of crude ginseng saponin were established and compared for their chemical composition. Those are hot MeOH extraction/n-BuOH fractionation (BuOH method) and hot MeOH extraction/Diaion HP-20 adsorption/MeOH elution (HP-20 method), which are already known methods, and additional three new methods: hot MeOH extraction/cation AG 50W $adsorption/H_2O$ elution/n-BuOH extraction (AG 50W method), cool MeOH extraction/Diaion HP-20 adsorption/MeOH elution (cool extraction method) and direct extraction with EtOAc/n-BuOH (direct extraction method). AG 50W method provided a crude saponin showing the highest content of ginsenosides of 61.5% and the lowest contents of protein and free amino acids of 0.93% and 0.19%, respectively. The protein content was the highest as 14.18% in the crude saponin by HP-20 method, while free sugar content was the highest as 13.5% by BuOH method, indicating that these are factors that lower the rate of ginsenoside in crude saponins by those methods. On the other hand, it was revealed that AG 50W method produced large amount of prosapogenins during the pass through the cation exchange resin (AG 50W) column being strongly acidic. Crude saponin from direct extraction method showed relatively higher composition of ginsenoside $Rg_1$ and Re. The results suggest that contents and composition of ginsenosides and other chemical components in crude ginseng saponin greatly depend on the condition of the extraction and fractionation.

  • The temperature fluctuations was investigated in cold distribution chain of radish sprout, typical of commercial practice. Although the temperature of distribution chain was maintained below 5$^{\circ}C$ in precooling and packaging steps, and 10$^{\circ}C$ in transporting, temperature of loading step increased up to 18$^{\circ}C$ at market. Based on this investigation, the simulated cold distribution conditions were consisted of precooling and packaging step; 5$^{\circ}C$ for 12 hours and transporting and loading steps; 5$^{\circ}C$, 10$^{\circ}C$, 20$^{\circ}C$ and $^{\circ}C$ for 6 hours, and storage and market steps; 5$^{\circ}C$ and 10$^{\circ}C$ for 17 days. The radish sprouts were cultivated at 25$^{\circ}C$ and dark condition for S days and placed in light condition for greening. They were packaged by 25 ${\mu}m$ ceramic film after precooling for 6 hours in 5$^{\circ}C$. The fresh weight loss and visual quality of radish sprout decreased with the increase of the temperature in transporting and loading steps. The carbon dioxide content of packages increased, but the oxygen content decreased rapidly in 1day after storage, as the temperature of transporting and loading steps increased. The ethylene content in packages increased fastest in higher temperature of transporting and loading steps treatment, and showed highest in 5$^{\circ}C$-30$^{\circ}C$-10$^{\circ}C$ treatment (temperature of precooling and packaging steps for 12 hours - temperature of transporting and loading steps for 6 hours - temperature of storage step for 14 days) followed by 5$^{\circ}C$-20$^{\circ}C$-10$^{\circ}C$ treatment. The high temperature of transporting and loading steps resulted in deterioration qualities and atmosphere conditions in packages of sprout. These results suggested that the temperature fluctuation in distribution should influence the shelf-life of radish sprouts, even thought the periods of fluctuation was just 6 hours.

  • This study was carried out to provide the information on seasonal variations of water content under highway asphalt pavements which influence on the dynamic behaviour and durability of pavements, and to assess the correlation between water content and soil or meteorological factors. Total eight sites for water content measurement which included fives sites in Kyungbu, two sites in Honam, and one site in Youngdong Highway were selected considering the variations in geology, topology and meteorology factors over all the country. Water contents under asphalt pavements were measured up to 170 cm depth every two week for total 13 months of August 1992 through September 1993 using neutron moisture meter(CPN-503DR). The range of water content ($\theta$$_{w}$) at the upper soils of above 50 cm depth was 7~12% and was not quite different regardless of sites, except for Iseo site. However, soil water contents below 60 or 70 cm depth were significantly different between the measurement sites, that is, the lowest water content was 5% at Kyungsan site and the highest water content was 20% at Iseo site. For all the sites, seasonal variations in water content during the experimental period were little, their range was within only 1 to 4%. Seasonal variations of water content in original or cutting area, which were 4% more or less, were slightly larger than in bedding areas, which were below 2%. Water contents under asphalt pavements had statistically significant positive correlations with silt and clay content in soil, but there were little correlations between water content and meteorological factors such as precipitation, relative humidity, mean air temperature, and wind velocity.

  • Although Semisulcospira libertina is generally regarded as a supplement for the alleviation of alcohol hangover, little is known about its effects on cell metabolism. Therefore, this study was conducted to analyze the constituents of the extracts prepared using different extraction methods and to compare their biochemical properties. The amino acid contents were found to be much higher in acidic and enzymatic hydrolysates than hot water extracts from S. libertina. DPPH radical scavenging activities in acidic and enzymatic hydrolysates were higher than those of hot water extracts. Three types of S. libertina hydrolysate was added to HepG2 cells damaged by acetaminophen (AAP), after which the survival rate of HepG2 cell were measured. In addition, lactate dehydrogenase (LDH) activities in the culture media were evaluated. The survival rates of HepG2 cells were $77.0{\pm}4.3%$ and $81.5{\pm}1.3%$ at 3 h and 5h enzymatic hydrolysates, respectively. These cell survival rates were higher compared to those of the negative control group ($67.8{\pm}4.3%$) treated only with acetaminophen. Cellular toxicities induced by treatment with AAP were also significantly alleviated in response to treatment with the extracts of S. libertina. In addition, the activities of 2 key enzymes that metabolize ethanol, alcohol dehydrogenase and aldehyde dehydrogenase, were upregulated by 4.7- and 2.7-fold respectively in response to treatment with a 3 h enzymatic hydrolysate of S. libertina. Taken together, these results provide biochemical evidence of the method by which S. libertina exerts its biological functions, including the alleviation of alcohol hangover and the protection of liver cells against toxic insults.

  • This study aimed to develop and apply guidelines for calculating greenhouse gas emissions to activate the contribution of the Korea Institute of Geoscience and Mineral Resources (KIGAM) for institutional-level research activities. In addition, we intended to improve awareness by identifying greenhouse gas emissions from KIGAM's basic research and development (R&D) activities in fiscal 2022. Herein, the research plan and budget contents of individual projects were analyzed, whilst the boundaries and scopes of greenhouse gas emissions were determined, with 22 cases being derived as either direct, indirect, or other sources of emissions. Subsequently, research activity emissions were calculated by emission source. The greenhouse gas emissions of KIGAM's 2022 basic project R&D activities were 2,041.506 tCO2eq, of which direct emissions were 793.235 tCO2eq (38.86%), indirect emissions comprised 305.647 tCO2eq (14.97%), whilst other emissions were 942.624 tCO2eq (46.18%). In particular, greenhouse gas emissions per 100 million won in the KIGAM's basic projects for fiscal 2022 (a total of 96.661 billion won) was calculated as 2.11 tCO2eq, whilst greenhouse gas emissions per participating researcher (was 4.800 tCO2eq. Such calculations should be carried out annually rather than once and accumulated for at least 5 years. Accordingly, it will be possible to standardize specific matters that influence emissions according to differences in research field characteristics and methods, thus guiding greenhouse gas emission reduction management in the future and evaluating the contributions of Environmental, Social and Governance (ESG) management to the environmental sector.
