Current status of wheat allergy research and prospect

  • Wheat is one of important food crop in the world. However, wheat has some negative things that are allergen. Wheat includes four different allergen kwon as alpha-, beta- and gamma gliadin and amylase inhibitor. Recently, people are interesting to gluten free food or low gliadin food. To study about wheat allergies and research trend is important for future research like as wheat breeding and allergy reduction technology. In this study, we analyzed that the reported on the research field of wheat allergy have been reported until now, and was to provide direction for future research. The analysis of 235 episodes of major papers published from 2007 to 2016. The last 5 years from 2012, wheat allergy appears increasing number of related papers. The ratio of national papers 13% and 60% were published by the United States and other European countries, respectively. The fields of wheat allergen-related preclinical technology and wheat allergen related to genome research for discovery technology were represented high rate by 58% and 26%, respectively. In the case of Korea, significantly account genetic and breeding areas. Recently, however, the research of glutenin protein which is closely linked to wheat allergy is in progress. So, we expected to increase wheat allergy of the research is in the future in Korea.

방사선 조사 고추장의 발효 특성 (Fermentation Properties of Irradiated Kochujang)

  • Kochujang, fermented hot pepper soybean paste, was prepared by traditional method, and irradiated with gamma ray of $^{60}Co{\;}up{\;}to{\;}15{\pm}1.5{\;}kGy$ for confirming main factor of kochujang fermentation. Nonirradiated (control) and irradiated samples kochujang were fermented at $25^{\circ}C$ and analyzed biochemical properties including enzyme activities and viable cell count during fermentation for 60 days. The total viable count in irradiated kochujang decreased to $10^4{\;}CFU/g$ which was $10^8{\;}CFU/g$ in the control. Because of a little changing enzyme activities of ${\alpha}{\cdot}{\beta}{\;}amylase$ and acid neutral protease by irradiation at the above level, amino type nitrogen which is the main quality reference of kochujang was comparable to the control. By irradiation, gas production was completely stopped which is one of biggest problems during distribution of kochujang. Total volume of gas produced at $25^{\circ}C$ from the control kochujang was 453 mL/100 g which was composed of over 90% of $CO_2$. The odor of irradiated kochujang was inferior to the control which seemed to be related to reduced microbial populations.

Bacillus subtilis HJ18-3과 KACC 15935를 이용하여 제조한 청국장의 품질특성과 isoflavone 함량의 변화 (Changes in isoflavone content and quality characteristics of Cheonggukjang prepared with Bacillus subtilis HJ18-3 and KACC 15935)

  • 청국장 제조 시 사용되는 균주인 Bacillus sp. HJ18-3과 B. subtilis KACC 15935균주를 청국장 제조 시 starter로 접종하여 발효시켜 품질특성과 isoflavone 비배당체 함량을 측정하였다. a-amylase와 cellulase활성은 control과 B. subtilis HJ18-3 균주를 접종한 청국장이 B. subtilis KACC 15935 균주를 접종한 청국장보다 각각 2.56, 2.59배 그리고 1.13, 1.23배 높은 결과를 보였다. 그러나 protease활성은 B. subtilis KACC 15935과 B. subtilis HJ18-3의 활성이 높은 효소활성을 보였다. 아미노태질소 함량은 Control, B. subtilis KACC 15935, B. subtilis HJ18-3의 함량이 각각 $88.20{\pm}13.86$, $129.62{\pm}1.15$, $118.40{\pm}48.65mg%$로 나타났으며 이는 protease 효소활성의 결과와 유사한 경향을 보였다. 환원당 함량은 control, KACC 15935, HJ18-3의 함량이 각각 $1.95{\pm}0.72$, $2.62{\pm}1.17$, $2.59{\pm}0.76%$로 전반적으로 control에 비해 처리구가 더 높은 경향을 나타냈다. 호기적 총균수와 미비한 차이로 B. subtilis HJ18-3의 균수가 적은 경향을 나타낸 것은B. cereus와Candida albicans에 대해 항균력이 우수하다고 보고된 영향으로 해석된다. control과B. subtilis KACC15935, B. subtilis HJ18-3의 isoflavone 비배당체 형태인 daidzein과 genistein, glycitein을 합한 함량은 각각 $158.20{\pm}6.55$, $131.60{\pm}4.00$, $200.79{\pm}7.54ug/g$으로 나타났다. 이는 초기 raw soybean의 aglycone함량이 3.96 ug/g인 것에 비하여 39~50배 전 환율을 보인다. 또한 ${\beta}$-glucosidase활성이 있는 B. subtilis HJ18-3균을 접종하여 제조한 청국장이 control과 Kacc15935균을 접종하여 제조한 청국장보다 1.27, 1.53배 더 높은 aglycone 전 환율을 보였다. 따라서 본 연구에서 사용한 메밀 속성 장으로부터 분리된 B. subtilis HJ18-3 균주는 amylase, protease, cellulase와 같은 세포의 효소분비능이 우수하며, 아미노태질소와 환원당 함량이 높고, isoflavone 비배당체 함량을 증가시키는 기능을 함유한 균주로 확인되었으며, 앞으로도 우리나라 전통 청국장의 세계화를 위해 유용 starter로 찾는 노력이 더 필요할 것이다.

CCK로 유발시킨 급성 췌장염에 대한 가미대황목단피탕의 효과 (Effects of Gami-Daehwangmokdanpi-Tang against CCK-induced acute pancreatitis)

  • Objective : Daehwangmokdanpi-Tang (DWT) has been frequently used as a remedy for antiinflamation. To evaluate effect of acute pancreatitis by DWT, we examined the effects of DWT on the cholecystokinin-octapeptide (CCK)-induced acute pancreatitis (AP) in rats. Methods : Male Wistar rats weighing 200 to 250 g were divided into three groups. Normal untreated group, in treatment with DWT group; DWT was administered orally, followed by $75\;{\mu}g/kg$ CCK subcutaneously three times, after 1, 3 and 5 h. This whole procedure was repeated for 5 days. In treatment with saline group, the protocol was the same as in treatment group with DWT. Results : The author determined the pancreatic weight/body weight ratio, the levels of pancreatic HSP (heat shock protein)60 and HSP72 and the secretion of pro-inflammatory cytokines. Repeated CCK treatment resulted in the typical laboratory and morphological changes of experimentally induced pancreatitis. DWT was significantly decreased the pancreatic weight/body weight ratio in CCK-induced AP. Futhermore, The author demonstrated that DWT increased HSP60 and HSP72 compared with CCK-induced AP. Additionally, the secretion of $IL-1{\beta}\;and\;TNF-{\alpha}$ and the levels of amylase and lipase were lower than that saline. Conclusions : These results suggested that DWT may has a protective effect against CCK-induced AP.

패장(敗醬)의 급성 췌장염 억제 효과 (Inhibitory Effect of Aacute Pancreatitis by Patriniae Herba)

  • Objective : Patriniae Herba (PH) has long been used as a remedy for treating infectious diseases in Korea. 타피오카와 겉보리 혼합원료의 당화 및 알코올 발효의 특성 연구 (Saccharification and alcohol fermentation characteristics of a mixture of tapioca and hulled barley)

  • 알코올 발효에 이용되는 타피오카와 겉보리 혼합원료의 효소당화액을 제조하여 알코올 발효 특성을 조사하였다. 타피오카와 겉보리 혼합원료를 내열성 액화효소인 Spezyme Fred를 0.04% 사용하여 액화하고 글루코아밀레이스(G), 단백질가수분해효소(P), 베타글루칸 가수분해효소(B)를 혼합한 당화효소제(GPB)를 사용하여 당화하였다. 타피오카와 겉보리 혼합원료(7:3, w/w)를 $50^{\circ}C$에서 150분간 당화하였을 때 당화액의 포도당 함량은 11.9%였고 점도는 26 cp로 나타났다. 베타글루칸 가수분해효소(B)의 첨가는 당화액의 포도당 수율의 증가와 점성의 저하에 도움을 주었다. 타피오카와 겉보리의 혼합비율을 7:3(w/w)으로 하였을 때 다른 비율의 당화액에 비하여 점도가 낮으며 포도당 함량이 높게 나타났다. 타피오카와 겉보리 혼합원료에 300%의 가수량으로 확립된 조건에서 당화하고 $30^{\circ}C$에서 72시간 알코올 발효를 수행한 결과 타피오카와 겉보리 혼합원료를 이용한 알코올 발효에 적합한 발효액의 알코올 및 포도당 함량은 각각 9.0과 0.02%였고, pH와 산도는 각각 pH 4.3과 0.3%이었다.

친환경 복합효소제 첨가가 이유자돈의 성장, 장내 형태학, 영양소 소화율에 미치는 영향 (Effects of Eco-friendly Multi-enzyme on Growth Performance, Intestinal Morphology, and Nutrient Digestibility of weaned Pigs)

  • This experiment was conducted to investigate the effects of multi-enzyme on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.1% multi-enzyme (Multi; mixture of ${\beta}-mannanase$, xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Measurements were growth performance, morphology of ileum, apparent ileal digestibility and apparent total tract digestibility of dry matter, crude protein, and energy of weaned pigs. There were no significant differences on growth performance during overall experimental period. No differences were found for the morphology of ileum and nutrient digestibility between CON and Multi groups. Therefore, the results in the current study indicated that multi-enzyme supplementation in diets had no effects on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs.

오미자(Schizandra chinensis) 추출물이 김치의 과숙억제에 미치는 영향 (Effect of Ethanolic Extract of Schizandra chinensis for the Delayed Ripening Kimchi Preparation)

  • 오미자 에탄올 추출물의 항균활성을 조사하고, 오미자 추출물에 함유된 유기산을 분석하여 오미자 추출물 및 유기산을 첨가하여 제조한 김치의 이화학적 변화와 미생물학적 특성을 살펴본 결과는 다음과 같았다. 1.시중에 유통되는 김치에서 분리한 CS6은 김치의 숙성과 부패에 관여하는 미생물인 Lactobacillus plantarum으로 동정되었으며, 이 균주에 대해 오미자 ethanol 추출물은 항균활성을 보였다. 최소 저해농도는 62.5mg/$m\ell$이었고, 액체배지의 경우 0.5$\mu\textrm{g}$/$m\ell$ 첨가시 미생물의 증식을 완전히 저해하였다. 2. 오미자 ethanol 추출물을 첨가한 김치에서 미생물의 생육을 저해하여 산도는 증가하지 않았다. 0.5% 이하 첨가시에 색상에 영향을 미치지 않으며, 0.4% 첨가시에 맛에 영향을 주지 않았다. 3. 오미자 ethanol 추출물에 succinic acid, malic acid, tartaric acid, oxalic acid등이 포함되어 있었고, 이 중 가장 강한 항균활성을 보인 유기산은 succinic acid였으며, 이의 항균활성은 오미자 ethanol 추출물과 비슷한 저해효과를 보여주었다. 4. Succinic acid를 첨가한 김치의 경우, 미생물의 생육저해로 김치의 신선도 유지에 효과적으로 밝혀졌고, 배양말기까지 일정한 경도를 유지하였고, 역시 0.4% 이하 첨가시 맛에 큰 영향을 주지 않았다. 5. 오미자 ethanol 추출물은 $\beta$-amylase에 대해서는 낮은 저해율을 보였지만 $\alpha$-amylase, glucoamlase에 대해서 강한 저해효과를 나타내어 조직의 연화방지에 효과가 있을 것으로 생각된다. 6. 오미자 ethanol 추출물의 Avicelase 저해효과는 CMCase에 대한 저해효과보다 더 강했으며, 과숙김치에서의 경도 유지효과가 기대된다. 7. 연화와 직접적으로 관련된 효소로 생각되는 polygalacturonase에 대한 저해효과는 오미자 ethanol 추출물 25$\mu\textrm{g}$/$m\ell$ 첨가시에 66.7%의 저해율을 보였고, 30$\mu\textrm{g}$/$m\ell$ 첨가시에는 92.6%의 저해율을 보였으나, pectinesterase의 경우 15~30$\mu\textrm{g}$/$m\ell$ 첨가시에 15~20%의 낮은 저해율을 보였다.

Effects of Exogenous Enzymes on Ruminal Fermentation and Degradability of Alfalfa Hay and Rice Straw

  • This study was conducted to evaluate the use of exogenous enzymes as a potential means of improving the ruminal digestion (i.e., degradability) of alfalfa hay and rice straw. Twenty six enzyme-additives were examined in terms of protein concentration and enzymic activities on model substrates. The exogenous enzymes contained ranges of endoglucanase, xylanase, ${\beta}$-glucanase, ${\alpha}$-amylase, and protease activities. Six of the enzyme additives were chosen for further investigation. The enzyme additives and a control without enzyme were applied to mature quality alfalfa hay substrate and subsequently incubated in rumen batch cultures. Five of the enzyme additives (CE2, CE13, CE14, CE19, and CE24) increased total gas production (GP) at 48 h of incubation compared to the control (p<0.05). The two additives (CE14 and CE24) having the greatest positive effects on alfalfa hay dry matter, neutral detergent fibre (NDF) and acid detergent fibre (ADF) degradability were further characterized for their ability to enhance degradation of low quality forages. The treatments CE14, CE24, a 50:50 combination of CE14 and CE24 (CE14+24), and control (no enzyme) were applied to mature alfalfa hay and rice straw. For alfalfa hay, application of the two enzyme additives, alone and in combination, increased GP compared to the control at 48 h fermentation (p<0.05), whereas only CE14 and CE14+24 treatments improved GP from rice straw (p<0.05). Rumen fluid volatile fatty acid concentrations throughout the incubation of rice straw were analyzed. Acetate concentration was slightly lower (p<0.05) for CE14${\times}$CE24 compared to the control, although individually, CE14 and CE24 acetate concentrations were not different from the control. Increases (p<0.05) in alfalfa hay NDF degradability measured at 12 and 48 h of incubation occurred only for CE14 (at 12 h) and for CE14+24 (at 12 and 48 h). Similarly, ADF degradability increased (p<0.05) with CE14 and CE14+24. As for rice straw, increased DM degradability was observed at 12 and 48 h of incubation for all enzyme treatments with an exception for CE14 at 12 h. The degradability of NDF was improved by all the enzyme treatments at either incubation time, while ADF degradability was only enhanced at 48 h. Overall, the enzymes led to enhanced digestion of mature alfalfa and there was evidence of improved digestibility of rice straw, an even lower quality forage.

Effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs

  • Soybean, one of most widely used swine feed component in the world, contains non-starch polysaccharides (NSP). The digestive system of weaned pigs is not yet fully developed, and thus weaned pigs cannot easily digest diets based on corn and soybean meal. Dietary exogenous enzymes supplementation has been intensively investigated to assist digestion of anti-nutritional factors, such as NSP. This experiment was conducted to investigate the effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; mixture of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Growth performance, morphology of ileum, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of dry matter, crude protein, and energy of weaned pigs were measured. No significant differences (p > 0.05) were observed for growth performance for the duration of the experimental period, and morphology of ileum, and nutrient digestibility between CON and Cocktail treatment groups. Therefore, the results from the current study indicated that enzyme cocktail supplementation in diets had no influence on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs.
