Development of Chiral Stationary Phases for the Gas Chromatographic Separation of Amino Acid Enantiomers New diamide chiral stationary phase (아미노산 광학이성질체 분리를 위한 가스크로마토그라피용 키랄 고정상의 개발 -새로운 diamide계 키랄 고정상의 응용-)

  • New diamide chiral stationary phases of four systematically substituted optically active N-(N-benzoyl-L-amino acid)-anilide synthesized from L-valine, L-leucine, L-isoleucine, and L-phenylalanine were described. The behaviors of these diamides as optically active stationary phases for the separation of N-trifluoroacetyl-D,L-amino acids were examined with respect to separation factors(${\alpha}$) and thermodynamic properties of interaction. The separation of twelve N-trifluoroacetyl-D,L-amino acid isopropyl esters were improved by the order of N-(N-benzoyl-L-leucyl)-anilide>N-(N-benzoyl-L-isoleucyl)-anilide>N-(N-benzoyl-L-valyl)-anilide>N-(N-benzoyl-L-phenylalanyl)-anilide. Eight amino acid derivatives with non-polar R-group and threonine, serine, aspartic acid, and glutamic acid enantiomers were separated on N-(N-benzoyl-L-leucyl)-anilide as chiral stationary phase with good separation factor between 1.07-1.25. The separation factors decreased with respect to increasing column temperature. Possible working temperature of diamide phase was between $130-190^{\circ}C$ for N-(N-benzoyl-L-phenylalanyl)-anilide and $130-180^{\circ}C$ for other three diamide phases. The differential Gibb's free energy (${\Delta}{\Delta}G$) of enantiomers was in the range of -100--180 cal/mol for ten amino acids and -40--60 cal/mol for alanine and aspartic acid.

Oxygen and Hydrogen Isotopic Compositions of the Hwacheon Granite (화천화강암의 산소와 수소 동위원소 조성)

  • Oxygen and hydrogen isotopic compositions of the Jurassic peraluminous Hwacheon granite were measured, and compared with those of other Jurassic peraluminous Daebo granite in Korea. $\delta$$\^$18/O values for quartz and feldspar of the Hwacheon granite range from 8.2 to 10.6 and 5.8 to 9.0$\textperthousand$, respectively. Whole rock $\delta$$\^$18/O values for banded biotite gneiss country rocks surrounding the Hwacheon granites range from 8.1 to 9.4$\textperthousand$. Whole rock and biotite $\delta$D Values for Hwacheon granite range from -84 to -113 and -107 to -113$\textperthousand$, respectively. Whole rock $\delta$D values for banded biotite gneiss country rocks range from -76 to -100$\textperthousand$. Both $\delta$$\^$18/O and $\delta$D values of the Hwacheon granite are characterized by low values compared to the 'normal' values for the fresh peraluminous granitic rocks. Low $\delta$$\^$18/O values of the Hwacheon granite resulted from fluid-rock interaction for a long period. Isotopic modelling result renders that a relatively low-$\delta$$\^$18/O fluid below -1$\textperthousand$ was involved in subsolidus isotopic exchange under a relatively high fluid/rock ratio (<-6). The fluid of meteoric origin has experienced a modification of oxygen isotopic composition as a result of fluid-rock interaction with the Hwacheon granite and surrounding metapelitic country rocks.

Thermally Stratified Hot Water Storage (태양열의 성층축열과 주택이용에 관한 연구(성층축열))

  • This paper deals with experimental research to increase thermal storage efficiency of hot water stored in an actual storage tank for solar application. The effect of increased energy input rate due to stratification has been discussed and illustrated through experimental data, which was taken by changing dynamic and geometric parameters. Ranges of the parameters were defined for flow rate, the ratio of diameter to height of the tank and inlet-exit water temperature difference. During the heat storage, when the flow was lower, the temperature difference was larger and the ratio of diameter to height of the tank was higher, the momentum exchange decreased. As for this experiment, when the flow rate was 8 liter/min, the temperature difference was $30^{\circ}C$ and the ratio of diameter to height of the tank was 3, the momentum exchange was minimized resulting in a good thermocline and a stable stratification. In the case of using inlet ports, if the modified Richardson number was less than 0.004, full mixing occured and so unstable stratification occured, which mean that this could not be recommended as storage through thermal stratification. Using a distributor was better than using inlet ports to form a sharp thermocline and to enhance the stratification. It was possible to get storage efficiency of 95% by using the distributor, which was higher than a storage efficiency of 85% obtained by using inlet ports in same operation condition. Furthermore, if the distributor was manufactured so that the mainpipe decreases in diameter toward the dead end to maintain constant static pressure, it might be predicted that further stable stratification and higher storage efficiency are obtainable(ie:more than 95%).

The Changes in Properties of Silk Fabrics Dyed with Safflower under Ultraviolet-Light (홍화 염색 견직물의 자외선에 의한 성능 변화 연구)

  • The purpose of this study was to investigate the changes in properties of silk fabrics dyed with safflower red and yellow colorants under Ultraviolet(UV)-light. Silk fabrics dyed with safflower red and yellow colorants were compared with each other after uv-light exposure in terms of K/S value, color changes(${\Delta}E$), morphology, and strength retention. The K/S value rapidly decreased with increasing exposure time for samples of both colorants, but the extent of decrease was less for the samples dyed with safflower yellow colorant, than that of the samples dyed with safflower red colorant. Regarding the color changes with increasing exposure time, $L^*$ and $b^*$ increased, $a^*$ decreased, and thereby ${\Delta}E$ increased in the red-dyed samples, whereas $L^*$ increased, $a^*$ and $b^*$ decreased, and so ${\Delta}E$ increased in the yellow-dyed samples, indicating that the UV-light induced fading and changes of hue, value and chroma value. However, the color change of the yellow-dyed samples was less than that of the red-dyed samples. SEM images showed a severe degradation by UV-exposure for both colorant samples. Tensile strength slowly decreased until 14 days, but rapidly decreased thereafter. Strength retention of the yellow-dyed samples was higher than that of red-dyed samples.

Genetic Organization of the hrp Genes Cluster in Erwinia pyrifoliae and Characterization of HR Active Domains in HrpNEp Protein by Mutational Analysis

  • The disease-specific (dsp) region and the hypersensitive response and pathogenicity (hrp) genes, including the hrpW, $hrpN_{Ep}$, and hrpC operons have previously been sequenced in Erwinia pyrifoliae WT3 [Shrestha et al. (2005a)]. In this study, the remaining hrp genes, including the hrpC, hrpA, hrpS, hrpXY, hrpL and hrpJ operons, were determined. The hrp genes cluster (ca. 38 kb) was comprised of eight transcriptional units and contained nine hrc (hrp conserved) genes. The genetic organization of the hrp/hrc genes and their orientation for the transcriptions were also similar to and collinear with those of E. amylovora, showing ${\geq}80%$ homologies. However, ORFU1 and ORFU2 of unknown functions, present between the hrpA and hrpS operons of E. amylovora, were absent in E. pyrifoliae. To determine the HR active domains, several proteins were prepared from truncated fragments of the N-terminal and the C-terminal regions of $HrpN_{Ep}$ protein of E. pyrifoliae. The proteins prepared from the N-terminal region elicited HR, but not from those of the C-terminal region indicating that HR active domains are located in only N-terminal region of the $HrpN_{Ep}$ protein. Two synthetic oligopeptides produced HR on tobacco confirming presence of two HR active domains in the $HrpN_{Ep}$. The HR positive N-terminal fragment ($HN{\Delta}C187$) was further narrowed down by deleting C-terminal amino acids and internal amino acids to investigate whether amino acid insertion region have role in faster and stronger HR activity in $HrpN_{Ep}$ than $HrpN_{Ea}$. The $HrpN_{Ep}$ mutant proteins $HN{\Delta}C187$ (D1AIR), $HN{\Delta}C187$ (D2AIR) and $HN{\Delta}C187$ (DM41) retained similar HR activation to that of wild-type $HrpN_{Ep}$. However, the $HrpN_{Ep}$ mutant protein $HN{\Delta}C187$ (D3AIR) lacking third amino acid insertion region (102 to 113 aa) reduced HR when compared to that of wild-type $HrpN_{Ep}$. Reduction in HR elicitation could not be observed when single amino acids at different positions were substituted at third amino acids insertion region. But, substitution of amino acids at L103R, L106K and L110R showed reduction in HR activity on tobacco suggesting their importance in activation of HR faster in the $HrpN_{Ep}$ although it requires further detailed analysis.

Continuous Production of ${\delta}-Aminolevulinate$ from Immobilized Cells of Rhodocyclus gelatinosus KUP-74 (Rhodocyclus gelatinosus KUP-74의 고정화균체로부터 ${\delta}-Aminolevulinate$의 연속생산)

  • An optimal condition for the continuous production of ${\delta}-aminolevulinate$(ALA) was investigated using high concentrated resting cells of Rhodocyclus gelatinosus KUP-74. The increase of the amount of extracellular ALA versus the concentration of resting cells showed rectangular hyperbolic pattern until 20 mg cells/ml, but no further increase in the ALA amount by increasing its concentration was occurred. The highest yield of the extracellular ALA was observed after 3 hr of incubation of 1 ml reaction system containing 20 mg cells, 4 mM levulinate and 5 mM L-glutamate. On the other hand, the immobilized cells prepared by Ca-alginate inclusive method needed to incubate for 6 hr with 6 mM levulinate and 10 mM L-glutamate to give maximal yield of the extracellular ALA. In addition, under these conditions the resulted continuous productivities of the ALA by immobilized cells and highly concentrated resting cells were appeared 50 percent decreases after incubations for 185 hr and 100 hr, respectively, and the method of the cells to be immobilized was more efficient to recover the extracellular ALA produced.

Translucency and Color Stability of Various Core Ceramics for All-Ceramic Restoration (전부도재수복물을 위한 수종의 코어 세라믹의 반투명도 및 색 안정성)

  • Purpose: This investigation was designed to determine the translucency and color stability of various core ceramics for all-ceramic restoration using the CIE $L^*a^*b^*$ system. Material and Methods: IPS e.max Press ceramic(Ivoclar-Vivadent, Liechtenstein), $LAVA^{TM}$ All Ceramic(3M-Espe, Germany), Cercon Smart Ceramic(Dentsply, Germany), and Z-match Ceramic(DentAim, Korea) were used for this study. For the specimens of zirconia oxide ceramics, the as-sintered cylindrical blanks($11.0{\times}25.0mm$) were machined into the shape of a disk(0.4, 0.8, 1.5 mm in thickness, 10 mm in diameters) with a diamond grind machine. The IPS e.max Press specimens ($0.8{\times}10mm$) were fabricated using the "lost wax" technique. CIE $L^*a^*b^*$ coordinates and light transmission were recorded for each specimen with a spectrophotometer(CM-2600d, Minolta, Japan). Color differences were calculated using the equation, ${\Delta}E^*ab=[({\Delta}L^*)2+({\Delta}a^*)2+({\Delta}b^*)2]1/2$. Results: The results were obtained as follows: 1. The most translucent group was IPS e.max Press ceramic that is a glass-ceramic, and $Lava^{TM}$ and Z-match ceramic were more translucent than Cercon Smart ceramic in zirconia ceramic group. 2. In the all groups, there was no significant translucent change after 6 times heat-treatments required to make a final restoration. 3. Colored zirconia was showed more yellowish and dark than uncolored zirconia. 4. After heat-pressing, IPS e.max Press ceramic was showed high ${\Delta}E^*ab$ value(4.1 of eM1, 6.8 of eM2) that means to be more whiter than before heat-pressing. However, there was no color change after additive heat treatments for final restoration. 5. In the colored zirconia groups, there was no significant color change after some heat-treatments required to make a final restoration.

SEM pictures showed a severe degradation by uv exposure, regardless of colorants type. Tensile strength slowly decreased until 14 days, and rapidly decreased until 28 days. Strength retention of the samples dyed with safflower yellow colorants was higher than that of the samples dyed with safflower red colorants.

COMPARATIVE EVALUATION OF LIGHT-CURED COMPOSITE RESINS BASED ON VITA SHADE BY SPECTROCOLORIMETER (분광색채계측기를 이용한 Vita Shade 광중합형 복합레진의 색상 비교)

  • To get a satisfactory result in the composite resin restorations, it is necessary to choose correct shade. At present, most of the commercial composite resins are based on the Vita Lumin shade guide or Bioform shade guide, but color differences might be expected even using the same shade in various materials. In this study, five kinds of light-cured composite resins with A2 and B3 shade were used to measure and compare the color each other while one porcelain served as a control. All composite resins (Spectrum TPH (SP), VeridonFil- Photo (VE), Z100 (Z100), Charisma (CH), Prodigy (PRO)) were filled in to the metal mold (12 mm diameter, 2 mm depth), followed by compression, polymerization and polishing with wet sandpaper. The specimens of porcelain were fabricated by using the refractory mold for porcelain. After 24 hours, the specimens were placed on the spectrocolorimeter and spectral reflectance were measured under CIE illuminant D65. After measuring the values of $L^*$, $a^*$, $b^*$ and ${\Delta}E^*$, following results were obtained; 1. The $L^*$, $a^*$ and $b^*$ values of both shade of porcelain specimens showed significantly higher than those of resin specimens(p<0.05). 2. In comparing the resin specimens of the A2 shade, differences were significant except $L^*$ values of SP-CH and PRO-VE, $a^*$ values of the VE-SP and $b^*$ values of the VE-Z100 and SP-PRO(p<0.05), 3. In comparing the resin specimens of the B3 shade, differences were significant except $L^*$ values of PRO-SP, $a^*$ values of the SP-PRO and Z100-VE and b* values of the PRO-SP(p<0.05). 4. In comparing the resin specimens of the A2 shade, color differences between materials (${\Delta}E^*$) showed the lowest value of 1.66, and the highest was 5.16. ${\Delta}E^*$ values of the materials of VE-PRO, CH-PRO, SP-PRO, SP-Z100 and SP-CH were lower than 3.3. 5. In comparing the resin specimens of the B3 shade, the lowest value of the ${\Delta}E^*$ was 0.57 and the highest was 5.92. ${\Delta}E^*$ values of Z100-CH and SP-PRO were lower than 3.3. The present study revealed there was perceptible color difference between materials even if they have the same designated shade based on Vita shade guide. The results of the present study suggested that it would be necessary to establish the reproducible and constant color specification system for an esthetic restoration.

  • We investigated the phase noise of an oscillator with a extremely high-Q resonator used as the resonant element. A TE$_{011}$ mode rutile-loaded resonator with high-temperature superconductive (HTS) $YBa_2Cu_3O_{7-\delta}$(YBCO) films used as the endplates is prepared for this purpose. At 23.5 K, the unloaded Q and the loaded Q are 863000 and 180000, respectively. The phase noise of -104.8 dBc/Hz at 1 KHz offset was observed for the oscillator having a resonator with $Q_{L}$ =180000 at the $TE_{01\delta$ mode resonant frequency of 8.545 GHz at 23.5 K Such oscillators with very low phase noise are expected to be used for building up communication systems capable of efficient use of the frequency band and high-speed data transmission as well as for Doppler radars. Frequency tuning could be realized for the resonator by using a piezoactuator Applicability of the tunable rutile resonator for fabricating tunable oscillators of high performances is discussed.
