감마선 조사에 의한 갈색 새우(Penaeus aztecus) 주요알러젠(Pen a 1)의 알러지성 및 항원성의 변화 (The Changes of Allergenic and Antigenic Properties of Major Allergen(Pen a 1) of Brown Shrimp(Penaeus aztecus) by Gamma Irradiation)

  • 새우 알러지를 감소시키기 위한 방법으로써 감마선 조사기술의 이용 가능성을 평가하였다. 새우로부터 분리한 열안정성 단백질(HSP)과 병원에서 임상진단용으로 사용하는 새우 단백질 추출액을 1, 3, 5, 7, 10 kGy의 선량으로 감마선을 조사하였다. 단백질 추출액 및 HSP의 알러지성과 항원성의 변화를 mAb와 환자의 IgE를 사용한 ELISA법과 immunoblotting법으로 측정하였고, 단백질의 구조변화는 gel permeation chromatography와 SDS-PAGE로 관찰하였다. 환자의 IgE는 감마선 조사된 단백질 추출액과 HSP용액 모두에서 감마선 조사선량이 증가함에 따라 감소하였다. GPC-HPLC와 sandwich ELISA법으로 HSP를 정량하였을 때도 역시 감마선 조사선량이 증가함에 따라 감소하였다. SDS-PAGE pattern 변화에서 HSP는 감마선 조사에 의해 변화되어 더 큰 분자량으로 전환되었다. 이러한 결과는 감마선 조사가 새우 알러지 억제에 적용 할 수 있다는 가능성을 시사하였다.

감마선 조사된 개량메주의 보존 중 품질특성 (Quality Characteristics of Gamma Irradiated-Grain Shape Improved Meju)

  • 전통메주의 대체제품으로 시판되고 있는 낱알형 개량메주의 보존성 향상을 목적으로 메주에 0, 5, 10, 20 kGy의 선량으로 감마선을 조사하고 $25^{\circ}C$에 6개월간 보존하면서 메주의 품질변화를 조사하였다. 실험 결과, 곰팡이와 산생성세균은 10 kGy의 감마선 조사로 완전사멸이 가능하였으며 Bacillus는 10 kGy의 조사선량에서 약 5 log cycle 정도의 감소를 나타내었다. 감마선 조사에 의한 미생물군의 $D_{10}$을 곰팡이, Bacillus, 산생성 세균에서 각각 0.79, 2.28, 1.02 kGy 수준이었으며 감마선 조사 후 생존한 미생물들도 보존기간에 따라 지속적으로 감소하였다. 메주의 품질열화 요소인 암모니아태질소 함량과 갈색화도의 중가는 감마선 조사에 의하여 유의적으로 억제되었고 메주의 생화학적 품질평가 지표인 protease의 활성과 pH는 감마선 조사에 의해서 영향을 받지 않았다. 따라서 감마선 조사는 메주의 품질을 유지하는데 유용한 방법이 될 것으로 평가되었다.

감마선 조사가 미역귀 조다당의 추출수율 및 미백활성 변화에 미치는 영향 (Effects of Gamma Irradiation on the Extraction Yield and Whitening Activity of Polysaccharides from Undaria pinnatifida Sporophyll)

  • 본 연구에서는 해조류에 대한 방사선의 영향 평가에 관하여 알아보기 위하여 감마선 조사된 미역귀로부터 추출된 조다당의 추출수율 및 미백활성에 관하여 관찰하였다. 미백활성은 melanin 합성에 중요하게 작용하는 tyrosinase 억제활성과 melanocyte의 melanin 생성 억제능을 평가하였다. 실험 결과, 방사선의 조사는 선량 의존적으로 조다당의 추출수율을 증가시키고, tyrosinase 억제 활성 및 ${\alpha}$-MSH로 melanin을 과생성 시킨 B16BL6 melanoma 세포내 melanin 생성 억제 활성을 유지시켜 주는 것으로 나타났다. 이러한 결과로 미루어 보아 감마선의 조사는 미백활성을 갖는 미역귀 조다당을 얻기 위한 효과적인 방법이며, 미백 산업 분야에서도 효과적으로 이용될 수 있을 것으로 사료된다.

감마선 조사된 장류 물추출 분획의 유전독성학적 안전성 평가 (Genotoxicological Safety on Water-Soluble Fraction of Gamma Irradiated Korean Soybean Fermentation Foods)

  • 우리나라 전통 장류인 간장, 된장, 고추장, 청국장의 위생화를 위한 방사선 조사기술의 이용 가능성을 검토할 목적으로 방사선 조사를 실시한 후 유전독성학적 안전성 실험의 일환으로 Ames test를 실시하였다. 감마선 조사 및 비조사된 간장, 된장, 고추장, 청국장 물추출 분획의 Salmonella typhimurium TA98, TA100, TA1535 및 TA1537에 대한 복귀변이 집락수를 조사한 결과, 대사활성계 도입 및 부재시 모두, 모든 시험균주에서 시험적용 농도인 10,000 $\mu\textrm{g}$/plate까지의 농도에서 복귀변이 집락수의 농도 의존적인 증가 혹은 감소를 보이지 않아 20 kGy의 고선량 감마선 조사된 간장, 된장, 고추장, 청국장은 돌연변이원성이 없는 것을 확인할 수 있었다.

Optimization of Pan Bread Prepared with Ramie Powder and Preservation of Optimized Pan Bread Treated by Gamma Irradiation during Storage

  • This study was conducted to develop an optimal composite recipe for pan bread with ramie powder that has high sensory approval with all age groups and to estimate the DPPH radical scavenging activity and the pan bread shelf life after gamma irradiation. The sensory evaluation results showed significant differences in flavor (p<0.05), appearance (p<0.01), color (p<0.01), moistness (p<0.01), and overall quality (p<0.05) based on the amount of ramie powder added. As a result, the optimum formulations by numerical and graphical methods were calculated to be as follows: ramie powder 2.76 g (0.92%) and water 184.7 mL. Optimized pan bread with ramie powder and white pan bread were irradiated with gamma-rays at doses of 0, 10, 15, and 20 kGy. The total bacterial growth increased with the longer storage time and the least amount of ramie powder added. Consequently, these results suggest that the addition of ramie powder to pan bread provides added value to the bread in terms of increased shelf life.

방사선 조사 인삼이 랫드의 수태능 및 일반 생식독성에 미치는 영향에 관한 연구 (Effects of Gamma-Irradiated Korean Ginseng on Fertility and General Reproductive Toxicity in Rats)

  • Korean ginseng products have been fumigated with ethylene oxide (EO) for sterilization and prolongation of storage periods. However, there had been controversies indicating that the consumption of food treated with EO might cause harmful effects in human. Since, in Korea the use of EO gas for food treatment was banned in 1991. Since then, irradiation technique has been developed as an alternative. This study was carried out to investigate the effects of irradiated ginseng on fertility, and reproductive and developmental toxicity. Either EO gas fumigated or gamma-irradiated ginseng was administered to male rats by oral gavage for 63 days during the premating period. Female rats were administered from 14 days before mating to day 20 of gestation or to day 21 of lactation. The exposure amount of irradiation used was 5, 10 and 30 kGy, respectively. There were no treatment related changes of darns in clinical signs, and parturition. No treatment related changes in food consumption, body/organ weights, male/female reproductive and fertility performances were observed. F1 fetuses showed no external abnormality. Reflex/sensory junctions, physical/behavioral development, and reproductive performance of F1 rats were not adversary affected. The results of this study show that gamma-irradiated ginseng, up to 30 kGy, has no adverse effects on the fertility, reproduction and development in Wistar rats.

방사선 조사 인삼이 랫드의 기형유발에 미치는 영향에 관한 연구 (Embryo and Fetal Developmental Toxicity Study on Gamma-Irradiated Korean Ginseng in Rats)

Since then, irradiation technique has been developed as an alternative. This study was carried out to evaluate the safety of irradiated ginseng on embryo and fetal developmental toxicity effects in rats. Either EO gas fumigated or $\gamma$-irradiated ginseng was administered to pregnant Wistar rats by oral gavage from gestational day 7 to 17. The amount of irradiation used in this study was 5, 10 and 30 kGy, respectively. There were no treatment related changes of dams in deaths, clinical signs, food consumption and body/organ weight. No treatment related changes in implantation ratio, litter size, sex ratio and body/organ weight of fetuses were observed. Also, no F1fetuses with external, visceral, head and skeletal mal-formation were observed. The results of this study showed that $\gamma$-irradiated ginseng, up to 30 kGy, has no adverse effects on fetal development of rats.

Isolation and Proteomic Analysis of a Chlamydomonas reinhardtii Mutant with Enhanced Lipid Production by the Gamma Irradiation Method

  • In this study, an enhanced lipid-producing mutant strain of the microalga Chlamydomonas reinhardtii was developed by gamma irradiation. To induce the mutation, C. reinhardtii was gamma irradiated at a dose of 400 Gy. After irradiation, the surviving cells were stained with Nile red. The mutant (Cr-4013) accumulating 20% more lipid than the wild type was selected. Thin-layer chromatography revealed the triglyceride and free fatty acid contents to be markedly increased in Cr-4013. The major fatty acids identified were palmitic acid, oleic acid, linoleic acid, and linolenic acid. Random amplified polymeric DNA analysis showed partial genetic modifications in Cr-4013. To ascertain the changes of protein expression in the mutant strain, two-dimensional electrophoresis was conducted. These results showed that gamma radiation could be used for the development of efficient microalgal strains for lipid production.

  • The gamma-radiation sensitivity of three kinds of pathogenic bacteria to fish was investigated. $D_{10}$ values (irradiation dose required to inactivate 90% of the population) of Edwardsiella tarda, Vibrio anguillarum and Streptococcus faecalis were 0.08, 0.10 and 0.44 kGy, respectively. The inactivation factors were 4.50 - 24.30 and 6.75 - 36.45 at radiation doses of 2 and 3 kGy. Raw moist pellets were inoculated with Edwardsiella tarda, Vibrio anguillarum or Streptococcus faecalis. Inoculated feed samples were packaged in air and irradiated at 5 kGy. This dose was effective in controlling the inoculated and general bacteria in fish feed. We consider gamma-radiation to be an effective method to sterilize pathogenic bacteria in fish feed.
