Gamakamide C and D as Two New Analogues of Bitter-Tasting Cyclic Peptide with Hydantoin Structure from Oyster Crassostrea gigas

  • Two new bitter-tasting cyclic peptides comprising six amino acids, namely gamakamide C and D, were isolated from cultured oysters Crassostrea gigas. Dimethylaminoazobenzene sulfonyl-amino acid analysis detected Val and Leu in gamakamide C and Ile and Leu in gamakamide D. The molecular formula of gamakamide C was determined as $C_{43}H_{60}N_{7}O_8S$ by high-resolution fast atom bombardment mass spectroscopy (HR FAB-MS) ($[M+H]^+m/z822.4200{\Delta}-2.4mmu$), and that of gamakamide D was determined as $C_{43}H_{62}N_7O_8S$ by HR FAB-MS ($[M+H]^+m/z836.4379{\Delta}-2.0mmu$). Comparison of amino acid analyses and fragment ions by MS/MS among gamakamide C, D, and E (known), the structures of gamakamide C and D were confirmed $as-{\small{L}}-Val-{\small{L}}-Met(SO)-{\small{L}}-NMe-Phe-{\small{L}}-Leu-{\small{D}}-Lys-{\small{L}}-Phe-$ and $-{\small{L}}-Ile-{\small{L}}-Met(SO)-{\small{L}}-NMe-Phe-{\small{L}}-Leu-{\small{D}}-Lys-{\small{L}}-Phe-$, respectively.

Glutathione Depletion by L-Buthionine-S,R-Sulfoximine Induces Apoptosis of Cardiomyocytes through Activation of PKC-δ

  • In the present study, we investigated the effect of intracellular glutathione (GSH) depletion in heart-derived H9c2 cells and its mechanism. L-buthionine-S,R-sulfoximine (BSO) induced the depletion of cellular GSH, and BSO-induced reactive oxygen species (ROS) production was inhibited by glutathione monoethyl ester (GME). Additionally, GME inhibited BSO-induced caspase-3 activation, annexin V-positive cells, and annexin V-negative/propidium iodide (PI)-positive cells. Treatment with rottlerin completely blocked BSO-induced cell death and ROS generation. BSO-induced GSH depletion caused a translocation of PKC-${\delta}$ from the cytosol to the membrane fraction, which was inhibited by treatment with GME. From these results, it is suggested that BSO-induced depletion of cellular GSH causes an activation of PKC-${\delta}$ and, subsequently, generation of ROS, thereby inducing H9c2 cell death.

Here, we provide a decomposition of the set X = {$h\;{\in}\;L^{\infty}(D)\;{\mid}\;lim_{n{\rightarrow}{\infty}}\;h\;*\;\tau^n$ exists}. Let $\tau_1$, ..., $\tau_n$ be measures on D with above mentioned properties. Here, we prove that if $f\;{in}\;L^{\infty}(D^n)$ satisfies an invariant volume mean value property with respect to $\tau_1$, ..., $\tau_n$, then f is n-harmonic.

  • Aminolevulinic acid(ALA) was shown to be synthesized via active pathways of either C4 or C5 ALA biosynthesis in cells of a photosynthetic bacterium, Rhodocyclus gelatinosus KUP-74, where the C5 pathway was appeared to be preferntially expressed in the cells. It was strongly suggested that L-glutamine might be utilized more effectively than L-glutamate to synthesize ALA via C5 pathway in this bacterium from the fact of relationship between the cellular uptake rates of glutamate and its Gamma-derivaties and corresponded ALA productivities in vitro and in vivo.

  • Apoemulsan is a biopolymer with potent emulsification activity, produced by Acinetobacter venetian us RAG-1 (RAG-1). The wee gene cluster is responsible for apoemulsan biosynthesis. The analysis of (i) a putative polysaccharide copolymerase mutant (${\Delta}wzc$), (ii) a putative polymerase mutant (${\Delta}wzy$), and (iii) an apoemulsan-deficient variant (${\Delta}2$) indicated that the wee gene cluster controls the synthesis of two polysaccharides: high molecular weight (HMW) and low molecular weight (LMW). LMW polysaccharide of wee origin was present in LPS isolated from RAG-1 cells, suggesting a link to the Lipid A-core of LPS molecules. SDS-PAGE analysis indicated that apoemulsan is copurified with LPS polysaccharide, with implications in the emulsification activity of RAG-1 polymer.

  • A novel system has been developed to produce ${\delta}-aminolevulinic$ acid (ALA) using the intact cells of late logarithmic phase of Rhodocyclus gelatinosus KUP-74. The system was shown optimum yield of extracellular ALA under a condition of anaerobic light irradiation (4 Klux) at $30^{\circ}C$ with no variation in cell mass. The rate of extracellular ALA formation was stimulated by low doses of either $C_4\;or\;C_5$ ALA biosynthetic precursors, where 5 mM ($C_4) and 3 mM ($C_5) of each precursors were appeared to generate the maximum rates of 3.3 and 4.0 nmoles of ALA per 0.35 mg cells per hr, respectively. Half-life of the system was 10 hr in a sense of an ability of portage transport of L-glutamate, and sequential dose of this compound was resulted in promising recovery of the ALA.

  • Stainless steel sheets are widely used as the structural material for the railroad cars and the commercial vehicles. These kinds structures used stainless steel sheets are commonly fabricated by using the gas welding. For fatigue design of gas welded joints such as fillet and plug type joint, it is necessary to obtain design information on stress distribution at the weldment as well as fatigue strength of gas welded joints. And also, the influence of the geometrical parameters of gas welded joints on stress distribution and fatigue strength must be evaluated. the ${\Delta}P-N_f$ curves were obtained by fatigue tests. Using these results, ${\Delta}P-N_f$ curves were rearranged in the ${\Delta}{\sigma}-N_f$ relation with the maximum stress at the edge of fillet welded joint.

  • The operating characteristics, such as, the threshold gain, lasing frequency, and longitudinal intensity profile, etc., of 1.55$\mu$m DFB laser diode with index and/or gain grating structures and with one side AR-coated mirror have been analyzed. From this analysis, the optimum design parameters have been shown that ${\Delta}{\Omega}$ (the phase difference between index grating and gain grating) is 0 or ${\pi}$, (xL)$_{r}$=1~3 and (xL)$_{i}$=0.5~0.9. It has been also shown that the modal selectivity and intensity uniformity of the DFB lasers with .DELTA..OMEGA.=0 are ~1.2 times better thatn those of the DFB lasers with ${\Delta}{\Omega}$= ${\pi}$.

  • Oxygen-plasma effects of single crystal and thin film samples of L $a_{0.7}$C $a_{0.3}$Mn $O_{3-}$$\delta$/ have been studied. Our resistivity measurements indicate that oxygen plasma treatment gives rise to oxygen diffusion into bulk regions, which results in a decrease of M $n^{3+}$ concentration in oxygen nonstoichiometric L $a_{0.7}$C $a_{0.3}$Mn $O_{3-}$$\delta$/ and in the activation energies of Holstein's small polarons in the paramagnetic region.n.egion.n.n.
