당근즙 및 ${\beta}$-Carotene의 섭취가 흡연자의 혈장 항산화 영양 상태에 미치는 영향 (The Effect of Carrot Juice, ${\beta}$-carotene Supplementation on Plasma Antioxidant Status of Korean Smokers)

  • 건강한 성인 남자 48명을 대상으로 각각 당근즙 섭취군과 ${\beta}$-carotene 섭취군, placebo 약 섭취군으로 나누어 8주동안 실시하였으며, 당근즙은 매일 300 mL을 주었고 정제된 항산화 비타민은 당근즙에 함유된 ${\beta}$-carotene과 같은 양을 주어 항산화 영양 상태에 대한 개선 효과를 비교하고자 하였다. 그 결과, 혈장 비타민 C, 비타민 E 및 ${\beta}$-carotene 함량은 당근즙 및 항산화 비타민 섭취 전보다 유의적으로 높게 나타났고, 그 증가 정도가 비슷하게 나타나 식품으로부터 항산화 영양소를 충분히 섭취하는 것이 흡연자의 산화적 손상과 관련된 질병을 예방하는 좋은 방법이 될 수 있을 것이라 사료된다.

생쥐의 난소내 스테로이드호르몬 농도에 미치는 ${\gamma}$-선의 영향 (${\gamma}-ray$ Effects on Steroid Hormone Concentration of Mouse Ovarian Follicles)

  • 생쥐의 난소에 미치는 방사선의 조직학적 및 기능적 영향을 조사하기 위하여 2.88Gy 및 7.2Gy의 방사선(Co-60)을 전신조사하였다. $7{\mu}m$ 두께의 조직절편을 제작하여 조직학적 관찰을 하였고, 방사면역측정 법을 이용하여 난소마쇄현탁액내 프로게스테론, 테스토스테론 및 에스트라디올을 정량하였다. 방사선조사로 유강소난포 및 무강소난포가 공히 높은 폐쇄율을 보였고, 난포내 프로게스테론의 농도가 증가되었으며 테스토스테론 및 에스트라디올의 농도는 감소되었다. 즉, 방사선을 조사함으로써 난소내 난포세포인 협막세포에 영향이 미친 결과 $3{\beta}-HSD(3{\beta}-hydroxysteroid\;dehydrogenase) $ 및 isomerase의 활성이 억제 혹은 둔화되어 테스토스테론 및 에스트라디올의 합성이 저하되었고 그 결과 난포의 폐쇄가 가속화되었다고 사료된다.

  • 방사성 제논 탐지는 공기 중 $^{131m}Xe$, $^{133}Xe$, $^{133m}Xe$$^{135}Xe$를 저준위 백그라운드 계측 시스템으로 검출하여 지하 핵실험 여부를 규명하는 핵심기술 중 하나이다. 방사성 제논 감시는 공기 포집, 제논 추출, 측정 및 분석을 통해 수행되며, $^{135}Xe$의 최소검출가능농도는 비교적 짧은 반감기로 인해 포집, 추출 및 측정시간에 따라 큰 차이를 보이게 된다. 본 연구에서는 방사성 제논 계측 시스템의 정해진 시료 포집 및 전처리 조건에서 최적의 방사성 제논 측정시간을 도출하기 위해 이론적 접근 및 SAUNA 시스템을 이용한 실험을 통해 최소의 MDC를 보이는 측정시간을 결정하고 이론적 계산과 실험결과에 대하여 비교 평가하였다.

MITOCHONDRIAL DNA DELETION AND IMPAIRMENT OF MITOCHONDRIAL BIOGENESIS ARE MEDIATED BY REACTIVE OXYGEN SPECIES IN IONIZING RADIATION-INDUCED PREMATURE SENESCENCE

  • Mitochondrial DNA (mtDNA) deletion is a well-known marker for oxidative stress and aging, and contributes to harmful effects in cultured cells and animal tissues. mtDNA biogenesis genes (NRF-1, TFAM) are essential for the maintenance of mtDNA, as well as the transcription and replication of mitochondrial genomes. Considering that oxidative stress is known to affect mitochondrial biogenesis, we hypothesized that ionizing radiation (IR)-induced reactive oxygen species (ROS) causes mtDNA deletion by modulating the mitochondrial biogenesis, thereby leading to cellular senescence. Therefore, we examined the effects of IR on ROS levels, cellular senescence, mitochondrial biogenesis, and mtDNA deletion in IMR-90 human lung fibroblast cells. Young IMR-90 cells at population doubling (PD) 39 were irradiated at 4 or 8 Gy. Old cells at PD55, and H2O2-treated young cells at PD 39, were compared as a positive control. The IR increased the intracellular ROS level, senescence-associated ${\beta}$-galactosidase (SA-${\beta}$-gal) activity, and mtDNA common deletion (4977 bp), and it decreased the mRNA expression of NRF-1 and TFAM in IMR-90 cells. Similar results were also observed in old cells (PD 55) and $H_2O_2$-treated young cells. To confirm that a increase in ROS level is essential for mtDNA deletion and changes of mitochondrial biogenesis in irradiated cells, the effects of N-acetylcysteine (NAC) were examined. In irradiated and $H_2O_2$-treated cells, 5 mM NAC significantly attenuated the increases of ROS, mtDNA deletion, and SA-${\beta}$-gal activity, and recovered from decreased expressions of NRF-1 and TFAM mRNA. These results suggest that ROS is a key cause of IR-induced mtDNA deletion, and the suppression of the mitochondrial biogenesis gene may mediate this process.

Ascospore Infection and Colletotrichum Species Causing Glomerella Leaf Spot of Apple in Uruguay

  • Glomerella leaf spot (GLS) caused by Colletotrichum spp. is a destructive disease of apple restricted to a few regions worldwide. The distribution and evolution of GLS symptoms were observed for two years in Uruguay. The recurrent ascopore production on leaves and the widespread randomized distribution of symptoms throughout trees and orchard, suggest that ascospores play an important role in the disease dispersion. The ability of ascospores to produce typical GLS symptom was demonstrated by artificial inoculation. Colletotrichum strains causing GLS did not result in rot development, despite remaining alive in fruit lesions. Based on phylogenetic analysis of actin, ${\beta}$-tubulin and glyceraldehyde-3-phosphate dehydrogenase gene regions of 46 isolates, 25 from fruits and 21 from leaves, C. karstii was identified for the first time causing GLS in Uruguay and C. fructicola was found to be the most frequent (89%) and aggressive species. The higher aggressiveness of C. fructicola and its ability on to produce abundant fertile perithecia could help to explain the predominance of this species in the field.

양파(Allium cepa L.) 음료의 콜린성 활성 증가 및 뇌신경세포 보호로 인한 Amyloid β Peptide 유도에 대한 인지장애 개선 효과 (Onion Beverages Improve Amyloid β Peptide-Induced Cognitive Defects via Up-Regulation of Cholinergic Activity and Neuroprotection)

  • 본 연구는 양파의 불쾌치를 저감화시킨 무취음료와 양파 과피 추출물을 첨가한 생리활성 성분 강화음료의 $H_2O_2$로 유도된 산화적 스트레스에 대한 뇌신경세포 보호 효과와 $A{\beta}$로 유도된 인지기능 장애 동물모델에서의 개선 효과를 검증하고자 수행되었다. 뇌신경세포 보호 효과에서는 상대적으로 강화음료에서 무취음료 대비 우수한 산화적 스트레스 억제효과 및 생존율을 나타내었다. $A{\beta}$로 유도된 인지기능 장애 동물모델에 있어 Y-maze, passive avoidance 및 Morris water maze test에서 강화음료가 상대적으로 우수한 학습 및 기억력 개선 효과를 나타내는 것을 확인할 수 있었다. 마우스의 뇌 조직에서 강화음료 그룹은 AChE 활성을 저해하고, 신경전달물질인 ACh의 함량을 증가시킴으로써 $A{\beta}$로 유도된 cholinergic system 장애에 있어 개선 효과를 나타내었다. 또한, 마우스 뇌에서 SOD 함량의 증가, oxidized GSH/total GSH와 MDA 함량을 감소시킴으로써 $A{\beta}$와 같은 산화적 스트레스 인자에 대한 뛰어난 항산화 효과를 나타내었다. 최종적으로 무취음료와 강화음료의 주요성분들을 Q-TOF UPLC/MS system을 통하여 분석한 결과, 강화음료의 경우 무취음료보다 생리활성을 가진 2개의 steroidal saponin과 6개의 phenolic 화합물 등이 추가 검출되었다. 이러한 결과들을 종합해볼 때 강화음료는 상대적으로 protocatechuic acid와 quercetin 같은 강력한 항산화 효과를 나타내는 phenolic 화합물과 steroidal saponin 계열에 의한 우수한 인지기능 개선 효과를 기반으로 한 고부가가치 식품으로 활용될 수 있는 산업적 가능성이 있다고 판단된다.

국민 건강보험 급여 한약 처방 56종의 치매 주요 생리지표 및 신경세포 변화에 대한 효능 비교 연구 (Screening of 56 Herbal formulas covered by the National Health Insurance Service on Dementia-related Factors)

  • Objectives: The purpose of this study was to investigate the effects of 56 herbal formulae covered by the National Health Insurance Corporation (NHIC) on dementia-related biomarkers and neuronal cell changes. Methods: The 56 herbal formulae were extracted with 70% ethanol at $100^{\circ}C$ for 2 h. The antioxidant properties was measured by radical scavenging assay using ABTS+ radical. The acetylcholinesterase (AChE) activity was tested by Ellman's assay and $amyloid-{\beta}$ ($A{\beta}$) aggregation was determined using fluorescence method. To estimate the inhibitory effects of herbal formulae on neuronal cell death and inflammation using HT22 hippocampal cells and BV-2 microglia, respectively. Results: Among the 56 herbal formulae, Dangguiyukhwangtang, Banhasasimtang, Samhwangsasimtang, Cheongwiesan, Hwangryunhaedoktang, Banhabaekchulchunmatang, Jaeumganghwatang, Cheongseoikgitang, and Hoechunyanggyuksan has a significant inhibitory effects on acetylcholinesterase (AChE) activity. Doinseunggitang and Samhwangsasimtang exerted the effect on the inhibition of $amyloid-{\beta}$ ($A{\beta}$) aggregation. Additionally, 10 herbal formulae affected AChE and $A{\beta}$ aggregation revealed antioxidant activity as well as neuroprotective and anti-neuroinflammation effects in neuronal cell lines. Conclusions: 10 herbal formulae that have been shown to be effective against the major dementia markers have been shown to have antioxidant activity, neuronal cell protection and inhibition of brain inflammation. Further investigation of these herbal formulae will need to be validated in dementia animal models.

Carotenoid Intake and Esophageal Cancer Risk: a Meta-analysis

  • This meta-analysis was conducted to evaluate the association between intake of carotenoids and risk of esophageal cancer. A systematic search using PubMed, Cochrane Library, Web of Science, Scopus, CNKI, and CBM (updated to 6 May 2012) identified ten articles meeting the inclusion criteria with 1,958 cases of esophageal cancer and 4,529 controls. Higher intake of beta-carotene, alpha-carotene, lycopene, beta-cryptoxanthin, lutein, and zeaxanthin reduced esophageal cancer risk with pooled ORs of 0.58 (95% CI 0.44, 0.77), 0.81 (95% CI 0.70, 0.94), 0.75 (95% CI 0.64, 0.86), 0.80 (95% CI 0.66, 0.97), and 0.71 (95% CI 0.59, 0.87), respectively. In subgroup analyses, beta-carotene showed protective effects against esophageal adenocarcinoma in studies located in Europe and North America. Alpha-carotene, lycopene, and beta-cryptoxanthin showed protection against esophageal squamous cell cancer. This meta-analysis suggested that higher intake of carotenoids (beta-carotene, alpha-carotene, lycopene, beta-cryptoxanthin, lutein, and zeaxanthin) is associated with lower risk of esophageal cancer. Further research with large-sample studies need to be conducted to better clarify the potentially protective mechanisms of carotenoid associations risk of different types of esophageal cancer.

Consumer perceptions on sustainable practices implemented in foodservice organizations in Korea

  • BACKGROUND/OBJECTIVES: Sustainable practices in foodservice organizations including commercial and noncommercial ones are critical to ensure the protection of the environment for the future. With the rapid growth of the foodservice industry, wiser usage of input sources such as food, utilities, and single use packaging should be reconsidered for future generations. Therefore, this study aims to investigate the customer's perceptions on sustainable practices and to identify the relationship among sustainable practices, social contribution and purchase intention. SUBJECTS/METHODS: The study was conducted using content analyses by reviewing articles on sustainable food service practices published domestically and abroad. Thereafter, data were collected with a face-to-face survey using a questionnaire and analyzed with factor analyses and multiple regressions. RESULTS: Sustainable practices classified with factor analysis consisted of 6 dimensions of green food material procurement, sustainable food preparation, green packaging, preservation of energy, waste management, and public relations on green activity, with a total of 25 green activities in foodservice operations. Consumers were not very familiar with the green activities implemented in the foodservice unit, with the lowest awareness of "green food material procurement (2.46 out of 5 points)", and the highest awareness of "green packaging (3.74)" and "waste management (3.28). The factors influencing the perception of social contribution by foodservice organizations among 6 sustainable practice dimensions were found to be public relations on green activity (${\beta}=0.154$), waste management (${\beta}=0.204$) and sustainable food preparation (${\beta}=0.183$). Green packaging (${\beta}=0.107$) and the social contribution of the foodservice organization (${\beta}=0.761$) had strong relationships with the image of the organization. The purchase intentions of customers was affected only by the foodservice image (${\beta}=0.775$). CONCLUSIONS: The results of this study suggest that sustainable practices by foodservice organization present a good image to customers and increase the awareness of valuable contributions that benefit the customer as well as the community.

결혼이주여성의 구강건강신념 및 자기효능감과 구강건강인식도와의 관련성 (The Relationship Between of Marriage Migrant Women's Oral Health Belief and Self-Efficacy on Oral Health Awareness)

  • 본 연구는 영남지역에 거주하고 있는 일부 결혼이주여성을 대상으로 구강건강신념 및 자기효능감과 구강건강인식도와의 관련성을 파악하여 구강건강 인식 개선과 태도 변화에 필요한 프로그램을 구축하는데 기초자료로 사용하고자 2013년 10월 1일부터 12월 15일까지 대구, 경북, 경남지역 8곳의 다문화 관련센터를 이용하는 결혼이주여성을 대상으로 면접조사방법을 통한 자기기입식 설문지를 이용하여 최종 256부를 분석 자료로 활용하였다. 통계분석은 SPSS 18.0과 AMOS 7.0을 사용하였고, 통계적 유의수준은 p<.05로 하였다. 구강건강신념과 자기효능감이 구강건강인식도에 영향을 미치는지 분석한 결과 치석제거(스케일링)경험은 구강건강신념요인 중 심각성(${\beta}$=.568)이 영향을 미치는 것으로 나타났고(p<.05), 구강건강상태는 구강건강신념요인 중 감수성(${\beta}$=-.391)이 영향을 미치는 것으로 나타났으며(p<.01), 구강건강관심도는 구강건강신념요인 중 감수성(${\beta}$=-.183), 심각성(${\beta}$=.172), 유익성(${\beta}$=.224)과 자기효능감(${\beta}$=.237)이 영향을 미치는 것으로 나타났다(p<.01). 결혼이주여성의 효율적인 구강건강관리를 위하여 구강건강신념과 자기효능감이 구강건강인식에 미치는 요인의 분석으로 향후 예방적 구강건강관리 및 구강보건교육 프로그램의 적극적인 방안의 모색과 다문화가정의 구강건강 향상을 위한 기본 정책의 마련이 필요할 것으로 사료된다.
