1,25-dihydroxyvitamin D3 affects thapsigargin-induced endoplasmic reticulum stress in 3T3-L1 adipocytes

  • BACKGROUND/OBJECTIVES: Endoplasmic reticulum (ER) stress in adipose tissue causes an inflammatory response and leads to metabolic diseases. However, the association between vitamin D and adipose ER stress remains poorly understood. In this study, we investigated whether 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) alleviates ER stress in adipocytes. MATERIALS/METHODS: 3T3-L1 cells were treated with different concentrations (i.e., 10-100 nM) of 1,25(OH)2D3 after or during differentiation (i.e., on day 0-7, 3-7, or 7). They were then incubated with thapsigargin (TG, 500 nM) for an additional 24 h to induce ER stress. Next, we measured the mRNA and protein levels of genes involved in unfold protein response (UPR) and adipogenesis using real-time polymerase chain reaction and western blotting and quantified the secreted protein levels of pro-inflammatory cytokines. Finally, the mRNA levels of UPR pathway genes were measured in adipocytes transfected with siRNA-targeting Vdr. RESULTS: Treatment with 1,25(OH)2D3 during various stages of adipocyte differentiation significantly inhibited ER stress induced by TG. In fully differentiated 3T3-L1 adipocytes, 1,25(OH)2D3 treatment suppressed mRNA levels of Ddit3, sXbp1, and Atf4 and decreased the secretion of monocyte chemoattractant protein-1, interleukin-6, and tumor necrosis factor-α. However, downregulation of the mRNA levels of Ddit3, sXbp1, and Atf4 following 1,25(OH)2D3 administration was not observed in Vdr-knockdown adipocytes. In addition, exposure of 3T3-L1 preadipocytes to 1,25(OH)2D3 inhibited transcription of Ddit3, sXbp1, Atf4, Bip, and Atf6 and reduced the p-alpha subunit of translation initiation factor 2 (eIF2α)/eIF2α and p-protein kinase RNA-like ER kinase (PERK)/PERK protein ratios. Furthermore, 1,25(OH)2D3 treatment before adipocyte differentiation reduced adipogenesis and the mRNA levels of adipogenic genes. CONCLUSIONS: Our data suggest that 1,25(OH)2D3 prevents TG-induced ER stress and inflammatory responses in mature adipocytes by downregulating UPR signaling via binding with Vdr. In addition, the inhibition of adipogenesis by vitamin D may contribute to the reduction of ER stress in adipocytes.

Identification of the sprU Gene Encoding an Additional sprT Homologous Trypsin-Type Protease in Streptomyces griseus

  • Cloning of a 6.6-kb BamHI digested chromosomal DNA from S. griseus IFO13350 revealed the presence of an additional gene encoding a novel trypsin-like enzyme, named SprU. The SprU protein shows a high homology ($79\%$ identity, $88\%$ similarity) with the SGT protease, which has been reported as a bacterial trypsin in the same strain. The amino acid sequence deduced from the nucleotide sequence of the sprU gene suggests that SprU is produced as a precursor consisting of an amino-terminal presequence (29 amino acid residues), prosequence (4 residues), and mature trypsin consisting of 222 amino acids with a molecular weight of 22.94 kDa and a calculated pI of 4.13. The serine, histidine, and aspartic acid residues composing the catalytic triad of typical serine proteases are also well conserved. When the trypsin activity of the SprU was spectrophotometrically measured by the enzymatic hydrolysis of the artificial chromogenic substrate, N-${alpha}$-benzoyl-DL-arginine-p-nitroanilide, the S. lividans transformant with pWHM3-U gave 3 times higher activity than that of control. When the same recombinant plasmid was introduced into S. griseus, however, the gene dosage effect was not so significant, as in the cases of other genes encoding serine proteases, such as sprA, sprB, and sprD. Although two trypsins, SprU and SGT, have a high degree of homology, the pI values, the gene dosage effect in S. griseus, and the gene arrangement adjacent to the two genes are very different, suggesting that the biochemical and biological function of the SprU might be quite different from that of the SGT.

살구 추출물의 항산화성, 항돌연변이성 및 세포독성 효과 (Antioxidative, Antimutagenic and Cytotoxic Effects of Prunus armeniaca Extracts)

  • 살구 에탄올 추출물의 항산화 활성은 RC50값이 살구씨와 살구과육 에탄을 추출물의 경우 각각 $48.3{\mu}g$$43.9{\mu}g$으로서 강한 항산화 활성을 나타내었다. 살구 에탄을 추출물의 항돌연변이 효과의 검토는 Salmonella typhimurium의 변이주인 TA98과 TA100을 이용한 Ames test로 확인하였다. 그 결과 살구씨 및 과육 에탄올 추출물 자체의 돌연변이원성은 없었고 직접변이원인 MNNG에 대해 시료농도 $200{\mu}g/plate$에서 살구씨와 살구과육 에탄올 추출물 각각에서 TA100이 69.4% 및 65.9%의 억제효과를 나타내었다. 같은 농도에서 4NQO에 대해서는 살구과육 에탄을 추출물의 경우TA98과TA100이 각각45.9% 및 44.3%의 억제효과를 나타내었다. 암세포 성장억제효과를 검토한 결과 살구씨 에탄올 추출물 4mg/mL첨가 시 A549, AGS, MCF-7, HeLa 및 Hep3B에서 각각 63.7, 56, 86.3, 78 및 53.7%의 억제 효과를 보였다. 살구과육 에탄을 추출물 4mg/mL 첨가시 위암세포 AGS에서 58.0% 억제효과를 보인 반면 모든 암세포에서 72.8%이상의 높은 억제효과를 나타내었다. 이러한 암세포에 대한 높은 억제 효과에 비해 인간정상신장세포 293에 대해서는 37.2% 이하의 생육 억제율을 나타냄으로서 정상세포에 대해서는 낮은 독성효과를 가짐을 알 수 있었다.

  • 수도저위(水稻低位) 생산력(生産力)의 원인구명(原因究明)에 관(關)한 영양생리학적연구(營養生理적硏究)로 여러 가지 생육조건하(生育條件下)에서 품종별(品種別) 양분흡수(養分吸收) 및 동화기능(同化機能)과 양분(養分)의 체내(體內)에서의 이행양상(移行樣相)을 추구(追究)하는 한편 고수량지(高收量地) 벼와 저수량지(低收量地) 벼에 대(對)한 생육시기별(生育時期別)로 양분(養分)의 흡수(吸收) 체내무기양분(體內無機養分)과 수량(收量) 및 수량구성요소(收量構成要素)와의 관계(關係) 등(等)을 추구(追究)하여 저수량지(低收量地) 벼의 영양생리적(營養生理的) 저수요인(低收要因)을 밝히고저 본연구(本硏究)를 수행(遂行)하였던 바 그 결과(結果)를 요약(要約)하면 다음과같다. 1. 양분(養分)의 흡수력(吸收力)은 품종(品種)에 따라 차이(差異)가 있으며 인산(燐酸) 및 가리(加里)의 흡수력(吸收力)이 강(强)한 품종(品種)은 구사부에, 진흥(振興)이며 낮은 품종(品種)은 팔달(八達)이었다. 2. 근(根)의 ${\alpha}-naphthylamine$의 산화력(酸化力)과 인산(燐酸)의 흡수력(吸收力)과는 유의적(有意的)인 상관관계(相關關係)가 있으며 산화력(酸化力)이 높으면 인산(燐酸)의 흡수(吸收)가 증대(增大)되었바. 3, 탄소동화력( 炭素同化力)은 품종(品種)에 따라 차이(差異)가 있으며 노인도(老人稻), 구대내조욱(九大耐潮組) 3호(號), 수원(水原)82호(號), 진흥(振興) 등은 강(强)하고 대구조(大脚租), 관옥(關玉), 육우(陸羽) 132호(號)가 약(弱)하였다. 4, 질소(窒素)의 증시(增施)는 소비(少肥)에 비(比)하여 동화량(同化量)은 증가(增加) 되나 등화산물(同化産物)의 이삭으로의 이전(移轉)은 적었다5. 정상(正常) 생육(生育)한 벼에 비(比)하여 인산(燐酸), 가리(加里) 및 마그네슘이 결핍(缺達)된 벼는 전탄소동화량이(全炭素同化量)이 정상(正常) 벼 100에 비(比)하여 40이하(以下)로 현저(顯著)히 낮으며 그 순서(順序)는 Mg>P>K이고, 동화물(同化物)의 이전(移轉)에 장해(障害)가 큰 것은 K>Mg>P의 순(順)이었다. 6. 고수량지(高收量地) 벼는 저수량지(低收量地) 벼에 비(比)하여 총건물(總乾物) 생산량(生産量)이 현저(顯著)하게 많고 특(特)히 유수형성기(幼穗形成期) 이후(以後)에 증가량(增加量)이 더욱 현저(顯著)하였다. 7. 고수량지(高收量地) 벼는 출수기(出穗期)에 옆면적(葉面積)이 최고(最高)에 달(達)하였다가 그 이후(以後) 완만(緩慢)하게 줄어지나 저수량지(低收量地) 벼는 출수이전(出穗以前)에 엽면적(葉面積)이 최고(最高)에 달(達)하였다가 출수이후(出穗以後)에 급격(急激)히 감소(減少)되며 그 원인(原因)은 하엽(下葉)의 조기고사(早期枯死)에 기인(起因)되었다. 8. 고수량지(高收量地) 벼는 저수량지(低收量地) 벼에 비(比)하여 생육초기(生育初期)에는 투과광율(透過光率)이 높고 생육후기(生育後期)에 낮았으나 투과광율대(透過光率對) 엽면적비(葉面積比)는 고수량지(高收量地) 벼에서 높았다. 9. 순동화량(純同化量)은 저수량지(低收量地) 벼에 비(比)하여 고수량지(高收量地)벼에서 많았으며 이는 또한 엽면적(葉面積)이 증가(增加)됨에 따라 감소(減少)되었다. .10. 고수량지(高收量地) 벼는 저수량지(低收量地) 벼에 비(比)하여 유수형성기(幼穗形成期) 이후(以後) 체내질소(體內窒素), 가리(加里) 및 규산(珪酸)의 함유율(含有率)이 높으면서 $K_2O/N$, $SiO_2/N$ 비(比)가 높았다.11. 저수량지(低收量地) 벼는 고수량지(高收量地) 벼에 비(比)하여 출수기(出穗期)에 보유(保有)된 질소(窒素), 인산(燐酸), 가리(加里) 및 마그네슘등이 출수이후(出穗以後) 이삭으로의 이행율(移行率)이 높았다. 이는 저수량지(低收量地) 벼가 생육후기(生育後期)에 양분(養分)의 흡술량(吸收量)이 부족(不足)하여 체내(體內) 저장양분(貯藏養分)이 재이동(再移動)되기 때문이며 따라서 하엽(下葉)은 양분부족(養分不足)으로 조길고사(早期桔死)가 많아지는 것으로추정(推定)되었다. 12. 고수량지(高收量地) 벼는 질소흡수(窒素吸收)가 전생육기간(全生育期間)을 통(通)하여 완만(緩慢)하며 출수이후(出穗以後)에도 흡수량(吸收量)이 많으나 저수량지(低收量總) 벼는 출수이전(出穗以前)에 많고 출수이후(出濾以後)는 거의 흡수(吸收)되지 않았다. 13. 출수기(出穗期)의 체내(體內) 질소(窒素), 가리(加里) 함유율(含有率)과 수량(收豊)과는 현저(顯著)한 정(正)의 상관(相關)이 성립(成立)되였으며 수확기(收獲期)의 가리(加里), 규산함유율(珪酸含有率) 그리고 $K_2O/N$, $SiO_2/N$과 수량(收量)과도 현저(顯著)한 정(正)의 상관관계(相關關係)가 성립(成立)되었다. 14. 출수기(出穗期)에 보유(保有)된 전분(澱粉)이 이삭으로의 이전(移轉)은 고수량지(高收量地) 벼가 약(約) 10%인데 반(反)하여 저수량지(低收量地) 벼는 약(約) 40%로 많았다. 따라서 저수량지(高收量地) 벼는 출수(出穗) 이후(以後) 동화의존도(同化依存度)가 높았다. 15. 고수량지(高收量地) 벼는 저수량지(低收量地) 벼에 비(比)하여 질소(窒素), 인산(燐酸), 가리(加里), 규산(珪酸) 및 망간의 흡수량(吸收量)이 현저(顯著)하게 많았고 생육시기별(生育時期別) 흡수량(吸收量)의 차이(差異)는 유수형성기(幼穗形成期) 전후(前後)에 현저(顯著)하였다. 16. 수확기(收穫期)의 질소(窒素), 인산(燐酸), 가리(加里), 석탄(石炭), 마그네슘, 규산(珪酸) 및 망간의 총흡수량(總吸收量)과 수량(收量)과는 고도(高度)의 정(正)의 상관관계(相關關係)가 인정(認定)되었다. 17. 수확기(收穫期)에 질소(窒素), 인산(燐酸), 가리(加里), 석탄(石炭), 마그네슘 및 규산(珪酸)의 흡수량(吸收量)과 수당입수간(穗當粒熱間)에, 질소(窒素) 및 가리(加里)의 흡수량(吸收量)과 수수(穗數)와는 유의적(有意的)인 정(正)의 상관관계(相關關係)가 성립(成立)되었다.

Saccharification of Foodwastes Using Cellulolytic and Amylolytic Enzymes from Trichoderma harzianum FJ1 and Its Kinetics

  • The study was targeted to saccharify foodwastes with the cellulolytic and amylolytic enzymes obtained from culture supernatant of Trichoderma harzianum FJ1 and analyze the kinetics of the saccharification in order to enlarge the utilization in industrial application. T. harzianum FJ1 highly produced various cellulolytic (filter paperase 0.9, carboxymethyl cellulase 22.0, ${\beta}$-glucosidase 1.2, Avicelase 0.4, xylanase 30.8, as U/mL-supernatant) and amylolytic (${alpha}$-amylase 5.6, ${\beta}$-amylase 3.1, glucoamylase 2.6, as U/mL-supernatant) enzymes. The $23{\sim}98\;g/L$ of reducing sugars were obtained under various experimental conditions by changing FPase to between $0.2{\sim}0.6\;U/mL$ and foodwastes between $5{\sim}20\%$ (w/v), with fixed conditions at $50^{\circ}C$, pH 5.0, and 100 rpm for 24 h. As the enzymatic hydrolysis of foodwastes were performed in a heterogeneous solid-liquid reaction system, it was significantly influenced by enzyme and substrate concentrations used, where the pH and temperature were fixed at their experimental optima of 5.0 and $50^{\circ}C$, respectively. An empirical model was employed to simplify the kinetics of the saccharification reaction. The reducing sugars concentration (X, g/L) in the saccharification reaction was expressed by a power curve ($X=K{\cdot}t^n$) for the reaction time (t), where the coefficient, K and n. were related to functions of the enzymes concentrations (E) and foodwastes concentrations (S), as follow: $K=10.894{\cdot}Ln(E{\cdot}S^2)-56.768,\;n=0.0608{\cdot}(E/S)^{-0.2130}$. The kinetic developed to analyze the effective saccharification of foodwastes composed of complex organic compounds could adequately explain the cases under various saccharification conditions. The kinetics results would be available for reducing sugars production processes, with the reducing sugars obtained at a lower cost can be used as carbon and energy sources in various fermentation industries.

지하수 관개에 의한 수도의 멸준양상과 그 방지책에 관한 연구 (Studies on the Rice Yield Decreased by Ground Water Irrigation and Its Preventive Methods)

  • The purposes of this thesis are to clarify experimentally the variation of ground water temperature in tube wells during the irrigation period of paddy rice, and the effect of ground water irrigation on the growth, grain yield and yield components of the rice plant, and, furthermore, when and why the plant is most liable to be damaged by ground water, and also to find out the effective ground water irrigation methods. The results obtained in this experiment are as follows; 1. The temperature of ground water in tube wells varies according to the location, year, and the depth of the well. The average temperatures of ground water in a tubewells, 6.3m, 8.0m deep are $14.5^{\circ}C$ and $13.1^{\circ}C$, respercively, during the irrigation period of paddy rice (From the middle of June to the end of September). In the former the temperature rises continuously from $12.3^{\circ}C$ to 16.4$^{\circ}C$ and in the latter from $12.4^{\circ}C$ to $13.8^{\circ}C$ during the same period. These temperatures are approximately the same value as the estimated temperatures. The temperature difference between the ground water and the surface water is approximately $11^{\circ}C$. 2. The results obtained from the analysis of the water quality of the "Seoho" reservoir and that of water from the tube well show that the pH values of the ground water and the surface water are 6.35 and 6.00, respectively, and inorganic components such as N, PO4, Na, Cl, SiO2 and Ca are contained more in the ground water than in the surface water while K, SO4, Fe and Mg are contained less in the ground water. 3. The response of growth, yield and yield components of paddy rice to ground water irrigation are as follows; (l) Using ground water irrigation during the watered rice nursery period(seeding date: 30 April, 1970), the chracteristics of a young rice plant, such as plant height, number of leaves, and number of tillers are inferior to those of young rice plants irrigated with surface water during the same period. (2) In cases where ground water and surface water are supplied separately by the gravity flow method, it is found that ground water irrigation to the rice plant delays the stage at which there is a maximum increase in the number of tillers by 6 days. (3) At the tillering stage of rice plant just after transplanting, the effect of ground water irrigation on the increase in the number of tillers is better, compared with the method of supplying surface water throughout the whole irrigation period. Conversely, the number of tillers is decreased by ground water irrigation at the reproductive stage. Plant height is extremely restrained by ground water irrigation. (4) Heading date is clearly delayed by the ground water irrigation when it is practised during the growth stages or at the reproductive stage only. (5) The heading date of rice plants is slightly delayed by irrigation with the gravity flow method as compared with the standing water method. (6) The response of yield and of yield components of rice to ground water irrigation are as follows: \circled1 When ground water irrigation is practised during the growth stages and the reproductive stage, the culm length of the rice plant is reduced by 11 percent and 8 percent, respectively, when compared with the surface water irrigation used throughout all the growth stages. \circled2 Panicle length is found to be the longest on the test plot in which ground water irrigation is practised at the tillering stage. A similar tendency as that seen in the culm length is observed on other test plots. \circled3 The number of panicles is found to be the least on the plot in which ground water irrigation is practised by the gravity flow method throughout all the growth stages of the rice plant. No significant difference is found between the other plots. \circled4 The number of spikelets per panicle at the various stages of rice growth at which_ surface or ground water is supplied by gravity flow method are as follows; surface water at all growth stages‥‥‥‥‥ 98.5. Ground water at all growth stages‥‥‥‥‥‥62.2 Ground water at the tillering stage‥‥‥‥‥ 82.6. Ground water at the reproductive stage ‥‥‥‥‥ 74.1. \circled5 Ripening percentage is about 70 percent on the test plot in which ground water irrigation is practised during all the growth stages and at the tillering stage only. However, when ground water irrigation is practised, at the reproductive stage, the ripening percentage is reduced to 50 percent. This means that 20 percent reduction in the ripening percentage by using ground water irrigation at the reproductive stage. \circled6 The weight of 1,000 kernels is found to show a similar tendency as in the case of ripening percentage i. e. the ground water irrigation during all the growth stages and at the reproductive stage results in a decreased weight of the 1,000 kernels. \circled7 The yield of brown rice from the various treatments are as follows; Gravity flow; Surface water at all growth stages‥‥‥‥‥‥514kg/10a. Ground water at all growth stages‥‥‥‥‥‥428kg/10a. Ground water at the reproductive stage‥‥‥‥‥‥430kg/10a. Standing water; Surface water at all growh stages‥‥‥‥‥‥556kg/10a. Ground water at all growth stages‥‥‥‥‥‥441kg/10a. Ground water at the reproductive stage‥‥‥‥‥‥450kg/10a. The above figures show that ground water irrigation by the gravity flow and by the standing water method during all the growth stages resulted in an 18 percent and a 21 percent decrease in the yield of brown rice, respectively, when compared with surface water irrigation. Also ground water irrigation by gravity flow and by standing water resulted in respective decreases in yield of 16 percent and 19 percent, compared with the surface irrigation method. 4. Results obtained from the experiments on the improvement of ground water irrigation efficiency to paddy rice are as follows; (1) When the standing water irrigation with surface water is practised, the daily average water temperature in a paddy field is 25.2$^{\circ}C$, but, when the gravity flow method is practised with the same irrigation water, the daily average water temperature is 24.5$^{\circ}C$. This means that the former is 0.7$^{\circ}C$ higher than the latter. On the other hand, when ground water is used, the daily water temperatures in a paddy field are respectively 21.$0^{\circ}C$ and 19.3$^{\circ}C$ by practising standing water and the gravity flow method. It can be seen that the former is approximately 1.$0^{\circ}C$ higher than the latter. (2) When the non-water-logged cultivation is practised, the yield of brown rice is 516.3kg/10a, while the yield of brown rice from ground water irrigation plot throughout the whole irrigation period and surface water irrigation plot are 446.3kg/10a and 556.4kg/10a, respectivelely. This means that there is no significant difference in yields between surface water irrigation practice and non-water-logged cultivation, and also means that non-water-logged cultivation results in a 12.6 percent increase in yield compared with the yield from the ground water irrigation plot. (3) The black and white coloring on the inside surface of the water warming ponds has no substantial effect on the temperature of the water. The average daily water temperatures of the various water warming ponds, having different depths, are expressed as Y=aX+b, while the daily average water temperatures at various depths in a water warming pond are expressed as Y=a(b)x (where Y: the daily average water temperature, a,b: constants depending on the type of water warming pond, X; water depth). As the depth of water warning pond is increased, the diurnal difference of the highest and the lowest water temperature is decreased, and also, the time at which the highest water temperature occurs, is delayed. (4) The degree of warming by using a polyethylene tube, 100m in length and 10cm in diameter, is 4~9$^{\circ}C$. Heat exchange rate of a polyethylene tube is 1.5 times higher than that or a water warming channel. 넙치(Paralichthys olivaceus), 참돔(Pagrus major) 및 조피볼락(Sebastes schlegeli)의 이화학적 성분 특성 (Physicochemical Properties of Olive Flounder Paralichthys olivaceus, Red Seabream Pagrus major and Jacopever Sebastes schlegeli)

  • 넙치, 참돔 및 조피볼락을 이용하여 새로운 가공식품을 개발하기 위한 기초적인 자료를 얻기 위해 이화학적 특성을 조사한 결과를 요약하면다음과 같다. 넙치, 참돔 및 조피볼락의 일반성분의 경우 수분함량은 각각 78.3, 73.2 및 79.2%, 조단백질함량은 14.8, 19.2 및 18.5%, 조지방함량은 1.5, 6.4 및 1.2%, 조회분은 0.6, 1.1 및 0.6%였다. pH는 각각 6.41, 6.32 및 6.53이었으며, 염도는 각각 0.6, 0.7 및 0.6이었다. 색차의 경우 넙치, 참돔 및 조피볼락의 명도(L값)는 각각 42.47, 41.70 및 43.09, 적색도(a값)는 각각 -3.07, 0.24 및 -1.87, 황색도(b값)는 각각 3.86, 6.25 및 3.59, 색차(${\Delta}E$값)는 각각 54.57, 55.40 및 53.83이었다. 넙치, 참돔 및 조피볼락의 총아미노산 함량은 각각 13,003.7, 17,126.0 및 16,398.3 mg/100 g이었고, glutamic acid가 각각 1,694.7(13.0%), 2,496.1(14.6%) 및 2,089.5(12.7%) mg/100 g으로 가장 많은 함량이었다. 시료의 총유리아미노산 함량은 각각 173.0, 195.5 및 162.9 mg/100 g이었으며, 넙치 및 조피볼락은 lysine이 각각 30.4(17.6%) 및 43.3(26.6%) mg/100 g, 참돔은 alanine이 40.2(20.6%) mg/100 g으로 가장 많은 함량이었다. 무기질의 경우 모든 시료에서 K이 각각 383.8, 404.3 및 335.8 mg/100g으로 가장 많았다. 넙치의 경우 포화지방산, 단일 불포화지방산 및 다가 불포화지방산이 각각 29.3, 19.1 및 51.1%, 참돔의 경우 각각 14.5, 15.7 및 69.8%, 조피볼락의 경우 각각 12.7, 15.0 및 72.3%를 차지하였다. 넙치, 참돔 및 조피볼락에서 n-3계 지방산인 ${\alpha}$-linolenic acid는 각각 0.7, 1.0 및 0.7%, EPA는 각각 6.8, 7.1 및 6.7%, DHA는 각각 15.2, 15.2 및 17.1%를 차지하였다. 관능검사 결과 넙치, 참돔 및 조피볼락의 색 및 냄새에서는 유의적인 차이가 없었으나, 맛 및 조직감에서는 유의적인 차이가 있었다.

홍삼추출액은 lipoteichoic acid로 자극된 소교세포에서 Akt 및 MAPK 의존적으로 heme oxygenase-1 발현을 유도함으로써 NO 생성을 억제함 (A Formulated Korean Red Ginseng Extract Inhibited Nitric Oxide Production through Akt- and Mitogen Activated Protein Kinase-dependent Heme Oxygenase-1 Upregulation in Lipoteichoic Acid-stimulated Microglial Cells)

  • 생삼을 쪄서 건조시킨 홍삼은 전통적으로 사용되고 있는 약재로서 면역, 내분비 및 중추신경계 작용을 증진시키며 염증을 억제하는 효과가 있는 것으로 알려져 있다. 본 연구에서는 그람 양성균의 세포벽 성분인 lipoteichoic acid (LTA)에 의한 염증반응에 홍삼추출액(RGE)이 항염증 효과를 가지는지 관찰하고 그 작용 기전을 연구하였다. BV-2 소교세포에서 RGE는 세포에 독성을 유도하지 않으면서 LTA로 인한 nitric oxide (NO)의 생성과 inducible nitric oxide synthase (iNOS) 발현을 억제하였으며, NF-kB p65의 핵으로의 이동과 IkB-a의 분해 또한 억제하였다. 한편, RGE는 농도의존적으로 heme oxygenase-1 (HO-1)의 발현을 유도하였으며, HO-1 siRNA를 처리했을 때는 RGE가 iNOS의 발현을 억제하지 못하였다. RGE는 HO-1의 발현에 관여하는 전사인자인 nuclear factor E2-related factor 2 (Nrf2)를 핵으로 이동을 촉진시켰다. 또한 RGE에 의한 HO-1의 발현은 phosphatidylinositol-3-kinase(PI-3K) 및 MAPK 억제제에 의해 감소되었으며, RGE가 Akt와 ERK, p38, JNK의 인산화를 유도하였다. 이상의 결과를 종합해보면, RGE는 PI-3K/Akt 및 ERK, p38, JNK 신호전달과정을 통해 HO-1의 발현을 유도함으로써 NO와 같은 염증매개물질의 생성을 억제한다는 것을 알 수 있다. 그러므로 홍삼추출액은 그람 양성균에 의한 신경염증과 염증관련 신경계 질환의 치료제로서 사용될 수 있을 것이라 사료된다.
